Developing an effective approach based on microwave distillation and extraction using deep eutectic solvents for multiple target analytes prepared from Eleutherococcus senticosus fruits

Xu Pan,Xinyi Zhang,Yue Meng,Ying Yang,Honglian Zhang,Tingting Liu,Xiaoyu Sui,Cheng Yuan

Deep eutectic solvents (DES) have emerged as an environmentally-friendly alternative for extracting active ingredients from botanical materials. The combination of the microwave extraction method and DES can enhance mass transfer, and DES are an excellent alternative solvent for microwave solid–liquid extraction, having a low volatility and thermal expansion coefficient. In the present study, we used the microwave-assisted extraction/distillation technique to help the upgraded DES extract essential oil fractions and unique active ingredients (different in polarity) from Eleutherococcus senticosus fruits at the same time. After single-factor trials and a Box–Behnken design (BBD) and experiments, the total yield of the three target constituents, eleutheroside B, eleutheroside E, and isofraxidin, was 30.11 ± 0.16 mg/g, that of anthocyanins was 6.29 ± 0.11 mg/g, and that of essential oil was 2.00 ± 0.13 mg/g. Compared with other methods, the proposed method ensures that DES are compatible with microwave processes and that several types of components can be extracted.

Effects of Chemical Modification on the Structure and Biological Activities of Polysaccharides Extracted from Inonotus Obliquus by Microwave

Hao Xie,Wenwen Li,Linghe Zhang,Nurmamat Eliyas

A novel polysaccharide, Inonotus obliquus polysaccharide (IOP), was extracted using a microwave extraction method and subsequently subjected to modifications through sulfation, carboxymethylation, phosphorylation, and acetylation. Its physical and chemical properties were analyzed using various chemical techniques, including high-pressure liquid chromatography, ultraviolet light, Fourier-transform infrared spectroscopy, X-ray diffraction, Congo red test, and scanning electron microscopy. The antioxidant capacity was assessed using DPPH, ABTS, and hydroxyl radical assays, as well as by measuring the reducing power. Additionally, hypoglycemic activity was evaluated through α-glucosidase and α-amylase assays. The results indicated that the chemical modifications effectively altered the physical and chemical properties, as well as the biological activities of IOP. Compared to the unmodified IOP, the derivatives exhibited reduced sugar content, uronic acid content, and molecular weight, while demonstrating varying levels of antioxidant and hypoglycemic capabilities. Notably, the carboxymethylated IOP (IOP−C) displayed lower molecular weight, higher ABTS free radical scavenging rate, greater reducing ability, and increased α-amylase inhibition rate. Therefore, IOP−C shows promise as a potential edible antioxidant and hypoglycemic agent. Graphical

Establishment of a Generalizable Industrial Crop Model for Microwave Extraction of Unsaturated Fatty Acids

Junyi Chen,Didi Lu,Shiqiang Chen,Song Liu,Yaqiu Zhang,Conghong Zhan

To explore the relationship between unsaturated fatty acid (UFA) content and parameters for microwave extraction, multimaterial and multiparameter testing was conducted in which five kinds of materials with different UFA contents (potato, wheat, corn, soybean, and peanut) were selected for the experiment. Four factors, namely, extraction temperature ( X 1 ), extraction time ( X 2 ), proportional volume of acetone ( X 3 ), and liquid-to-solid ratio ( X 4 ), were screened for their significant effects by using Prob > | T | values from the Plackett–Burman experiment. A microwave extraction orthogonal experiment with four factors and five levels was designed separately using Design-Expert 8.05 software and them concentrated. Microwave-accelerated methyl esterification was then performed, and the UFA content was determined via gas chromatography (flame ionization detector). The optimal extraction conditions ( X 1 , X 2 , X 3 , X 4 ) and the optimal UFA content of potato were 80.68°C, 10.74 min, 0.80, 3.25 mL × g −1 , 1.08%; wheat: 80.81°C, 10.54 min, 0.80, 20.91 mL × g −1 , 2.26%; corn: 81.18°C, 9.93 min, 0.80, 50.94 mL × g −1 , 6.89%; soybean: 82.07°C, 9.07 min, 0.80, 93.87 mL × g −1 , 15.81%; and peanut: 83.12°C, 8.11 min, 0.80, 168.70 mL × g −1 , 33.07%. Then, the optimization results for the five kinds of materials were synthesized by Origin 8.0 software, the fitting degree of the cubic model with the four extraction factors was the highest, the determination coefficients were 0.9984, 0.9991, 0.8953, and 0.9989, the residual sums of squares were 0.2888, 0.1587, 0.8265, and 0.1864, and the correlation coefficients are ideal. The stability and accuracy of the model were verified by the orthogonal experiment of UFA extraction from rice, and the correlation coefficient between the predicted value and the actual value of the orthogonal experiment was 0.9998. This study systematically collates the optimal parameters for microwave extraction of UFA content in different crops from the perspective of multimaterial and multiparameter, which can provide a large amount of detailed basic data for microwave extraction technology.

Extraction, Characterization, Antioxidant and Hypoglycemic of Pectic Polysaccharides from Cantaloupe (Cucumis melo L.) Peels

Hui Sun,Ruizhan Chen,Fanlei Meng,Helong Bai,Li Tian,Juan Lu,Chunlong Bai,Dongxue Li,Wenjing Wu,Yongtang Wang,Mingze Gong

In this study, microwave extraction (ME) of pectic polysaccharides (M-CPPs) from cantaloupe peels (CPs) is optimized using response surface methodology (RSM) with a Box–Behnken design (BBD). The optimum ME parameters are liquid–solid ratio 26.04 mL g −1 , microwave power 543.04 W, irradiation time 6.38 min, and pH 1.5. Compared heat reflux extraction polysaccharides (H-CPPs), the yield, the contents of neutral sugars (TSC), uronic acids (UAC), total flavonoids (TFC), total phenolics (TPC), sulfate groups (SGC) of M-CPPs increased by 156.06%, 15.33%, 7.27%, 1019.05%, 1.75%, and 57.89%, but the molecular weight (Mw) and degree of esterification (DE) reduced by 29.08% and 24.81%, respectively. M-CPPs exhibit superior antioxidant and hypoglycemic activities than H-CPPs, which may be attributed to its higher UAC, TFC and SGC, lower Mw, and DE. There is almost no change in the monosaccharide types of isolated polysaccharides, only a change in molar ratio. Results proved that ME is an efficient technique for the extraction and modification of pectic polysaccharides (CPPs) from CPs with high yield, strong antioxidant, and hypoglycemic activities for applications in medical and food industries.

Qingfen Zhang,Chenglin Li,Shengcheng Mi,Xiaojie Xu,Zhiqiang He,Lin Wang,Zhonghua Tang,Qi Lu

The primary purpose of this study is to develop a highly efficient extraction method to meet the growing demand for Kaempferia galanga essential oil (KGEO) in various industries. In this study, an innovative approach for extracting essential oil from fresh K. galanga rhizomes using salt solution-enhanced homogenization pretreatment and microwave extraction (HP-SOME) was proposed. A kinetic study was conducted on the extraction process of essential oil. The key factors were screened using the Plackett–Burman Design and optimized using the Box-Behnken Design. Under the optimization parameters: sodium chloride concentration of 1.5 %, microwave power of 475 W, and microwave time of 23 min, the actual yield was 5.26 ± 0.10 %. The HP-SOME method exhibited a significant increase of 58.91 % in essential oil yield compared to that of the hydrodistillation method (3.31 ± 0.10 %). In addition, the composition of the KGEO was determined using Fourier transform infrared spectroscopy, Raman spectroscopy, and gas chromatography-mass spectrometry. The KGEO obtained by HP-SOME had a 6.26 % increase in the main component ethyl p-methoxycinnamate and a 4.13 % increase in ethyl cinnamate compared to that obtained by the HD method. Investigations into antimicrobial activity revealed that KGEO extracted by the HP-SOME method exhibited superior and more comprehensive antimicrobial characteristics. In conclusion, the HP-SOME method demonstrated remarkable capabilities in obtaining KGEO with notable efficiency, yield, and quality. This method holds great promise in the field of essential oil extraction.

Solvent-free microwave extraction of essential oil and hydrosol from fresh leaves of Cinnamomum burmannii (Nees et T. Nees) Blume after screw extrusion treatment and evaluation of pancreatic lipase inhibitory activities in vitro compared to its main components

Zehao Wang,Ru Zhao,Hongfei Gao,Yang Yang,Xinyu Yang,Huiyan Gu,Lei Yang,Xiaonan Zhang,Tingting Liu

One of the most significant sources of commercial camphor oil is the essential oil of Cinnamomum burmannii (Nees et T. Nees) Blume leaves, which has been manufactured using a variety of distillation techniques, including hydrodistillation and microwave-assisted hydrodistillation. In this study, we employed solvent-free microwave extraction (SFME) to recover hydrosol and essential oil from fresh C. burmannii leaves that had undergone screw extrusion. An optimum yield of 12.35 ± 0.62 mL/kg DW of essential oil was obtained by optimizing the major SFME contributing parameters using a face-centered central composite design (FCCCD). By using SFME, the influence of microwave irradiation power on the yield and quality of the essential oil of fresh leaves of C. burmannii was examined. With the aid of first- and second-order kinetic equations, SFME was contrasted with hydrodistillation and dry distillation. A total of 32 components were detected in the essential oil, and 22 components were detected in the hydrosol obtained by SFME under the optimized conditions. The main components of the essential oil were 1,8-cineole (21.67%), borneol (17.37%) and α -terpineol (7.45%), in descending order, and the main components of the hydrosol were trans -2-hydroxycinnamic acid (21.45%), borneol (22.07%), 1,8-cineole (13.65%) and α -terpineol (11.93%). 1,8-Cineole and borneol were the two components with the highest influence on hierarchical clustering, according to principal component analysis and heatmap analysis. Moreover, box-whisker plots were used to determine and compare the inhibitory activities of the essential oil and its three primary constituents against pancreatic lipase . The outcomes demonstrated that 1,8-cineole played a critical role in the essential oil's ability to inhibit pancreatic lipase, and they also demonstrated a positive correlation between the concentration and the amount of inhibition activity.

An eco-friendly and effective method for isolating essential oil from fresh Kaempferia galanga L.: Process optimization based on entropy weight method and response surface methodology

Qingfen Zhang,Dan Ge,Shengcheng Mi,Qi Lu

The bioactive properties of essential oil are strongly associated with their chemical composition. In this work, a novel method of salt-enhanced homogenization pretreatment coupled with solvent-free microwave extraction (HP-SSFME) was developed for extracting essential oil (KGEO) from fresh Kaempferia galanga rhizomes, which significantly improved the extraction efficiency. Gas chromatography was employed to measure the contents of the main active components, ethyl p -methoxycinnamate (EPMC) and ethyl cinnamate (EC) in the essential oil. The extraction process was optimized by the Box-Behnken Design using the comprehensive scores of the yields of EPMC, EC, and KGEO obtained by the entropy weight method. The optimal parameters were determined as follows: amount of sodium chloride was 1.60 %, homogenization speed of 15000 r/min, homogenization time of 4 min, microwave irradiation power of 300 W, microwave irradiation time of 23 min. Under these conditions, the KGEO yield was 5.10 %, EPMC yield was 3.81 %, EC yield was 0.89 %. Compared with the conventional methods, the HP-SSFME method not only significantly increased the extraction yield of KGEO but also remarkably enhanced the yields of EPMC and EC. Additionally, the HP-SSFME method has low CO 2 emission and energy consumption, highlighting its environmental friendliness. Moreover, in terms of antibacterial activity, the HP-SSFME outperformed other methods, demonstrating a more powerful and extensive inhibitory effect against a range of bacteria. This study provides a valuable reference for the efficient extraction and quality control of essential oils from Kaempferia galanga rhizomes.

Combined microwave and enzymatic treatment improve the release of insoluble bound phenolic compounds from the grapefruit peel insoluble dietary fiber

Guanyi Peng,Jiapan Gan,Ruihong Dong,Yi Chen,Jianhua Xie,Ziyan Huang,Yue Gu,Danfei Huang,Qiang Yu

Increasingly attention has been paid to the active substances in grapefruit peel. In this study, insoluble bound phenolic compounds (BP) were released from grapefruit peel insoluble dietary fiber (GP-IDF) by microwave extraction (M-BP), enzymatic hydrolysis (E-BP) and combined microwave and enzymatic treatment (ME-BP). The results showed that the yield of ME-BP was the highest (148.856 ± 0.620 GAE mg/100 g), and conditions of microwave treatment (microwave power, temperature, time) were 600 W, 85 °C, and 37 min, while that of enzymatic treatment were 8% addition, 60 °C, and 2 h. Scanning electron microscopy displayed that GP-IDF became looser by ME. ME mainly resulted in the removal of lignin in IDF from X-ray diffraction, which was consistent with FT-IR. Twenty seven BP were identified by UPLC-ESI-QTOF-MS/MS. The high content of gallic acid (42.50 ± 3.21 μg/g DW), ferulic acid (18.46 ± 1.65 μg/g DW) and protocatechuic acid (6.16 ± 0.72 μg/g DW) was quantified by LC-ESI-QqQ-MS/MS. Furthermore, ME-BP showed stronger antioxidant capacities than that of others (ABTS, oxygen and DPPH free radical scavenging ability), and showed good inhibition of α-glucosidase. Our results demonstrated that combined microwave and enzymatic treatment was an effective treatment for improving the release of BP.

Enhanced electromagnetic shielding and absorption in Ti3C2Tx MXene films via hydroxyl functionalization and interlayer spacing control

Jin Duan,Zhe Liu,Xiuchen Wang

Developing flexible materials with combined electromagnetic interference (EMI) shielding and microwave absorption capabilities holds significant application potential. Ti 3 C 2 T x MXene has emerged as a flexible EMI shielding material due to its thinness, adhesion, flexibility, and dual shielding and absorption properties. High conductivity-induced reflection and layered self-stacking challenges hinder Ti 3 C 2 T x MXene performance enhancement, posing challenges demanding solutions. This study first uses simulations to analyze how functional group modifications and interlayer spacing affect Ti 3 C 2 T x MXene's electromagnetic properties. Hydroxyl groups are proposed as key regulators of Ti 3 C 2 T x MXene's properties. Subsequently, Ti 3 C 2 T x MXenes were treated with a low-concentration NaOH gradient to modulate hydroxyl groups. Ultrasonic-microwave extraction resolved delamination, enabling ultra-thin film preparation. The treatment caused hydroxyl groups to initially increase and then decrease, with interlayer spacing first contracting and then expanding, achieving synchronous modulation of Ti 3 C 2 T x MXene's surface functional groups and interlayer spacing. The EMI shielding and microwave absorption of ultrathin films were analyzed, with shielding efficiency correlating positively with hydroxyl group concentration. Ti 3 C 2 T x MXene treated with 0.2 mol/L NaOH exhibited maximum hydroxyl concentration, achieving 48.965 dB shielding at 0.016 mm thickness (SE/t = 3060.320 dB/mm), demonstrating hydroxyl groups' role in improving shielding performance. Wave absorption increased with higher NaOH concentrations, correlating with surface hydroxyl groups. The 0.3 mol/L NaOH-treated MXene showed a 12.883 dB absorption increase versus untreated samples. Additionally, the green shielding index (g s ) of Ti 3 C 2 T x MXene films increased by 32.9 % with higher NaOH concentration. Results demonstrate that surface hydroxyl modification and layer spacing adjustment significantly enhance the EMI shielding and wave absorption of Ti 3 C 2 T x MXene. Meanwhile, the prepared thin, flexible products have potential applications in aerospace, defense, and flexible electronics.

Antioxidant Activity and Phenolic Acids Profiles of Artemisia Selengensis Turcz Extracted with Various Methods by HPLC-QTOF-MS/MS

Lu Zhang,Zong-Cai Tu,Hui Wang,Qing-Hui Wen,Zhi-Feng Fu,Xing Xie

Effects of dynamic high pressure microfluidization (DHPM), microwave, ultrasound, maceration and high temperature, on the phenolic acids and antioxidant activity of Artemisia selengensis Turcz (AST) were investigated. Microwave exhibited the best extraction efficacy on phenolic compounds and flavonoids with the content of 40.30 μg GAE/mg Extract, and 37.75 μg RTE/mg Extract, respectively. Microwave extract (MWE) also showed the strongest DPPH· and HO· scavenging ability (IC 50 value of 0.23 and 0.50 mg/mL, respectively), and reducing power (Ab 0.3 value of 0.23 mg/mL). Quinic acid, 3 caffeoylquinic acids (CQAs), 2 coumaroylquinic acids, 4 dicaffeoylquinic acids (diCQAs) were identified as the major phenolic acids in AST using HPLC-QTOF-MS/MS, and 3-CQA (63.71 μg/mL) showed the highest content followed by 1,5-diCQA (17.91 μg/mL) and 3,5-diCQA (13.50 μg/mL). MWE had the highest 3-CQA and diCQAs. While, high temperature could promote the degradation of diCQAs, 3- and 4-caffeoyl are more liable to decompose as compared with 1- and 5-caffeoyl. Practical Application Over production of reactive oxygen species (ROS) in body will induce various chronic diseases. AST is an underutilized vegetable with many health benefits. This research indicates that AST possessed appreciating antioxidant activities and abundant phenolic acids, it is could be a potentail antioxidants resource and for prevention of chronic diseases associated with ROS. It also suggested that microwave could be a promising method for extracting the antioxidants in AST. Therefore, this research could lay a foundation for the high-value application of AST in food, health care and natural therapy industry, and provide some methodological references for the development of antioxidants in AST.

Temperature-responsive deep eutectic solvents as eco-friendly and recyclable media for microwave extraction of flavonoid compounds from waste onion (Allium cepa L.) skins

Shang Xian-chao,Zhang Yu-qin,Zheng Yan-fen,Li Yiqiang

Polyphenols and flavonoids are abundant in spent onion skins (SOSs), being present in amounts are 2–sixfold greater times of than those in the inner bulb. In this study, an eco-friendly and efficient extraction method based on temperature-responsive deep eutectic solvents (TRDESs) was used to obtain natural flavonoids from SOSs. Microwave-assisted extraction yields of quercetin, kaempferol, luteolin, and quercetin-3-O-β-D-glucoside were evaluated to determine the efficiencies of a series of TRDESs. An extraction system composed of ethanolamine and m -cresol at ratio of 1:1 (20% water in TRDES system) was found to possess the highest efficiency. The extraction conditions were screened and optimized for efficiency using a Box-Behnken design. As a result, an extraction power of 554 W, extraction temperature of 76 °C, extraction time of 16 min, and solid–liquid ratio of 14 mg/mL were determined to be the optimal conditions. Under these extraction conditions, the total flavonoid extraction yields reached 47.83 ± 0.21 mg/g. The macroporous resin HPD 600 was selected for the separation and recovery of the TRDESs extraction mixture and the experimental measurements revealed that it allowed recovery rate more than 90%. In vitro activity testing of the flavonoid extract demonstrated excellent FRAP assay, DPPH assay and ABTS assay results for the SOSs extract prepared using a TRDES, and its inhibitory effects on S. aureus , E. carotovora , and B. subtilis were marked.
