Rich polysaccharides were directly observed in the bulbs of Fritillaria unibracteata var. wabuensis (FUW) using the periodic acid–Schiff (PAS) method and microexamination. An acidic water-soluble heteropolysaccharide (FWPS1-1) was isolated from FUW through ethanol precipitation, decoloration, deproteinization, dialysis and separation using a DE-52 anion-exchange column and a Sepharose G-150 gel filtration column. FWPS1-1 (average molecular weight: ~7.44 kDa) has many branches and long side chains; holds the triple-helix conformation; was composed of mannose (Man), galacturonic acid (GalA), galactose (Gal), xylose (Xyl) and arabinose (Ara) with a molar ratio of 2.62:5.59:10.00:0.76:9.38; and features side chains that may be composed of Ara, Man, Gal and GalA, while the backbone may be composed of Xyl, Ara and Gal. In addition, the backbone of FWPS1-1 mainly consists of α-type glycosidic bonds. Bioactivity tests in vitro showed that the polysaccharide exhibited weak 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and low ferric reducing antioxidant power (FRAP) but high 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)l (ABTS) radical scavenging activity, good Fe(II)-chelating ability and remarkable DNA damage protective activity. FWPS1-1 was the first heteropolysaccharide purified from FUW and showed good antioxidant activity and DNA protective effect. The results confirmed that macromolecule is also bioactive ingredient that requires attention like the small-molecule active compounds in FUW.

As an important antitussive and expectorant herbal medicine, Fritillariae cirrhosae bulbus (FCB) is rare and expensive, leading to massive adulterations in the market. Herein, we report a solid colorimetric sensor array (CSA) method for the rapid identification of FCB and its adulterations. This CSA was fabricated by printing nine pH indicators on a mixed cellulose ester membrane via a commercially available inkjet printer filled with polyethylene glycol-containing ink. This CSA displayed color changes to various analytes, as their solution of varied pH values triggers chemical reactions with the nine pH indicators (and other potential interactions). The digital database of these colorimetric response patterns was collected automatically using a homemade program. The outstanding identification capability of the CSA was confirmed via the analysis of 1,2-diols and catechols. The principal component analysis result demonstrated that this CSA could distinguish pure FCB powder from its six common adulterations. The linear discriminant analysis models were established for the qualitative identification of adulterated FCB samples with 89.58 %− 100 % accuracy. Furthermore, partial least squares regression models were developed for the quantitation of adulterant percentages with correlation coefficients higher than 0.87. We expect that the inkjet-printed CSA provided an effective on-site screening method for identifying adulterated FCB.

Fully understanding the chemicals in an herbal medicine remains a challenging task. Molecular networking (MN) allows to organize tandem mass spectrometry (MS/MS) data in complex samples by mass spectral similarity, which yet suffers from low coverage and accuracy of compound annotation due to the size limitation of available databases and differentiation obstacle of similar chemical scaffolds. In this work, an enhanced MN-based strategy named diagnostic fragmentation-assisted molecular networking coupled with in silico dereplication (DFMN-ISD) was introduced to overcome these obstacles: the rule-based fragmentation patterns provide insights into similar chemical scaffolds, the generated in silico candidates based on metabolic reactions expand the available natural product databases, and the in silico annotation method facilitates the further dereplication of candidates by computing their fragmentation trees. As a case, this approach was applied to globally profile the steroidal alkaloids in Fritillariae bulbus, a commonly used antitussive and expectorant herbal medicine. Consequently, a total of 325 steroidal alkaloids were discovered, including 106 cis -D/E-cevanines, 142 trans -D/E-cevanines, 29 jervines, 23 veratramines, and 25 verazines. And 10 of them were confirmed by available reference standards. Approximately 70% of the putative steroidal alkaloids have never been reported in previous publications, demonstrating the benefit of DFMN-ISD approach for the comprehensive characterization of chemicals in a complex plant organism.

In this study, a new strategy combining mass spectrometric (MS) techniques with partial least squares regression (PLSR) was proposed to identify and quantify closely related adulterant herbal materials . This strategy involved preparation of adulterated samples, data acquisition and establishment of PLSR model. The approach was accurate, sensitive, durable and universal, and validation of the model was done by detecting the presence of Fritillaria Ussuriensis Bulbus in the adulteration of the bulbs of Fritillaria unibracteata. Herein, three different MS techniques, namely wooden-tip electrospray ionization mass spectrometry (wooden-tip ESI/MS), ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS) and UPLC-triple quadrupole tandem mass spectrometry (UPLC-TQ/MS), were applied to obtain MS profiles for establishing PLSR models. All three models afforded good linearity and good accuracy of prediction, with correlation coefficient of prediction ( r p 2 ) of 0.9072, 0.9922 and 0.9904, respectively, and root mean square error of prediction (RMSEP) of 0.1004, 0.0290 and 0.0323, respectively. Thus, this strategy is very promising in tracking the supply chain of herb-based pharmaceutical industry , especially for identifying adulteration of medicinal materials from their closely related herbal species.

Medicinal food homology substances have served as a traditional means of overcoming many health complications, and gained increasing attention. Fritillaria Bulbus is used as an edible herbal food with medicinal food homology; however, its antioxidant efficacy substantially varies among different species. In this study, an integrated approach of UHPLC-Q-Exactive Orbitrap MS/MS-based metabolomics, network pharmacology, and molecular docking was used to compare the antioxidant activities of four FBs and identify the active compounds and potential mechanisms. A total of 143 metabolites were identified, predominantly comprising alkaloids. In vitro antioxidant tests revealed a positive correlation between alkaloids and their antioxidant capabilities. Network pharmacology predicted 17 differential and 44 shared compounds. Molecular docking assessed that seven alkaloids (peimisine、imperialine、peimine、solasodine、veratramine、cyclopamine and jervine) exhibited good affinity for the antioxidant targets. Key therapeutic targets AKT1 and ESR1 mediate antioxidant mechanisms through PI3K/AKT signaling. These findings provide insights into interspecies differences in FB’s antioxidant properties.

ABSTRACT Ethnopharmacological relevance Fritillaria cirrhosa D. Don (FCD) is a traditional Chinese medicine used to treat respiratory disorders, known for its effects in clearing heat, moistening the lungs, resolving phlegm, and relieving cough. Additionally, the total alkaloids extracted from FCD can alleviate asthma symptoms and reduce airway inflammation. However, no studies have investigated the effects of total alkaloids on lung macrophages. Aim of the study This study explored whether the total alkaloids of FCD (TAs-FCD) reduce M2 macrophage polarization and, consequently, attenuate airway remodeling in asthmatic mice. This study further elucidated its mechanism of action in treating allergic asthma. Materials and methods The extracted TAs-FCD was analyzed for its composition using UPLC-Q-TOF/MS. Network pharmacology was employed to identify the active ingredients and potential mechanisms of TAs-FCD in the treatment of allergic asthma. A mouse model of ovalbumin-induced allergic asthma was established, adopted, and validated through in vivo experiments. Hematoxylin-eosin staining (H&E), immunohistochemistry (IHC), immunofluorescence staining (IF), enzyme-linked immunosorbent assay (ELISA), Western blotting (WB), and real-time fluorescence quantitative polymerase chain reaction (q-PCR) were used to investigate the role of TAs-FCD in inhibiting M2 macrophage polarization in the context of allergic asthma. Results A total of 66 active ingredients were screened from 116 compounds using SWISSADME. The targets of these 66 compounds were predicted by SwissTargetPrediction, resulting in 808 unique drug targets after excluding duplicates. Additionally, 1,756 targets related to allergic asthma were identified from the DisGeNET, Genecard, and OMIM databases. This led to 267 cross-targets between the active ingredient targets and allergic asthma targets, including interleukin (IL)-1β, tumor necrosis factor (TNF), and STAT3. Animal experiments demonstrated that TAs-FCD improved histopathological injury in mouse lungs, reduced peri-airway collagen fiber accumulation, airway mucus secretion, and airway smooth muscle proliferation. TAs-FCD also lowered IL-1β, TNF-α and IL-4 levels in lung tissues and alleviated airway inflammation. Furthermore, TAs-FCD significantly reduced levels of Arg1 and CD206, which are closely associated with M2 macrophages, and downregulated the expression of p-STAT3 and p-JAK2. Conclusion TAs-FCD may inhibit M2 macrophage polarization by regulating the JAK2/STAT3 pathway, thereby alleviating airway remodeling and inflammation in allergic asthmatic mice.

Aim: Imperialine is an effective compound in the traditional Chinese medicine chuanbeimu (Bulbus Fritillariae Cirrhosae ) that has been used as antitussive/expectorant in a clinical setting. In this study we investigated the absorption characteristics of imperialine in intestinal segments based on an evaluation of its physicochemical properties. Methods: Caco-2 cells were used to examine uptake and transport of imperialine in vitro , and a rat in situ intestinal perfusion model was used to characterize the absorption of imperialine. The amount of imperialine in the samples was quantified using LC-MS/MS. Results: The aqueous solubility and oil/water partition coefficient of imperialine were determined. This compound demonstrated a relatively weak alkalinity with a pKa of 8.467±0.028. In Caco-2 cells, the uptake of imperialine was increased with increasing pH in medium, but not affected by temperature. The apparent absorptive and secretive coefficient was (8.39±0.12)×10 −6 cm/s and (7.78±0.09)×10 −6 cm/s, respectively. Furthermore, neither the P-glycoprotein inhibitor verapamil nor Niemann-Pick C1-Like 1 transporter inhibitor ezetimibe affected the absorption and secretion of imperialine in vitro . The in situ intestinal perfusion study showed that the absorption parameters of imperialine varied in 4 intestinal segments (duodenum, jejunum, ileum and colon) with the highest ones in the colon, where a greater number of non-ionized form of imperialine was present. Conclusion: The intestinal absorptive characteristics of imperialine are closely related to its physicochemical properties. The passive membrane diffusion dominates the intestinal absorption of imperialine.
