Amino acid coating enables micromotor operation in physiological conditions

Jia Sun,Yusen Ding,Yicheng Ye,Fei Wang,Hao Tian,Jiamiao Jiang,Huaan Li,Junbin Gao,Haixin Tan,Fei Peng,Jinyao Tang,Yingfeng Tu

Physiological environment with high ionic strength will quench the propulsion of micro/nanomotors (MNMs) by suppressing electric double layers, especially for those motors based on electrolyte diffusiophoresis and electrophoresis. Herein, we demonstrate an efficient, general, and simple strategy to improve the ion tolerance of light-driven titanium dioxide (TiO2) micromotors with amino acid surface modification. Compared to the bare TiO2 counterpart, L-arginine (Arg)-treated TiO2 micromotors display over 200 times higher ion tolerance, which is mainly attributed to the increased surface conductivity. This simple ion tolerance improvement strategy can also be applicable to other motors driven by self-electrophoresis. As TiO2 is an efficient sonosensitizer, we combined the light-guiding ability with ultrasound to generate reactive oxygen species to effectively induce in situ tumor apoptosis. We envision that this simple amino acid surface modification can not only provide a solution for MNMs to tolerate the ionic environment but also open up opportunities for further biomedical and translational research of MNMs.

Flexible fabrication of lipophilic-hydrophilic micromotors by off-chip photopolymerization of three-phase immiscible flow induced Janus droplet templates

Kailiang Zhang,Yukun Ren,Tianyi Jiang,Hongyuan Jiang

Self-propelled microparticles are promising for lots of applications ranging from analytical detection to water treatment . Herein, we present an effective approach to fabricate lipophilic-hydrophilic micromotors via the photocuring of three-phase immiscible flow induced droplet templates. In the microfluidic system, two immiscible inner fluids, the lipophilic 1, 6-Hexanediol diacrylate (HDDA), and the hydrophilic poly (ethylene glycol) diacrylate (PEGDA), are simultaneously injected into a theta-shaped cylindrical capillary from two separate inlets, and they are emulsified into Janus drops when encountering the outer immiscible silicone oil. Because of the immiscible feature of droplet templates, off-chip photopolymerization strategy has been used, which can significantly decrease the blocking chance of microdevice. And also, the lipophilic-hydrophilic structure of droplets is convenient for the loading of cargos with different characteristics. More importantly, the size and configuration of droplet templates can be flexibly regulated by changing the flow rates of three different phases. Accordingly, multifunctional micromotors can be fabricated by adding different nanoparticles and materials into the HDDA or PEGDA phase first and then photocuring the droplets. Taking the bubble-propelled micromotors for example, we prepare microswimmers by loading Ag, TiO 2 and Fe 3 O 4 nanoparticles into the PEGDA phase. The swimming behaviors of micromotors in H 2 O 2 solution are systematically investigated, finding that the proportion of PEGDA phase and the concentration of H 2 O 2 both positively affect the moving speed. Furthermore, the applicability of motor particles on water treatment is successfully demonstrated by using neutral red solution as the model pollutant. And the micromotors can be recycled using magnets after the catalytic degradation process. Therefore, this micromotor generation technique and this kind of micromotor can be attractive for many applications.

Visible light-regulated BiVO4-based micromotor with biomimetic ‘predator-bait’ behavior

Chen Zongchen,Jiang Jiwei,Wang Xin,Zhang Hui,Song Bo,Dong Bin

Inspired by the swarming behavior of various bacteria in nature, light-driven micromotor with collective behavior has attracted a great deal of attention, which will be a valuable tool to understand the cooperation and communication between bacterial populations and will contribute to the creation of reconfigurable microrobots and the cooperative transportation of large cargoes. However, it is still challenging to modulate the micromotors’ collective behavior based on local interaction and communication. Herein, we develop a light-powered BiVO 4 -based micromotor with self-propulsion and dynamic ‘predator-bait’ collective behavior, which is based on the photocatalytic reaction of BiVO 4 . The BiVO 4 -based micromotor has great potential in the field of organic pollutant degradation with enhanced apparent rate constant (0.076 min −1 ) which originates from the dynamic ‘predator-bait’ behavior.

Precisely controlled and deeply penetrated micro-nano hybrid multifunctional motors with enhanced antibacterial activity against refractory biofilm infections

Haiwei Ji,Haolu Hu,Qu Tang,Xiaoxia Kang,Xiaodi Liu,Lingfeng Zhao,Rongrong Jing,Mingmin Wu,Guo Li,Xiaobo Zhou,Jinxia Liu,Qi Wang,Hui Cong,Li Wu,Yuling Qin

The biofilm resistance of microorganisms has severe economic and environmental implications, especially the contamination of facilities associated with human life, including medical implants, air-conditioning systems, water supply systems, and food-processing equipment, resulting in the prevalence of infectious diseases. Once bacteria form biofilms, their antibiotic resistance can increase by 10–1,000-fold, posing a great challenge to the treatment of related diseases. In order to overcome the contamination of bacterial biofilm, destroying the biofilm’s matrix so as to solve the penetration depth dilemma of antibacterial agents is the most effective way. Here, a magnetically controlled multifunctional micromotor was developed by using H 2 O 2 as the fuel and MnO 2 as the catalyst to treat bacterial biofilm infection. In the presence of H 2 O 2 , the as-prepared motors could be self-propelled by the generated oxygen microbubbles. Thereby, the remotely controlled motors could drill into the EPS of biofilm and disrupt them completely with the help of bubbles. Finally, the generated highly toxic •OH could efficiently kill the unprotected bacteria. This strategy combined the mechanical damage, highly toxic •OH, and precise magnetic guidance in one system, which could effectively eliminate biologically infectious fouling in microchannels within 10 min, possessing a wide range of practical application prospects especially in large scale and complex infection sites.

Magnesium-based micromotors for electrochemical detection of dopamine in blood

Pengshen Jing,Qian Chen,Haifeng Ke,WenHui Xu,Yanhong Ni,Chun Mao,Runzhi Deng

In the fields of neuroscience and medicine, the accurate capture and quantitative analysis of dopamine (DA) levels in the blood are of great clinical significance for the early diagnosis of neurological dysfunctions and other pathological conditions. In this study, we developed a laccase (La) sensor based on micromotor technology to accurately detect DA content in blood. Firstly, the magnesium-based micromotor was used to endow the sensor with autonomous motion performance, which significantly increased the contact frequency between La and DA molecules in blood. Secondly, the micromotor-loaded Fe 3 O 4 /PPY/La magnetic nanoparticles provide an efficient electrical signal transmission interface for La, while the presence of polypyrrole (PPY) prevented electrode passivation caused by La. In addition, we used magnetic to collect Fe 3 O 4 /PPY/La after capture and tested it by differential pulse voltammetry. Finally, thanks to the excellent catalytic performance and high selectivity of La, the effective recognition and detection of DA in blood samples were realized, and an efficient electrochemical detection method was developed. This innovative detection scheme has a wide linear detection range and high sensitivity, which is expected to provide a more accurate diagnostic tool for health problems related to DA abnormalities, so as to better cope with and prevent related diseases.

Novel catalytic micromotor of porous zeolitic imidazolate framework-67 for precise drug delivery

Linlin Wang,Hongli Zhu,Ying Shi,You Ge,Xiaomiao Feng,Ruiqing Liu,Yi Li,Yanwen Ma,Lianhui Wang

Micromotors hold promise as drug carriers for targeted drug delivery owing to the characteristics of self-propulsion and directional navigation. However, several defects still exist, including high cost, short movement life, low drug loading and slow release rate. Herein, a novel catalytic micromotor based on porous zeolitic imidazolate framework-67 (ZIF-67) synthesized by a greatly simplified wet chemical method assisted with ultrasonication is described as an efficient anticancer drug carrier. These porous micromotors display effective autonomous motion in hydrogen peroxide and long durable movement life of up to 90 min. Moreover, the multifunctional micromotor ZIF-67/Fe3O4/DOX exhibits excellent performance in precise drug delivery under external magnetic field with high drug loading capacity of fluorescent anticancer drug DOX up to 682 μg mg−1 owing to its porous nature, high surface area and rapid drug release based on dual stimulus of catalytic reaction and solvent effects. Therefore, these porous ZIF-67-based catalytic micromotors combine the domains of metal–organic frameworks (MOFs) and micomotors, thus developing potential resources for micromotors and holding great potential as label-free and precisely controlled high-quality candidates of drug delivery systems for biomedical applications.

A Light-Driven Micromotor with Complex Motion Behaviors for Controlled Release

Yunyu Sun,Ye Liu,Bo Song,Hui Zhang,Ruomeng Duan,Dafeng Zhang,Bin Dong

Micromotor that exhibits various movement behaviors has attracted more attention in recent years. However, most researchers focus on its motion behavior and few have studied the fluid flow surrounding it. In this work, a novel micromotor with both controlled motion behavior and surrounding fluid flow, which is driven by near-infrared (NIR) light based on the unique Marangoni effect caused by the light irradiation of its constituent, i.e., polypyrrole (PPy), is reported. Interestingly, these motion behaviors and the surrounding fluid flow can be easily switched by adjusting the incident angle of the NIR laser. This unique property may have potentials in different fields. Among others, its application in remotely controlled targeted delivery and release is demonstrated.

Enhanced Propulsion of Urease-Powered Micromotors by Multilayered Assembly of Ureases on Janus Magnetic Microparticles

Ming Luo,Shouli Li,Jieshuo Wan,Chenglin Yang,Beidi Chen,Jianguo Guan

Enzyme-powered micro/nanomotors propelled by biocompatible fuels generally show a weak propulsive force, which greatly limits their applications in complex biological environments. Herein, we have developed a novel and versatile approach to significantly enhance the propulsion of enzyme-powered micromotors by multilayered assembly of enzymes. As an example, multilayers of biotinylated ureases (BU) were asymmetrically immobilized on biotinylated Janus Au/magnetic microparticles (MMPs) with the assistance of streptavidin (SA). When the mass ratio of BU into SA and the amount of BU used in the assembly process are increased, the amount of urease immobilized on the biotinylated Janus Au/MMPs increased monotonously while the migration speed of the micromotor was augmented gradually until a saturated value. The as-optimized micromotors can be self-propelled with an average speed up to about 21.5 ± 0.8 μm/s at physiological urea concentrations (10 mM), which is five times faster than that of the monolayered counterparts and two times faster than that of the previously reported values. Owing to the enhanced thrust, the micromotors can move in liquids with viscosities similar to that of blood. In addition, with the inherent magnetic property of MMPs, the micromotors can exhibit fast magnetic separation and controllable motion direction by external magnetic fields. Our results provide a new pathway for designing high-efficient enzyme-powered micro/nanomotors and thereby promote their biomedical applications.

Micromotor-assisted fluorescence detection of Hg2+ with bio-inspired AO–Mn2O3/γ-AlO(OH)

Tingting Li,Yangsai Lyu,Jia Li,Chengyan Wang,Ningning Xing,Jie Yang,Min Zuo

Micromotors have held great promise for environmental treatment due to their unique self-propelled movement that can significantly accelerate the chemical process in solution. Herein, we have developed a new AO–Mn2O3/γ-AlO(OH) fluorescent micromotor using renewable and uniquely hollow kapok fibers as biological templates. The AO–Mn2O3/γ-AlO(OH) micromotor was powered by the asymmetric decomposition of hydrogen peroxide fuel catalyzed by Mn2O3, reaching a maximum speed of 139.6 μm s−1 in 3% H2O2 solution. The micromotor revealed a markedly enhanced fluorescence effect compared to both pure AO solution and non-micromotor counterparts and exhibited excellent selectivity and sensitivity for Hg2+ detection. The detection range and limit for Hg2+ were 5 × 10−8–5 × 10−5 M and 2.25 × 10−8 M, respectively. The enhanced Hg2+ sensing benefited from the synergy of enhanced fluorescence via the inhibition of the dimerization of AO, fluorescence quenching induced by charge transfer and the enhanced contact between the micromotor and Hg2+ ions resulted from the self-propelled motion. This micromotor-assisted fluorescence detection idea provided a new strategy for the development of sensors with high sensitivity.

Stomatocyte structural color-barcode micromotors for multiplex assays

Cai Lijun,Wang Huan,Yu Yunru,Bian Feika,Wang Yu,Shi Keqing,Ye Fangfu,Zhao Yuanjin

Artificial micromotors have a demonstrated value in the biomedical area. Attempts to develop this technology tend to impart micromotors with novel functions to improve the values. Herein, we present novel structural color-barcode micromotors for the multiplex assays. We found that, by rapidly extracting solvent and assembling monodispersed nanoparticles in droplets, it could form stomatocyte colloidal crystal clusters, which not only showed striking structural colors and characteristic reflection peaks due to their ordered nanoparticles arrangement, but also provided effective cavities for the integration of functional elements. Thus, the micromotors with catalysts or magnetic elements in their cavities, as well as with the corresponding structural color coding, could be achieved by using the platinum and ferric oxide dispersed pre-gel to fill and duplicate the stomatocyte colloidal crystal clusters. We have demonstrated that the self-movement of these structural color-barcode micromotors could efficiently accelerate the mixing speed of the detection sample and greatly increase the probe–target interactions towards faster and more sensitive single or multiplex detection, and the magnetism of these barcode micromotors enables the flexible collection of the micromotors, which could facilitate the detection processes. These features make the stomatocyte structural color-barcode micromotors ideal for biomedical applications.

Chemotactic Zn micromotor for treatment of high blood ammonia-associated hepatic encephalopathy

Feng Ye,Gao Chao,Peng Xiuyun,Chen Bin,Ding Miaomiao,Du Dailing,Rong Jinghui,Lv Qi,Wilson Daniela A.,Tu Yingfeng,Peng Fei

Hepatic fibrosis involves hepatocyte damage, causing blood ammonia accumulation, which exacerbates liver pathology and crosses the blood-brain barrier, inducing hepatic encephalopathy. It is meaningful to construct a therapeutic platform for targeted ammonia clearance. In this work, a biocompatible water-powered Zn micromotor is constructed as an ammonia chemotaxis platform, which can be actuated by the water splitting reaction and the self-generated Zn 2+ gradient. It can propel towards NH 3 ·H 2 O source through the formation of complex ions [Zn(NH 3 ) 1 ](OH) + and [Zn(NH 3 ) 2 ](OH) + , representing a generalizable chemotaxis strategy via coordination reaction. In vivo, biomimetic collective behavior allows precise navigation and reduction of the intrahepatic ammonia level, reshaping the pathological microenvironment. This mechanism, operating in a green, zero-waste manner, facilitates integration of these micromotors into the domain of biological regulation. Such environment environment-adaptive platform is favorable for targeted treatment of hepatic fibrosis and hepatic encephalopathy caused by hyperammonemia, which is expected to provide inspiration for future personalized and precision medicine.

Dual-responsive micromotors pill for targeted retention in intestines in vivo

Zitong An,Enguang Lin,Zhiguang Wu,Yongming Kang

Recent advances in synthetic micro/nanomotors in numerous biofluids have garnered widespread hobby because of their capability biomedical programs. However, present micro/nanomotor systems for shipping inside the gastrointestinal (GI) tract are inefficient in propulsion. Here, we present a micromotor-tableted tablet that achieves lively retention in the GI tract in vivo. The drug-loaded magnesium-based totally absolutely Janus micromotors, enveloped in an enteric polymer-protected magnetic tablet, stay sturdy within the stomach. The movement of the micromotor tablets (MPs) is facilitated by means of the use of an outside low-frequency gradient magnetic difficulty, directing them to focused regions inside the intestines. A excessive-frequency alternating magnetic concern induces water-powered propulsion of the micromotors via the magnetocaloric effect, attaining lively retention in GI tissue. The integration of this newly evolved MP tool lets in energetic retention of micromotors in vivo, promising more suitable drug transport for GI treatment.
