Transferable Self-Assembled Quantum Dot Microarrays for High-Resolution Luminescent Applications

Dacheng Xia,Jianping Li,Qiuyi Zhang,Shuwen Shen,Chenxu Sheng,Chunxiao Cong,Zhi-Jun Qiu,Laigui Hu,Ran Liu

With tunable and exceptional optical properties, quantum dots (QDs) have shown promise as valuable luminescent materials for various visual applications. However, achieving the economical and facile manufacturing of QD patterns still remains a challenge for high-resolution applications. QD arrays with pixel sizes down to micrometers are difficult to achieve with conventional techniques. In this work, a low-cost dewetting-assisted self-assembly approach for patterning QDs was demonstrated using oleophilic/oleophobic patterns on different substrates treated by lithography and surface modification. High-resolution ordered droplet arrays of UV-curable QD ink were formed on prepatterned templates. Resultant large-area QD arrays with a pixel size down to 5 μm exhibited a high resolution of 2540 pixels per inch (PPI). Furthermore, dual-color microarrays with a resolution exceeding 4 kPPI were achieved by transfer-printing red and green QD microdot patterns onto a receiving substrate. High-resolution fluorescent anticounterfeiting labels were then successfully demonstrated, implying an avenue for low-cost high-resolution QD patterns for luminescent applications, such as optical anticounterfeiting, information encryption, and displays.

Development of Sphere-Polymer Brush Hierarchical Nanostructure Substrates for Fabricating Microarrays with High Performance

Xia Liu,Rongrong Tian,Dianjun Liu,Zhenxin Wang

In this work, a sphere-polymer brush hierarchical nanostructure-modified glass slide has been developed for fabricating high-performance microarrays. The substrate consists of a uniform 160 nm silica particle-self-assembled monolayer on a glass slide with a postcoated poly(glycidyl methacrylate) (PGMA) brush layer (termed PGMA@3D(160) substrate), which can provide three-dimensional (3D) polymer brushes containing abundant epoxy groups for directly immobilizing various biomolecules. As a typical example, the interactions of three monosaccharides (4-aminophenyl β-d-galactopyranoside, 4-aminophenyl β-d-glucopyranoside, and 4-aminophenyl α-d-mannopyranoside) with two lectins (biotinylated ricinus communis agglutinin 120 and biotinylated concanavalin A from Canavalia ensiformis) have been assessed by PGMA@3D(160) substrate-based carbohydrate microarrays. The carbohydrate microarrays show good selectivity, strong multivalent interaction, and low limit of detection (LOD) in the picomolar range without any signal amplification. Furthermore, the proposed sphere-polymer brush hierarchical nanostructure substrates can be easily extended to fabricate other types of microarrays for DNA and protein detection. PGMA@3D(160) substrate-based microarrays exhibit higher reaction efficiencies and lower LODs (by at least 1 order of magnitude) in comparison to those of two-dimensional microarrays, which are fabricated on planar epoxy substrates, making it a promising platform for bioanalytical and biomedical applications.

A microdots array-based fluoremetric assay with superwettability profile for simultaneous and separate analysis of iron and copper in red wine

Chenchen Xu,Qingqing Liu,Su Chu,Pan Li,Fengxiang Wang,Yanmei Si,Guojiang Mao,Choufei Wu,Hua Wang

A microdots array-based fluoremetric method with superwettability profile has been developed for the simultaneous and separate detection of Fe 3+ and Cu 2+ ions in red wine samples. A wettable micropores array was initially designed with high density by using polyacrylic acid (PAA) and hexadecyltrimethoxysilane (HDS), followed by the NaOH etching route. Zinc metal organic frameworks (Zn-MOFs) were fabricated as the fluorescent probes to be immobilized into the micropores array to obtain the fluoremetric microdots array platform. It was found that the fluorescence of Zn-MOFs probes could decrease significantly in the presence of Fe 3+ and/or Cu 2+ ions towards their simultaneous analysis. Yet, the specific responses to Fe 3+ ions could be expected if using histidine to chelate Cu 2+ ions. Moreover, the developed Zn-MOFs-based microdots array with superwettability profile can enable the accumulation of targeting ions from the complicated samples without any tedious pre-processing. Also, the cross-contamination of different samples droplets can be largely avoided so as to facilitate the analysis of multiple samples. Subsequently, the feasibility of simultaneous and separate detection of Fe 3+ and Cu 2+ ions in red wine samples was demonstrated. Such a design of microdots array-based detection platform may promise the wide applications in analyzing Fe 3+ and/or Cu 2+ ions in the fields of food safety, environmental monitoring, and medical diseases diagnostics.

A photofabricated honeycomb micropillar array for loss-free trapping of microfluidic droplets and application to digital PCR

Yu He,Zefan Lu,Hongliang Fan,Tao Zhang

Droplet microfluidics is a promising platform for various biological and biomedical applications. Among which, droplet-based digital PCR (ddPCR) is one of the most challenging examples, with practical issues involving possible fusion/fission of droplets during PCR thermocycling and difficulties of indexing them for real-time monitoring. While spatially trapped droplet arrays may be helpful, they currently are either of low trapping density or suffer from high droplet loss. In this paper, we, for the first time, report a photofabricated honeycomb micropillar array (PHMA) for high-density and loss-free droplet trapping. By rationally designing high-aspect-ratio micropillars into a honeycomb configuration, droplets can be captured at a density of 160–250 droplets per mm2 and, more interestingly, without any loss. The PHMA device can be fabricated from several photocurable materials, with one gasproof photopolymer being optimally selected herein to enable the simple design to avoid sample evaporation and tedious surface modification, thereby making the fabrication very convenient. Moreover, by using a photocurable oil as a continuous phase, the trapped droplets can be further immobilized, and thus, become more stable even in PCR thermocycling. With these features, the proposed PHMA has shown promising potential in ddPCR, and is expected to find a wide range of applications in various biological and biomedical research.

DNA microarray fabricated on poly(acrylic acid) brushes-coated porous silicon by in situ rolling circle amplification

Cuie Wang,Xue-Mei Jia,Chuan Jiang,Guang-Nan Zhuang,Qin Yan,Shou-Jun Xiao

Microarrays hold considerable promise in large-scale biology on account of their analytical, massive and parallel nature. In a step toward further enabling such a capability, we describe the application of rolling circle amplification (RCA) for a sensitive and multiplex detection of nucleic acid targets on oligonucleotide-conjugated polymer brushes covalently grown from porous silicon. Both RCA and polymer brushes have been taken to increase the loading quantity of target molecules and thus improve the detection sensitivity without loss of multiplexing. Besides, polymer brushes were employed to protect porous silicon and to provide biologically simulated environments, making the attached biomolecules maintain bioactivity. This approach can reach a detection limit of 0.1 nM target analytes and three orders of magnitude dynamic range of 0.1–100 nM, with a fluorescence scanner. A two-colour DNA microarray was achieved via RCA of two kinds of circular DNA targets on one chip simultaneously. The porous silicon chip-based RCA technique is promising for the multiplex detection of deoxynucleic acids on microarrays.

Development of a sandwiched microarray platform for studying the interactions of antibiotics with Staphylococcus aureus

Xia Liu,Zhen Lei,Dianjun Liu,Zhenxin Wang

It still confronts an outstanding challenge to screen efficient antibacterial drugs from millions of potential antibiotic candidates. In this regard, a sandwiched microarray platform has been developed to culture live bacteria and carry out high-throughput screening antibacterial drugs. The optimized lectin-hydrogel microarray can be used as an efficient bacterial capturing and culturing platform, which is beneficial to identify spots and collect data. At the same time, a matching drug-laden polyacrylamide microarray with Luria–Bertani (LB) culture medium can be generated automatically and accurately by using a standard non-contacting procedure. A large number of microscale culture chambers (more than 100 individual samples) between two microarrays can be formed by linking two aligned hydrogel spots using LB culture medium, where live bacteria can be co-cultured with drug candidates. Using Staphylococcus aureus ( S. aureus ) and four well-known antibiotics (amoxicillin, vancomycin , streptomycin and chloramphenicol) as model system, the MIC (minimum inhibitory concentration) values of the antibiotics can be determined by the drug induced change of bacterial growth, and the results demonstrate that the MIC values of amoxicillin , vancomycin and streptomycin are 1.7 μg mL −1 , 3.3 μg mL −1 and 10.3 μg mL −1 , respectively.

Paper-based immune-affinity arrays for detection of multiple mycotoxins in cereals

Li Li,Chen Hongpu,Lv Xiaolan,Wang Min,Jiang Xizhi,Jiang Yifei,Wang Heye,Zhao Yongfu,Xia Liru

Mycotoxins produced by different species of fungi may coexist in cereals and feedstuffs, and could be highly toxic for humans and animals. For quantification of multiple mycotoxins in cereals, we developed a paper-based mycotoxin immune-affinity array. First, paper-based microzone arrays were fabricated by photolithography. Then, monoclonal mycotoxin antibodies were added in a copolymerization reaction with a cross-linker to form an immune-affinity monolith on the paper-based microzone array. With use of a competitive immune-response format, paper-based mycotoxin immune-affinity arrays were successfully applied to detect mycotoxins in samples. The detection limits for deoxynivalenol, zearalenone, T-2 toxin, and HT-2 toxin were 62.7, 10.8, 0.36, and 0.23 μg·kg -1 , respectively, which meet relevant requirements for these compounds in food. The recovery rates were 81–86% for deoxynivalenol, 89–117% for zearalenone, 79–86% for T-2 toxin, and 78–83% for HT-2 toxin, and showed the paper-based immune-affinity arrays had good reproducibility. In summary, the paper-based mycotoxin immune-affinity array provides a sensitive, rapid, accurate, stable, and convenient platform for detection of multiple mycotoxins in agro-foods. Solution-Processed Selective Area Homoepitaxial Growth of Suspended MAPbX3 (X = Cl, Br) Perovskite Micro-Arrays

Jinshuai Zhang,Chun Li,Yin Liang,Jiepeng Song,Qiuyu Shang,Liyun Zhao,Huifeng Tian,Lei Liu,Qing Zhang

Suspended nano/microstructures on semiconductor surfaces that can manage light are attractive for various on-chip and efficiency-enhanced optoelectronics. Hybrid halide perovskites have shown considerable potential for optoelectronic and photonic applications. However, the ionic and soft lattices of perovskites are sensitive to polar solvents, photons, and heat, limiting the post-processing of suspended microstructures. Herein, a solution-processed selective area homoepitaxial growth (SAEG) method is introduced to create well-ordered, suspended microstructure arrays on the surface of MAPbX 3 (X = Br, Cl) single-crystalline thin films. The suspended micro-arrays exhibit single crystalline structures and 11.2 fold photoluminescence enhancement compared to the thin-film substrates. The success of SAEG lies in the making use of perovskite supersaturated HX aqueous solutions that not only repair surface defects induced by lithography but also satisfy homoepitaxial growth kinetics. This study paves a pathway for the fabrication of cost-effective perovskite surface-suspended microstructures for efficiency-enhanced and on-chip optoelectronic devices.

Phenylboronic acid polymer brush-enabled oriented and high density antibody immobilization for sensitive microarray immunoassay

Yingshuai Liu,Yuanyuan Zhang,Yanan Zhao,Jie Yu

There still has a big challenge for low-abundance protein detection with antibody microarrays. In this work, phenylboronic acid (PBA) polymer brush-enabled oriented, high density, and covalent antibody immobilization was realized for sensitive antibody microarrays. PBA-enabled oriented antibody attachment via carbohydrate at Fc portion keeps antigen-binding sites fully expose to their corresponding antigens, resulting in higher antibody–antigen (Ab–Ag) binding efficiency. Sandwich immunoassay with rabbit IgG as model analyte was performed on poly(glycidyl methacrylate)-amino-phenylboronic acid-coated glass slide (PGMA-APBA-slide). One order improvement of LOD was achieved as compared with that on poly(glycidyl methacrylate) glass slide (PGMA-slide). The improvement is mainly attributed to PBA-assisted high density and oriented antibody immobilization. This work provides a versatile and effective strategy to develop high sensitive antibody microarrays for low-abundance protein analysis in various proteomic applications.

High-throughput assessment of corrosion inhibitor mixtures on carbon steel via droplet microarray

Chenhao Ren,Lingwei Ma,Xiejing Luo,Chaofang Dong,Taijiang Gui,Bo Wang,Xiaogang Li,Dawei Zhang

A high-throughput screening method for rapid laboratory evaluation of corrosion inhibitors was developed based on microarray consisting of up to 200 droplets of corrosion inhibitor solutions on one carbon steel substrate. The extent of corrosion on the steel under each single droplet was studied by tracking the variation of the 3D morphology of the corrosion product. Using this high-throughput screening platform, we evaluated the corrosion inhibition effect of benzotriazole and Ce(NO 3 ) 3 mixture and identified their best mixing ratio. The enhanced inhibition of the inhibitor mixture was discussed based on the corrosion experimental data and the molecular dynamic simulation.

Fabrication of paper-based enzyme immobilized microarray by 3D-printing technique for screening α-glucosidase inhibitors in mulberry leaves and lotus leaves

Guo Shangxin,Lin Xiaotong,Wang Yi,Gong Xingchu

Background The discovery of bioactive compounds in traditional Chinese medicine (TCM) has become an important field in TCM modernization. Ligand fishing is a suitable method for discovery of bioactive compounds in complex mixtures such as TCM with high selectivity. Because of unique advantage of low cost and convenience, paper-based microdevices can be good carriers for enzyme immobilized ligand fishing. Methods As an important enzyme for glucose metabolism, α-glucosidase was immobilized on polycaprolactone–chitosan-modified paper to prepare the microdevice with unique microfluid structure generated by 3D printing technology, which can be easily applied to screen active compounds in herbal extracts. The preparation conditions of the paper microarray were optimized. The activity of immobilized α-glucosidase was verified by colorimetric reactions which can be easily monitored by cellphone. The paper microarray with α-glucosidase immobilized was used to screen active compounds in the water extracts of mulberry leaves and lotus leaves. Results Several key parameters including Na 2 CO 3 solution concentration, Na 2 CO 3 solution volume, glutaraldehyde concentration, crosslinking time of glutaraldehyde and time of α-glucosidase immobilization were optimized. The proposed paper-based microarray was successfully applied in screening active compounds in two herbal extracts. Four compounds including chlorogenic acid, quercetin-3- O -glucuronide, isoquercetin, and quercetin were identified as α-glucosidase inhibitors. The compounds with significant non-specific adsorption caused by chitosan, such as isoquercitrin, astragalin, quercetin, were also found to be active compounds. Conclusions An enzyme immobilized paper microarray was designed and fabricated in this work. Polycaprolactone and chitosan were used to modify filter paper to prepare paper microarrays. Parameters of paper device preparation were optimized. Our findings suggested that 3D-printing paper-based microarrays can be a simple and low-cost approach for discovery of active compounds of TCM.

Highly Sensitive Microarray Immunoassay for Multiple Mycotoxins on Engineered 3D Porous Silicon SERS Substrate with Silver Nanoparticle Magnetron Sputtering

Jing Yang,Wei Li,Hao Li,Xiu Wang,Kaisong Xu,Qianjin Li,Tiesong Zheng,Jianlin Li

A high-throughput, rapid, and highly sensitive surface-enhanced Raman spectroscopy (SERS) microarray for screening multiple mycotoxins has been developed on a three-dimensional silver nanoparticle porous silicon (3D AgNP-Psi) SERS substrate, which was easy to be engineered by electrochemical etching and magnetron sputtering technology. The etching current density, etching waveform, and target material for magnetron sputtering have been investigated to obtain an optimal 3D SERS substrate. The optimized 3D AgNP-Psi SERS substrate showed an enhancement factor of 2.3 × 107 at 400 mA/cm2 constant current density etching for 20 s and Ag target magnetron sputtering for 200 nm thickness on the surface of Psi. The simulation electric field distribution showed the near-field enhancement can reach 3× higher than that of AuNPs. A protein microarray has been designed to screen multiple mycotoxins by AuNP Raman tags and a competitive immunoassay protocol on the surface of the 3D SERS substrate. The SERS protein microarray displayed wide linear detection ranges of 0.001–100 ng/mL for ochratoxin A, 0.01–100 ng/mL for aflatoxin B1, 0.001–10 ng/mL for deoxynivalenol, along with pg/mL low limit of detection, good recovery rates, repeatability, and reproducibility. The 3D SERS protein microarray is easily engineered and has a great potential application in medicine, environment, and food industry fields.
