Polysaccharide from Agrocybe cylindracea has been demonstrated to exhibit excellently biological functions, but insufficient understanding of its physicochemical property and fine structure limits its wide application and development. In the present study, two water-soluble heteroglycans, a high molecular weight (SACP-40) and a low molecular weight (SACP-80), were isolated from the fruit bodies of A. cylindracea by hot water extraction and gradient ethanol precipitation. There was little difference in their physicochemical properties, including major chemicals and monosaccharide compositions. Rheological measurements revealed SACP-80 was a typical pseudoplastic fluid with shear thinning behavior at the concentration of 3.0%, whereas exhibited a near-Newtonian fluid at lower concentration. SEM observation suggested SACP-80 showed micro-spherical shapes loosely attached to fibrous filament in its surface. Besides, methylation and 1D/2D NMR analysis indicated that SACP-80 was a branched heteroglycan with a backbone composed of β-(1 → 6)-glucopyranosyl, α-(1 → 6)-galactopyranosyl and α-(1 → 2,6)-glucopyranosyl residue at a ratio of 3:1:1. This work provides a theoretical basis for gaining insight into the relationship between the fine structure and viscosity of polysaccharide from A. cylindracea , which could further guide its functional studies and advance the potential application in food and biomedicine industries.

A novel heteroglycan TLH-G was successfully isolated from Tricholoma lobayense cultivated in south China. TLH-G was composed of five monosaccharides, namely, mannose, rhamnose, glucuronic acid, glucose and galactose, and consisted of →1)-β-D-Glcp-(6→ and →1)-α-D-Galp-(3→ motifs. TLH-G had excellent scavenging activities towards 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals. Structure-activity relationship analysis demonstrated that glycosidic bond types, monosaccharide composition, and branching degree were more important for the antioxidant activity of polysaccharides from Tricholoma lobayense than other structural factors. All the above results showed that the growth environment played a crucial role in the structure and antioxidant activity of polysaccharides from Tricholoma lobayense.

A fundamental study was undertaken to determine the general role of heteropolysaccharides during the production of TEMPO-oxidized cellulose nanofibrils (TOCNs). Four major fiber resources, viz. , fully bleached kraft pulps of softwood and hardwood varieties (pine, eucalyptus) and non-woods (bamboo, bagasse) were used because of their substantial morphological differences and relative abundance. The effect of heteropolysaccharides during TEMPO-mediated oxidation and high-pressure homogenization for TOCNs production was investigated under constant conditions. Most galactoglucomannans were removed during oxidation, whereas the majority of xylans were retained. The galactoglucomannans, however, non-beneficially consumed NaClO, the terminal TEMPO oxidant, while xylans adversely affected carboxylate group formation by limiting chemical accessibility to cellulose. However, lower xylans content led to more transparent and processable suspensions, while during mechanical processing, heteropolysaccharides supported nanofibrillation. The average length of the final TOCNs from eucalyptus, bamboo, bagasse, and pine were 290, 350, 360 and 370 nm, respectively, with average widths of ∼4 nm.

Heteropolysaccharides, the principal bioactive constituents of the esteemed medicinal foodTremella aurantialba, remain poorly understood in both structure and function. Herein, we describe a novel heteropolysaccharide, designated TAP-2a, isolated from the fruiting bodies ofT. aurantialbavia multi-step column chromatography. With a molecular weight of 16.95 kDa, TAP-2a is dominated by the pyranose forms of ᴅ-galactose (ᴅ-Galp), ᴅ-glucose (ᴅ-Glcp) and ᴅ-mannose (ᴅ-Manp), accompanied by minor proportions of ᴅ-xylose (ᴅ-Xylp), ʟ-fucose (ʟ-Fucp) and glucuronic acid. Methylation-GC-MS and exhaustive 1D/2D NMR analyses revealed a backbone assembled from →6)-α-Galp-(1→, →6)-β-Glcp-(1→, and →3)-α-Manp-(1→residues, branched at →2,6)-β-Galp-(1→, →3,6)-α-Galp-(1→, and →2,3)-α-Manp-(1→residues, and terminated by β-Glcp-(1→, α-Fucp-(1→, and β-Xylp-(1→. This intricate glycosidic architecture generates an exceptionally complex mannoglucogalactan in which a Gal→Man domain is substituted atO-3 of Gal by t-β-Glcpside chains and atO-2 of Man by t-α-Fucpstubs; additionally, a discrete fragment comprising t-β-Glcp-(1→3)-β-Glcp-(1→ was identified, along with a minor branch in which t-β-Xylpis attached toO-2 of a mannose residue. Functionally, TAP-2a proved to be a potent immunomodulator, markedly enhancing the secretion of nitric oxide, interleukin-1β, interleukin-6 and tumour necrosis factor-α while concurrently up-regulating the corresponding mRNA transcripts and augmenting phagocytic capacity. These findings establish the highly elaborate heteropolysaccharides ofT. aurantialbaas powerful immunomodulators that underpin the fungus’s renowned medicinal efficacy.
