ACC SYNTHASE4 inhibits gibberellin biosynthesis and FLOWERING LOCUS T expression during citrus flowering

Chu Le-Le,Zheng Wei-Xuan,Liu Hai-Qiang,Sheng Xing-Xing,Wang Qing-Ye,Wang Yue,Hu Chun-Gen,Zhang Jin-Zhi

Flowering is an essential process in fruit trees. Flower number and timing have a substantial impact on the yield and maturity of fruit. Ethylene and gibberellin (GA) play vital roles in flowering, but the mechanism of coordinated regulation of flowering in woody plants by GA and ethylene is still unclear. In this study, a lemon (Citrus limon L. Burm) 1-aminocyclopropane-1-carboxylic acid synthase gene (CiACS4) was overexpressed in Nicotiana tabacum and resulted in late flowering and increased flower number. Further transformation of citrus revealed that ethylene and starch content increased, and soluble sugar content decreased in 35S:CiACS4 lemon. Inhibition of CiACS4 in lemon resulted in effects opposite to that of 35S:CiACS4 in transgenic plants. Overexpression of the CiACS4-interacting protein ETHYLENE RESPONSE FACTOR3 (CiERF3) in N. tabacum resulted in delayed flowering and more flowers. Further experiments revealed that the CiACS4–CiERF3 complex can bind the promoters of FLOWERING LOCUS T (CiFT) and GOLDEN2-LIKE (CiFE) and suppress their expression. Moreover, overexpression of CiFE in N. tabacum led to early flowering and decreased flowers, and ethylene, starch, and soluble sugar contents were opposite to those in 35S:CiACS4 transgenic plants. Interestingly, CiFE also bound the promoter of CiFT. Additionally, GA3 and 1-aminocyclopropanecarboxylic acid (ACC) treatments delayed flowering in adult citrus, and treatment with GA and ethylene inhibitors increased flower number. ACC treatment also inhibited the expression of CiFT and CiFE. This study provides a theoretical basis for the application of ethylene to regulate flower number and mitigate the impacts of extreme weather on citrus yield due to delayed flowering.

Pectin Metabolism Influences Phloem Architecture and Flowering Time in Arabidopsis Thaliana

Qing Zhang,Juan Du,Shuang Hao,Zhengmei Leng,Weiguo Liu,Chengjian Zhao,Yue Yu,Longquan Chen,Charles T. Anderson,Chaowen Xiao

Cell wall matrices undergo continuous remodeling during plant growth and development. The methyl- and de-methyl-esterification of pectic homogalacturonan dynamically influence the physicochemical and mechanical properties of cell walls, and consequently regulate organ morphogenesis. Phloem plays a crucial role in the transport of photoassimilates and other organic molecules from source to sink tissues, and the walls of phloem cells are rich in pectins. However, how pectin metabolism orchestrates phloem architecture and function is unclear. This work demonstrates that the methyl-esterification status and subsequent degradation of pectic homogalacturonan affect flowering time by modulating phloem development and transport capacity, which control the transport of a mobile florigen, FLOWERING LOCUS T protein, in the phloem stream. During this process, auxin signaling is stimulated by pectin de-methyl-esterification to influence vasculogenesis. Together, this data provide new insights into the mechanisms by which pectin chemistry is coordinated with auxin signaling to mediate phloem development, long-distance transport and flowering time.

Onion cryptochrome 1 (AcCRY1) regulates photomorphogenesis and photoperiod flowering in Arabidopsis and exploration of its functional mechanisms under blue light

Qingwei Jia,Yuqing Yin,Shuting Gai,Lu Tian,Zhihao Zhu,Lei Qin,Yong Wang

Cryptochromes (CRYs), as blue-light photoreceptors, play a crucial role in regulating flowering time and hypocotyl and cotyledon development. Their physiological functions have been extensively studied in various plant species. However, research on onions remains limited. In this study, we identified AcCRY1 and conducted preliminary investigations into its function. Our results demonstrate that AcCRY1 possesses a conserved domain typical of cryptochromes with high homology to those found in monocots. Furthermore, we examined the expression level of AcCRY1 in onion. The green tissues is significantly higher compared to non-green tissues, and it exhibits a significant response to blue-light induction. AcCRY1 demonstrates cytoplasmic localization under blue-light conditions, while it localizes in the nucleus during darkness, indicating a strong dependence on blue-light for its subcellular distribution. In comparison to cry1 , overexpression of AcCRY1 leads to a significant shorten in seedling hypocotyl length, notable expansion of cotyledons, and acceleration of flowering time. The yeast two-hybrid experiment demonstrated the in vitro interaction between AcCRY1, AcCOP1, and AcSPA1. Additionally, BIFC analysis confirmed their interaction in Onion epidermis. Notably, under blue-light conditions, a significantly enhanced binding activity was observed compared to dark conditions. These findings establish a functional foundation for the regulatory role of AcCRY1 in important physiological processes of onion and provide initial insights into the underlying molecular mechanisms.

An integrated strategy for rapid discovery and prediction of nucleobases, nucleosides and amino acids as quality markers in different flowering stages of Flos Chrysanthemi using UPLC–MS/MS and FT-NIR coupled with multivariate statistical analysis

Xiangwei Chang,Dandan Wei,Shulan Su,Sheng Guo,Sien Qian,Hui Yan,Ming Zhao,Erxin Shang,Dawei Qian,Xiaodong Sun,Jin-ao Duan

Nucleobases, nucleosides, nucleotides and amino acids are important nutritional compounds, and some of them are positively correlated with the umami taste in scented tea. Taking these nutrients of Flos Chrysanthemi from three different flowering stages as the research object, an integrated strategy of ultra high-performance liquid chromatography coupled with triple-quadrupole linear ion-trap tandem mass spectrometry (UPLC–MS/MS), Fourier transform near infrared spectroscopy (FT-NIR) and multivariate statistical analysis was proposed to set up a systematic quality evaluation system for Flos Chrysanthemi from discovery to the rapid prediction of quality markers. First, a rapid, sensitive and accurate UPLC–MS/MS method was established to simultaneously determine 40 nucleobases, nucleosides, nucleotides and amino acids in three different flowering stages of Flos Chrysanthemi. The results indicated that Flos Chrysanthemi from three different flowering stages are rich in 40 nutritional compounds detected above and could be developed as functional products possessed nutraceutical and bioactive properties. Next, ten potential quality markers for three flowering stages of Flos Chrysanthemi were screened using partial least squares discriminant analysis (PLS-DA) and the plot of variable importance for projection (VIP), and the related metabolic pathways were identified for the first time. Furthermore, three of the ten potential quality markers, namely, glutamate, asparagine and aspartate were selected as key quality markers. Finally, three best PLS regression models constructed with FT-NIR were developed to predict the contents of the three key quality markers using the optimal spectral preprocessing methods and spectral interval combinations screened by synergy interval-PLS (Si-PLS), which yielded excellent prediction performance: 0.9766 ≤ R 2 p (coefficient of determination for prediction) ≤ 0.9858 and 6.3059 ≤ RPD (ratio of prediction to deviation) ≤ 7.3613. To sum up, the integrated quality marker discovery and FT-NIR quantitation strategy was suitable for fast quality evaluation of Flos Chrysanthemi.

Strigolactone represses the synthesis of melatonin, thereby inducing floral transition in Arabidopsis thaliana in an FLC-dependent manner

Zixin Zhang,Qian Hu,Yanan Liu,Peilei Cheng,Hua Cheng,Weixin Liu,Xiaojuan Xing,Zhiyong Guan,Weimin Fang,Sumei Chen,Jiafu Jiang,Fadi Chen

The transition from vegetative to reproductive growth is a key developmental event in a plant's life cycle. The process is mediated by a combination of phytohormones, including melatonin (MT) and strigolactone (SL). Here, the Arabidopsis mutants, d14-1 and max4-1 , which are compromised with respect to either SL synthesis or signaling, were shown to flower earlier than wild types. The tissue MT content in both mutants was higher than in wild types, as a result of the up-regulation of various genes encoding enzymes involved in MT synthesis. The abundance in the mutants of transcripts derived from each of the genes SPLs , AP1 , and SOC1 was reduced with exogenously supplied MT, while FLC was induced. Plants exposed to a high concentration of MT did not flower earlier than wild types. The tissue MT content of a mutant unable to synthesize caffeic acid O -methyltransferase was less than that of wild type and flowered earlier than did wild types. The suggestion is that the flowering time of Arabidopsis is altered if the tissue content of MT is either higher than ~ 8 ng/g F.W, or lower than ~ 0.9 ng/g. Within this range, SL acts to determine flowering time by its regulation of SPL gene s . The application of exogenous SL reduces tissue MT content. The flowering time of the flc-3 mutant was unaffected by exogenously supplying either MT or/and SL. It is proposed that MT acts downstream of SL to activate FLC , inducing a delay to flowering if its concentration lies outside a certain range.

Genome-Wide Identification of the PEBP Family Gene in Three Cymbidium Species and Its Expression Patterns Analysis in C. ensifolium

Jinliao Chen,Fei Wang,Yangting Zhang,Ruiyue Zheng,Xiaopei Wu,Ye Ai,Sagheer Ahmad,Zhongjian Liu,Donghui Peng

ThePEBPgene family is involved in many biological processes in plants, including plant growth and development, flowering regulation, light response, and abiotic stress response. But there is little information about the role of thePEBPgene family inCymbidiumspecies. In this study, we identified 11, 9, and 7PEBPgenes inC. ensifolium,C. sinense, andC. goeringii, respectively, and mapped them to the chromosomes. We also studied the physicochemical characteristics of the proteins encoded by thesePEBPs and analyzed their intra-species collinearity, gene structure, conserved motifs, and cis-acting elements. Furthermore, a total of forty PEBP genes fromC. sinense,C. ensifolium,C. goeringii,Phalaenopsis, andArabidopsiswere divided into three clades based on the phylogenetic tree. The expression patterns of 11PEBPgenes in different tissues and organs ofC. ensifoliumwere analyzed based on transcriptome data, indicating that theCePEBPs might play an important role in the growth and development, especially in the flower bud organs (1–5 mm).CePEBP5plays an indispensable role in both the vegetative and reproductive growth cycles ofC. ensifolium.CePEBP1is essential for root development, whileCePEBP1,CePEBP3,CePEBP5, andCePEBP10regulate the growth and development of different floral organ tissues at various stages. The findings of this study can do a great deal to understand the roles of thePEBPgene family inCymbidium.

Transcriptional Regulatory Network of GA Floral Induction Pathway in LA Hybrid Lily

Wenqi Li,Yubing Yong,Yue Zhang,Yingmin Lyu

Background: The LA hybrid lily ‘Aladdin’ has both excellent traits of Longiflorum hybrids and Asiatic hybrids—such as big and vivid flower, strong stem, high self-propagation coefficient, and shorter low temperature time required to release bulb dormancy in contrast to Oriental hybrids. A genome-wide transcriptional analysis using transcriptome RNA-Seq was performed in order to explore whether there is a gibberellin floral induction pathway in the LA hybrid lily. Subsequently, gene co-expression network analysis was used to analyze the possible interactions of key candidate genes screened from transcriptome data. At the same time, a series of physiological, biochemical, and cultivation tests were carried out. Results: The content of five endogenous hormones changed sharply in the shoot apex during the treatment of 200 mg/L exogenous gibberellin and the ratio of ABA/GA3dropped and stayed at a lower level after 4 hours’ treatment from the higher levels initially, reaching a dynamic balance. In addition, the metabolism of carbohydrates in the bulbs increase during exogenous gibberellin treatment. A total of 124,041 unigenes were obtained by RNA-seq. With the transcriptome analysis, 48,927 unigenes and 48,725 unigenes respectively aligned to the NR database and the Uniprot database. 114,138 unigenes, 25,369 unigenes, and 19,704 unigenes respectively aligned to the COG, GO, and KEGG databases. 2148 differentially expression genes (DEGs) were selected with the indicators RPKM ≥ 0, FDR ≤ 0.05 and |log2(ratio)| ≥ 2. The number of the upregulated unigenes was significantly more than the number of the downregulated unigenes. Some MADS-box genes related to flowering transformation—such asAGL20,SOC1, andCO—were found to be upregulated. A large number of gibberellin biosynthesis related genes such asGA2ox,GA3ox,GA20ox,Cytochrome P450,CYP81, and gibberellin signal transduction genes such asDELLA,GASA, andGID1were significantly differentially expressed. The plant hormones related genes such asNCED3and sugar metabolism related genes such asα-amylase,sucrose synthase hexokinase, and so on were also found expressing differentially. In addition, stress resistance related genes such asLEA1,LEA2,LEA4,serine/threonine protein kinase,LRR receptor-like serine/threonine protein kinase,P34 kinase, histidine kinase 3and epigenetic related genes in DNA methylation, histone methylation, acetylation, ubiquitination of ribose were also found. Particularly, a large number of transcription factors responsive to the exogenous gibberellin signal includingWRKY40, WRKY33, WRKY27, WRKY21, WRKY7, MYB, AP2/EREBP, bHLH, NAC1, NAC2, andNAC11were found to be specially expressing. 30 gene sequences were selected from a large number of differentially expressed candidate genes for qRT-PCR expression verification (0, 2, 4, 8, and 16 h) and compared with the transcriptome expression levels. Conclusions: 200mg/L exogenous GA3can successfully break the bulb’s dormancy of the LA hybrid lily and significantly accelerated the flowering process, indicating that gibberellin floral induction pathway is present in the LA lily ‘Aladdin’. With the GCNs analysis, two second messenger G protein-coupled receptor related genes that respond to gibberellin signals in the cell were discovered. The downstream transport proteins such asAMT,calcium transport ATPase, andplasma membrane ATPasewere also discovered participating in GA signal transduction. Transcription factors includingWRKY7,NAC2,NAC11, andCBFspecially regulated phosphorylation and glycosylation during the ubiquitination degradation process ofDELLAproteins. These transcription factors also activated in abscisic acid metabolism. A large number of transcription factors such asWRKY21,WRKY22,NAC1,AP2,EREB1,P450, andCYP81that both regulate gibberellin signaling and low-temperature signals have also been found. Finally, the molecular mechanism of GA floral induction pathway in the LA hybrid lily ‘Aladdin’ was constructed. Keywords:GA floral induction pathway;LA hybrid lily;transcriptome RNA-Seq;GCN;qRT-PCR
