Dried ginger, a well-known medicine and food homologous production, has been widely circulated in China with high health benefits and economic value. Currently, there is still a lack of quality assessment on whether dried ginger in China exhibits chemically and biologically distinct properties, which creates a barrier to its quality control in commercial circulation. In this study, the chemical characteristics of 34 batches of common dried ginger samples in China were first explored using non-targeted chemometrics based on the UPLC-Q/TOF-MS analysis, leading to the identification of 35 chemicals that contributed to clustering into two categories, with sulfonated conjugates being the key chemically distinct components. By comparing the samples before and after sulfur-containing treatment and the further synthesis of a key differentiating component of [6]-gingesulfonic acid, it was then demonstrated that sulfur-containing treatment was the primary cause of the formation of sulfonated conjugates, as opposed to regional or environmental factors. Furthermore, the anti-inflammatory efficacy of dried ginger with high presence of sulfonated conjugates was significantly decreased. Consequently, for the first time, UPLC-QqQ-MS/MS was used to develop a targeted quantification method for 10 characteristic chemicals in dried ginger, allowing researchers to quickly determine whether dried ginger has been processed with sulfur and quantitatively evaluate the quality of dried ginger. These results provided an insight into the quality of commercial dried ginger in China and a suggested method for its quality supervision as well.

Gancao Ganjiang decoction (GGD) is one of the classic formulas in China known for its effective therapeutic effects in treating various diseases. Component research before and after the compatibility of Gancao and Ganjiang revealed that 6-gingerol content considerably changed. Compared with the decoction of Ganjiang, 6-gingerol in GGD demonstrated improved solubility. It was assumed that the partially hydrophilic glycyrrhizic acid and 6-gingerol self-assembled into nanoparticles, which then improved the solubility of 6-gingerol. The self-assembled nanoparticles in GGD were separated and characterized. Subsequently, 6-gingerol–glycyrrhizic acid nanoparticles (6-Gin-Gly NPs) were further prepared and characterized via dynamic light scattering (DLS) and transmission electron microscope (TEM). The formation mechanism of the self-assembled nanoparticles was examined using ultraviolet–visible spectroscopy (UV–vis), Fourier transform infrared spectroscopy (FTIR), electrospray ionization–mass spectrometry (ESI-MS), and molecular docking. The in vitro antioxidant activity of the 6-Gin-Gly NPs was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino bis(3-ethylbenzothiazoline-6- sulfonic acid) + (ABTS + ) free radical scavenging assays, which confirmed that the nanoparticles considerably enhanced their in vitro antioxidant capacity. In this study, the solubility of insoluble components was improved by the formation of self-assembled nanoparticles, which provided substantial reference data for the study of complicated multi-component interaction in herbal medicine decoction.

Background Chronic obstructive pulmonary disease (COPD) is a chronic respiratory disease. Licorice and dried ginger decoction (LGD) is traditional Chinese medicine prescription with multiple effects. Glycyrrhetinic acid (GA) is the main bioactive components of LGD, which has been proven to have a relieving effect on various inflammatory diseases. Siglec-1 is a cell surface sialoadhesin and has been confirmed to be overexpressed in COPD and facilitate inflammatory reaction. This study is aimed to probe the interaction between LGD, GA, and siglec-1. Methods Cigarette smoke (CS) combined with lipopolysaccharide (LPS) treatment was utilized to construct a COPD rat model. Cigarette smoke extract (CSE) was utilized to induce alveolar macrophage NR8383 to construct a COPD cell model. HE staining was applied for measuring histopathological changes of COPD rats. Enzyme-linked immunosorbent assay (ELISA), reverse transcription real-time polymerase chain reaction (RT-qPCR), and western blot were applied for testing the concentrations and expressions of proinflammatory factors. High performance liquid chromatography-tandem mass spectrometry (HPLC-MS) analysis was utilized to determine the combination between siglec-1 and GA. JC-1 assay was utilized to evaluate mitochondrial function. Reactive oxygen species (ROS) production was tested by dichloro-dihydro-fluorescein diacetate (DCFH-DA) staining. Results LGD treatment notably alleviated lung injury and inflammatory response in COPD rats. In CSE-induced cells, LGD treatment suppressed the contents of tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-6, and IL-8. Sialic-acid-binding Ig-like lectin 1 (Siglec-1) expression induced by CS was decreased after LGD treatment. Furthermore, we proved that GA could target siglec-1 to regulate the inflammatory response in COPD rats and cells . Additionally, GA could reduce ROS production and alleviate mitochondrial dysfunction to suppress COPD progression. Conclusion LGD inhibits inflammation and alleviates mitochondrial dysfunction in COPD by targeting siglec-1.

Xixin–Ganjiang herb pair (XGHP) is a classic combination for warming the lungs to dissolve phlegm and is often used to treat a variety of chronic lung diseases; it can treat the syndrome of cold phlegm obstruction of lungs. First, ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS) was used to examine the composition of XGHP, and network pharmacology was used to predict its potential core targets and signaling pathways in the current study. Second, a rat model of chronic obstructive pulmonary disease (COPD) was established for assessing the anti-COPD activity of XGHP, and metabolomics was used to explore the biomarkers and metabolic pathways. Finally, the sample was validated using molecular docking and Western blotting. The integration of metabolomics and network pharmacology results identified 11 targets, 3 biomarkers, 3 pathways, and 2 metabolic pathways. Western blotting showed that XGHP effectively regulated the expression of core proteins via multiple signaling pathways (downregulation of toll-like receptor 4 [ TLR4 ] and upregulation of serine/threonine-protein kinase 1 [ p-AKT1 ] and nitric oxide synthase 3 [ NOS3 ]). Molecular docking results showed that the 10 potentially active components of XGHP have good affinity with tumor necrosis factor-alpha ( TNF-α ), interleukin-6 ( IL-6 ), matrix metalloproteinase 9 ( MMP-9 ), TLR4 , p-AKT1 , and NOS3 . Our findings suggest that XGHP may regulate glucolipid metabolism, improve energy supply, and inhibit inflammatory responses ( TNF-α , IL-6 , and MMP-9 ) via the PI3K-Akt signaling pathway and HIF-1 signaling pathway in the management of COPD.

Xixin-Ganjiang Herb Pair (XGHP), a classic combination treatment to warm the lungs and dissolve phlegm, is widely used in the treatment of various pulmonary diseases. Chronic obstructive pulmonary disease (COPD) refers to a group of chronic obstructive airway diseases that can seriously harm human health. However, the effective components, targets, and pathways that underlie XGHP in the treatment of COPD remain unclear. Therefore, this study initially identified the effective components of XGHP through the use of UPLC–MS/MS and pharmacologic methods of traditional Chinese medicine. Secondly, transcriptomic analysis of the lung tissues of rats revealed the pharmacodynamic transcripts of each group, and metabolomics analysis revealed the differential metabolites associated with XGHP treatment. Finally, molecular docking of effective components with transcriptome genes was performed and western blotting was performed in order to determine the expression of related proteins in rat lung tissue. Overall, 30 effective components of XGHP were identified, including L-asarinin, 6-gingerol, sesamin, kaempferol, and quercetin. Transcriptomic studies demonstrated that expression of 386 genes recovered after XGHP treatment, and that they were mainly enriched in the oxidative phosphorylation and AMPK signaling pathways. According to the metabolomics studies, expression of eight metabolites differed between the COPD and the XGHP groups. These metabolites were mainly involved the biosynthesis of unsaturated fatty acids. Finally, the transcriptomic and metabolomics data were integrated. FASN and SCD in AMPK signaling pathway were directly linked to certain metabolites, including linoleic acid, palmitic acid, and oleic acid. These results indicate that XGHP can inhibit pAMPK expression and negatively regulate FASN and SCD expression during treatment of COPD in order to enhance the biosynthesis of unsaturated fatty acids and maintain energy homeostasis.
