Controllable Direct-Writing of Serpentine Micro/Nano Structures via Low Voltage Electrospinning

Feiyu Fang,Xin Chen,Zefeng Du,Ziming Zhu,Xindu Chen,Han Wang,Peixuan Wu

Micro/nanofibers prepared by direct-writing using an electrospinning (ES) technique have drawn more attention recently owing to their intriguing physical properties and great potential as building blocks for micro/nanoscale devices. In this work, a wavy direct-writing (WDW) process was developed to directly write serpentine micro/nano structures suitable for the fabrication of micro devices. This fabrication ability will realize the application of electrospun-nanofiber-based wiring of structural and functional components in microelectronics, MEMS, sensor, and micro optoelectronics devices, and, especially, paves the way for the application of electrospinning in printing serpentine interconnector of large-area organic stretchable electronics.Keywords:electrospinning;direct-writing;serpentine structure;micro pattern

Rational design of dual-functional surfaces on polypropylene with antifouling and antibacterial performances via a micropatterning strategy

Liu Jiang,Niuniu Qin,Shunli Gu,Wancheng Zhu,Changhao Wang,Yashao Chen

The hydrophobicity and inertness of the polypropylene (PP) material surface usually lead to serious biofouling and bacterial infections, which hamper its potential application as a biomedical polymer. Many strategies have been developed to improve its antifouling or antibacterial properties, yet designing a surface to achieve both antifouling and antibacterial performances simultaneously remains a challenge. Herein, we construct a dual-function micropatterned PP surface with antifouling and antibacterial properties through plasma activation, photomask technology and ultraviolet light-induced graft polymerization. Based on the antifouling agent poly(2-methacryloyloxyethyl phosphate choline) (PMPC) and the antibacterial agent quaternized poly(N,N-dimethylamino)ethyl methacrylate (QPDMAEMA), two different micropatterning structures have been successfully prepared: PP-PMPC-QPDMAEMA in which QPDMAEMA is the micropattern and PMPC is the coating polymer, and PP-QPDMAEMA-PMPC in which PMPC is the micropattern and QPDMAEMA is the coating polymer. The composition, elemental distribution and surface morphology of PP-PMPC-QPDMAEMA and PP-QPDMAEMA-PMPC have been thoroughly characterized by Fourier transform infrared (FT-IR) spectroscopy, X-ray photoelectron spectroscopy (XPS) and scanning electron microscopy (SEM), respectively. Compared with pristine PP, the two types of micropatterned PP films exhibit good surface hydrophilicity as characterized by water contact angle measurements. The results of anti-protein adsorption, platelet adhesion and antibacterial evaluation showed that PP-PMPC-QPDMAEMA and PP-QPDMAEMA-PMPC had good anti-protein adsorption properties, especially for lysozyme (Lyz). They can effectively prevent platelet adhesion, and the anti-platelet adhesion performance of PP-QPDMAEMA-PMPC is slightly better than that of the PP-PMPC-QPDMAEMA sample. The sterilization rate of S. aureus and E. coli is as high as 95% for the two types of micropatterned PP films. Due to the rational design of micropatterns on the PP surface, the two classes of dual-functional PP materials realize both the resistance of protein and platelet adhesion, and the killing of bacteria at the same time. We anticipate that this work could provide a design strategy for the construction of multifunctional biomedical polymer materials.

Efficient Differentiation of hiPSCs into hMSC-like Cells under Chemically Defined Conditions on Temperature-Sensitive Micropatterned Surfaces

Pengxia Zhang,Maoying Liu,Suying Pei,Hongxin Huang,Zhengyan Zhao,Ling Yang,Wen Pan,Siyi Li,Qifeng Bai,Rui Zhang,Ping Zhou

The fairness of long-term self-renewal and robust cell proliferation limits the applications of human mesenchymal stem cells (hMSCs) in regenerative medicine. Inducing hMSCs from human-induced pluripotent stem cells (hiPSCs), which have the advantages of autogenous and no cell number issues, is highly valuable. However, current induction methods using FBS-containing mesenchymal culture medium have problems, including immunogenicity, microbial contamination, and low efficiency. To solve these problems, we propose a chemically defined induction protocol incorporating transforming growth factor-beta 1 and retinoic acid (RA) additives in serum-free E6 medium for the suspension induction of embryoid bodies in hiPSCs. Additionally, microgroove-patterned polydimethylsiloxane surfaces coated with temperature-sensitive N-isopropylacrylamide (PNIPAAm) were prepared for efficient harvesting and purification of induced hiPSC-derived hMSCs (hiPSC-MSCs). The results showed that both supplementation with RA and patterned microgrooves with a width of 20 μm could accelerate the induction of hiPSC-MSCs, realizing the promising scalable production of homogeneous cells. This study successfully established a chemically defined induction protocol and utilized patterned surfaces to obtain clinically applicable hiPSC-MSCs, which show great promise in tissue engineering, gene therapy, and cell transplantation.

Generation of a co-culture cell micropattern model to simulate lung cancer bone metastasis for anti-cancer drug evaluation

Huixiang Zhong,Liuyang Xuan,Dandan Wang,Jianhua Zhou,Yan Li,Qing Jiang

Lung cancer bone metastases usually involve multiple interactions between cancer cells and host organs. To mimic such a structure, a cell co-culture micropattern of A549 lung cancer cells and osteoblast cells (A549/OB) was developed using a μ-eraser strategy for anti-cancer drug evaluation and also understanding cell–cell communication. When applying the μ-eraser strategy, a PDMS stamp was pressed to induce cell lysis. Hence, the minimum pressure required to induce cell lysis was first quantified. Then the pressure and pressing time for the micropatterning process were optimized to obtain cell micropatterns of high fidelity and repeatability. For different types of cells (A549 and human mesenchymal stem cells) and different substrates (TCP and PLGA nanofiber sheets), the optimized pressure was also different (i.e., for A549 on TCP, a pressure of 24.5 kPa was pressed for 10 s). According to a live/dead assay and Alamar Blue assay, cell viability and proliferation potential were not affected by the micropatterning process. There were several advantages for the μ-eraser strategy: substrates were not pre-treated; cell micropatterns mainly relied on the pattern of the PDMS stamp along with the micropatterning process; cells in the micropattern were not restricted in specified regions. Thus the A549/OB co-culture micropattern on TCP was used to evaluate the efficacy of a therapeutic agent (doxorubicin). In the co-culture micropattern, the efficacy of doxorubicin decreased when the expression of ALP in OB was elevated. The co-culture micropattern model showed the potential to be used for new anti-cancer drug development.

Printable and filament-drawable PDMS based adhesive assisted manufacturing of highly conductive copper micro-patterns

Jing Tian,Sanying Hou,Lingjie Mao,Xin Xu,Humeizi Cao,Xinyi Duan,Lingsisi Li,Zhiling Zhou,Yaqiang Ji,Jin-Qi Xie

Manufacturing of copper micro-patterns is crucial in electronics for its utilization as high conductivity transparent conductive films (TCFs) and circuits. In the preparation process of current TCFs, a plethora of materials have emerged that can replace traditional indium tin oxide (ITO). However, even for the most promising metal-based nanowire materials, there are issues such as high cost, complex welding, and high contact resistance. To address these problems, this paper proposes a printable and filament-drawable polydimethylsiloxane (PDMS)-based adhesive, which, through a novel additive patterning technology, efficiently and economically manufactures self-welding copper micro-meshes and circuits. The adhesive can be processed into micro-patterns through printing and filament drawing, on which ionic Ag can be in situ reduced and anchored, thereby eliminating the need for tedious pre- and post-treatment steps. The fully exposed Ag particles dramatically minimize the usage of precious metal catalyst, thus efficiently catalyzing electroless copper deposition (ECD) reaction. Highly conductive (1.03 × 10 7 S m −1 ) copper circuits can be fabricated on the printed adhesive patterns, exhibiting versatile applicability to diverse substrates. Highly precise copper micro-meshes (∼50 μm) can be fabricated on the filament networks drawn by the adhesive. The copper meshes undergo complete self-welding at junctions during the ECD process, thus exhibiting ultra-low square resistance of 0.45 Ω sq −1 while maintaining a high transmittance of 82.2 %. This is far superior to most of TCFs in published literature.

Fabrication of robust metallic micropatterns on glass surfaces by selective metallization in laser-induced porous surface structures

Jiangyou Long,Jianguo Li,Miaoni Li,Xiaozhu Xie

Conductive metallic micropatterns on transparent glass substrates are desired in lots of applications, e.g. electrofluidic devices and micrototal analysis systems. Due to the significant differences in material properties between metal and glass, the fabricated metallic micropatterns prone to show limited bonding strength to the glass substrates. In this paper, microgrooves with porous edge areas are prepared by a laser-induced backside wet etching (LIBWE) method. In the subsequent electroless plating process, this peculiar surface structure enables fast deposition of copper on these areas without further treatment of the surface. Moreover, due to the strong anchor effect caused by the porous microstructures, the obtained copper micropatterns show high bonding strength to the glass substrate. No damage or peeling is observed under 20-min sonication in water and repeated scotch tape test. These micropatterns can even survive from surface polishing by sandpapers. We also demonstrated that this copper micropatterns with low resistivity (1.9 μΩ·cm) could be used for rapid heating of localized surface areas, which may be a potential choice for various microsystems.

Rare earth complexes using azobenzene-containing poly(aryl ether)s with different absorption wavelengths as macromolecular ligands: synthesis, characterization, fluorescence properties and fabrication of fluorescent holographic micropatterns

Yuxuan Zhang,Shanyong Chen

In this paper, two novel azobenzene-containing poly(aryl ether)s with different absorption wavelengths were synthesized via Ullmann coupling and Sonogashira coupling, respectively. The obtained polymers were characterized and evaluated by elemental analysis, IR, 1H NMR, UV-vis, DSC and TGA. Rare earth complexes were prepared by using the two novel azobenzene-containing poly(aryl ether)s as macromolecular ligands. The obtained rare earth complexes were characterized by elemental analysis, IR and WAXD. The influence of the absorption wavelength of azobenzene chromophores on the fluorescent properties was investigated. The polymer whose absorption wavelength was far from the excitation wavelengths of the rare earth complexes showed a much larger fluorescence intensity. By exposing the films of the rare earth complexes to two interference laser beams, SRGs can be formed on the films and can also be detected by fluorescence microscopy measurement.

Ultrafast Fabrication of Large-Area Colloidal Crystal Micropatterns via Self-Assembly and Transfer Printing

Xuan Li,Lei Chen,Yuan Ma,Ding Weng,Zhaoxin Li,Lele Song,Xuanhe Zhang,Guoxu Yu,Jiadao Wang

Colloidal crystals have brought the promise of revolution to modern engineering, yet commonly used fabrication technologies are still limited by the small preparation area, time-consuming process, and dependence on sophisticated equipment. Here, a surface tension gradient-driven self-assembly strategy is proposed for the ultrafast fabrication of large-area colloidal crystals. The hydrogel loaded with sodium dodecyl sulfate is devised to construct a stable and continuous liquid-air interfacial tension gradient, and the resulting Marangoni effect can drive the micro-nano particles to instantaneously form (within several seconds) highly ordered colloidal crystals. Benefiting from the long range of surface tension gradients, the fabrication area of colloidal crystal films is demonstrated to exceed an astonishing 1000 cm 2 without compromising their quality, showing great potential in scale-up manufacture. Moreover, particles of a wide variety of sizes, materials, and functionalities can form close-packed self-assembly monolayers and be transferred to various substrates without damage, exhibiting great versatility. Inspired by ink microprinting, an ultrafast nanoparticle transfer printing method is further proposed to convert the close-packed nanoparticle monolayers into large-area conformal micropatterns with single-nanoparticle resolution. The great potential of nanoparticle micropatterns is demonstrated in flexible micro-electronics/skin electronics. This user-friendly, efficient self-assembly, and micropatterning strategy provide promising opportunities for academic and real industrial applications.

Robust metallic micropatterns fabricated on quartz glass surfaces by femtosecond laser-induced selective metallization

Yajun Huang,Yajun Huang,Xiaozhu Xie,Xiaozhu Xie,Xiaozhu Xie,Jiaqi Cui,Wenqian Zhou,Wenqian Zhou,Jianqiang Chen,Jianqiang Chen,Jiangyou Long,Jiangyou Long

Quartz glass has a wide range of application and commercial value due to its high light transmittance and stable chemical and physical properties. However, due to the difference in the characteristics of the material itself, the adhesion between the metal micropattern and the glass material is limited. This is one of the main things that affect the application of glass surface metallization in the industry. In this paper, micropatterns on the surface of quartz glass are fabricated by a femtosecond laser-induced backside dry etching (fs-LIBDE) method to generate the layered composite structure and the simultaneous seed layer in a single-step. This is achieved by using fs-LIBDE technology with metal base materials (Stainless steel, Al, Cu, Zr-based amorphous alloys, and W) with different ablation thresholds, where atomically dispersed high threshold non-precious metals ions are gathered across the microgrooves. On account of the strong anchor effect caused by the layered composite structures and the solid catalytic effect that is down to the seed layer, copper micropatterns with high bonding strength and high quality, can be directly prepared in these areas through a chemical plating process. After 20-min of sonication in water, no peeling is observed under repeated 3M scotch tape tests and the surface was polished with sandpapers. The prepared copper micropatterns are 18 µm wide and have a resistivity of 1.96 µΩ·cm (1.67 µΩ·cm for pure copper). These copper micropatterns with low resistivity has been proven to be used for the glass heating device and the transparent atomizing device, which could be potential options for various microsystems.

Light-induced dynamically tunable micropatterned surface for the regulation of the endothelial cell alignment

Jiao Wang,Chengzhen Chu,Yang He,Tao Xiang,Shaobing Zhou

The surface topography has a great effect on the behaviour of adherent cells. We report an efficient method to prepare shape memory surfaces with dynamically changed micropatterns, which can be induced by near-infrared (NIR) light by varying the power density. After polydopamine (PDA) was coated on the cross-linked polyethylene glycol-poly(ɛ-caprolactone), the surface temperature increases by 40°C at room temperature when 808 nm light with 1.0 W/cm 2 is used because of the photothermal properties of PDA. This temperature increase is enough for the shape recovery of the pressed micropatterns. The depth of the recovered micropatterns is controllable by adjusting the power density of the 808 nm light. The NIR-induced micropatterns efficiently regulate the morphology and alignment of endothelial cells. Therefore, NIR-induced shape memory surfaces have the potential to be used in remote-controlled devices.

Single cell migration dynamics mediated by geometric confinement

Hua Zhang,Ruixia Hou,Peng Xiao,Rubo Xing,Tao Chen,Yanchun Han,Penggang Ren,Jun Fu

The migration dynamics of cells plays a key role in tissue engineering and regenerative medicine. Previous studies mostly focus on regulating stem cell fate and phenotype by biophysical cues. In contrast, less is known about how the geometric cues mediate the migration dynamics of cells. Here, we fabricate graphene oxide (GO) microstripes on cell non-adhesive PEG substrate by using micromolding in capillary (MIMIC) method. Such micropatterns with alternating cell adhesion and cell resistance enable an effective control of selective adhesion and migration of single cells. The sharp contrast in cell adhesion minimizes the invasion of cells into the PEG patterns, and thereby strongly confines the cells on GO microstripes. As a result, the cells are forced to adapt highly polarized, elongated, and oriented geometry to fit the patterns. A series of pattern widths have been fabricated to modulate the extent of cell deformation and polarization. Under strong confinement, the cytoskeleton contractility, intracellular traction, and actin filament elongation are highly promoted, which result in enhanced cell migration along the patterns. This work provides an important insight into developing combinatorial graphene-based patterns for the control of cell migration dynamics, which is of great significance for tissue engineering and regenerative medicine.

Anti-Scar Effects of Micropatterned Hydrogel after Glaucoma Drainage Device Implantation

Yiling Han,Qiangwang Geng,Aimeng Dong,Menglu Jiang,Jingyi Ma,Wulian Song,Pan Fan,Yuanyuan Li,Jiawen Gao,Fenghua Zhang,Jinsong Leng,Huiping Yuan

Excessive fibrosis is the primary factor for the failure of glaucoma drainage device (GDD) implantation. Thus, strategies to suppress scar formation in GDD implantation are crucial. Although it is known that in implanted medical devices, microscale modification of the implant surface can modulate cell behavior and reduce the incidence of fibrosis, in the field of ophthalmic implants, especially the modification and effects of hydrogel micropatterns have rarely been reported. Here, we designed the patterned gelatin/acrylamide double network hydrogel and developed an innovative GDD with micropattern to suppress inflammatory and fibroblast activation after GDD implantation. Pattern topography suppressed F-actin expression and mitigated actin-dependent nuclear migration of myocardin-related transcription factor A (MRTF-A) during the proliferative phase after GDD implantation. Ultimately, the expression of α-smooth muscle actin (α-SMA), a key fibrosis-related gene product, was suppressed. Moreover, the modified GDD effectively controlled intraocular pressure (IOP), mitigated fibrous formation, and remodeled extracellular matrix (ECM) collagen distribution in vivo. Therefore, the novel GDD with surface patterning interventions provides a promising strategy to inhibit scar formation after GDD implantation and raise the efficacy of GDD implantation.
