Enhanced enzymatic degradation of viscous marine polysaccharide by the self-propulsion of urease-driven micromotor

Yihong Chen,Lili Yin,Hao Dong,Yang Hu,Xiangzhao Mao

Efficient enzymatic degradation of marine polysaccharides is hindered by the limited mass transfer induced from the high viscosity. In this study, taking viscous chitosan as an example, a urease-powered hollow mesoporous silica micromotor (UHMSMM) was prepared as the carrier to immobilize chitosanase (Chi), aiming at improving the degradation of chitosan by self-propulsion. At a urea concentration of 300 mM, Chi@UHMSMM exhibited the maximum motion behavior, confirmed by the doubled diffusion coefficient, even in viscous chitosan. The enhanced motion behavior improved the enzymatic performance. The catalytic efficiency was significantly increased, confirmed by the enhancement (3.8 times) of k c at / K m value. It is demonstrated that the autonomous movement could induce the micromixing of fluid in the microenvironment around Chi according to the simulated result, contributing to improved substrate affinity and catalytic efficiency. Also, the immobilization of Chi on UHMSMM induced a common improvement of enzyme immobilization, including enhanced thermo-stability and acid resistance. As a result, Chi@UHMSMM could work as an efficient platform for degrading chitosan, with a 125.75 % yield enhancement within 40 min. In this work, the reported strategy, which accelerates the enzymatic degradation of chitosan by the self-propulsion of enzyme-driven micro/nanomotors offers a novel approach for improving enzyme reactions in high-viscosity systems.

Responsive Janus Structural Color Hydrogel Micromotors for Label-Free Multiplex Assays.

Huan Wang,Lijun Cai,Dagan Zhang,Luoran Shang,Yuanjin Zhao

Micromotors with self-propelling ability demonstrate great values in highly sensitive analysis. Developing novel micromotors to achieve label-free multiplex assay is particularly intriguing in terms of detection efficiency. Herein, structural color micromotors (SCMs) were developed and employed for this purpose. The SCMs were derived from phase separation of droplet templates and exhibited a Janus structure with two distinct sections, including one with structural colors and the other providing catalytic self-propelling functions. Besides, the SCMs were functionalized with ion-responsive aptamers, through which the interaction between the ions and aptamers resulted in the shift of the intrinsic color of the SCMs. It was demonstrated that the SCMs could realize multiplex label-free detection of ions based on their optical coding capacity and responsive behaviors. Moreover, the detection sensitivity was greatly improved benefiting from the autonomous motion of the SCMs which enhanced the ion-aptamer interactions. We anticipate that the SCMs can significantly promote the development of multiplex assay and biomedical fields.

Efficient demulsification of oil-in-water emulsion triggered by pollen-based micromotors

Mengyin Tan,Chuan Shi,Chen Cui,Enhui Ma,Qi Liu,Ke Dong,Hong Wang

For oil-in-water emulsion wastewaters, the primary challenge in treatment lies in demulsification due to the presence of numerous small oil droplets stabilized by emulsifiers. Toward effective demulsification, this study utilizes the naturally abundant sunflower pollen with a spiny structure and low-cost MnO 2 catalyst to construct a self-driven micromotor for rapid oil-water separation. The hollow structure of treated pollen with an opening formed during carbonization is ideal for the formation and ejection of bubbles, as evidenced by the bubble generation process inside the cavity and the bubble-propelled motion in H 2 O 2 . Surface modification with APTES and oleic acid endows the pollen-based micromotors with hydrophobic properties, enabling them to interact with the oil droplets. Leveraging self-propelled motion and micro/nanobubble production, the pollen-based micromotors can serve as active demulsifiers to bring oil droplets out from the aqueous phase and form a continuous oil layer. Influence factors including pH, temperature, and micromotor dosage are optimized, and a demulsification efficiency of over 97 % is achieved within 30 min. The proposed bio-derived micromotors offer a new platform for efficient demulsification of emulsified wastewater.

BioTemplated Fe3+-Doped g-C3N4 Heterojunction Micromotors for the Degradation of Tetracycline through the Photo-Fenton Reaction

Qingbao Gan,Jianwei Zhang,Jinglin Wang,Yuntian Wei,Shikun Chen,Shuguang Cai,Xueqing Xiao,Chan Zheng

The excessive discharge of antibiotics into aquatic systems is a major issue in many countries worldwide and poses a threat to human health and the sustainable development of society. Hence, developing efficient treatment methods and purification technologies to degrade antibiotics is essential. Herein, we present the synthesis of low-cost, self-propelled tubular Fe3+-incorporated graphitic carbon nitride (g-C3N4-Fe@KF) micromotors using kapok fibers (KFs) as templates and their application as photo-catalysts for the photo-Fenton degradation of tetracycline (TC) under visible-light irradiation. The g-C3N4-Fe@KF micromotors moved rapidly when being propelled by oxygen bubbles generated in a hydrogen peroxide (H2O2) solution as a result of a photo-assisted Fenton reaction. The motion behavior of the g-C3N4-Fe@KF micromotors was dependent on the concentration of H2O2and the length of the micromotors. The propulsion mechanism was discussed in detail. The micromotors efficiently degraded antibiotics via the photo-Fenton process. Photo-Fenton degradation efficiency was attributed to the synergistic effects of the doped Fe3+and g-C3N4under visible-light irradiation and self-propulsion of the micromotors. In addition, the micromotors possessed good reusability, thereby efficiently realizing multiple cycles of degradation. The current work offers an avenue for the design of micromotors, using inexpensive approaches, for various potential environmental applications.

Self-propelled jet carbon micromotor enhanced photocatalytic performance for water splitting

Tengguo Dong,Menglong Sun,Kan Hu,Qian Wang,Chunhua Lu,Jiahui Kou

In the research of photocatalysis , the effect of mass transfer has been seriously ignored, though its importance to photocatalytic performance has been already proved. In this work, self-propelled jet carbon micromotors , carbon bottle (CB), are combined with photocatalysts to enhance the photocatalytic performance, based on the infrared light utilization and active mass flow transfer, the so-called “on-the-fly” working mode. Compared with the traditional Janus motors, the environmentally friendly CB motors, which can be prepared through one-step hydrothermal route, has the advantages of simple synthesis, low price and not containing precious metal. The CB motor has excellent motion characteristics in pure water without additional chemicals as fuels. The photocatalytic activity of 10 wt% CB/g-C 3 N 4 and 10 wt% CB/P25 are 2.1 and 2.3 times than that of pure g-C 3 N 4 and P25, respectively. This work provides a new strategy for the development of photocatalytic technology by utilizing the self-propulsion of micromotors.

Multimodal capsule micromotors with magnetic actuation, ultrasound imaging, and NIR-triggered drug release

Yaqi Liu,Xugen Chen,Likai Hou,Tianxiang Chen,Xuehua Ma,Yujie Zhang,Qiang Li,Aiguo Wu,Changyong Gao

Micro/nanomotors have gained substantial interest for their potential to enable precision therapy. However, achieving reliable in vivo control, particularly precise navigation, targeted accumulation within tumors, and spatiotemporally regulated drug release, remains a major challenge. In this study, we present a multifunctional capsule micromotor to overcome these barriers and enhance targeted therapy for bladder cancer. Using a microfluidic fabrication strategy, we engineered composite capsule micromotors that integrate magnetic nanoparticles, photothermal agents, and chemotherapeutic drugs. This design enables ultrasound-guided real-time imaging, magnetically directed locomotion, and near-infrared (NIR)-triggered drug release. In vitro studies revealed that the capsule micromotors rapidly accumulated on bladder tumor spheroids under magnetic field propulsion. Subsequent NIR irradiation induced pronounced local heating, resulting in capsule shell breakup and efficient drug release. The synergistic combination of magnetic navigation and photothermal activation significantly enhanced tumor cell killing. This system provides a promising strategy for achieving efficient, controllable, and minimally invasive tumor treatment within complex biological microenvironments.

Zirconium Metal–Organic Frameworks as Micromotors with Enzyme-like Activity for Glutathione Detection

Xiaoqing Li,Gui Wang,Dongyang Zhou,Ziwei Lan,Fengling Jiang,Dickon H. L. Ng,Jia Li

Herein, we report a novel UiO-67-Co(bpy)0.35 micromotor synthesized by a facile postsynthesis metalation via introducing cobalt salts ligated with 2,2′-bipyridine-5,5′dicarboxylic acid into UiO-67-bpy0.35 framework. The Co2+ active sites can decompose H2O2 to generate bubbles to power UiO-67-Co(bpy)0.35. Meanwhile, the UiO-67-Co(bpy)0.35 micromotor exhibits robust peroxidase-like activity through catalyzing H2O2 to generate •OH under neutral conditions. Based on this, a sensing platform was constructed for the colorimetric detection of GSH. Due to the synergy of self-driven motion and excellent peroxide-like activity, UiO-67-Co(bpy)0.35 micromotor can sensitively detect GSH with a low analytic limitation as 0.13 μM for GSH detection. This study provides a new sight of using the postsynthesis metalation method to prepare Zr(Co)-MOF micromotor for highly selective, sensitive, and facile detection of GSH.

Synergistic photothermal-bubbly driving effect on MnO2/carbon microsphere micromotors assisted by photocatalytic H2O2 decomposition

Yacong Yang,Ya Zhao,Fengyan Shi,Zitao Chen,Weiguang Zhao,Yue Gao,Yingchun Miao,Yuning Huo

Micromotors with autonomous motion by multiple driving forces have attracted wide attentions. In this work, we successfully constructed MnO 2 -carbon microspheres (Mn-Cs) composite micromotors with synergistic photothermal-bubbly driving effect. The uniform MnO 2 loading and the stable combination of MnO 2 with Cs in Mn-Cs composite facilitated to improve the photothermal conversion performance. The regular morphology and large specific surface area of Mn-Cs were beneficial for the in-situ photocatalytic decomposition of H 2 O 2 under light irradiation. Notably, the photothermal conversion effect of Mn-Cs could further promote the decomposition of H 2 O 2 . The low thermal conductivity of air was beneficial for expanding the temperature gradient between the micromotor surface and the environment. As a result, the speed of optimal 1.0-Mn-Cs sample under the light irradiation and addition of H 2 O 2 (75.2 μm/s) was 14.3 and 5.88 times higher than that individual light irradiation (5.27 μm/s) and addition of H 2 O 2 (12.8 μm/s), respectively. It was indicative of the great significance of synergistic photothermal-bubbly driving effect. This study provided a new strategy for developing synergistically driven micromotors to reduce chemical fuel dependence, and showed a significant prospect in green, economic and sustainable applications.

Shape-controlled Mn–Fe PBA derived micromotors for organic pollutant removal

Jing Huang,Qi Shan,Yanan Fang,Ning Zhao,Xiaomiao Feng

A new simple strategy to prepare Mn–Fe Prussian blue analogue (PBA) derived oxide micromotors through co-precipitation and heat treatment is demonstrated. The morphology of the precursors could be precisely controlled from cubic-like to spherical-like by introducing citrate. Because of the synergistic catalytic reaction of bimetallic Mn–Fe oxides, micromotors with different shapes could be powered efficiently in H2O2 solution. In particular, the obtained cubic Mn–Fe oxide micromotors showed strong propulsion at a speed of 71.21 μm s−1 in 1% H2O2 solution. With dual mechanisms of adsorption bubble separation (ABS) and Fenton oxidation, the cubic Mn–Fe oxide micromotors showed extraordinary removal ability for Rhodamine B (RhB) as a simulated pollutant and could remove up to 95.2% of RhB within 30 minutes. Magnetic Fe3O4 contained in the component not only could degrade organic pollutants, but also could be recycled by magnetic field and reused with high degradation efficiency. These dual-functional metal oxide catalytic micromotors provide a practical and environmentally friendly strategy for environmental restoration.

Micromotor-Enabled Active Hydrogen and Tobramycin Delivery for Synergistic Sepsis Therapy

Yanzhen Song,Ruotian Zhang,Hanfeng Qin,Wenxin Xu,Jia Sun,Jiamiao Jiang,Yicheng Ye,Junbin Gao,Huaan Li,Weichang Huang,Kun Liu,Yunrui Hu,Fei Peng,Yingfeng Tu

Sepsis is a highly heterogeneous syndrome normally characterized by bacterial infection and dysregulated systemic inflammatory response that leads to multiple organ failure and death. Single anti-inflammation or anti-infection treatment exhibits limited survival benefit for severe cases. Here a biodegradable tobramycin-loaded magnesium micromotor (Mg-Tob motor) is successfully developed as a potential hydrogen generator and active antibiotic deliverer for synergistic therapy of sepsis. The peritoneal fluid of septic mouse provides an applicable space for Mg-water reaction. Hydrogen generated sustainably and controllably from the motor interface propels the motion to achieve active drug delivery along with attenuating hyperinflammation. The developed Mg-Tob motor demonstrates efficient protection from anti-inflammatory and antibacterial activity both in vitro and in vivo. Importantly, it prevents multiple organ failure and significantly improves the survival rate up to 87.5% in a high-grade sepsis model with no survival, whereas only about half of mice survive with the individual therapies. This micromotor displays the superior therapeutic effect of synergistic hydrogen-chemical therapy against sepsis, thus holding great promise to be an innovative and translational drug delivery system to treat sepsis or other inflammation-related diseases in the near future.

Magnesium-based micromotors for the detection of carcinoembryonic antigen in whole blood

Yue Li,Di Shi,Tiantian Liang,Jiabo Zhang,Yanhong Ni,Pengsheng Jing,Chun Mao,Runzhi Deng

The rapid and accurate detection of tumor marker carcinoembryonic antigen (CEA) is of great significance for the early diagnosis of cancer. Here, we design an electrochemical detection platform based on zwitterionic surface modification, nanomotor and magnetic separation technology. Specifically, magnesium-based micromotors (magnesium/ferroferric oxide/poly(carboxybetaine methacrylate)/ carcinoembryonic antigen antibody, Mg/Fe 3 O 4 /PCBMA/anti-C) were firstly prepared using asymmetric surface modification technology, which could react with water to form active motion to promote the capture of the target detection substance CEA. Furthermore, after the Mg-based micromotor was completely consumed in the test solution, a self-made funnel device was used to collect the magnetic nanoparticles that captured CEA onto the surface of the magnetic electrode. Then, the electrochemical testing was performed. When CEA bound with anti-CEA, the resulting immune complex hindered the electrochemical probe Fe(CN)₆³⁻/⁴⁻ from reaching the electrode surface, thereby causing a decrease in the electrochemical signal. By measuring the change in the electrochemical signal, the concentration of CEA could be quantitatively determined. The experimental results indicate that Mg-based micromotors display good blood compatibility and could actively capture CEA in whole blood. This detection platform was very sensitive for CEA detection, with a detection range from 0.001   ng   mL -1 to 100   ng   mL -1 and a detection limit of 2.58×10 -4   ng   mL -1 . This multi-technology composite application of whole blood antigen detection is expected to provide a new technological platform for early diagnosis of cancer.

Nanoarchitectonics on Oxygen-Rich Vacancy C/OVs-Ni-Cu-LDHs@MnO2 Tubular Micromotors for Sensitive Detection and Efficient Catalytic Degradation

Xiaoqing Li,Min Zuo,Ziwei Lan,Fengling Jiang,Dickon H. L. Ng,Jia Li

C/OVs-Ni-Cu-LDHs@MnO2 tubular micromotors with enzyme-like activity and abundant oxygen vacancies (OVs) were synthesized hydrothermally using kapok as a biotemplate. MnO2 acted as the micromotor’s “driving engine”, catalyzing the generation of H2O2 bubbles to provide driving force for the motor, enabling efficient motion (144.45 ± 3.97 μm/s in 5% H2O2). The micromotors also exhibited enzyme-like activity, oxidizing 3,3′,5,5′-tetramethylbenzidine (TMB) via reactive radicals. A sensor for the colorimetric detection of GLY (lowest detection limit 0.63 μM) was developed by utilizing the enzyme inhibition induced by Cu2+-glyphosate (N-(phosphonomethyl)glycine, GLY) chelation. Meanwhile, the micromotor catalyzes H2O2 and activates peroxymonosulfate (PMS), efficiently generating hydroxyl radicals (•OH), superoxide radicals (O2•–), and single-linear oxygen (1O2). Under the synergistic effect of self-driven propulsion and efficient catalysis, achieving 93.71% GLY degradation in 40 min. This study provides an intelligent strategy for developing dynamic sensors with dual functions of simultaneous detection and degradation of GLY.
