Direct Writing of Shape-Gradient Magnetic Alloy Microwire Arrays with Meniscus-Confined Electrodeposition Process

Weiye Nie,Yu Lei,Yifan Zhang,Qingqing Gao,Jiangang Chen,Xianyun Zhang,Lifeng Yuan,Yuchuan Cheng,Aihua Sun,Gang Wang,Gaojie Xu,Jianjun Guo

Gradient magnetic micro/nanowire arrays have attracted widespread attention due to their interesting properties. However, fabricating such an ordered array of gradient micro/nanowires with controllable diameters and compositions is still a great challenge to most of the current methods. Here, meniscus-confined electrodeposition (MCED) technique is adopted for the rapid prototyping of the shape-gradient magnetic Cu/Co-alloy microwire arrays by adjusting the printing speed continuously, which provides a novel idea for the preparation and performance research of shape-gradient magnetic alloy microwire arrays with well-defined structures. It is found that the key to fabricating controllable gradient alloy micro/nano structures by increasing the printing speed is to continuously stretch the meniscus within the stable range of the meniscus. In the range of incremental speed in this study, the shape-gradient magnetic alloy wires with stable and uniform compositions and dense internal structures can be successfully prepared, and the gradient ratio can be adjusted from 0 to about 0.025. Compared with the uniform-diameter array, the shape-gradient magnetic alloy array shows an improvement in remanence and coercive force.

Functionalized Microwires-Based Bundle Electrodes for Detection of Multiplexed Metabolites in Interstitial Space

Shuang Huang,Chuanjie Yao,Mengyi He,Xinshuo Huang,Zhengjie Liu,Jiayi Chen,Lelun Jiang,Hui-jiuan Chen,Xi Xie

Metabolic substances play a pivotal role in maintaining the body's regular physiological functions. When these processes are disrupted, it can lead to metabolic disorders which may cause severe damage to various organs. Diabetes mellitus, a prevalent metabolic disorder, arises from disturbances in sugar metabolism among other substances. Consequently, there's a pressing need to monitor metabolite levels for early diagnosis. To address this, in this study, a semi-implantable metabolite sensing system developed around functionalized microwires-based bundle electrodes (FMBE) is introduced. This FMBE device tracks glucose, hydrogen peroxide (H 2 O 2 ), and uric acid (UA) levels in vivo in real time, sensitively, and continuously. The FMBE is coated with Au nanoclusters and carbon nanotube–2D carbides and nitride (MXene) nanocomposites to enhance sensing surface area. In vitro characterizations affirm the FMBE's linear responsivity, detection sensitivity, and selectivity toward glucose, H 2 O 2 , and UA sensing. In addition, in in vivo testing in healthy and diabetic rats, it is demonstrated that FMBE is able to continuously monitor interstitial glucose, H 2 O 2 , and UA concentrations after implantation. The FMBE system, thus, stands out as a promising platform for real-time, in situ monitoring of metabolite concentrations, potentially assisting in the diagnosis of diabetes and associated complications.

Talin promotes integrin activation accompanied by generation of tension in talin and an increase in osmotic pressure in neurite outgrowth

Yifan Wang,Xiaolong Zhang,Jilai Tian,Jinjun Shan,Yunfeng Hu,Yiqian Zhai,Jun Guo

Neuronal polarization depends on the interaction of intracellular chemical and mechanical activities in which the cytoplasmic protein, talin, plays a pivotal role during neurite growth. To better understand the mechanism underlying talin function in neuronal polarization, we overexpressed several truncated forms of talin and found that the presence of the rod domain within the overexpressed talin is required for its positive effect on neurite elongation because the neurite number only increased when the talin head region was overexpressed. The tension in the talin rod was recognized using a Förster resonance energy transfer-based tension probe. Nerve growth factor treatment resulted in inward tension of talin elicited by microfilament force and outward osmotic pressure. By contrast, the glial scar-inhibitor aggrecan weakened these forces, suggesting that interactions between inward pull forces in the talin rod and outward osmotic pressure participate in neuronal polarization. Integrin activation is also involved in up-regulation of talin tension and osmotic pressure. Aggrecan stimuli resulted in up-regulation of docking protein 1 (DOK1), leading to the down-regulation of integrin activity and attenuation of the intracellular mechanical force. Our study suggests interactions between the intracellular inward tension in talin and the outward osmotic pressure as the effective channel for promoting neurite outgrowth, which can be up-regulated by integrin activation and down-regulated by DOK1.—Wang, Y., Zhang, X., Tian, J., Shan, J., Hu, Y., Zhai, Y., Guo, J. Highly Crystallized Tin Dioxide Microwires toward Ultraviolet Photodetector and Humidity Sensor with High Performances

Hui Liu,Chaolei Zuo,Ziliang Li,Xinya Liu,Xiaosheng Fang

The rapid development of artificial intelligent and internet of things calls for high-performance multifunctional devices for synchronous detection of a wide variety of environmental signals, such as gas, light, and humidity. Herein, highly crystallized tin dioxide (SnO 2 ) microwires (MWs) with low density of point defects are synthesized by the chemical vapor deposition method and constructed into a multifunctional device for photo and humidity sensing. The device shows excellent photoelectric performances, for example, ultralow dark current of ≈10 −13 A, ultrahigh on–off ratio of >10 7 , UV/visible rejection ratio of R 300 nm / R 400 nm > 10 7 , specific detectivity ( D *) of 1.16 × 10 15 Jones, linear dynamic range (LDR) of 152 dB, high responsivity of 18 A W −1 , and fast photoresponse speed of t rise / t decay = 2.7 µs/2.5 ms at 5 V bias. Furthermore, the p-CuI/n-SnO 2 heterojunction shows outstanding self-powered properties, such as responsivity of 8.98 mA W −1 , specific detectivity of 1.98 × 10 12 Jones and LDR of 106 dB at 0 V bias. Additionally, the SnO 2 MWs also show high sensitivity to ambient humidity changes. Therefore, the SnO 2 MWs show high potential for multifunctional applications, such as UV photodetector and humidity sensor.

CK-666 protects against ferroptosis and renal ischemia-reperfusion injury through a microfilament-independent mechanism

Qian Hu,Yanan Zhao,Wan-yang Sun,Zexian Ou,Wentao Duan,Zeyu Qiu,Yuanlong Ge,Daolin Tang,Tianfeng Chen,Xiang Cheng,Rong-rong He,Shu Wu,Zhenyu Ju

Ferroptosis is a type of regulated cell death driven by iron-dependent accumulation of lipid peroxidation, exhibiting unique morphological changes. While actin microfilaments are crucial for various cellular processes, including morphogenesis, motility, endocytosis, and cell death, their role in ferroptosis remains unclear. Here, our study reveals that actin microfilaments undergo remodeling and disassembly during ferroptosis. Interestingly, inhibitors that target actin microfilament remodeling do not affect cell sensitivity to ferroptosis, with the exception of CK-666 and its structural analogue CK-636. Mechanistically, CK-666 attenuates ferroptosis independently of its canonical function in inhibiting the Arp2/3 complex. Further investigation revealed that CK-666 modulates the ferroptotic transcriptome, prevents lipid degradation, and diminishes lipid peroxidation. In addition, CK-666 does not impact the labile iron pool within cells, nor does the inhibition of FSP1 impact its anti-ferroptosis activity. Notably, the results of DPPH assay and liposome leakage assay suggest that CK-666 mitigates ferroptosis by directly eliminating lipid peroxidation. Importantly, CK-666 significantly ameliorated renal ischemia-reperfusion injury and ferroptosis in renal tissue, underscoring its potential therapeutic impact.

Vector Analysis of Cytoskeletal Structural Tension and the Mechanisms that Underpin Spectrin-Related Forces in Pyroptosis

Tingting Chen,Yichen Guo,Jinjun Shan,Jiarui Zhang,Xu Shen,Jun Guo,Xiaoguang Margaret Liu

Aims: Pyroptotic cells are characterized by plasma swelling, membrane blebbing, and disintegration of the cell membrane mediated by spectrin-based membrane skeleton and intercellular competitive tension activities. The spectrin-based membrane skeleton is involved in membrane organization through the regulation of intercellular tension. Using genetically encoded tension sensors to attain noninvasive force measurements in structural proteins, we investigated how cytoskeletal structural tension influences changes in plasma morphology during pyroptosis and the regulatory mechanism of cytoskeletal structural tension that underpins pyroptosis. Results: The results indicate that increasing spectrin tension is caused by osmotic swelling. Hightened tension of spectrin was closely associated with the shrink tension transmitted synergistically by microfilaments (MFs) and microtubules (MTs). However, the increment of spectrin tension in pyroptotic cells was controlled antagonistically by MF and MT forces. Different from MF tension, outward MT forces participated in the formation of membrane blebs. Spectrin tension caused by inward MF forces resisted pyroptosis swelling. Stabilization of MF and MT structure had little influence on intracellular tension and pyroptosis deformation. Pyroptosis-induced cytoskeletal structural tension was highly dependent on calcium signaling and reactive oxygen species generation. Blocking of membrane pores, nonselective ion flux, or elimination of caspase-1 cleavage resulted in the remission of structural forces associated with pyroptosis failure.Innovation and Conclusions: The data suggest that subcellular tension, in terms of magnitude and vector, is integral to pyroptosis through the mediation of swelling and blebbing and the elimination of structural tension, especially MT forces, may result in pyroptosis inhibition.

Inward Tension of Talin and Integrin-related Osmotic Pressure are involved Synergetically in the Invasion and Metastasis of Non-small Cell Lung Cancer

Ying Song,Chen Li,Yahan Fu,Qiu Xie,Jun Guo,Guangming Li,Huiwen Wu

The integrin receptor protein talin plays vital roles in intracellular chemical and mechanical activities, and it is implicated in the high invasion and poor prognosis of non-small cell lung cancer (NSCLC). To better understand the mechanism underlying the function of talin in NSCLC invasion and metastasis, a few newly designed tension probe based on Förster resonance energy transfer was used for real-time observation of tension changes in A549 cells. High NSCLC cell aggressiveness was found to be accompanied with inward talin and outward glial fibrillary acidic protein (GFAP) tensions, which are closely associated with microfilament (MF) force and intracellular osmotic potential. The increased osmotic pressure resulted from the production of intracellular protein nanoparticles and the related ion influx. Furthermore, integrin activation was found to adjust the talin and GFAP tensions. Disruption of the interaction between talin and MFs blocked the mechanical source of talin, reducing both talin tension and osmotic pressure and thus inhibiting NSCLC cell invasion and migration. Consequently, our study demonstrates that talin is involved in NSCLC invasion and migration via its inward tension and that the integrin pathway is correlated closely with protein-nanoparticle-induced outward osmotic pressure.

The mechanical mechanism of angiotensin II induced activation of hepatic stellate cells promoting portal hypertension

Yiheng Zhang,Mulan Xing,Fansheng Meng,Ling Zhu,Qingchuan Huang,Tianle Ma,Huihua Fang,Xujing Gu,Suzhou Huang,Xinyu Wu,Gaohong Lv,Jun Guo,Li Wu,Xin Liu,Zhipeng Chen

In the development of chronic liver disease, the hepatic stellate cell (HSC) plays a pivotal role in increasing intrahepatic vascular resistance (IHVR) and inducing portal hypertension (PH) in cirrhosis. Our research demonstrated that HSC contraction, prompted by angiotensin II (Ang II), significantly contributed to the elevation of type I collagen (COL1A1) expression. This increase was intimately associated with enhanced cell tension and YAP nuclear translocation, mediated through α-smooth muscle actin (α-SMA) expression, microfilaments (MF) polymerization, and stress fibers (SF) assembly. Further investigation revealed that the Rho/ROCK signaling pathway regulated MF polymerization and SF assembly by facilitating the phosphorylation of cofilin and MLC, while Ca 2+ chiefly governed SF assembly via MLC. Inhibiting α-SMA-MF-SF assembly changed Ang II-induced cell contraction, YAP nuclear translocation, and COL1A1 expression, findings corroborated in cirrhotic mice models. Overall, our study offers insights into mitigating IHVR and PH through cell mechanics, heralding potential breakthroughs.

In situ-formed micro silk fibroin composite sutures for pain management and anti-infection

Xiaoxiao Li,Ying Luo,Fengbo Yang,Guoping Chu,Lingqiao Li,Ling Diao,Xiaoli Jia,Chunjing Yu,Xiaozhuo Wu,Wen Zhong,Malcolm Xing,Guozhong Lyu

Surgical sutures close wounds and promote healing. Sutures need to meet appropriate mechanical properties and biocompatibility, so complex and high-cost braiding processes have been developed for better mechanical properties. However, they still cannot work ideally in some scenarios, such as pain and infection. Pain is the most common symptom in almost all injuries, infections, and diseases. Inadequate management of pain leads to untoward physical and psychological outcomes in patients. For these purposes, we designed a multifunctional composite suture of micro silk fibroin fiber (SF). It was extracted in situ ultra-long and ultra-fine SFs where a single thread can reach hundreds of meters from a cocoon. The suture can adsorb anti-infection vancomycin by turning pH and then coated with methacrylated gelatin as a sheath to load a pain-killer ibuprofen (SMG-VC-IB). SMG-VC-IB can withstand a tension force of nearly 18 N, two times the Mersilk® suture. SMG-VC-IB can maintain prolonged release for efficient pain management and anti-infection. We applied PET/CT to investigate objective metabolic data in brain regions and found that the pain-killer reached efficacy in our built pain model of quadriceps femoris incision in rats. We found SMG-VC-IB reduced metabolic levels in pain-related brain regions, including the classical pain matrix. It was also verified that the pain-killer ibuprofen from sutures helps to pass through the postoperative inflammatory period to a comfortable recovery period. Our research presented an eco-friendly manufacturing solution for designing multifunctional composite sutures for pain management and infection control.
