High-Performance T1–T2 Dual-Modal MRI Contrast Agents through Interface Engineering

Dan Zhao,Shibo Peng,Hanzhang Xiao,Qilong Li,Yahong Chai,Hongxia Sun,Ruping Liu,Li Yao,Lin Ma

Iron oxide nanoparticles (IONPs) have been developed as contrast agents for T1- or T2-weighted magnetic resonance imaging (MRI) on account of their excellent physicochemical and biological properties. However, general strategies to improve longitudinal relaxivity (r1) often decrease transverse relaxivity (r2), thus synchronously strengthening the T1 and T2 enhancement effect of IONPs remains a challenge. Here, we report interface regulation and size tailoring of a group of FePt@Fe3O4 core–shell nanoparticles (NPs), which possess high r1 and r2 relaxivities. The increase of r1 and r2 is due to the enhancement of the saturation magnetization (Ms), which is a result of the strengthened exchange coupling across the core–shell interface. In vivo subcutaneous tumor study and brain glioma imaging revealed that FePt@Fe3O4 NPs can serve as a favorable T1–T2 dual-modal contrast agent. We envision that the core–shell NPs, through interface engineering, have great potential in preclinical and clinical MRI applications.

Changes in the relaxivity of water-organic solvent miscible systems: Hidden forces beyond metal ions

Shiman Wu,Chaochao Wang,Junjie Xia,Dijia Sun,Pu-Yeh Wu,Yue Wu,Zhenwei Yao,Yan Ren,Qi Zhang

Background The relaxivity of magnetic resonance imaging (MRI) contrast agents is primarily attributed to metal ions such as gadolinium (Gd) and iron. However, the impact of organic solutes on relaxivity, particularly through alterations in water molecule dynamics, has not been thoroughly investigated. This research was aimed to explore how organic solutes affect the relaxivities of water and Gd-based contrast agents (GBCAs), potentially revealing new aspects for the development of contrast agents. Purpose To investigate the effects of different proportions of water-soluble organic solvents mixed with pure water and GBCA on T 1 and T 2 relaxivities. Methods Ethanol, dimethyl sulfoxide (DMSO), 1,4-dioxane, glycerin, ethylene glycol, diethylene glycol, polyethylene glycol (PEG) 200, and PEG 400 were mixed with ultrapure water in various ratios (100:0, 90:10, 80:20, 70:30, 60:40, 50:50, 40:60, 30:70, 20:80, 10:90, 0:100). GBCA was added to these mixtures at a concentration of 0.05 mmol/L. The mixtures underwent T 1 and T 2 mapping scans using a 3.0 Tesla MRI machine, and the relaxivities R 1 (1/T 1 ) and R 2 (1/T 2 ) were calculated and compared. Results The relaxivity R 1 of ethanol, 1,4-dioxane, and DMSO mixtures with water or GBCA, as well as R 2 of DMSO mixtures with GBCA, initially increased and then decreased. Conversely, R 1 and R 2 increased significantly with higher proportions of diethylene glycol, PEG 200, and glycerin in the mixtures with GBCA. The increase in relaxivity R 1 was correlated with greater viscosity. When the proportion of DMSO and diethylene glycol exceeded 20%, minimal variability in R 2 was observed as the water content decreased. Conclusions Adding organic solvents to water and paramagnetic relaxation reagents could alter T 1 and T 2 relaxivities, suggesting potential new directions for modifying current contrast agents. Additionally, increasing the viscosity of the contrast agent was found to enhance the relaxivity.

Engineered Graphene Quantum Dots as a Magnetic Resonance Signal Amplifier for Biomedical Imaging

Zhongtao Li,Guiqiang Qi,Guangyue Shi,Meng Zhang,Haifeng Hu,Liguo Hao

The application of magnetic resonance imaging (MRI) nano-contrast agents (nano-CAs) has increasingly attracted scholarly interest owing to their size, surface chemistry, and stability. Herein, a novel T1 nano-CA (Gd(DTPA)−GQDs) was successfully prepared through the functionalization of graphene quantum dots with poly(ethylene glycol) bis(amine) and their subsequent incorporation into Gd-DTPA. Remarkably, the resultant as-prepared nano-CA displayed an exceptionally high longitudinal proton relaxivity (r1) of 10.90 mM−1s−1(R2= 0.998), which was significantly higher than that of commercial Gd-DTPA (4.18 mM−1s−1, R2= 0.996). The cytotoxicity studies indicated that the Gd(DTPA)−GQDs were not cytotoxic by themselves. The results of the hemolysis assay and the in vivo safety evaluation demonstrate the outstanding biocompatibility of Gd(DTPA)−GQDs. The in vivo MRI study provides evidence that Gd(DTPA)−GQDs exhibit exceptional performance as T1-CAs. This research constitutes a viable approach for the development of multiple potential nano-CAs with high-performance MR imaging capabilities. Keywords:graphene quantum dots;magnetic resonance imaging;gadolinium;enhanced relaxivity

Topology-Dependent T2 Relaxivity in Fe3O Cluster-Based MOFs for Enhanced Tumor Monitoring via MRI

Qiao Wang,Yimin Gong,Jianing Li,Xin Zeng,Dan Luo,Yun Ling,Yaming Zhou,Zhenxia Chen

Metal-organic frameworks (MOFs) are crystalline porous materials with tunable structures, where metal ions or clusters serves as magnetic centers and organic ligands offer spatial separation. These characteristics, combined with their diverse topologies, make MOFs promising candidates for contrast agents (CAs) in magnetic resonance imaging (MRI). Herein we synthesized four MOFs based on the same triangular Fe3O clusters with different topologies: MIL-101(Fe) (mtn net), MIL-100(Fe) (moo net), MIL-59(Fe) (pcu net), and MIL-88B(Fe) (acs net). To clarify the relationship between topologies and T2 relaxivities, the MOFs were tailored into uniform, nanoscale spherical morphologies. Notably, the value of T2 relaxivity for MIL-88B(Fe) with acs topology is nearly three times of that for MIL-101(Fe) with mtn topology at 7.0 T. By comparing with the magnetic properties of Fe3O molecular clusters and Ga-doped MIL-88B(Fe), our analysis demonstrated the significant advantage of MOFs with fixed arrays, adjustable component and diverse topologies in enhancing magnetic relaxation. Cellular MRI experiments further revealed that MIL-88B(Fe) could differentiate between M1 and M2 macrophages, highlighting its potential for monitoring tumor progression. These findings offer valuable insights into how MOF topology can be strategically utilized to enhance T2 relaxivities for MRI applications.

An ultra-sensitive T2-weighted MR contrast agent based on Gd3+ ion chelated Fe3O4 nanoparticles

Jing Chen,Hui-Hui Xiang,Zu-Zhi Zhao,Yun-Kai Wu,Meng-Yu Fei,Meng-Meng Song

An ultra-sensitive T2-weighted MR imaging contrast agent was prepared based on Fe3O4 nanoparticles and Gd3+ ions (Fe3O4@Gd). Amino modified Fe3O4 nanoparticles were conjugated to diethylenetriamine pentaacetic acid, and finally coordinated with Gd3+ ions. The nanoparticles had a uniform morphology with a size of 100 nm and a Gd/Fe mass ratio of 1/110. The r2 (transverse relaxivity) of the Fe3O4 nanoparticles increased from 131.89 mM−1 s−1 to 202.06 mM−1 s−1 after coordination with Gd3+ ions. MR measurements showed that the aqueous dispersion of Fe3O4@Gd nanoparticles had an obvious concentration-dependent negative contrast enhancement. Hepatoma cells were selected to test the cytotoxicity and MR imaging effect. The application of Fe3O4@Gd nanoparticles as contrast agents was also exploited in vivo for T2-weighted MR imaging of rat livers. All the results showed the effectiveness of the nanoparticles in MR diagnosis.

Facile synthesis of Silicon quantum dot-Gadolinium: A potential fluorescent/T1-T2 multimodal imaging agent

Fangnan Xiao,Yue Xiao,Fangman Chen,Xiaolin Liu,Chentao Lin,Jianxin Chen,Yunkun Wu

Highly stable and multifunctional fluorescent quantum dots are particularly attractive in practical applications. Here, a new kind of ultra-small-sized silicon quantum dot-gadolinium (SiQD-Gd) was successfully fabricated by a newly-designed facile hydrothermal growth and chelating method. The obtained SiQD-Gd exhibited outstanding water dispersibility, stability and good fluorescent property with the quantum yield of 11.6%. SiQD-Gd displayed a low cytotoxicity in normal cell lines (HELF, HEK293F) and tumor cell lines (H1299, A549). Meanwhile, SiQD-Gd showed excellent magnetic resonance response with r 1 relaxation rate of 10.5 mmol L −1 ·s −1 and r 2 relaxation rate of 47.5 mmol L −1 ·s −1 , which are 2.5 and 7.4 times enhanced comparing to that of the commercial MR agent Magnevist. In vivo studies showed significant contrast enhancement effect of its T 1 - and T 2 -weighted MR imaging. In addition, in vivo fluorescent imaging for mice and zebrafish indicated its potential applications in fluorescent tracking. Thus, the excellent multimodal imaging capacity and biocompatibility of SiQD-Gd make it a potential imaging agent for clinic applications.

Binuclear gadolinium(III) complex based on DTPA and 1,3-bis(4-aminophenyl)adamantane as a high-relaxivity MRI contrast agent

Fuxian Wan,Lipeng Wang,Wenxin Xu,Changcheng Li,Ying Li,Chunbao Zhang,Lin Jiang

A novel dimeric gadolinium(III) complex, bis(4-Gd-DTPA-aminophenyl)adamantane ( 5 ), was successfully synthesized using bis(4-aminophenyl)adamantane to link two Gd-DTPA units. All the intermediates and bis(4-Gd-DTPA-aminophenyl)adamantane ( 5 ) have been characterized by 1 H NMR, 13 C NMR, ES-API-MS, HR-ESI-MS, FT-IR and elemental analysis (EA). The longitudinal relaxivity of bis(4-Gd-DTPA-aminophenyl)adamantane is 8.43 mM −1  Gd −1  s −1 at 0.43 T (32 °C, pH 7.0), and 4.89 mM −1  Gd −1  s −1 at 3.0 T (32 °C, pH 7.4). The T 1 -weighted imaging in vitro showed that bis(4-Gd-DTPA-acetylaminophenyl)adamantane can enhance the contrast of images distinctly. But most of all, the kinetic inertness of bis(4-Gd-DTPA-aminophenyl)adamantane ( 5 ) was comparable to that of [Gd(DTPA)(H 2 O)] 2− (Magnetvist®). Moreover, cytotoxicity tests indicated that the bis(4-Gd-DTPA-acetylaminophenyl)adamantane exhibited low toxicity to healthy liver cells (L-02 cells). These results demonstrate that the Gd(III) complex 5 may be a promising MRI contrast agent candidate.

An MRI contrast agent based on a zwitterionic metal-chelating polymer for hepatorenal angiography and tumor imaging

Peisen Zhang,Zihua Wang,Yiyang Wang,Yuqing Wang,Chunyan Liu,Kai Cao,Yijie Lu,Leila Behboodpour,Yi Hou,Mingyuan Gao

MRI contrast agents such as paramagnetic Gd(III)-chelates, can improve the ability of MRI in differentiating diseased and healthy tissues, and have been widely used in clinical diagnosis. However, the enhancement effect of small molecular MRI contrast agents is unsatisfied due to their relative high rotation rates. Furthermore, the small molecular contrast agents also suffer from the short blood half-life and nonspecific extracellular diffusion in tissues, which also restricts their applications. To address these issues, we developed a macromolecular MRI contrast agent based on a zwitterionic metal-chelating polymer. Poly(acrylic acid) (PAA) was chosen as the main chain, and diethylenetriamine pentaacetic acid (DTPA) as the metal-chelating group was coupled through the carboxyl groups of PAA using diethylenetriamine (DET) as a linker. The macromolecular MRI contrast agent constructed by chelating with Gd3+ (Gd-PAA) exhibited a much higher longitudinal relaxation rate (r1) than the clinical contrast agent Gd-DTPA. Importantly, due to the stealth ability of the zwitterionic structure, Gd-PAA can reside in the blood long enough without any microvascular leakage in the extracellular space of normal tissues, which allows it to be used for precise blood MR imaging, such as hepatorenal angiography, but also for tumor imaging because of the enhanced permeability and retention (EPR) effecta. Besides, the result of long-term toxicity tests highlights the safety feature of the current contrast agent. Hence, the current contrast agent overcomes the defect of traditional small molecular Gd(III)-based T1-weighted contrast agents and shows great prospects for future clinical applications.

Synthesis of Metal–Organic Framework Nanosheets with High Relaxation Rate and Singlet Oxygen Yield

Yuewu Zhao,Ye Kuang,Min Liu,Jine Wang,Renjun Pei

A novel gadolinium–porphyrin metal–organic framework (MOF) nanosheet was synthesized by coordination chelation between gadolinium(III) ions and 4,4,4,4-(porphine-5,10,15,20-tetrayl) tetrakis (benzoic acid) (TCPP). The mechanism and molecular structure of the prepared Gd-TCPP products were analyzed through the comparison of 4,4,4,4-(porphine-5,10,15,20-tetrayl) tetrakis (benzenesulfonic acid) (TPPS) and TCPP. The Gd-TCPP MOF nanosheets exhibit a high relaxation rate of 40.8 mM–1 s–1, which is better than the rates for most reported contrast agents. In addition, the photosensitive activity of the Gd-TCPP nanosheets was enhanced compared to TCPP owing to the special periodic porous structure of the MOF, indicating that the Gd-TCPP nanosheets could produce the singlet oxygen (1O2) more effectively under irradiation by visible light. Performance enhancement was observed for both metal ions and organic ligands in the Gd-TCPP MOF nanosheets. This study presents a new concept in the development bifunctional MOF nanomaterials with enhanced performance in magnetic resonance imaging and photodynamic therapy.

Hybrid Dextran-gadolinium Nano-suitcases as High-relaxivity MRI Contrast Agents

Wang Hao,Dai Ting-Ting,Lu Bo-Lun,Li Sheng-Li,Lu Qing,Mukwaya Vincent,Dou Hong-Jing

Dextran-poly(glycidyl methacrylate) (Dex-PGMA) nano-suitcases were synthesized efficiently via a graft copolymerization induced self-assembly (GISA) approach. On this basis, the Dex-PGMA nano-suitcases were modified with hydrazide, and the attachment of multiple chelated Gd(III) ions to the interior of the nano-suitcases affords nanoscale MRI contrast agents with high relaxivity values. The highly fenestrated dextran shell of the nano-suitcases assures water exchange which readily occurs between the surrounding environment and the Gd(III) ions encapsulated within the hybrid nano-suitcases. The complexation between the hydrophilic hydrazide interior of the nano-suitcases and Gd(III) ions results in an impressive Gd payload at 22.6 wt% in the hybrid nano-suitcases. The longitudinal relaxivity (r1) of the hybrid nano-suitcases is reported as 44.4 L/(mmol·s), which is 9–14 folds of that of commercial Gd-DTPA agents. In vivo MRI studies demonstrate that the hybrid nano-suitcases accumulated in the lymph node of the rat due to their nanoscale dimensions and displayed strong signals in vivo . The results indicated that the hybrid nano-suitcases provide a promising platform for the diagnosis of lymph node related diseases.

Zeolitic imidazolate framework nanoparticles loaded with gadolinium chelate as efficient T1 MRI contrast agent

Zhou Weixiu,Shen Jiyun,Lin Jiaomin,An Miao,An Lu,Tian Qiwei,Yang Shiping

Gd–chelate magnetic resonance imaging (MRI) contrast agents used in conventional clinical medicine still have some shortcomings, such as their low relaxation rates and poor imaging sensitivity. In this work, a Gd–chelate (Gd–DTPA), with a comparison made to the commercial contrast agent Magnevist was loaded into a zeolitic imidazolate framework (ZIF-8) to construct the efficient T 1 MRI contrast agent Gd–DTPA@ZIF-8. At 0.5 T, the longitudinal relaxivity value ( r 1 ) of Gd–DTPA@ZIF-8 reaches 29.60 mM −1  s −1 , which is about 6 times higher than that of Gd–DTPA. In terms of the retention of Gd–DTPA in ZIF-8, no obvious Gd 3+ ions are released in water and complete culture solution over a long time period. Haemolysis and cytotoxicity experiments revealed that Gd–DTPA@ZIF-8 has good biocompatibility. In addition, the in vivo MRI results prove that Gd–DTPA@ZIF-8 can significantly enhance the T 1 -weighted MRI contrast effect of the tumour site. These advantages make Gd–DTPA@ZIF-8 a potential candidate for biomedical applications.

Copper Sulfide-Decorated Gd2O3 Nanorods: A Promising Probe for Magnetic Resonance Imaging and Photothermal Therapy

Dan Wang,Yunhong Xie,Lingfan Xu,Yixuan Zhou,Kai Li,Zhongfeng Li

The development of bifunctional nanoplatform-based theranostics has garnered considerable attention in cancer therapy research. In this study, we engineer Gd2O3@SiO2-CuS nanorods as an integrated diagnostic-therapeutic system, simultaneously harnessing photothermal conversion and paramagnetic properties for MRI-assisted photothermal intervention. Hydrothermally synthesized Gd2O3 nanorods demonstrate superior T1-weighted contrast performance, achieving a longitudinal relaxivity (r1) of 4.2 mM–1·s–1, which compares favorably with that of commercial contrast agents. Furthermore, the strategically integrated CuS component exhibits remarkable photothermal conversion efficiency (19.6%) and sustained therapeutic stability. These attributes pave the way for achieving the application objective of magnetic resonance imaging-assisted photothermal therapy. Critically, comprehensive biosafety evaluations, including hemocompatibility assays and cytotoxicity profiling, confirm the nanocomposite’s favorable biocompatibility. The findings underscore the substantial potential of Gd2O3@SiO2-CuS nanorods for image-guided photothermal therapy applications.
