Inoculation of single cells into separate culture chambers is one of the key requirements in single-cell analysis. This paper reports an innovative microfluidic chip integrating two pneumatic microvalves to screen and print single cells onto a well plate. The upper and lower size limits of cells can be dynamically controlled by regulating the deformation of two adjacent microvalves. Numerical simulations were employed to systematically study the influence of membrane dimensions and pressure on the deflection of a valve. A mathematical model was then modified to predict the size of cells captured by a microvalve at various pressures. The membrane deflection was further studied using confocal imaging. The critical pressure trapping beads of various sizes was experimentally determined. These experiments validated the accuracy of both numerical simulations and the mathematical model. Furthermore, single beads and endothelial cells with the desired size range were screened using dual valves and printed onto well plates with 100% efficiency. Viability studies suggested that the screening process had no significant impact on cells. This device enables dynamic regulation of both the lower and the upper size limits of cells for printing. It has significant application potential in inoculating cells with desired sizes for various fields such as clonal expansion, monoclonality development and single-cell genomic studies.

Printing single cells into individual chambers is of critical importance for single-cell analysis using traditional equipment, for instance, single-cell clonal expansion or sequencing. The size of cells can usually be a reflection of their types, functions, and even cell cycle phases. Therefore, printing individual cells within the desired size range is of essential application potential in single-cell analysis. This paper presents a method for the development of a microfluidic chip integrating pneumatic microvalves to print single cells with appropriate size into standard well plates. The reported method provided essential guidelines for the fabrication of multi-layer microfluidic chips, control of the membrane deflection to screen cell size, and printing of single cells. In brief, this paper reports: • the manufacturing of the chip using standard soft lithography; • the protocol to dynamically screen both the lower and the upper size limit of cells passing through the valves by deflection of the valve membrane; • the screening and dispensing of suspended human umbilical vein endothelial cells (HUVECs) into 384-well plates with high viability.

Near-infrared (NIR) responsive hydrogels are of significance in remote control microvalves. However, the development and the application of NIR responsive hydrogels were either limited by high cost or limited by long response time (on the order of minutes) and modest volume change. To address this problem, in this work, a graphene oxide@polyaniline (GO@PANI) hybrid nanocomposite with nanorod array hierarchical structure was fabricated by dilute polymerization. The as-synthesized hierarchical GO@PANI nanosheets exhibited high NIR photothermal conversion efficiency (PTCE) (49%), which was approximately 188% the magnitude of that of GO sheets. Due to its high photothermal conversion efficiency, when GO@PANI was incorporated into temperature-responsive poly(N-isopropylacrylamide) (PNIPA), a highly responsive hydrogel with rapid and significant NIR-induced volume change (over 50%) was obtained. When this material was applied as a remote-controllable microvalve, it exhibited a fast NIR response (∼30 s).

The real-time detection of biomarkers in saliva is of great significance for early non-invasive diagnostic of related diseases. In this work, molecular imprinting technology was used to prepare a lactic acid monolithic column, which could realize specific adsorption and elution of lactic acid, achieving solid phase microextraction (SPME) of lactic acid in samples, and purify the analytes. Lactic acid monolithic column was then embedded into the microfluidic channel, and passive microfluidic structures such as stop valve, retention valve, trigger valve and capillary pump were utilized to construct the SPME self-driven microfluidic chip. A series of complex operations such as lactic acid adsorption, rinse and elution on the monolithic column could be realized automatically without external power. Since the purification of the analytes was achieved by the monolithic column, the low-cost nano copper oxide modified electrode was used to achieve the specific electrochemical analysis of lactic acid. The screen-printed electrode was designed and fabricated, and the integrated electrochemical detection circuit was used to realize the portable analysis of Na + , K + and lactic acid simultaneously. The microfluidic chip, screen-printed electrode and miniaturized detection circuit are integrated into a Point-of-care testing (POCT) device, which realizes the automatically real-time detection of saliva. By adjusting the template molecule in monolithic column polymerization, the application of this physiological fluid POCT device can be extended, and may provide some implications for the development of portable non-invasive health monitoring devices.
