Microneedle arrays (MAs) are among the most promising transdermal drug delivery systems in the last decades due to its minimally invasive nature, convenient operation, and first-pass-metabolism avoidance. However, most MA fabrication methods are difficult to operate, need multiple steps, or require expensive equipment. A novel magnetorheological drawing lithography approach was proposed to rapidly fabricate a flexible microneedle array (FMA) for transdermal drug delivery. A 3D structural liquid MA was drawn in one step from the droplets of curable magnetorheological fluid and maintained its shape under an external magnetic field. The liquid MA was subsequently solidified and sputter-coated with the Ti/Au film to fabricate FMA. FMA morphology, mechanical properties, and transdermal drug delivery performance in vitro were experimentally investigated. FMA consisted of a 5 × 5 cone-shaped microneedle array on a PET flexible substrate. FMA exhibited good strength and excellent penetration performance. It could easily penetrate into skin without breakage, creating microchannels for the promotion of skin permeability. Drugs could be well permeated and diffused in the skin along the microchannels created by FMA. Finally, a dissolvable microneedle array (DMA) was also fabricated by a micromolding technique using FMA as a master template. The DMA exhibited good dissolvable and permeable performance in the agarose block.

We developed a modified micromolding method for the mass production of a novel tip-hollow microneedle array (MA). The tip-hollow MA was fabricated by tuning of the vacuum degree at −80 kPa for 60 s during the micromolding process. Subsequently, a tip-dissolvable MA encapsulated with drugs in the microcraters was fabricated from tip-hollow MA using repeated dipping and the freeze-drying process. Both the tip-hollow and tip-dissolvable MAs could easily penetrate in the rabbit skin without breakage, while the tip-hollow MA can just create a shallow loop hole in the skin. The drug-loaded tip-dissolvable MA can rapidly dissolve, releasing and diffusing the drug in the skin. The tip-dissolvable MA exhibited the best drug permeation ability in that the corresponding flux through the punctured skin using tip-dissolvable MA loaded with Rhodamine B is about 1.7- and 5.8-fold of that through the punctured skin using solid MA and the intact skin, respectively. The tip-dissolvable MA loaded with 5 IU insulin was fabricated to in vivo treat the type 1 diabetic SD rats. The tip-dissolvable MA had a good hypoglycemic effect and exhibited longer normoglycemic period in comparison with subcutaneous injection (5 IU). Therefore, our tip-dissolve MA is a promising medical device for transdermal drug delivery.

Microneedle arrays have emerged as an alternative method for transdermal drug delivery. Although micromolding using a centrifugation method is widely used to prepare microneedles in laboratory, few researchers were focused on manufacturing processes capable of facile scale-up. A novel female mold was initially designed in this study, namely double-penetration female mold (DPFM) with the pinpoints covered by waterproof breather membrane which was beneficial to reduce the influence of gas resistance and solution viscosity. In addition, DPFM-based positive-pressure microperfusion technique (PPPT) was proposed for the scale-up fabrication of dissolving microneedle arrays (DMNA). In this method, polymer solution and base solution were poured into the DPFM by pressure difference, followed by drying and demolding. The results of optimal microscopy and SEM revealed that the obtained microneedles were uniformly distributed conical-shaped needles. The skin penetration test showed that DMNA prepared using PPPT were able to penetrate the rat skin with a high penetration rate. To realize the transition of microneedles fabrication from laboratory to industry, an automatic equipment was further designed in this study. Different from micromolding method using centrifugation, the equipment based on PPPT and DPFM has superiorities in the scale-up fabrication of microneedles in a highly effective, controllable, and scalable way.

Timely administration of key medications toward patients with sudden diseases is critical to saving lives. However, slow transport of first-aid therapeutics and the potential absence of trained people for drug usage can lead to severe injuries or even death. Herein, an unmanned aerial vehicle (UAV)-mediated first-aid system for targeted delivery (uFAST) is developed. It allows unattended administration of emergency therapeutics-loaded transdermal microneedle (MN) patches toward patients to relieve symptoms by a contact-triggered microneedle applicator (CTMA). The implementability and safety of the uFAST for first aid is demonstrated in a severe hypoglycemic pig model by automatically delivering a glucagon patch with immediate and bioresponsive dual release modes. This platform technique may facilitate the development of UAV-mediated first-aid treatments for other sudden diseases.

s Vaccines play a crucial role in managing pandemic diseases like SARS and emerging coronaviruses. Efficient and quick detection methods for antigen-specific antibodies could significantly aid in designing vaccines intelligently. Presently, the detection of these antibodies typically involves the collection of blood and subsequent enzyme-linked immunosorbent assay, lateral flow immunoassay, and similar assay techniques. However, blood collection causes patient discomfort and pain, requiring professional handling and often resulting in poor patient compliance. This work is based on the establishment of a rapid vaccine antibody detection technique based on microneedle technology, in which microneedles modified with bio-recognition elements (such as antigens) penetrate the stratum corneum to extract interstitial fluid through capillary action and selectively capture protein biomarkers (such as antibodies) in the ISF in a concentration-dependent manner. The protein biomarkers attached to the microneedles are subsequently identified using an in vitro fluorescence immunoassay. This study merges the specificity inherent in antigen-antibody binding with the porous characteristics of microneedles to swiftly capture and detect antibodies within the skin's interstitial fluid. Compared to conventional testing methods, microneedle technology doesn't necessitate blood collection, rendering the technique non-invasive, minimally uncomfortable, and highly efficient. Moreover, it enables rapid antibody detection, offering a promising and straightforward approach to the efficient detection of various antibodies.

Interest in transdermal delivery methods for stimulating hair regrowth has been increasing recently. The microneedle approach can break the barrier of the stratum corneum through puncture ability and improve drug delivery efficiency. Herein, we report a dissolvable microneedle device for the co-delivery of rapamycin and epigallocatechin gallate nanoparticles that can significantly promote hair regeneration. Compared with the mice without any treatment, our strategy can facilitate hair growth within 7 days. Higher hair shaft growth rate and hair follicle density with inconspicuous inflammation were exhibited in C57BL/6 mice, elucidating its potential for clinical application. Keywords:hair regrowth;microneedles;nanoparticles;rapamycin

As a novel painless and minimally invasive transdermal drug delivery method, microneedles have solved the challenges of microbial infection and tissue necrosis associated with multiple subcutaneous injections in patients with diabetes. However, traditional soluble microneedles cannot switch drug release on and off according to the patient's needs during long-term use, which is one of the most critical elements of diabetes treatment. Herein, an insoluble thermosensitive microneedle (ITMN) that can control the release of insulin by adjusting the temperature, enabling the precise treatment of diabetes is designed. Thermosensitive microneedles are produced by in situ photopolymerization of the temperature-sensitive compound N-isopropylacrylamide with the hydrophilic monomer N-vinylpyrrolidone, which is encapsulated with insulin and bound to a mini-heating membrane. ITMN are demonstrated to have good mechanical strength and temperature sensitivity, can release significantly different insulin doses at different temperatures, and effectively regulate blood glucose in type I diabetic mice. Therefore, the ITMN provides a possibility for intelligent and convenient on-demand drug delivery for patients with diabetes, and when combined with blood glucose testing devices, it has the potential to form an integrated and precise closed-loop treatment for diabetes, which is of great importance in diabetes management.

Gastrointestinal-retentive drug delivery devices represent a novel oral drug delivery approach capable of significantly enhancing patient compliance and improving local drug delivery efficiency compared to conventional oral formulations. However, developing gastrointestinal-retentive drug delivery devices with high retention capability based on compatible material systems presents significant challenges. Here, we introduce silk-based double-sided magnetic microneedles (SDMM) designed to facilitate targeted oral drug delivery with precise local retention in the gastrointestinal tract. The SDMM platform demonstrates robust mechanical properties, site-specific degradation, and compatibility with various capsule geometries, facilitating seamless integration into existing oral delivery systems. In vitro analyses demonstrate region-dependent drug release from the SDMM, supporting its potential for localized delivery of drugs in the gastrointestinal tract. The incorporation of iron oxide nanoparticles (IONPs) confers magnetically responsive targeting, offering a significant advantage over conventional approaches by ensuring precise retention and enhancing the targeted delivery potential under external magnetic fields. Notably, SDMM retains its positions under fluid flow rates up to 550 mL/min (stomach) and 400 mL/min (small intestine), respectively, underscoring its superior gastrointestinal retention. Furthermore, the in vivo retention experiments also showed significantly enhanced retention in the gastrointestinal tract, with the retention time of SDMM reaching almost 15 hours with magnet assistance, representing significantly prolonged retention compared to typical transit times in these regions. These findings establish SDMM as a promising platform for precise, efficient, and patient-friendly oral drug delivery.

Psoriasis is a chronic inflammatory skin condition with high oxidative stress and immune dysregulation. Current therapies are limited by side effects and inefficacy over long-term use, highlighting the need for targeted treatments. This study investigates a reactive oxygen species (ROS)-responsive hydrogel (HP)-dissolving microneedle system, loaded with Momordin Ic, designed for controlled, site-specific release to alleviate psoriasis. We hypothesized that this system could target psoriatic inflammation by reducing pro-inflammatory cytokines and oxidative stress. The Franz diffusion cell assay confirmed that Momordin Ic release was accelerated under elevated ROS conditions, demonstrating the stimuli-responsive nature of the hydrogel microneedles system. An imiquimod (IMQ)-induced psoriasis-like mouse model was used to evaluate the therapeutic efficacy of the Momordin Ic/HP microneedle system. The HP microneedles exhibited controlled Momordin Ic release under elevated ROS, and in vitro studies showed reduced ROS levels and blocked hyperproliferation in HaCaT keratinocytes. In vivo , the microneedle treatment alleviated psoriasis symptoms, reducing erythema, scaling, and epidermal thickness while downregulating inflammatory markers (IL-17A, TNF-α). These findings suggest that Momordin Ic/HP microneedles provide a promising therapeutic approach to treating inflammatory skin diseases like psoriasis by combining anti-inflammatory and ROS-scavenging functions.

Coated microneedles have been paid much attention recently, and several coating strategies have been developed to address the problems during coating process. However, there are still some unresolved issues, such as, precise control requirements, microneedle substrate contamination and high processing temperature. The purpose of this study was to develop a simple and controllable method to make uniform coatings on microneedles at room temperature. This novel method avoids the contamination of microneedle substrate by providing both the adsorption force of thickener and micro-scale coating film produced by a newly design device. Thickeners were screened to enhance the mass of coatings. The parameters that influence the coatings were tested systematically, which made coating process controllable. Finally, three model drugs were coated onto microneedles to prove the method is applicable more broadly. In addition, insertion experiments were carried out to test the drug delivery feasibility of the coated microneedles. In conclusion, this study presents a simple and controllable method to coat microneedles with small molecular chemical drugs or large proteins for rapid skin drug delivery.

Hypertensive emergency is a clinical syndrome characterized by abrupt and substantial elevation of blood pressure in hypertensive patients due to specific triggers, leading to potentially life-threatening consequences. Intravenous injection of sodium nitroprusside is commonly used to rapidly lower blood pressure. However, its specialized nature limits its applicability for first aid or self-help in emergency situations. To address this issue, we have developed a self-powered and controllable microneedle drug delivery system that is easy to use and effective for lowering blood pressure. The system mainly consists of two parts: a piezoelectric thin film self-powered module made of polyvinylidene fluoride and carbon nanotubes, and a drug delivery module composed of polylactic acid-Au microneedles and polylactic acid-Au-polypyrrole microneedles. The results show that different pressures applied to the self-powered module generate different voltages, thereby controlling the drug release efficiency of the microneedles. Compared with conventional active devices, it exhibits a higher drug release rate and effectiveness. Furthermore, the system was successfully utilized to reduce and control blood pressure in spontaneously hypertensive rats. Compared with the injection of sodium nitroprusside, this system can more effectively lower and control blood pressure levels, and its biological safety has been verified. This study presents a controllable and user-friendly rapid blood pressure lowering system suitable for hypertensive emergencies, which has important clinical application prospects.

Hypocrellin A (HA) is an efficient photosensitizer for photodynamic therapy in recent years. In order to improve the delivery of this photosensitizer, dissolving HA microneedles were established. Firstly, dissolving microneedles were prepared from HA by a two-step solvent casting method. Then, the dissolving HA microneedle were characterized by stereomicroscope, laser confocal microscope, scanning electron microscope, differential scanning calorimeter (DSC), X-ray diffractometer (XRD) and fourier infrared spectrometer (FTIR). Finally, the transdermal cumulative penetration experiments were conducted. The results showed that the dissolving HA microneedle has a conical shape with an average height of 650 μm. The mechanical strength endure a force of 44.78 N, resulting in 0.20 N per needle. The in vitro transdermal experiment showed that the dissolving HA microneedle almost completely dissolved in 5 minutes after insertion into the mouse skin. The depth of drug penetration through the skin can reach 601.36 μm. Moreover, the dissolving HA microneedle significantly improved the cumulative transdermal penetration efficiency of the photosensitizer through the skin. Therefore, the present study successfully established a dissolving microneedle of HA, which can quickly dissolve and significantly improve the transdermal delivery efficiency of HA.
