A novel electrochemical biosensor based on AuNPs/Ti 3 C 2 MXene three-dimensional nanocomposite for sensitive miRNA-155 detection by exonuclease III (Exo III)-aided cascade target recycling is reported. Ti 3 C 2 MXene is synthesized and used as a support to immobilize Au nanoparticles (AuNPs) due to its special layered structure and metallic properties. The three-dimensional (3D) structure of the AuNPs/Ti 3 C 2 MXene nanocomposite for the biosensor platform leverages the integrated advantages of a large specific surface area, excellent electrical conductivity and electrocatalytic properties . AuNPs are used to immobilize capture DNA (C-DNA) by Au S chemical bonds. The C-DNA was modified with methylene blue (MB) at the 3′ end, resulting in an initial DPV electrochemical signal ( I d ). In the presence of target, miRNA-155 and C-DNA formed a double-stranded structure by complementary base pairing. Subsequently, Exo III digested the 3′ end of the C-DNA in the double-stranded structure and caused the electrochemical signal to “turn off” (the final signal denoted as I h ). By Exo III-aided cascade cleavage of the target, one miRNA-155 chain causes the release of several C-DNAs under certain conditions, which significantly amplifies the peak current difference on the fabricated biosensor (Δ I = I d ﹣ I h ). The electrochemical sensor achieved a detection limit of 0.35 fM (S/N = 3) with a linear range from 1.0 fM to 10 nM. This sensor also shows desirable stability, reproducibility and specificity.

Despite numerous efforts, the accurate determination of trace biomolecules with zeptomolar sensitivity remains elusive. Here, a 3D carbon nanomaze (CAM) electrode for the ultrasensitive detection of trace biomolecules such as nucleic acids, proteins, and extracellular vesicles is reported. The CAM electrode consists of an interlaced carbon fiber array on which intercrossed graphene sheets are vertically tethered in situ, permitting local confinement of trace molecules to increase molecular hybridization efficiency. Furthermore, a self-assembled DNA tetrahedron array adopts a rigid spatial conformation to guarantee the controllable arrangement of immobilized biological probes, facilitating analytical sensitivity and reproducibility. In a proof-of-concept experiment on detecting microRNA-155, a linearity of 0.1 aM to 100 nM and a sensitivity of 0.023 aM (23 zM) are achieved. With the optimal parameters, the proposed nanoelectrode demonstrates encouraging consistency with quantitative real-time polymerase chain reaction during clinical sample detection. Through simple functionalization by appending various biomolecular probes of interest, the developed CAM platform with ultrahigh sensitivity can be exploited as a versatile tool in environmental, chemistry, biology, and healthcare fields.

The imbalanced expression of microRNA is related to many diseases and tumors, which can be used as a noninvasive biomarker to assist in the diagnosis, prognosis, and monitoring of cancer. A target conversion strategy-based electrochemical biosensor was developed by DNAzyme-cleavage dual cyclic signal amplification and the strand displacement reaction (SDR) to produce high electrochemical signals for microRNA-155 (miRNA-155) analysis. Thereinto, VSe2/multi-walled carbon nanotubes (MWCNTs) with high conductivity were applied as the electrode surface material to provide more active sites and accelerate electron transportation. Then, the excellent immobilization efficiency and large specific surface area of the ZnO@CuS nanoprobes were applied as a recognition molecule carrier. Furthermore, hexaammineruthenium(III) chloride (RuHex), with superior electrochemical redox activity, acted as the electroactive indicator by intercalating into double-helix DNA through electrostatic interaction, which produced high current responses in the presence of the target. Therefore, this miRNA-155 biosensing platform exhibited a relatively low limit of detection of 0.11 fM, ranging from 0.5 fM to 5 nM, and demonstrated excellent repeatability and stability. Hence, this platform appeared to possess favorable human serum detection capacity and be suitable for early cancer diagnostics.

The microRNA family shows great promise for the diagnosis and prediction of different cancers as biomarkers, in which miRNA-155 (miR155) is recognized as a biomarker for lung cancer and thus has drawn much attention. Therefore, the development of specific and accurate methods to detect miR155 has become a potential research hotspot. In this work, we presented a fluorescence resonance energy transfer (FRET) technique by utilizing core–shell CdSeTe/ZnS quantum dots (QDs) as a signal reporter and a target recycling strategy as a signal amplifier for the detection of miR155. In the presence of miR155, part of the hairpin probe (HP) immobilized onto the surface of gold nanoparticles (AuNPs)@SiO2, could hybridize with it. Then, numerous H1-AuNPs@SiO2 could be produced with the aid of exonuclease III. With the linkage of the assistant chain A2, H1-AuNPs@SiO2 combined with A1-QDs and then the fluorescence (FL) signal of QDs was quenched by AuNPs@SiO2via FRET. The more miR155 present in solution, the weaker the detected FL signal. A linear response range of 0.01 fM to 1 nM and low detection limit of 3 aM were obtained using the proposed method. Furthermore, this method showed high specificity and satisfactory recovery between 93 and 105% in real human serum samples, illustrating its great potential for practical applications.

A promising nanocarrier of the complementary DNA (cDNA) toward miRNA-155 was synthesized on the basis of the nanohybrid type of Ti 3 C 2 T x MXene nanosheets decorated using iron phthalocyanine quantum dots (FePc QDs) (denoted as Ti 3 C 2 T x @FePcQDs) to construct a novel ultrasensitive impedimetric aptasensing system for microRNA-155 (miRNA-155). Owing to π–π* stacking interaction between Ti 3 C 2 T x nanosheets and FePc QDs, the homogeneous nanostructure of the Ti 3 C 2 T x @FePcQD nanohybrid that comprises mixed-valence states (Ti 2+ /Ti 3+ and Fe 2+ /Fe 3+ ), multicomponent (Ti-O and Ti-C), and various N-related groups was achieved. The constructed Ti 3 C 2 T x @FePcQDs-based aptasensor displayed an ultrahigh sensitivity for detecting miRNA-155 with a low detection limit of 4.3 aM (S/N = 3) within the miRNA-155 concentration ranging from 0.01 fM to 10 pM. Compared with the individual component-based aptasensors and other reported miRNA-155 aptasensors, the proposed impedimetric aptasensing system exhibited substantial merits of a feasible preparation process, non-use of labels or electrochemical indicators, fast response time, and comprehensive sensing performances for detecting miRNA-155. This strategy for determining miRNAs can extensively be applied as the platform for anchoring other kinds of aptamers in detecting diverse targets, thus indicating its great potential application for the early diagnosis of cancer biomarkers.

Owing to the excellent structural rigidity and programmable reaction sites, DNA nanostructures are more and more widely used, but they are limited by high cost, strict sequence requirements, and time-consuming preparation. Herein, a general signal amplifier based on a micelle-supported entropy-driven circuit (MEDC) was designed and prepared for sensitive quantification of biomarkers. By modifying a hydrophobic cholesterol molecule onto a hydrophilic DNA strand, the amphiphilic DNA strand was first prepared and then self-assembled into DNA micelles (DMs) driven by hydrophobic effects. The as-developed DM showed unique advantages of sequence-independence, easy preparation, and low cost. Subsequently, amplifier units DMF and DMTD were successfully fabricated by connecting fuel strands and three-strand duplexes (TDs) to DMs, respectively. Finally, the MEDC was triggered by microRNA-155 (miR-155), which herein acted as a model analyte, resulting in dynamic self-assembly of poly-DNA micelles (PDMs) and causing the recovery of cyanine 3 (Cy3) fluorescence as the DMTD dissociated. Benefiting from the “diffusion effect”, the MEDC herein had a nearly 2.9-fold increase in sensitivity and a nearly 97-fold reduction in detection limit compared to conventional EDC. This amplifier exhibited excellent sensitivity of microRNAs, such as miR-155 detection in a dynamic range from 0.05 to 4 nM with a detection limit of 3.1 pM, and demonstrated outstanding selectivity with the distinguishing ability of a single-base mismatched sequence of microRNAs. Overall, the proposed strategy demonstrated that this sequence-independent DNA nanostructure improved the performance of traditional DNA probes and provided a versatile method for the development of DNA nanotechnology in biosensing.

Antisense oligonucleotides (ASOs) are promising tools for gene silencing and have been exploited as therapeutics for human disease. However, delivery of therapeutic ASOs to diseased tissues or cells and subsequent escape from the endosomes and release of ASO in the cytosol remain a challenge. Here, we reported a neutrophil-membrane-coated zeolitic imidazolate framework-8 (ZIF-8) nanodelivery platform (AM@ZIF@NM) for the targeted transportation of ASOs against microRNA-155 (anti-miRNA-155) to the endothelial cells in atherosclerotic lesions. Neutrophil membrane could improve plaque endothelial cells targeting through the interaction between neutrophil membrane protein CD18 and endothelial cell membrane protein intercellular adhesion molecule-1 (ICAM-1). The ZIF-8 “core” provided high loading capacity and efficient endolysosomal escaping ability. Delivery of anti-miR-155 effectively downregulated miR-155 expression and also saved the expression of its target gene BCL6. Moreover, RELA expression and the expression of its downstream target genes CCL2 and ICAM-1 were correspondingly reduced. Consequently, this anti-miR-155 nanotherapy can inhibit the inflammation of atherosclerotic lesions and alleviate atherosclerosis. Our study shows that the designed biomimetic nanodelivery system has great application prospects in the treatment of other chronic diseases.
