Simple SummarySeveral plant species have diterpene compounds with juvenile hormone (JH) disruptor activity in insects. Demethoxycurcumin (DMC) and bisdemethoxycurcumin (BDMC) are two curcuminoid components of turmeric, interfering with the JH-mediated formation of the JH receptor complex. In vitro, DMC and BDMC also inhibit larval and pupal development inDrosophila melanogaster. The results suggest that both DMC and BDMC are JH disruptors that affect insect growth and development by regulating JH-mediated gene expression.AbstractJuvenile hormones (JHs) play a central role in insect development, reproduction, and various physiological functions. Curcuminoids generally exhibit a wide range of biological activities, such as antioxidant, anti-inflammatory, antibacterial, and insecticidal, and they exhibit insect growth inhibitory effects. However, research on insecticidal properties of curcuminoids has been limited. Moreover, to the best of our knowledge, studies on JHs of insects and curcuminoids are lacking. Therefore, this study aimed to identify the substances that act as JH disruptors (JHDs) from edible plants. Demethoxycurcumin (DMC) and bisdemethoxycurcumin (BDMC), two curcuminoids from the turmeric plantCurcuma longaL. inhibited the formation of a methoprene-tolerant (Met)–Taiman (Tai) heterodimer complex inDrosophila melanogaster, as shown through in vitro yeast two-hybrid assays. An artificial diet containing 1% (w/v) DMC or BDMC significantly reduced the number ofD. melanogasterlarvae in a concentration-dependent manner; larval development was disrupted, preventing the progression of larvae to pupal stages, resulting in an absence of adults. Building on the results obtained in this study on curcuminoids, researchers can use our study as a reference to develop eco-friendly pesticides. Keywords:juvenile hormone;methoprene-tolerant;diterpene;juvenile hormone disruptor;curcuminoids;demethoxycurcumin;bisdemethoxycurcumin

To better understand the application prospect of cyantraniliprole against the Asian corn borer, Ostrinia furnacalis, a diet-incorporation bioassay was adopted to determine the toxicity of cyantraniliprole against the fourth instar of O. furnacalis. Moreover, other experiments were conducted to examine effects of sublethal levels of cyantraniliprole on larval development, nutrient utilization, and reproduction. In this study, cyantraniliprole showed a high toxicity to fourth-instar larvae of O. furnacalis at dosages of 0.05 μg/g (LC5), 0.11 μg/g (LC20), 0.20 μg/g (LC40), and 0.26 μg/g (LC50) administered through artificial diet. At three sublethal dosages, cyantraniliprole inhibited larval feeding processes, decreased the relative growth rate, the relative consumption rate, the efficiency of food ingestion, and the efficiency of food digestion, as well as decreased the contents of nutrients such as proteins, lipids, and carbohydrates. All these effects go against to the normal growth of O. furnacalis, including reductions in larval and pupal weight, the extension of the larval and pupal period, the prolongation of the adult preoviposition period, total preoviposition period, and mean generation time, and the reduction of adult longevity, oviposition period, and eggs laid by female adults. Together, these changes resulted in the reduction of the intrinsic rate of increase. Hence, even at sublethal concentrations, cyantraniliprole can affect population dynamics by reducing the survival rate, fecundity, and population parameters. This result provides useful information for developing control strategies for O. furnacalis.

Sodium benzoate (SB), the sodium salt of benzoic acid, is widely used as a preservative in foods and drinks. The toxicity of SB to the human body attracted people’s attention due to the excessive use of preservatives and the increased consumption of processed and fast foods in modern society. The SB can inhibit the growth of bacteria, fungi, and yeast. However, less is known of the effect of SB on host commensal microbial community compositions and their functions. In this study, we investigated the effect of SB on the growth and development of Drosophila melanogaster larvae and whether SB affects the commensal microbial compositions and functions. We also attempted to clarify the interaction between SB, commensal microbiota and host development by detecting the response of commensal microbiota after the intervention. The results show that SB significantly retarded the development of D. melanogaster larvae, shortened the life span, and changed the commensal microbial community. In addition, SB changed the transcription level of endocrine coding genes such as ERR and DmJHAMT. These results indicate that the slow down in D. melanogaster larvae developmental timing and shortened life span of adult flies caused by SB intake may result from the changes in endocrine hormone levels and commensal microbiota. This study provided experimental data that indicate SB could affect host growth and development of D. melanogaster through altering endocrine hormone levels and commensal microbial composition.

Summary Drosophila has long been a successful model organism in multiple biomedical fields. Spatial gene expression patterns are critical for the understanding of complex pathways and interactions, whereas temporal gene expression changes are vital for studying highly dynamic physiological activities. Systematic studies in Drosophila are still impeded by the lack of spatiotemporal transcriptomic information. Here, utilizing spatial enhanced resolution omics-sequencing (Stereo-seq), we dissected the spatiotemporal transcriptomic changes of developing Drosophila with high resolution and sensitivity. We demonstrated that Stereo-seq data can be used for the 3D reconstruction of the spatial transcriptomes of Drosophila embryos and larvae. With these 3D models, we identified functional subregions in embryonic and larval midguts, uncovered spatial cell state dynamics of larval testis, and revealed known and potential regulons of transcription factors within their topographic background. Our data provide the Drosophila research community with useful resources of organism-wide spatiotemporally resolved transcriptomic information across developmental stages.

G protein-coupled receptors (GPCRs) constitute a diverse and crucial family of membrane receptors, regulating a wide array of physiological processes. Although the involvement of GPCR signaling pathways in modulating key genes associated with insecticide resistance has been documented in various insect species, the molecular mechanisms underlying GPCR-mediated resistance in Cydia pomonella remain largely unknown. To elucidate the molecular basis of lambda-cyhalothrin (LCT) resistance in C. pomonella , we performed comparative transcriptome sequencing on whole-body, head, and midgut tissues of larvae from a susceptible population (SS) and a laboratory-selected LCT-resistant population (LCR) derived from SS. Five GPCR candidates ( CpGPCRs ) exhibiting overexpression in LCR were identified, including two members of Family A ( CpGPCR4 and CpGPCR55 ), two of Family B ( CpGPCR68 and CpGPCR76 ), and one of Family F ( CpGPCR94 ). Spatiotemporal expression profiling revealed that these genes were predominantly expressed during the first-instar larval and pupal-adult stages, with notably high expression levels in the larval heads. Notably, all genes exhibited significantly elevated expression in the midgut of LCR larvae compared to SS larvae. RNA interference (RNAi)-mediated knockdown of CpGPCR4 , CpGPCR55 , and CpGPCR68 increased larval susceptibility to LCT, while knockdown of CpGPCR4 and CpGPCR55 significantly reduced larval survival rates. Furthermore, larvae treated with dsGPCR55 exhibited molting defects and a decline in 20-hydroxyecdysone (20E) titers. These findings demonstrate that CpGPCRs play essential roles in the survival and development of C. pomonella and are critically involved in mediating resistance to LCT. This study enhances our understanding of the molecular mechanisms driving insecticide resistance and identifies potential targets for the development of novel pest management strategies against C. pomonella .
