The effects of exogenous amino acids on production of microcystin variants in Microcystis aeruginosa

Yaxin Guo,Xiating Zhao,Zongcheng Yao,Zongyao Qian,Yuting Wang,Qiming Xian

Dissolved free amino acids are a significant component of dissolved organic nitrogen (DON) in natural waterbodies. The effects of four amino acids (glutamic acid, phenylalanine, leucine, and arginine) on the growth and microcystins (MCs) production of  Microcystis aeruginosa  were studied in batch culture. The profiles of five MCs variants and the expression levels of target genes involved in MCs biosynthesis and nitrogen metabolism were measured. When amino acids were used as the sole nitrogen source instead of nitrate at different levels (0.5, 2.0 and 8.0 mg/L based on N) in BG-11 medium, algal cell growth and intracellular MCs quotas were inhibited slightly by the treatments with glutamic acid and arginine. The treatments with phenylalanine and leucine, on the other hand, had a strong inhibitory effect on algal cell growth and MCs production. Moreover, the concentrations of Chlorophyll a , phycocyanin and allophycocyanin in cells cultured in glutamic acid, leucine and phenylalanine were lower than those in the control group with nitrate as nitrogen source. The existence of leucine or phenylalanine can lead to a significant increase in the relative abundance of MCs variants structured with the corresponding amino acids. The expression of microcystin-producing gene mcyD was downregulated while the gene pipX associated with nitrogen metabolism was upregulated during the cultivation of M. aeruginosa with amino acids as sole nitrogen source. M. aeruginosa undergoes significant alterations due to exogenous amino acids and exhibits advanced strategies for MCs production.

Impact of nanoplastics on the algicide efficacy of CuSO4 against Microcystis aeruginosa and the release of microcystin: A physiological and metabolomic analysis

Xiayida Mamatimin,Wei Song,Mihebai Yilimulati,Tiandou Zhang,Nuzahat Habibul

CuSO 4 is frequently employed as a control measure for the emergency treatment of cyanobacterial harmful algal blooms. Concurrently, nano-plastics (NPs) have emerged as pervasive pollutants exacerbating the threat of eutrophication to aquatic ecosystem. However, there is little known regarding the influence of NPs on the efficacy of copper sulfate (CuSO 4 ) in controlling cyanobacterial harmful algal blooms (cyanoHABs). Here, we demonstrate that the co-existence of polystyrene nanoplastics (PS NPs) exacerbates the negative effects of CuSO 4 to photosynthetic activity of Microcystis aeruginosa . This is caused via the differences in disrupting light energy absorption, inactivation of the reaction center, as well as disruptions in electron transport and metabolic pathways, which collectively induce oxidative stress and damage cell membrane. Treatment with either NPs or CuSO 4 alone results in the downregulation of carbon-fixation, the tricarboxylic acid cycle, and pyruvate metabolism. Co-existence of NPs significantly influenced ABC transporters, carbon fixation, and the two-component system in M . aeruginosa . Notably, while single CuSO₄ treatment increased microcystin- leucine arginine (MC-LR) levels by 243.3 % relative to controls, combining CuSO₄ with NPs (PS, PS-NH₂, PS-COOH) significantly reduced MC-LR concentrations by 73.5 %, 90.2 %, and 70.7 %, respectively (p < 0.05). This study demonstrated that NPs exacerbated the oxidative stress induced by CuSO 4 on M . aeruginosa cell, while concurrently reducing MCs release. Furthermore, the underlying molecular mechanisms were systematically investigated. These findings provided novel insights into the crucial importance of considering NPs impact prior to the application of CuSO 4 for the management of (cyanoHABs).

Unraveling the effects of allelopathic algicide on Microcystis population: microcystin pollution risk assessment and proteomic-metabolomic insights into the sensitivity of toxic and non-toxic strains

Zhonghui Guo,Jiaqi Hu,Jieming Li,Xinyue Xia

The global expansion of Microcystis -dominated cyanobacterial blooms (MCBs) threatens aquatic ecosystems health, where Microcystis comprises coexisting microcystin (MC)-producing (MC + ) and MC-free (MC – ) strains. Luteolin, an allelopathic algicide, has shown potential in inhibiting MC + Microcystis growth and MC-production/-release. This study investigated luteolin's effects on co-cultured MC + and MC – Microcystis ( Microcystis population), comparing their sensitivity and elucidating mechanisms via MC-production/-release, proteomic, and metabolomic. Results showed that luteolin inhibited Microcystis population growth by 41.10% to 58.37%, with MC + strains exhibiting greater sensitivity, as indicated by stronger growth inhibition ratio (30.43% to 62.61%) and more severe cellular damage. Integrated proteomic and metabolomic analyses further revealed the underlying mechanisms of such sensitivity difference: MC + Microcystis exhibited severe impairments in photosynthetic activity, cell membrane composition, and antioxidant systems, alongside disruptions in protein biosynthesis, nucleotide degradation, and hindered DNA replication and repair. In contrast, MC – Microcystis experienced similar but milder disturbances, while promptly initiating compensatory responses. It enhanced carbohydrate metabolism and ATP synthase activity, promoted DNA replication and protein biosynthesis, and activated inflammatory responses to counteract luteolin stress. Additionally, although luteolin significantly decreased MC levels in the aqueous phase to 14.04   μg/L, it increased MC synthesis and retention within co-cultured MC + Microcystis cells, potentially exacerbating eco-risks due to prolonged MC release. This study clarifies the differential sensitivity of MC + and MC – Microcystis to allelopathic algicides and their molecular mechanisms, offering insights for developing targeted management plans based on each strain's survival strategies to control MCBs.

Effective removal of Microcystis aeruginosa and microcystins by integrated pre-oxidation and coagulation: an environmental and economical way

Song Q.,Zhang D.,Niu X.,Li Y.,Ma J.,Fan Y.,Zhou S.

The frequent occurrence of cyanobacteria blooms has gained worldwide concern. The conventional pre-oxidation methods are not only less effective in removing Microcystis aeruginosa cells, but also tend to trigger secondary pollution with an inevitable release of intracellular organic matter. In this study, Fe 2+ /persulfate process was applied to generate sulfate radicals and simultaneously form in situ Fe (III) for integrated oxidation–coagulation, leading to a notable enhancement in M. aeruginosa removal without cell breakage and better control of microcystins. Results showed that the Fe 2+ /persulfate mass ratio of 1:1 and the FeSO 4 dosage of 0.1 mmol/L were efficient in M. aeruginosa removal. With the optimal FeSO 4 dosage, up to 90.1% of M. aeruginosa were removed intact with the cell zeta potential of − 0.814. After settling down of M. aeruginosa within 30 min, the chlorophyll-a removal efficiency reached 98.9% and microcystins decreased remarkably. Furthermore, the degradation of algal organic matters was significantly enhanced with a relatively lower residual Fe. Protein-like substances and dissolved microbial metabolites were preferentially oxidized. The cell lysis only occurred to a small number of M. aeruginosa during the Fe 2+ /persulfate process, preventing from the environmental risk posed. This work demonstrates that an appropriate Fe 2+ /persulfate ratio and FeSO 4 addition is a feasible and promising method for removing M. aeruginosa in an environmental-friendly and cost-effective way.

Role of divalent metals Cu2+ and Zn2+ in Microcystis aeruginosa proliferation and production of toxic microcystins

Dong Ao,Zhen Lei,Mawuli Dzakpasu,Rong Chen

Cu 2+ and Zn 2+ , two ubiquitous metals in water environments, can widely trigger algae blooms at favourable environmental conditions. This paper elucidates the roles of Cu 2+ and Zn 2+ in the proliferation of Microcystis aeruginosa ( M. aeruginosa ) and synthesis of Microcystins (MCs). Findings indicate significant influences of increasing Cu 2+ and Zn 2+ concentrations on cell proliferation at limited available phosphorus concentrations of less than 0.1 mg/L. By contrast, Cu 2+ and Zn 2+ notably affected MCs production at all the inoculated phosphorus concentrations. The critical concentrations of 1 μg/L and 5 μg/L for Cu 2+ and Zn 2+ , respectively, are determined to trigger rapid cell proliferation and MCs production. Furthermore, the impacts of Cu 2+ and Zn 2+ on nitrogen absorption and, subsequently, on amino acids (AAs) formation in cells, is likely key in MCs synthesis. The two AAs Alanine (Ala) and glutamic acid (Glu) demonstrate the most notable variations with the concentrations of Cu 2+ & Zn 2+ .

Microcystis aeruginosa co-exposure to bisphenol AF and perfluorooctanoic acid promotes cell division and increases microcystin and nitrous oxide emissions

Hao Ming,Jinhua Mou,Carol Sze Ki Lin,Liandong Zhu

The global rise in cyanobacterial blooms risk parallels the frequent detection of endocrine-disrupting compounds (EDCs) in freshwater, raising the unexplored possibility that EDCs may covertly trigger and sustain these events. This study investigated how bisphenol AF (BPAF) and perfluorooctanoic acid (PFOA) modulated the physiology of Microcystis aeruginosa under different exposure concentrations and sequences. Single-cell transcriptomic and proteomic analyses revealed that low-level co-exposure (0.5 μM) to these chemicals enhanced cell division by 28.99 % via FtsZ upregulation and increased microcystin release by 20.11 %, accompanied by elevated tryptophan- and tyrosine-like fluorescence peaks. High-concentration exposure (5 μM) induced oxidative stress, disrupted cell division, and triggered cell enlargement and secretion of dissolved organic matters. BPAF stimulated nitrous oxide (N₂O) production to 885.11 ng/(10 6 cells·h), whereas PFOA suppressed emissions, compared to 8.93 ng/(10 6 cells·h) in the control group. The nitrogen regulator NtcA was identified as a central node linking nitrogen metabolism, oxidative stress and photosynthetic redox reactions with N₂O synthesis. Molecular docking supported stable interactions between NtcA and both compounds. These findings uncovered distinct regulatory strategies in Microcystis aeruginosa under chemical stress, offering mechanistic insights into how endocrine-disrupting pollutants reshape microbial growth and greenhouse gas emissions in aquatic ecosystems.

Higher sensitivity of MC-producing Microcystis to two algaecides compared to co-cultured non-MC-producers

Ying Wei,Yunni Gao,Jing Wu,Jing Dong,Jingxiao Zhang,Xiaofei Gao,Huatao Yuan,Xuejun Li,Michele A Burford

Targeting and selectivity are essential for assessing the effectiveness of ecofriendly algaecides in controlling toxic cyanobacterial blooms. The dose- and time-dependent effects of hydrogen peroxide (H 2 O 2 ), pyrogallol and their sustained-release microspheres (HRM, PRM) on proportion of MC-producing cell numbers and genotypes of the co-existing MC- and non-MC-producing Microcystis , as well as microcystins (MCs) concentrations were investigated in the present study. Two MC-producing and two non-MC-producing Microcystis strains were used in mono- and co-culture test systems. The findings revealed that the MC-producing Microcystis strains were more sensitive to both forms of H 2 O 2 and pyrogallol in comparison with the non-MC-producing strains. Inhibition of all Microcystis strains by H 2 O 2 and pyrogallol was found to be dose- and time-dependent, with H 2 O 2 exhibiting stronger effects. Additionally, both pure and granular forms of H 2 O 2 and pyrogallol reduced the relative abundance of MC-producing genotypes in co-culture conditions, along with total MC concentrations. The HRM and PRM demonstrated more potent and prolonged effects than their pure forms, indicating they are optimal for targeted reduction of Microcystis bloom toxicity. The release rate of MCs into the extracellular environment of Microcystis may exceed their degradation rate during H 2 O 2 and pyrogallol treatments. It resulted in elevated extracellular MC levels after treatment. Therefore, ongoing monitoring of environmental MCs is necessary following the inhibition of Microcystis population growth by algaecides.

Impact of butylparaben on growth dynamics and microcystin-LR production in Microcystis aeruginosa

Zhong-Hong Zhang,Jian-Wei Zheng,Si-Fen Liu,Ting-Bin Hao,Wei-Dong Yang,Hong-Ye Li,Xiang Wang

The presence of butylparaben (BP), a prevalent pharmaceutical and personal care product , in surface waters has raised concerns regarding its impact on aquatic ecosystems . Despite its frequent detection, the toxicity of BP to the cyanobacterium Microcystis aeruginosa remains poorly understood. This study investigates the influence of BP on the growth and physiological responses of M. aeruginosa . Results indicate that low concentrations of BP (below 2.5 mg/L) have negligible effects on M. aeruginosa growth, whereas higher concentrations (5 mg/L and 10 mg/L) lead to significant growth inhibition. This inhibition is attributed to the severe disruption of photosynthesis , evidenced by decreased Fv/Fm values and chlorophyll a content. BP exposure also triggers the production of reactive oxygen species (ROS), resulting in elevated activity of antioxidant enzymes . Excessive ROS generation stimulates the production of microcystin-LR (MC-LR). Furthermore, lipid peroxidation and cell membrane damage indicate that high BP concentrations cause cell membrane rupture , facilitating the release of MC-LR into the environment. Transcriptome analysis reveals that BP disrupts energy metabolic processes, particularly affecting genes associated with photosynthesis , carbon fixation , electron transport , glycolysis, and the tricarboxylic acid cycle . These findings underscore the profound physiological impact of BP on M. aeruginosa and highlight its role in stimulating the production and release of MC-LR, thereby amplifying environmental risks in aquatic systems.

Efficiency and mechanisms of simultaneous removal of Microcystis aeruginosa and microcystins by electrochemical technology using activated carbon fiber/nickel foam as cathode material

Huilan Lian,Ping Xiang,Yinghao Xue,Yuzhu Jiang,Mengying Li,Jingyu Mo

The significant removal efficiency of microcystis aeruginosa was presented using Pt/Ti anode and activated carbon fiber/nickel foam (ACF/Ni) cathode by addition of Fe 2+ slightly in a wide range of initial pH (3–9). Results showed that about 93% of the Microcystis aeruginosa cells were removed within 15 min for Pt/Ti-ACF/Ni–Fe 2+ system. Dosage of Fe 2+ , current density, and initial pH had remarkable effects on the removal efficiency of microcystis aeruginosa. The mechanism of algae removal in the Pt/Ti-ACF/Ni–Fe 2+ electrochemical system was revealed by the comparison between Pt/Ti-ACF/Ni–Fe 2+ process and classical Fenton process, the analysis on Microcystis aeruginosa and ACF/Ni by SEM, the specific surface area and pore size analysis of ACF, and the determination of UV 254 , OD 620 and microcystin-LR (MC-LR). Results showed that the main mechanism of this system was the electro-Fenton process, which was accompanied by electro-adsorption, electro-floatation, and electro-coagulation process. And the cooperation mechanism on the electrochemical removal system was further speculated. With the breakdown of algal cells during the electrolysis, the MC-LR and other substances released from the cells were effectively degraded. Besides, the new cathode exhibited favorable and stable reusability . This study built up a high-efficiency algae removal system, which broke through the limits of narrow working pH range and large consumption of exogenous chemicals in electro-Fenton process.

Efficient Immobilization of CMlrA on Mesoporous Carriers for Synchronized Harmful Microcystis Blooms Suppression and Microcystin Degradation

Yu Wu,Linna Shao,Cai Cheng,Xianyong Kou,Shi Huang,Liexiong Zhang,Jie Yu,Xuehan Xu,Xingyu Xiang,Xiaoliang Guo,Lingling Feng

Harmful Microcystis blooms (HMBs) and their hepatotoxic metabolites microcystins (MCs) pose escalating threats to aquatic biodiversity and public health due to their recalcitrance to conventional remediation approaches. To address this dual challenge, mesoporous silica (MSN) and hollow mesoporous organosilica (HMON) nanostructures were engineered via structure-guided design principles, enabling the development of the crude extract microcystinase A (CMlrA)-tailored immobilized platform (CMlrA@MSN and CMlrA@HMON) for synchronized HMBs suppression and MCs detoxification. Systematic optimization of carrier physicochemical properties (including specific surface area, hydrophilicity, and pore size) significantly enhanced the immobilization efficiency and catalytic durability of CMlrA. Kinetic modeling revealed 201.3% and 44.1% K cat / K m enhancements in MC-LR degradation for CMlrA@MSN and CMlrA@HMON relative to free CMlrA. Immobilized enzyme systems achieved 77.06% and 83.03% growth inhibition rates in Microcystis aeruginosa FACHB-905 after six-day treatment. This biocatalytic immobilization strategy establishes an eco-efficient framework for synergistic HMB suppression and MC degradation, providing the technical groundwork for practical water remediation systems.

Inactivation of Microcystis aeruginosa by peroxydisulfate activated with single‐atomic iron catalysis: Efficiency and mechanisms

Hangjun Zhang,Bingzhi Yu,Xizi Li,Yan Li,Yuchi Zhong,Jiafeng Ding

Harmful algal blooms caused by cyanobacteria in drinking water have posed serious threats to destroy aquatic ecosystems , and the toxins they produce are harmful to human and animal health. Herein, the feasibility of removing Microcystis aeruginosa ( M. aeruginos ) and microcystin-LR ( MC-LR ) by free radicals produced by simple-atomic iron-doped g-C 3 N 4 (Fe-g-C 3 N 4 )-activated peroxydisulfate (PDS) was studied. Pristine g-C 3 N 4 and three kinds of Fe-g-C 3 N 4 with different compositions were successfully synthesized and characterized. When the initial algae density was 2.5 × 10 6 cells·mL −1 , approximately 96.25 % of M. aeruginosa cells were inactivated with 2.0 g·L −1 Fe-g-C 3 N 4 and 4.0 mM PDS under the conditions of pH 7.0, and MC-LR could also be effectively degraded. The activated system can effectively remove M. aeruginosa, and MC-LR was evaluated for its morphological characteristics, cell membrane integrity and microcystin degradation. In addition to the widely recognized reaction species (SO 4 • − and • OH), singlet oxygen ( 1 O 2 ) was another predominant radical for destroying the cell structure and removing MC-LR . The materials can be well recycled, and a possible removal mechanism was proposed. This work shows that the Fe-g-C 3 N 4 -activated PDS system is an environmentally friendly and efficient technology for algae removal and algal toxin reduction.

Understanding the inhibitory mechanism of antialgal allelochemical flavonoids from genetic variations: Photosynthesis, toxin synthesis and nutrient utility

Shumiao Yu,Chao Li,Caicai Xu,Kokoette Effiong,Xi Xiao

Flavonoids are natural polyphenolic compounds from plants. As a new biotechnological algaecide, the molecular mechanism of plant flavonoids on the inhibition of Microcystis aeruginosa is still unknown. Therefore, in this study, we analyzed the variation of expressions of photosynthesis-related genes, microcystin synthesis-related genes and the genes involved in N and P acquisition in M. aeruginosa under the flavonoids stress. The results showed that the expression of psb D1, psa B and rbc L related to photosynthesis were influenced by three flavonoids but with different changing tendencies. The transcription of mcy A, mcy D and mcy H related to microcystin synthesis were decreased after 5-d of exposure, which could block microcystin synthesis. Meanwhile, flavonoids treatments resulted in the inhibition of N and P acquisition related genes transcription to affect the absorption of N and P in algal cells, and further influenced the physiological metabolic process of M. aeruginosa .
