The cell invasion preference of Varroa destructor between the original and new honey bee hosts

Wenfeng Li,Yi Zhang,Hui Peng,Ruonan Zhang,Zhengwei Wang,Zachary Y. Huang,Yan Ping Chen,Richou Han

Honey bees ( Apis ) are important pollinators for food crops and wild plants, but are facing great threats from pathogens and parasites, especially an obligate ectoparasitic mite, Varroa destructor . Cell invasion is a key step for V. destructor to reproduce, and the parasite displays remarkable host preference in this process. Varroa destructor made its host-shift from its original host, the Asian honey bee Apis cerana, to the new host, the European honey bee Apis mellifera several decades ago. However, it remains largely unstudied whether V. destructor shows a cell invasion preference between the two host species. Using cell invasion bioassays on a modified four-well arena, we showed that V. destructor significantly preferred to invade the worker and drone larvae of A. mellifera rather than A. cerana , suggesting that the new host is much more attractive to the parasite than the original one. Using gas chromatography-mass spectrometry (GC-MS), we revealed significant differences between the cuticular hydrocarbon (CHC) profiles of worker and drone larvae of the two bee hosts. The amounts of methyl-branched alkanes and alkenes (unsaturated CHCs), but not n -alkanes, were significantly different, and A. mellifera worker and drone larvae were found to express significantly higher amounts of methyl-alkanes, while A. cerana larvae produced higher amounts of alkenes. Cell invasion bioassays with glass dummies showed that the mites preferred the glass dummies coated with the CHCs of A. mellifera worker or drone larvae, which indicates a role of larval CHCs in mediating the preferential cell invasion of Varroa . The findings from this study extend our understanding of the host preference of V. destructor , and can potentially contribute to the development of effective strategies for mite control.

Quick shift in volatile attraction between the third and fifth instar larvae of Endoclita signifier

Ping Hu,Zhisong Qiu,Yujing Zhang,Yuan Xu,Zhende Yang

BACKGROUND Endoclita signifer is a polyphagous lepidopteran species of eucalyptus that selects its hosts in the third-instar larval period. To understand how it adapts to its host during development, we studied the olfactory responses of late-stage (fifth)-instar larvae to the dynamic chemical environment they encounter. RESULTS Thirty-two volatiles from eucalyptus trunk and soil were identified, among which 14 showed electroantennal activity and five were identified as new. Further behavioral bioassay showed that both β-pinene and the imitation ratio of six key volatile in eucalyptus trunk were attractive to the fifth-instar larvae, but both eliminated and increased β-pinene in the mixture decreased the choice ratio and showed no attraction. Although E. signifer larvae shifted their attraction from o -cymene at the third-instar stage to β-pinene at the fifth-instar stage in a single volatile, the appropriate ratio of the main compounds in eucalyptus trunk volatile is the key to the behavior choice of fifth-instar larvae. CONCLUSION The switch in olfactory attraction to different compounds between fifth- and third-instar larvae indicated an olfactory plasticity between third- and fifth-instar larvae. And the particular blend mediated the highly specialized communication interactions specificity between fifth-instar larvae and eucalyptus trunk volatile indicated the specialized host adaptation to fifth-instar larvae. This enhances understanding of how the primitive lepidopteran E. signifier , as a native pest, adapts to introduced eucalyptus. Moreover, this study provides knowledge for the screening and development of target volatiles for trapping and managing E. signifer larvae. © 2022 Society of Chemical Industry.

Green liquid marble-based hydrogels as pesticidal pyrethrin slow-release carriers

Qin Li,Changhong Wang,Jiayuan He,Dandan Yang,Ting Li,Huixian Xu,Weifeng Shen,Lian-Di Zhou,Saimeng Jin,Qi-Hui Zhang,James H. Clark

Pyrethrin are effective agents against mosquito larval but their rapid degradation rate in water reduces the deinsectization efficiency and increases the cost of deinsectization. Based on liquid marble,a biodegradable slow-release hydrogel carrier is prepared using green and low-cost materials for the slow release of natural pyrethrin on water surfaces to kill mosquito larvae. Green carriers with high stability and loading of high concentrations of pyrethrin are synthesized by cross-linking Ca2+ in 5% CaCl2 solution with sodium alginate. The superior stability of liquid marble at the water surface and the electrostatic interaction between alginate and gelatin can effectively reduce the degradation rate of pyrethrin in water. Furthermore, the carrier system displayed long sustained-release time (>144 h) and maintained larvicidal performance after 168 h. Good floating stability is demonstrated for the liquid marble hydrogels. The larvicidal activity of obtained drug carrier reached 100% with different water volume but same dosage. This work provides a strategy to highly efficiently exterminate mosquito larvae by preparing a green hydrogel drug slow-release carrier, which provides a new approach to expanding the application of liquid marbles in green chemistry.

Larvicidal Activity of Two Rutaceae Plant Essential Oils and Their Constituents Against Aedes albopictus (Diptera: Culicidae) in Multiple Formulations

Jian Rongchao,Lin Yuan,Li Yuling,Wu Weifeng,Ren Xiaofei,Liang Zhanyuan,Kong Lingjia,Cai Jinglin,Lao Canyao,Wu Min,Chen Wenhua,Chen Jing,Hong Weiqian David,Sheng Zhaojun

Aedes albopictus (Skuse) is a vector of several arboviruses, such as dengue, chikungunya, West Nile, and Zika viruses. At present, the use of synthetic insecticides is the main vector control strategy. However, the widespread and long-term use of insecticides has aroused several problems, including insecticide resistance, environmental pollution, and non-target species effects, thereby encouraging researchers to search for new alternatives derived from natural products. In recent decades, essential oils (EOs) as natural alternatives to control mosquitoes have received increasing attention. In the initial larvicidal activity screen, two Rutaceae plants (Citrus aurantium and Citrus paradisi) EOs were selected and evaluated for killing Ae. albopictus larvae. The LC50 values of C. aurantium and C. paradisi EOs against Ae. albopictus were 91.7 and 100.9 ppm, respectively. The main components of C. aurantium EO include diethyl o-phthalate (37.32%), limonene (10.04%), and methyl dihydrojasmonate (6.48%). The main components of C. paradisi EO include limonene (60.51%), diethyl o-phthalate (11.75%), linalool (7.90%), and styralyl acetate (6.28%). Among these main components of the two EOs, limonene showed potent larvicidal activity, with the LC50 value of 39.7 ppm. The nanoemulsions of limonene were prepared and characterized. The duration of larvicidal activity was greater in the limonene nanoemulsions than when limonene was applied in solvent. This study demonstrates that EOs of plants in family Rutaceae are a potential resource to develop new larvicides, and nanoemulsification is an effective method for improving the physicochemical properties and efficacy of natural products as larvicides.

The Herbicide Glyphosate Negatively Affects Midgut Bacterial Communities and Survival of Honey Bee during Larvae Reared in Vitro

Pingli Dai,Zhenxiong Yan,Shilong Ma,Yang Yang,Qiang Wang,Chunsheng Hou,Yanyan Wu,Yongjun Liu,Qingyun Diao

Effects of glyphosate on survival, developmental rate, larval weight, and midgut bacterial diversity of Apis mellifera were tested in the laboratory. Larvae were reared in vitro and fed diet containing glyphosate 0.8, 4, and 20 mg/L. The dependent variables were compared with negative control and positive control (dimethoate 45 mg/L). Brood survival decreased in 4 or 20 mg/L glyphosate treatments but not in 0.8 mg/L, and larval weight decreased in 0.8 or 4 mg/L glyphosate treatments. Exposure to three concentrations did not affect the developmental rate. Furthermore, the intestinal bacterial communities were determined using high-throughput sequencing targeting the V3–V4 regions of the 16S rDNA. All core honey bee intestinal bacterial phyla such as Proteobacteria (30.86%), Firmicutes (13.82%), and Actinobacteria (11.88%) were detected, and significant changes were found in the species diversity and richness in 20 mg/L glyphosate group. Our results suggest that high concentrations of glyphosate are deleterious to immature bees.

Laboratory Evaluation of Larvicidal Activity of the Essential oil of Allium tuberosum Roots and its Selected Major Constituent Compounds Against Aedes albopictus (Diptera: Culicidae)

Liu Xin Chao,Zhou Ligang,Liu Qiyong,Liu Zhi Long

The aim of this research was to evaluate larvicidal activity of the essential oil of Allium tuberosum Rottler ex Sprengle roots and its constituents against larval mosquitoes (Aedes albopictus Skuse). Essential oil of A. tuberosum was obtained by hydrodistillation and analyzed by gas chromatography and gas chromatography–mass spectrometry. The major constituents of the oil were found to be sulfur-containing compounds, including allyl methyl trisulfide (35.19%), diallyl disulfide (28.31%), diallyl trisulfide (20.91%), and dimethyl trisulfide (12.33%). The essential oil of A. tuberosum exhibited larvicidal activity against the fourth-instar larvae of Ae. albopictus, with an LC50 value of 18 μg/ml. The constituents compounds—diallyl trisulfide (LC50 = 4 μg/ml) and diallyl disulfide (LC50 = 6 μg/ml)—possessed stronger larvicidal activity than allyl methyl trisulfide (LC50 = 27 μg/ml) and dimethyl trisulfide (LC50 = 35 μg/ml) against the fourth-instar larvae of Ae. albopictus. The results indicated that the essential oil of A. tuberosum and its major constituents have good potential as a source for natural larvicides.

Effects of Different Nitrogen Sources and Ratios to Carbon on Larval Development and Bioconversion Efficiency in Food Waste Treatment by Black Soldier Fly Larvae (Hermetia illucens)

Yan Lu,Shouyu Zhang,Shibo Sun,Minghuo Wu,Yongming Bao,Huiyan Tong,Miaomiao Ren,Ning Jin,Jianqiang Xu,Hao Zhou,Weiping Xu

Simple SummaryBlack soldier fly larvae (BSFL) have received global research interest and industrial application due to their high performance on the organic waste treatment. However, the substrate C/N property, which may affect larvae development and the waste bioconversion process greatly, is significantly less studied. The current study focused on the food waste treatment by BSFL, compared the nitrogen supplying effects of 9 nitrogen species (i.e., NH4Cl, NaNO3, urea, uric acid, Gly, L-Glu, L-Glu:L-Asp (1:1,w/w), soybean flour, and fish meal), and further examined the C/N effects on the larval development and bioconversion process. We found that NH4Cl and NaNO3led to poor larval growth and survival, while 7 organic nitrogen species exerted no harm to the larvae. Urea was further chosen to adjust the C/Ns. Results showed that lowering the C/N from the initial 21:1 to 18:1–14:1 improved the waste reduction and larvae production performance, and C/N of 18:1–16:1 was further beneficial for the larval protein and lipid bioconversion, whereas C/N of 12:1–10:1 resulted in a significant performance decline. Therefore, the C/N range of 18:1–16:1 is likely the optimal condition for food waste treatment by BSFL and adjusting food waste C/N with urea could be a practical method for the performance improvement.AbstractBiowaste treatment by black soldier fly larvae (BSFL,Hermetia illucens) has received global research interest and growing industrial application. Larvae farming conditions, such as temperature, pH, and moisture, have been critically examined. However, the substrate carbon to nitrogen ratio (C/N), one of the key parameters that may affect larval survival and bioconversion efficiency, is significantly less studied. The current study aimed to compare the nitrogen supplying effects of 9 nitrogen species (i.e., NH4Cl, NaNO3, urea, uric acid, Gly, L-Glu, L-Glu:L-Asp (1:1,w/w), soybean flour, and fish meal) during food waste larval treatment, and further examine the C/N effects on the larval development and bioconversion process, using the C/N adjustment with urea from the initial 21:1 to 18:1, 16:1, 14:1, 12:1, and 10:1, respectively. The food wastes were supplied with the same amount of nitrogen element (1 g N/100 g dry wt) in the nitrogen source trial and different amount of urea in the C/N adjustment trial following larvae treatment. The results showed that NH4Cl and NaNO3caused significant harmful impacts on the larval survival and bioconversion process, while the 7 organic nitrogen species resulted in no significant negative effect. Further adjustment of C/N with urea showed that the C/N range between 18:1 and 14:1 was optimal for a high waste reduction performance (73.5–84.8%,p< 0.001) and a high larvae yield (25.3–26.6%,p= 0.015), while the C/N range of 18:1 to 16:1 was further optimal for an efficient larval protein yield (10.1–11.1%,p= 0.003) and lipid yield (7.6–8.1%,p= 0.002). The adjustment of C/N influenced the activity of antioxidant enzymes, such as superoxide dismutase (SOD,p= 0.015), whereas exerted no obvious impact on the larval amino acid composition. Altogether, organic nitrogen is more suitable than NH4Cl and NaNO3as the nitrogen amendment during larval food waste treatment, addition of small amounts of urea, targeting C/N of 18:1–14:1, would improve the waste reduction performance, and application of C/N at 18:1–16:1 would facilitate the larval protein and lipid bioconversion process. Keywords:nitrogen source;carbon to nitrogen ratio;food waste;urea;black soldier fly larvae;Hermetia illucens

Yeast enrichment facilitated lipid removal and bioconversion by black soldier fly larvae in the food waste treatment

Hongxu Lu,Yanxia Liu,Wenyue Shen,Yang Zhou,Xiangwei Ma,Shibo Sun,Xiaoying Dong,Fengyun Ji,Huiyan Tong,Jianqiang Xu,Gaohong He,Weiping Xu

Food waste can be converted into insectile fatty acids (FAs) by the larvae of black soldier fly (BSFL), Hermetia illucens , for use in the feed sector or as a source of biodiesel. However, waste oil was less decomposed than carbohydrate or protein in frass due to the limitation of larval lipid metabolism. In this study, 10 yeast strains were screened, corresponding to six species, to examine their capacity of improving lipid transformation performance by BSFL. The species of Candida lipolytica was superior to the other five species, which exhibited significantly higher lipid reduction rate (95.0–97.1 %) than the control (88.7 %), and the larval FA yields achieved 82.3–115.5 % of the food waste FA matters, suggesting that BSFL not only transformed waste oil but also biosynthesized FAs from waste carbohydrate and other substances. Further, the CL2 strain of Candida lipolytica was examined for treating food waste containing high lipid content (16–32 %). The lipid removal rate was found improved from 21.4 to 42.3 % (control) to 80.5–93.3% in the waste containing 20–32 % lipid. The upper limit of lipid content that could be endured by BSFL was ≈16 %, and the CL2-enrichment elevated the upper limit to ≈24 %. Fungal community analysis indicated that Candida spp. accounted for the lipid removal improvement. The Candida spp. CL2 strain may facilitate the lipid reduction and transformation by BSFL through microbial catabolizing and assimilation of waste FAs. Altogether, this study suggests that yeast enrichment is feasible in improving lipid transformation by BSFL especially for food waste exhibiting high lipid content.

Carvacrol acts on the larval midgut of Spodoptera frugiperda by destroying the tissue structure and altering the bacterial community

Jingjing Zhang,Cuiting Liu,Xinyi Xie,Xuanyue Yu,Yuting Huang,Rui Tang,Jian Zeng,Benshui Shu

The fall armyworm, Spodoptera frugiperda, has become a global invasive pest in recent years, threatening agricultural production. Carvacrol (CAR) is a multifunctional plant-derived compound and exhibited significant bioactivities against many pests. Our previous study found that CAR inhibited growth and development in S. frugiperda . However, the effects of CAR on the midgut of this pest remain unknown. In this study, the actions of CAR on the larval midgut of S. frugiperda were investigated. The results found that 1.0 and 2.0 g/kg CAR treatments destroyed the structure of the larval midgut based on hematoxylin-eosin staining and transmission electron microscope observation. In addition, 2.0 g/kg CAR exposure significantly altered the larval midgut bacterial community composition and structure. The results of the linear discriminant analysis effect size (LEfSe) analysis suggest that six bacterial genera contributed to the different structures of the larval bacterial community. Furthermore, PICRUSt2 analysis found that 16 bacterial function categories were altered by CAR treatment, of which 8 were significantly increased. Our results provided new insight into the toxicological mechanisms of CAR against S. frugiperda larvae and laid the foundation for the field application of S. frugiperda control.

Property of midgut α-amylase from Mythimna separata (Lepidoptera: Noctuidae) larvae and its responses to potential inhibitors in vitro

Wenping Xu,Qingchun Huang,Xiwei Wu,Xiaoqin Yu,Xuexiao Wang,Liming Tao

Midgut α-amylase is an important digestive enzyme involved in larval energy metabolism and carbohydrate assimilation. In this article, the properties of midgut α-amylase from the Oriental armyworm, Mythimna separata (Lepidoptera: Noctuidae), larvae were characterized, and its in vitro responses to chemical inhibitors were also determined. The kinetic parameters Km and Vmax of midgut α-amylase were 0.064 M, 4.81 U mg pro(-1) in phosphate buffer, and 0.128 M, 1.96 U mg pro(-1) in barbiturate-acetate buffer; α-amylase activity linearly increased as starch concentration increased. α-Amylase activity was not influenced by amino acids such as Pro, Met, Try, His, Ala, and Phe but was strongly activated by antioxidants. Reduced glutathione, 1,4-dithiothreitol, β-mercaptoethanol, and ascorbic acid improved the activity of α-amylase about 2.06, 3.46, 3.37, and 6.38 times, respectively, relative to the control. Ethylenediaminetetraacetic acid, sodium dodecyl sulfonate, and N-bromosuccinimide (NBS) strongly inhibited α-amylase. α-, β-, and γ-cyclodextrin were not the preferred substrates for α-amylase. Kinetic analysis showed that IC50 value of NBS against α-amylase was 1.52 (±0.26) µM, and the mode of action of NBS with Ki as 2.53 (0.35) µM was a mixed-type inhibition that indicated a combination of partial competitive and pure noncompetitive inhibition. The midgut α-amylase of armyworm larvae may be a potential target for novel insecticide development and pest control.

Identification of Larvicidal Constituents of the Essential Oil of Echinops grijsii Roots against the Three Species of Mosquitoes

Mei Ping Zhao,Qi Zhi Liu,Qiyong Liu,Zhi Long Liu

The screening of Chinese medicinal herbs for insecticidal principles showed that the essential oil ofEchinops grijsiiHance roots possessed significant larvicidal activity against mosquitoes. The essential oil was extracted via hydrodistillation and its constituents were determined by gas chromatography-mass spectrometry (GC-MS) analysis. GC-MS analyses revealed the presence of 31 components, with 5-(3-buten-1-yn-1-yl)-2,2′-bithiophene (5-BBT, 27.63%), α-terthienyl (α-T, 14.95%), 1,8-cineole (5.56%) andcis-β-ocimene (5.01%) being the four major constituents. Based bioactivity-directed chromatographic separation of the essential oil led to the isolation of 5-BBT, 5-(4-isovaleroyloxybut-1-ynyl)-2,2′-bithiophene (5-IBT) and α-T as active compounds. The essential oil ofE. grijsiiexhibited larvicidal activity against the fourth instar larvae ofAedes albopictus,Anopheles sinensisandCulex pipiens pallenswith LC50values of 2.65 μg/mL, 3.43 μg/mL and 1.47 μg/mL, respectively. The isolated thiophenes, 5-BBT and 5-IBT, possessed strong larvicidal activity against the fourth instar larvae ofAe. albopictus(LC50= 0.34 μg/mL and 0.45 μg/mL, respectively) andAn. sinensis(LC50= 1.36 μg/mL and 5.36 μg/mL, respectively). The two isolated thiophenes also had LC50values against the fourth instar larvae ofC. pipiens pallensof 0.12 μg/mL and 0.33 μg/mL, respectively. The findings indicated that the essential oil ofE. grijsiiroots and the isolated thiophenes have an excellent potential for use in the control ofAe. albopictus,An. sinensisandC. pipiens pallenslarvae and could be used in the search for new, safer and more effective natural compounds as larvicides. Keywords:Echinops grijsii;Aedes albopictus;Anopheles sinensis;Culex pipiens pallens;larvicidal activity
