A Facile Single-Phase-Fluid-Driven Bubble Microfluidic Generator for Potential Detection of Viruses Suspended in Air

Jia Man,Luming Man,Chenchen Zhou,Jianyong Li,Shuaishuai Liang,Song Zhang,Jianfeng Li

Microfluidics devices have widely been employed to prepare monodispersed microbubbles/droplets, which have promising applications in biomedical engineering, biosensor detection, drug delivery, etc. However, the current reported microfluidic devices need to control at least two-phase fluids to make microbubbles/droplets. Additionally, it seems to be difficult to make monodispersed microbubbles from the ambient air using currently reported microfluidic structures. Here, we present a facile approach to making monodispersed microbubbles directly from the ambient air by driving single-phase fluid. The reported single-phase-fluid microfluidic (SPFM) device has a typical co-flow structure, while the adjacent space between the injection tube and the collection tube is open to the air. The flow condition inside the SPFM device was systematically studied. By adjusting the flow rate of the single-phase fluid, bubbles were generated, the sizes of which could be tuned precisely. This facile bubble generator may have significant potential as a detection sensor in detecting viruses in spread droplets or haze particles in ambient air. Keywords:microfluidic device;single-phase fluid;bubbles;air detection

A Multi-Region Magnetoimpedance-Based Bio-Analytical System for Ultrasensitive Simultaneous Determination of Cardiac Biomarkers Myoglobin and C-Reactive Protein

Zhen Yang,Huanhuan Wang,Pengfei Guo,Yuanyuan Ding,Chong Lei,Yongsong Luo

Cardiac biomarkers (CBs) are substances that appear in the blood when the heart is damaged or stressed. Measurements of the level of CBs can be used in course of diagnostics or monitoring the state of the health of group risk persons. A multi-region bio-analytical system (MRBAS) based on magnetoimpedance (MI) changes was proposed for ultrasensitive simultaneous detection of CBs myoglobin (Mb) and C-reactive protein (CRP). The microfluidic device was designed and developed using standard microfabrication techniques for their usage in different regions, which were pre-modified with specific antibody for specified detection. Mb and CRP antigens labels attached to commercial Dynabeads with selected concentrations were trapped in different detection regions. The MI response of the triple sensitive element was carefully evaluated in initial state and in the presence of biomarkers. The results showed that the MI-based bio-sensing system had high selectivity and sensitivity for detection of CBs. Compared with the control region, ultrasensitive detections of CRP and Mb were accomplished with the detection limits of 1.0 pg/mL and 0.1 pg/mL, respectively. The linear detection range contained low concentration detection area and high concentration detection area, which were 1 pg/mL–10 ng/mL, 10–100 ng/mL for CRP, and 0.1 pg/mL–1 ng/mL, 1 n/mL–80 ng/mL for Mb. The measurement technique presented here provides a new methodology for multi-target biomolecules rapid testing.Keywords:myoglobin;C-reactive protein;Dynabeads;magnetoimpedance;microfluidic device

New Single-Layered Paper-Based Microfluidic Devices for the Analysis of Nitrite and Glucose Built via Deposition of Adhesive Tape

Peng Yu,Muhan Deng,Yi Yang

A simple, low-cost technique has been developed for the rapid fabrication of single-layered paper-based microfluidic devices (μPADs). This technique, for the first time, made use of the deposition of patterned adhesive tape into the filter paper to construct hydrophobic barriers, with the help of toluene. Unlike other reported multi-layered μPADs that merely made use of adhesive tape as a separate layer for sealing or fluid flow controlling, the patterned adhesive tape was simultaneously dissolved and penetrated into the filter paper, which resulted in the successful transfer of the pattern from the tape to the filter paper. To demonstrate the effectiveness of this approach, nitrite and glucose were individually measured; detection limits as low as 0.015 ± 0.004 mM and 0.022 ± 0.006 mM were reported for nitrite and glucose, respectively. Multiplexed analysis of both analytes was also carried out with respective detection limits of 0.048 ± 0.005 mM and 0.025 ± 0.006 mM for nitrite and glucose. The application of the method was demonstrated by measuring nitrite and glucose in spiked artificial urine samples and satisfied recovery results were obtained.Keywords:paper-based microfluidic devices;colorimetry;adhesive tape;nitrite;glucose

A simple capillary-based open microfluidic device for size on-demand high-throughput droplet/bubble/microcapsule generation

Liping Mei,Mingliang Jin,Shuting Xie,Zhibin Yan,Xin Wang,Guofu Zhou,Albert van den Berg,Lingling Shui

We report an easily-established capillary-based open microfluidic device (COMD) as a simple and robust method for size on-demand generation of monodisperse droplets of various fluidic materials with controllable volume. A device is set up in which a capillary is positioned with its tip close to a flat surface with a precise gap distance in a container. The continuous phase remains static in the container, and the dispersed phase is pumped through the capillary and forms droplets at the exit of the gap. Monodisperse droplets, bubbles and microcapsules of various fluids with diameters of 10–300 μm (picoliter to nanoliter) and generation frequency of 1–1000 Hz are obtained by controlling the gap distance in the range of 5–500 μm. The droplet formation is caused by capillarity-induced narrowing of the dispersed phase at the capillary exit, with droplet size being determined by the gap volume and fluid flow. We find that, at low flow rate, using the same COMD, the generated droplet size is constant, being determined by the gap size; however, at higher flow rate, droplet size increases with the flow rate. Droplet types can be managed by fluids and surface modification of the capillary and bottom surfaces. High throughput droplet generation is achieved by in-parallel integration of multiple capillaries in one device. Such a COMD is simple and easy-to-build without complex microfabrication requirements; however, it is highly robust, flexible and easy-to-operate for size on-demand droplet generation. It offers an opportunity for common laboratories to perform droplet-based assays, and has high potential for high throughput industrial emulsification applications as well.

Measurement of Carcinoembryonic Antigen in Clinical Serum Samples Using a Centrifugal Microfluidic Device

Zhigang Gao,Zongzheng Chen,Jiu Deng,Xiaorui Li,Yueyang Qu,Lingling Xu,Yong Luo,Yao Lu,Tingjiao Liu,Weijie Zhao,Bingcheng Lin

Carcinoembryonic antigen (CEA) is a broad-spectrum tumor marker used in clinical applications. The primarily clinical method for measuring CEA is based on chemiluminescence in serum during enzyme-linked immunosorbent assays (ELISA) in 96-well plates. However, this multi-step process requires large and expensive instruments, and takes a long time. In this study, a high-throughput centrifugal microfluidic device was developed for detecting CEA in serum without the need for cumbersome washing steps normally used in immunoreactions. This centrifugal microdevice contains 14 identical pencil-like units, and the CEA molecules are separated from the bulk serum for subsequent immunofluorescence detection using density gradient centrifugation in each unit simultaneously. To determine the optimal conditions for CEA detection in serum, the effects of the density of the medium, rotation speed, and spin duration were investigated. The measured values from 34 clinical serum samples using this high-throughput centrifugal microfluidic device showed good agreement with the known values (average relative error = 9.22%). These results indicate that the high-throughput centrifugal microfluidic device could provide an alternative approach for replacing the classical method for CEA detection in clinical serum samples. Keywords:centrifugal microfluidic device;CEA detection;density medium;fluorescent chemiluminescence

A PDMS–Agar Hybrid Microfluidic Device for the Investigation of Chemical–Mechanical Associative Learning Behavior of C. elegans

Jinchi Zhu,Yu Wang,Shuting Tang,Huiying Su,Xixian Wang,Wei Du,Yun Wang,Bi-Feng Liu

Associative learning is a critical survival trait that promotes behavioral plasticity in response to changing environments. Chemosensation and mechanosensation are important sensory modalities that enable animals to gather information about their internal state and external environment. However, there is a limited amount of research on these two modalities. In this paper, a novel PDMS–agar hybrid microfluidic device is proposed for training and analyzing chemical–mechanical associative learning behavior in the nematodeCaenorhabditis elegans. The microfluidic device consisted of a bottom agar gel layer and an upper PDMS layer. A chemical concentration gradient was generated on the agar gel layer, and the PDMS layer served to mimic mechanical stimuli. Based on this platform,C. eleganscan perform chemical–mechanical associative learning behavior after training. Our findings indicated that the aversive component of training is the primary driver of the observed associative learning behavior. In addition, the results indicated that the neurotransmitter octopamine is involved in regulating this associative learning behavior via the SER-6 receptor. Thus, the microfluidic device provides a highly efficient platform for studying the associative learning behavior ofC. elegans, and it may be applied in mutant screening and drug testing. Keywords:associative learning behavior;microfluidic device;Caenorhabditis elegans;octopamine

Numerical Simulation and Experimental Validation of Liquid Metal Droplet Formation in a Co-Flowing Capillary Microfluidic Device

Qingming Hu,Tianyi Jiang,Hongyuan Jiang

A two-phase flow axisymmetric numerical model was proposed to understand liquid metal droplet formation in a co-flowing capillary microfluidics device based on a phase field model. The droplet detachment processes were observed in the experiment and are in good agreement with the simulation method. The effects of the viscosities and flowrates of the continuous phase fluid, interfacial tension as well as the wetting property of the metallic needle against the bulk liquid metal on the droplet formation and production rate were numerically investigated. It was found that the droplet diameter decreased with the increment of the viscosities and flowrates of the outer phase carrier fluid. The dispersed phase fluid with high interfacial tension tended to prolong the time for equilibrium between the viscous drag force and interfacial tension on the liquid–liquid fluid surface, delaying the droplet to be pinched off from the capillary orifice and causing large droplet diameter. Finally, the wetting performance of the metallic needle against the liquid metal was explored. The result indicate that the droplet diameter became less dependent on the contact angle while the size distribution of the liquid metal droplet was affected by their wetting performance. A more hydrophilic wetting performance were expected to prepare liquid metal droplet with more monodispersity. The numerical model and simulation results provide the feasibility of predicting the droplet formation with a high surface tension in a glass capillary microfluidic device.

Study of the mechanism of embolism removal in xylem vessels by using microfluidic devices

Lihua Guo,Yuanchang Liu,Li Liu,Penghe Yin,Chong Liu,Jingmin Li

Determining the mechanism that effects embolism repair in the xylem vessels of plants is of great significance in predicting plant distribution and the screening of drought-resistant plants. However, the mechanism of perforation plates of xylem vessels in the acceleration of embolism repair is still not clear by using conventional methods of anatomy and visualization technology. Microfluidic devices have shown their ability to simulate physiological environments and conduct quantitative experiments. This work proposes a biomimetic microfluidic device to study the mechanism of perforation plates of xylem vessels in the acceleration of embolism repair. The results proffered that the perforation plates increase the rate of embolism removal by increasing the pressure differential through the vessel, and the rate of embolism removal is related to the structural parameters of the perforation plate. A combination of the perforation size, the vessel diameter and the perforation plate angle can be optimised to generate higher pressure differentials, which can accelerate the process of embolism repair. This work provides a new method for studying the mechanism of microstructures of natural plants. Furthermore, the mechanism that perforation plates accelerate embolism repair was applied to an electrochemical flow sensor for online determination of heavy metal ions. Test results of this application indicate that the mechanism can be applied in the engineering field to solve the problems of reduced sensitivity of devices, inaccuracy of analysis results and poor reaction performance caused by bubbles that are generated or introduced easily in microdevices, which paves the way for applying the theory to engineering.

An Integrated Electrochemiluminescence Microfluidic Device For Detecting Ultra-low Concentration of Ceftriaxone

Xiansu Liu,Hanwen Ren,Biao Wang,DongDong Zhao,Haolin Xiao,Qunfeng Tang,Hongli Huang,Feijun Zhao,Zhencheng Chen

Ceftriaxone sodium, a widely used third-generation cephalosporin antibiotic, poses significant health risks when residues accumulate in clinical or environmental settings. However, conventional detection techniques, including high-performance liquid chromatography and capillary electrophoresis, are limited by complex procedures, poor sensitivity, and a lack of real-time capability. To address these challenges, we report a sensitivity-enhanced electrochemiluminescence (ECL) microfluidic device for the rapid and label-free detection of ultra-low concentrations of ceftriaxone. Au 25 (Capt) 18 gold nanoclusters were served as a novel ECL luminophore to boost detection sensitivity, while a 3D-printed circulating microfluidic channel was designed to enrich ceftriaxone via inertial centrifugation. The device achieved a wide detection range of 4–400 μg/mL, a low detection limit of 0.522 μg/mL, and an average recovery of 99.1% in real sample tests. This integrated platform offers a promising strategy for simple, sensitive, and on-site monitoring of ceftriaxone contamination.

Synthesis of ZIF-8/PVA microspheres with the assistance of a microfluidic device and their controlled drug release properties

Changfeng Zeng,Jie Li,Jie Gong,Lixiong Zhang,Liang Yu

ZIF-8/PVA microspheres with an even size were synthesized with the assistance of a simple microfluidic device. The ZIF-8/PVA microdroplets were first generated in a simple co-flow microfluidic device using a PVA aqueous solution dispersed with ZIF-8 nanoparticles as the dispersed phase and fatty acid methyl ester (FAME) as the continuous phase. Subsequently, the ZIF-8/PVA microdroplets were further extracted to form ZIF-8/PVA microspheres. The influence of the type of continuous phase on the sphericity of microspheres was investigated and the effect of ZIF-8 content was studied. Further studies showed that decreasing the extraction rate and using additives of NaCl in the dispersed phase could improve the sphericity of the ZIF-8/PVA microspheres. Finally, the synthesized ZIF-8/PVA microspheres were used to study the loading and release of tetracycline. The results showed that the loading of ZIF-8/PVA microspheres was more than 5 times higher than that of pure PVA microspheres. The drug release time of ZIF-8/PVA microspheres was much longer than that of the PVA microspheres and the release rate was significantly affected by the pH of the environment.

Microfluidic one-step and large-scale production of silica and titania nanofluids toward phase-change heat transfer intensification of power electronic devices

Xiong Zhao,Lei Huang,Junsheng Hou,Zihan Ding,Li Ma,Junjie Wu,Dongyu Li,Yilong Yao,Zhenzhen Chen,Nanjing Hao

The nanoparticles with uniform size and excellent stability scattered in fluid (termed as nanofluid) hold the potential to extricate high-performance electronics from the heat crisis. However, the simple, general, and large-scale production of nanofluids with high quality still remains a challenge. Here, we present a delicate design of microfluidic apparatus with extremely high throughput for silica (SiO 2 ) and titania (TiO 2 ) nanofluids synthesis to enhance flow boiling heat transfer. The microfluidic apparatus was composed of a three-microreactor array, and the spiral microchannel in the microreactor can efficiently mix the reagents by the secondary vortex. With the apparatus, 1 L of nanofluids can be generated in 20 min, and the synthesized nanofluids had a narrow size distribution below 100 nm and outstanding long-term stability at room (25 °C) and high (75 °C) temperature. The established heat transfer platform tested the heat transfer performance of SiO 2 /TiO 2 nanofluids that the enhancement increased with the flow rate and decreased with the nanofluid concentration. The SiO 2 nanofluid always performed better than the TiO 2 nanofluid in the same condition. Maximumly, the critical heat flux (CHF) and heat transfer coefficient (HTC) were improved by 77 % and 96 %, respectively, for SiO 2 nanofluid, and they were raised by 44 % and 56 %, respectively, for TiO 2 nanofluid. The underlying mechanism of the difference was analyzed from the deposition on the surface and the bubble’s coalescence. These results not only shed light on the industrial manufacture of various high-performance nanofluids, but also promote the thermal management of high-power electronics.

Wound healing assay on a gravity-driven unidirectional perfusion microfluidic device

Yirong Chen,Yuanyuan Xie,Wen Sun,Ke Ning,Lingke Feng,Xiaoqing Chen,Yan Li,Ling Yu

Cell migration plays a pivotal role in a multitude of physiological and pathological processes, including tissue development, wound healing, immune response, and cancer metastasis. While traditional two-dimensional (2D) cell culture assays, such as the wound healing (scratch) test, have provided valuable insights into cell migration mechanisms, they are unable to replicate the complex in vivo environment. This study introduces a gravity-driven, pump-free microfluidic device to simulate in vivo flow conditions and to investigate the effects of flow rate and flow direction on wound healing assays. The regulation of the flow rate was achieved through adjustments to the tilt angle of the rocking plate. To evaluate the fluid dynamics and cell migration behavior under different flow conditions, numerical simulations and experimental analyses were conducted. The results showed that cell migration and wound healing were markedly influenced by the direction of fluid flow and the magnitude of flow rate. Specifically, compared to the static group, the wound healing rate increased by 76%±6% when the flow direction formed a 45° angle with the wound scratch axis at a -flow rate of 3.0 mL/min, corresponding shear stress of 0.38 dyne/cm 2 . Furthermore, cells exhibited enhanced alignment and migration in the direction of the fluid flow, with the most pronounced effects observed at angles of 45° and 90° between the scratch pattern and the flow direction. Analysis of the cell numbers within the wound healing area and measurements of individual cell area revealed that while the flow conditions did not enhance cell movement, they promoted cell spreading, making the collective wound healing region more pronounced. This gravity-driven, pump-free microfluidic device provides a cost-effective and physiologically relevant platform for studying cell migration, with potential applications in cancer research, wound healing, and developmental biology.
