Development of a BDDE-crosslinked hyaluronic acid based microneedles patch as a dermal filler for anti-ageing treatment

Jia Nan Zhang,Bo Zhi Chen,Mohammad Ashfaq,Xiao Peng Zhang,Xin Dong Guo

Hyaluronic acid (HA) has many applications in human medicine and cosmetic industry. This study describes the fabrication and evaluation of cross-linked HA (cHA) based microneedles (HA-cHA-MNs) for anti-ageing treatment. The results suggested that the increasing content of cHA gel particles decrease the mechanical properties of MNs. The in-vitro degradation profile and in-vivo epidermal expansion in mice were also carried out. The data suggested that the proportion of cHA particles decreased the rate of MNs degradation. The prepared HA-cHA-MNs have prolonged effectiveness with high swelling retention time or epidermal expansion time in mice up to 6   days or more.

HAMA/GelMA-based hydrogel microneedle patch via DLP 3D bioprinting for high-efficiency ISF extraction

Lujie Zhang,Qinghua Song,Peijing Yang,Zhanqiang Liu

Microneedle (MN) technology is a promising minimally invasive tool for interstitial skin fluid (ISF) extraction. Hydrogel MN patches enable efficient ISF extraction. However, the traditional polydimethylsiloxane (PDMS) photopolymerization method has limitations in morphology control, accuracy, and susceptibility to process parameter interference, affecting MN strength and functional consistency. To address these challenges, we propose a composite hydrogel MN patch based on Digital Light Processing (DLP) 3D bioprinting of hyaluronic acid methacryloyl (HAMA)/gelatin methacryloyl (GelMA) for high-efficiency ISF transdermal extraction. The structure and performance of the MNs were improved by optimizing the bio-ink ratio and printing parameters. Adjusting the ratio of GelMA and HAMA improved the cross-linking structure, optimizing the mechanical properties and swelling performance of the composite hydrogel. Optimizing the light intensity and exposure time ensured consistent MN morphology, resulting in well-defined and evenly distributed needle tips. Controlled UV exposure avoided defects such as uneven cross-linking or brittleness. The results showed that when the HAMA-to-GelMA ratio was 8:2, the light intensity was 9 mW/cm 2 , and the exposure time was 6 s, the MN patches demonstrated excellent microstructures and mechanical properties, with each MN enduring over 1.28 N and achieving a swelling rate above 1200 %. In vitro experiments verified effective penetration and ISF extraction, obtaining 8.5 mg of PBS in 1 min and successfully quantifying glucose concentration. In summary, this study establishes a robust MN patch with superior swelling performance and enhanced ISF extraction efficiency and provides a promising platform for minimally invasive diagnosis and real-time health monitoring.

Double-layered microneedle patch for melanoma chemo/photothermal co‑therapy and wound healing

Yisheng Hu,Ya Wang,Yuqing Cai,Xiaodong Xu,Liping Zhao,Jing Zhang

Melanoma is a leading cause of cancer-related deaths. Surgical excision, the standard treatment, often results in incomplete removal and recurrence. A double-layered microneedle patch was developed for chemo-photothermal treatment of melanoma. The patch, based on zwitterionic poly(sulfobetaine methacrylate) (SBMA), includes a slow-degrading hair particles/curcumin (HMP/Cur) layer and a fast-dissolving ZIF-8/doxorubicin/ Panax notoginseng polysaccharide (ZIF-8@DOX/PNPS) layer, with an asiaticoside/ HMP (AS-HMP) substrate. Prepared via a three-step casting process, the microneedles exhibit excellent photothermal properties and tumor synergism under near-infrared (NIR) light. Upon skin insertion, the fast-dissolving layer releases ZIF-8@DOX within 5 min, which then releases DOX in the tumor's acidic environment. The HMP/Cur layer heats to 56.52 °C within 60 s under NIR, aiding in tumor elimination. In addition, the AS-HMP layer covers the wound and promotes tissue regeneration and collagen deposition, enhancing skin regeneration. The double-layered microneedles platform can simultaneously eliminate the tumor and accelerate wounding healing, making it a promising for clinical melanoma therapy.

Biodegradable multifunctional hyaluronic acid hydrogel microneedle band-aids for accelerating skin wound healing

Zhao Erman,Tang Xiuling,Zhao Minggao,Yang Le

Wound healing for various diseases and wounds such as diabetes and burns remains a major biomedical challenge. Conventional monotherapy is ineffective, and the efficacy of drug delivery is limited by the depth of drug penetration. In this study, we develop a novel, multifunctional, dissolvable hyaluronic acid (HA) microneedle patch (MN-LTT). Microneedling is biocompatible and delivers the drug in a painless and non-invasive manner. Lidocaine and thrombin are mixed with HA hydrogel and loaded onto the needle tips of the MN-LTT, which facilitates wound repair by continuously delivering the drug deep into the dermis for rapid analgesia and hemostasis. In addition, the backing layer of the MN-LTT is composed of tetracycline hydrochloride and HA hydrogel, and its excellent antimicrobial properties further accelerate wound healing. In a mouse full-thickness skin wound model, MN-LTT accelerated cell proliferation and granulation tissue growth, reduced inflammatory-factor levels, and restored collagen deposition, resulting in complete wound healing within seven days. Thus, the proposed microneedle delivery system achieved rapid hemostatic, analgesic, and bactericidal effects, providing a safer and more effective strategy for wound healing. These features make the multifunctional HA microneedle patch potentially valuable for clinical applications. Graphical This self-assembled multifunctional microneedle (MN-LTT) can deliver drugs into the wound tissue in a painless and minimally invasive manner, quickly achieving the functions of hemostasis, analgesia, and antibacterial effects to jointly promote wound healing. In addition, the rapid degradation of MN-LTT can avoid secondary damage to the wound during dressing changes. This combined therapy is a potential strategy for achieving high-quality wound healing

An integrated colorimetric biosensing platform containing microneedle patches and aptasensor for histamine monitoring in seafood

Wenjing Wang,Rui Feng,Kaiyue Wei,Jiashuo Xu,Wenhui Dong,Jiao Li,Jianan Sun,Sai Wang,Xiangzhao Mao

Excessive histamine in spoiled seafood poses considerable health hazards to consumers, yet its detection is challenging due to complicated sample preparation and detection methodologies. Herein, an integrated colorimetric platform containing Poly (vinyl alcohol) (PVA)/ hyaluronic acid (HA) microneedle patchs-assisted extraction and aptasensor-based detection was reported. The developed PVA/HA microneedle patches facilitated on-site histamine extraction from seafood through a two-minute press-and-peel procedure. To enhance detection efficacy, strategies for generating high-affinity aptamers with specific terminal-fixed structures and constructing AuNPs@FeP-chitosan oligosaccharide (COS) nanozyme boosting catalytic efficiency were proposed. Utilizing the aptamer HIS3-T2 in conjunction with the nanozyme, a colorimetric aptasensor was developed. Integrated with the patches, the aptasensor achieved high sensitivity and selectivity, detecting histamine within a range of 2~800   nM with a limit of detection (LOD) of 1.89   nM. Validated on real-world salmon and shrimp samples, this integrated system promises rapid and accurate histamine monitoring, offering great reference for similar applications in food quality control.

Glutamic acid-loaded separable microneedle composite for long-acting hair regeneration treatment

Mbituyimana Bricard,Fu Lina,Wang Hao,Qi Fuyu,Jiang Yuchi,Meng Fanbo,Wu Min,Shi Zhijun,Yang Guang

Androgenetic alopecia (AGA) is the most common type of hair loss frequently observed in clinics. Right now, there are no effective treatments for AGA. Here, we developed a biodegradable microneedle (MN) patch made of polylactic-co-glycolic acid (PLGA) loaded with glutamic acid (GA) that improves sustainable drug release and effective hair regeneration treatment. Once the patch has penetrated the skin, the GA-loaded MNs (GA-MNs) are quickly separated from their base and enter the skin. These MNs then serve as drug storage tanks inside the skin, releasing the therapeutics gradually for over 4 weeks. The sustained release of GA from long-acting biodegradable needles could lead to endocytosis by dermal papilla cells (DPCs), promoting the proliferation of cells. Compared with topical minoxidil, which requires daily treatment, animal studies have shown that GA-MNs can increase hair regeneration more effectively with lower dose frequency; the number of hair follicles in the GA-MNs group reached 83.68 ± 4.52%, which was significantly higher than that of the minoxidil group (56.93 ± 3.21%). This transdermal technology of biodegradable MNs shows considerable promise in clinical applications and offers a straightforward, safe, and effective therapy approach for clinical hair regeneration treatment.

Claw-inspired microneedle patches with liquid metal encapsulation for accelerating incisional wound healing

Xiaoxuan Zhang,Guopu Chen,Lingyu Sun,Fangfu Ye,Xian Shen,Yuanjin Zhao

Microneedles are providing an emerging strategy for promoting wound healing. Attempts to develop this technology tend to impart them with additional functions. Herein, inspired by the claws of eagles, we present a liquid metal (LM)-encapsulated microneedle patch by employing a top-down multiple-mold-guided photolithography method for incisional wound healing. The microneedle patch is composed of two tilted hydrogel parts and ventilatory gauze as the connection. Such microneedle tips form a claw-like clamping structure, enabling the patch to stick to the skin, fix the wounded area and avoid secondary dehiscence. As the embedded W-patterned LM connects the tips of each microneedle part and links the microneedle part to an external power source, the microneedle patch can provide a stable spatial electric field around the wounds, guide cell migration and accelerate wound healing. It is demonstrated that the microneedle patch can perform well in treating wounded SD rats. These results indicate that the present microneedle patch is practically valuable and may find various applications in wound-healing related biomedical fields.

Development of a novel hyaluronic acid/alginate/RANKL degradable microneedle patch for accelerating bone remodeling and orthodontic tooth movement through promoting osteoclastogenesis

Yue Shan,Yu Jin,Xiaoqi Zhang,Yufei Tang,Wenli Lai,Jinfeng Liao,Mengjie Wu,Hu Long

The prolonged duration of orthodontic treatment remains a significant concern for both orthodontists and patients. In this study, we developed a degradable microneedle (MN) patch composed of hyaluronic acid (HA) and sodium alginate (SA) for the delivery of receptor activator of nuclear factor-kappa B ligand (RANKL) to accelerate tooth movement. This MN patch that was crosslinked by calcium chloride (CaCl 2 ) exhibited adequate mechanical properties and favorable in vitro mucosal insertion ability. Moreover, the MN patch can achieve sustained release of RANKL and maintain the biological stability of RANKL protein after one month of storage at −20 °C, 4 °C, or 37 °C. In vitro experiments using RAW 264.7 cells indicated that the HA/SA/RANKL MN possessed excellent biocompatibility and could effectively induce osteoclast differentiation. In vivo application of the HA/SA/RANKL MN in rat models showed a remarkable effect in promoting osteoclast formation and accelerating tooth movement. These findings suggest that the degradable HA/SA/RANKL MN holds significant potential for enhancing tooth movement efficiency.

Photothermal Nanozyme-Encapsulating Microneedles for Synergistic Treatment of Infected Wounds

Zesheng Chen,Lin Zhang,Ye Yang,Wenwen Cheng,Lingfeng Tu,Zijian Wang,Yun Chen,Weikang Hu

Photothermal therapy (PTT) is widely used in wound healing because it produces heat under near-infrared irradiation, eliminating bacterial infections. However, excessive production of reactive oxygen species (ROS) occurs during PTT, which causes oxidative stress damage. In this study, a nanozyme, PDA-MnO 2 (PM), is synthesized and encapsulated in the needle tip layer of a bi-layer GelMA microneedle patch (PbMP), which has excellent mechanical properties and can easily penetrate the skin. In a study involving an SD rat wound model infected with S. aureus , the PbMP significantly accelerated the healing process of bacterially challenged wounds, resulting in better blood vessel growth, cell proliferation, and collagen biosynthesis than other wound dressings. The experiment shows that the PM nanoparticles in PbMP improve photothermal conversion efficiency and activate SOD and CAT, resulting in an efficient cascade enzyme reaction system to eliminate ROS during the PTT process. This study describes a highly effective nanozyme hydrogel microneedle platform with photothermal synergistic antibacterial effects on wound healing.

Microneedle Patch for Transdermal Sequential Delivery of KGF-2 and aFGF to Enhance Burn Wound Therapy

Huacheng He,Wen Huang,Shihui Zhang,Jie Li,Jian Zhang,Bingxin Li,Jie Xu,Yuting Luo,Huiling Shi,Yue Li,Jian Xiao,Odinaka Cassandra Ezekiel,Xiaokun Li,Jiang Wu

Severe burn wounds usually destroy key cells’ functions of the skin resulting in delayed re-epithelization and wound regeneration. Promoting key cells’ activities is crucial for burn wound repair. It is well known that keratinocyte growth factor-2 (KGF-2) participates in the proliferation and morphogenesis of epithelial cells while acidic fibroblast growth factor (aFGF) is a key mediator for fibroblast and endothelial cell growth and differentiation. However, thick eschar and the harsh environment of a burn wound often decrease the delivery efficiency of fibroblast growth factor (FGF) to the wound site. Therefore, herein a novel microneedle patch for sequential transdermal delivery of KGF-2 and aFGF is fabricated to enhance burn wound therapy. aFGF is first loaded in the nanoparticle (NP aFGF ) and then encapsulated NP aFGF with KGF-2 in the microneedle patch (KGF-2/NP aFGF @MN). The result shows that KGF-2/NP aFGF @MN can successfully get across the eschar and sequentially release KGF-2 and aFGF. Additional data demonstrated that KGF-2/NP aFGF @MN achieved a quicker wound closure rate with reduced necrotic tissues, faster re-epithelialization, enhanced collagen deposition, and increased neo-vascularization. Further evidence suggests that improved wound healing is regulated by significantly elevated expressions of hypoxia-inducible factor-1 alpha (HIF-1ɑ) and heat shock protein 90 (Hsp90) in burn wounds. All these data proved that KGF-2/NP aFGF @MN is an effective treatment for wound healing of burns.

Iron (III) cross-linked alginate microneedle patch for xerosis treatment based on controllable photodegradation

Jiarong Xu,Qi Zeng,Xiuli Chen,Honglian Guo,Yusheng Gong,Rengui Xu,Peng Cao,Chunyu Wei,Fei Xiao,Dawei Jiang,Wei Chen

Dry skin, known as the xerosis, would induce deep cracks and fissures, leading to bleeding and increased risk of allergens and pathogens penetrating the skin, which may trigger allergic and irritant contact dermatitis as well as the severe infection. High-frequent administration of traditional emulsion or moisturizer often results in significantly reduced patient compliance/adherence and limited therapeutic efficacy. Microneedle (MN) patches have been extensively investigated due to their deep penetration through the epidermis for sustained release in a patient-friendly manner. In this study, an iron (III) cross-linked alginate microneedle patch (ICAMP) has been developed for long-term treatment of xerosis by delivering the ammonium lactate for local management. In order to achieve on-demand treatment, the microneedles were cross-linked with iron (III) which could be photodegraded into iron (II) by exposure to light at 405 nm, triggering microneedles to dissolve in the interstitial fluid for payload release. In such a way, the drug could be released in a well-tunable fashion by using a light-based controller, suggesting a regulated manner to treat xerosis especially under intense light radiation.

Synthesis and characterization of ionic liquid microneedle patches with different carbon chain lengths for antibacterial application

Xuan Chao,Chu Zhang,Xiaodan Li,Hongqian Lv,Guixia Ling,Peng Zhang

The prevention of bacterial infection is becoming more and more important in clinical medicine. Ionic liquids (ILs) can change the structure in an almost infinite way to actively antagonize pathogenic microorganism strains. The current biological materials of skin dressings inevitably have the shortcomings of single drug delivery form and low drug loading, which limit the practical application of skin dressings. Therefore, it is particularly important to develop drug delivery forms that can meet different conditions. The addition of ILs into crosslinked microneedle (MN) patches is a novel design scheme of MNs. The broad-spectrum antibacterial properties of imidazolium salt ILs ensure that the wound skin is sterile after the use of MN patches on the skin to open channels for drug delivery. In this study, imidazole IL monomers with different carbon chain lengths and the corresponding IL-MN patches were designed and synthesized. By comparing the antibacterial properties of four imidazolium salt IL monomers with different carbon chain lengths and the corresponding ionic liquid microneedle patches, we found that the antibacterial properties of IL monomers and IL-MN patches increased with the increase of substituent carbon chain lengths. Imidazole IL monomers have excellent antibacterial properties, which may be caused by the electrostatic interaction between the cations in the IL monomers and the anions in the bacterial membrane and the hydrophilic and hydrophobic interactions between the IL monomers.
