Turn-on mode probe based on the sustainable xanthohumol extract for the efficient viscosity response in a liquid system

Lingfeng Xu,Xinya Liu,Jingyi Zhao,Xinmin Deng,Hui Peng

Viscosity is a typical physical parameter and plays an important role in nutrient transferring, diffusion process regulating and safety warning. Aberrant mitochondrial viscosity is closely associated with an imbalance in a liquid system. Nevertheless, there is currently a lack of convenient and efficient tools for the mutation of viscosity detection at the molecular level. Herein, a natural product xanthohumol (XTH) was extracted from Humulus lupulus and used to measure the microenvironmental viscosity. Due to the existence of carbonyl and phenolic hydroxyl groups, a typical twisted intramolecular charge transfer (TICT) was formed. The conjugated single and double bonds can be employed as the rotatable site. Consequently, a turn-on method based on viscosity response is developed. High sensitivity (x = 0.56) with a remarkable enhancement (55-fold) toward viscosity and a visualized fluorescent signal can be found. In addition, it displays a single selectivity with excellent photostability and pH stability in the complex liquid system. Using the extracted XTH, a typical application toward the liquid spoilage process was performed and a positive correlation was noted. Given the comprehensive properties of XTH, liquid safety inspection at a molecular level with natural source-extracted products can be obtained.

Dual role of BSA for synthesis of MnO2 nanoparticles and their mediated fluorescent turn-on probe for glutathione determination and cancer cell recognition

Qi Wang,Yuan Zhang,Xiaodong Wang,Yuehuan Wu,Chuan Dong,Shaomin Shuang

A MnO2 nanoparticle (MnO2 NP)-mediated fluorescent turn-on probe for sensitively and selectively detecting glutathione (GSH) and recognizing cancer cells was established in this work. MnO2 NPs were synthesized simply and quickly through an in situ redox reaction by mixing bovine serum albumin (BSA) and KMnO4, in which BSA served the dual roles of template and reductant. It was found that the MnO2 NPs served as an effective energy acceptor and quenched the fluorescence intensity of carbon dots (CDs), owing to the fluorescence resonance energy transfer (FRET) process. Further, the addition of GSH triggered the decomposition of MnO2, breaking the FRET between MnO2 NPs and CDs and thus restoring the fluorescence intensity of CDs. Based on this mechanism, quantitative determination of GSH was performed. Under optimal conditions, a satisfactory linear range of 0.05–90 μM and limit of detection of 39 nM were obtained, and GSH content in human serum samples was detected. Moreover, taking advantage of the higher levels of GSH in cancer cells than in normal cells, the MnO2 NP-CD probe was applied to distinguish SMMC-7721 cancer cells from L02 normal cells. The FRET was interrupted by GSH in cancer cells, and strong fluorescence was observed. This work provides a facile approach for synthesizing MnO2 NPs, and this rapid, low-cost method with no need for reductants makes synthesis green and convenient. The MnO2 NP-mediated fluorescent turn-on response to GSH could improve the MnO2 nanomaterial-based biochemical analysis applications.

Simultaneous Detection of Antibiotic Resistance Genes on Paper-Based Chip Using [Ru(phen)2dppz]2+ Turn-on Fluorescence Probe

Bofan Li,Xiaoming Zhou,Hongxing Liu,Huaping Deng,Ru Huang,Da Xing

Antibiotic resistance, the ability of some bacteria to resist antibiotic drugs, has been a major global health burden due to the extensive use of antibiotic agents. Antibiotic resistance is encoded via particular genes; hence the specific detection of these genes is necessary for diagnosis and treatment of antibiotic resistant cases. Conventional methods for monitoring antibiotic resistance genes require the sample to be transported to a central laboratory for tedious and sophisticated tests, which is grueling and time-consuming. We developed a paper-based chip, integrated with loop-mediated isothermal amplification (LAMP) and the “light switch” molecule [Ru(phen)2dppz]2+, to conduct turn-on fluorescent detection of antibiotic resistance genes. In this assay, the amplification reagents can be embedded into test spots of the chip in advance, thus simplifying the detection procedure. [Ru(phen)2dppz]2+ was applied to intercalate into amplicons for product analysis, enabling this assay to be operated in a wash-free format. The paper-based detection device exhibited a limit of detection (LOD) as few as 100 copies for antibiotic resistance genes. Meanwhile, it could detect antibiotic resistance genes from various bacteria. Noticeably, the approach can be applied to other genes besides antibiotic resistance genes by simply changing the LAMP primers. Therefore, this paper-based chip has the potential for point-of-care (POC) applications to detect various gene samples, especially in resource-limited conditions.

Design mitochondria-specific fluorescent turn-on probes targeting G-quadruplexes for live cell imaging and mitophagy monitoring study

Meng-Ting She,Jia-Wei Yang,Bo-Xin Zheng,Wei Long,Xuan-He Huang,Jun-Ren Luo,Ze-Xin Chen,Ao-Lu Liu,Dong-Peng Cai,Wing-Leung Wong,Yu-Jing Lu

A series of new mitochondrial-selective fluorescent probe was designed and synthesized based on the integration of two planar molecular scaffolds of benzo-indole/indole with a p -substituted styrene moiety. The small-sized probes are flexible and rotatable via an ethylene bridge. The restriction of free rotation upon interaction with targeting biomolecules of G-quadruplexes in mitochondria generates strong emission in visible range (575–615 nm). Cell imaging study showed that the ligands are targeting mitochondria but not nucleus. Competition experiments showed that the ligand BYM is highly selective towards mitochondrial G4-DNA structures against other non-G4 nucleic acid structures including single-/double-stranded DNA and hairpin. The equilibrium binding constant ( K eq ) of BYM interacting with mitochondrial G4-DNA (mt6363, K eq  = 10.8 × 10 6 M −1 ) is almost 1000-fold higher than that of mitochondrial double-stranded DNA ( K eq  = 0.01 × 10 6 M −1 ). The probe also showed high sensitivity (LOD = 1.52 nM) and good linear relationship (R 2  = 0.9983) with the mitochondrial G4-DNA. The delivery of BYM to mitochondria is not mitochondrial membrane potential dependent but mainly through the permeability transition pore on the mitochondrial inner membrane. In addition, BYM exhibits low cytotoxicity against a number of human cancer and noncancerous cells, indicating that BYM could be a good ligand for bioorthogonal study in live cells by targeting the mitochondrial G4-DNA structures. In the present study, BYM was demonstrated in monitoring the dynamic process of mitochondrial autophagy. The small-sized fluorescent probe possessing high photostability, selectivity and sensitivity targeting mitochondrial G-quadruplexes may able to provide a useful chemical tool for real-time study of mitochondrial functions in live cells.
