Micro-CT Imaging of RGD-Conjugated Gold Nanorods Targeting Tumor In Vivo

Xiaochao Qu,Xiaoxiao Li,Jingning Liang,Yanran Wang,Muhan Liu,Jimin Liang

Gold nanomaterials as computed tomography (CT) contrast agents at lower X-ray dosage to get a higher contrast have advantages of longer imaging time and lower toxic side effects compared to current contrast agents. As a receptor for Cyclo (Arg-Gly-Asp-D-Phe-Lys) (RGD) peptide, integrin is overexpressed on some tumor cells and tumor neovasculature. In this paper, we conjugated the RGD peptide on the surface of gold nanorods (AuNRs), designated as RGD-AuNRs, a promising candidate in applications such as tumor targeting and imaging capability for micro-CT imaging. Integrin -positive U87 cells and integrin -negative HT-29 cells were chosen to establish animal models relatedly and then texted the tumor targeting ability and imaging capability of RGD-AuNRs in vitro and in vivo. The MTT assay and stability measurement showed that RGD-conjugation eliminated their cytotoxicity and improved their biocompatibility and stability. Dark-field imaging of U87 cells and HT-29 cells testified the binding affinities and uptake abilities of RGD-AuNRs, and the results showed that RGD-AuNRs were more specifical to U87 cells. The enhanced micro-CT imaging contrast of intramuscular and subcutaneous injection illustrated the feasibility of RGD-AuNRs to be contrast agents. Furthermore, the micro-CT imaging of targeting U87 and HT-29 tumor models verified the targeting abilities of RGD-AuNRs.

SiO2 Coated Up-Conversion Nanomaterial Doped with Ag Nanoparticles for Micro-CT Imaging

Wei Zhang,Yanli Lu,Yang Zang,Jinhui Han,Qingyun Xiong,Jinping Xiong

In this study, a new method for synthesizing Ag-NaYF4:Yb3+/Er3+@ SiO2nanocomposites was introduced. Using a hydrothermal method, the synthesized Yb3+- and Er3+-codoped NaYF4up-conversion luminescent materials and Ag nanoparticles were doped into up-conversion nanomaterials and coated with SiO2up-conversion nanomaterials. This material is known as Ag-UCNPs@SiO2, it improves both the luminous intensity because of the doped Ag nanoparticles and has low cytotoxicity because of the SiO2coating. The morphology of UCNPs was observed using scanning electron microscopy (SEM), and the mapping confirmed the successful doping of Ag nanoparticles. Successful coating of SiO2was confirmed using transmission electron microscopy (TEM). Fluorescence spectra were used to compare changes in luminescence intensity before and after doping Ag nanoparticles. The reason for the increase in luminescence intensity after doping with Ag nanoparticles was simulated using first-principles calculations. The cytotoxicity of Ag-UCNPs@SiO2was tested via the cell counting kit-8 (CCK-8) method, and its imaging ability was characterized using the micro-CT method.Keywords:up-conversion;nanomaterials;CT imaging

Animal Experimental Study on Delayed Implantation in a Severely Atrophic Alveolar Ridge Reconstructed Using a 3D-Printed Bioactive Glass Scaffold: A Pilot Study

Lei Deng,Liya Ai,Runxu Li,Wusheng Xu,Lingling Zheng,Chao Wang,Haitao Huang

In this study, a scaffold was designed using 3-Matic software 12.0 (Materialise, Leuven, Belgium) and fabricated via Digital Light Processing (DLP) 3D printing technology, followed by a mechanical property evaluation. The scaffold was bilaterally implanted into mandibular bone defect models in four Beagle dogs to facilitate guided alveolar bone regeneration. After 12 weeks, samples were harvested from two dogs for radiographic and histopathological evaluations. In the remaining two dogs, two dental implants were placed into the scaffold sites. After an additional 12 weeks, samples were harvested for further radiographic and histopathological assessments. (1) Compression testing of the scaffold demonstrated a compressive strength of 24.77 ± 2.36 MPa. (2) Three of the implantation sites exhibited poor wound healing and exposure of the bone grafts early post-surgery (4 weeks), with an exposure rate of 37.5%. (3) Micro-CT imaging revealed a uniform distribution of newly formed bone within the scaffold, with an average bone height of 4.05 ± 0.55 mm and a bone volume fraction of 43.93 ± 4.68%. Histopathological analysis demonstrated the presence of vascularized tissue, non-calcified bone, and newly calcified bone within the scaffold. Additionally, newly formed calcified bone and vascularized tissue were observed at the interface between the implant and the scaffold. These findings suggest that DLP 3D-printed A-W bioactive glass scaffolds represent a promising approach for guided alveolar bone regeneration in dental implant applications.

Regulation of gut microbiome with redox responsible bacterial cellulose hydrogel for precision chemo-radiotherapy of intestinal cancer

Lin Shi,Haijun Lv,Chuntao Chen,Fengmei Cui,Lei Zhang,Jianping Cao,Remo Proietti Zaccaria,Qi Zhang,Dongping Sun

Although various types of radiosensitive nanoplatforms are explored to enhance radiotherapy , digestible and radiosensitive nanofiber hydrogel with sensitive responsiveness to the intestinal environment, greatly amplified radiation-induced intestinal tumor destruction, as well as the ability to regulate gut microbiome is still demanded. Herein, sulfhydryl crosslinked bacterial cellulose hydrogel (SulBC gel) was synthesized through silylation and oxidation reaction. Anticancer drug, cis -dichlorodiamineplatinum (CDDP) is loaded into the hydrogel ( [email protected] gel) for precision radiotherapy of intestinal cancer. The [email protected] gel selectively degrades and consequently exhibits an accelerated drug release behavior in reductive environments. Meanwhile, the [email protected] gel can increase X-ray-induced double-strand breaks, induce G2/M phase arrest, and enhance the radio-sensitivity to intestinal tumor cells. Upon intragastric administration of [email protected] gel, the hydrogel is noninvasively monitored in the gastrointestine by micro-CT imaging contributed to the relative high Z -element of silicon and shows an excellent anti-tumor efficacy under X-ray-irradiation of the human intestinal cancer xenograft nude mice. Additionally, the hydrogel can also elevate the ratio of Bacteroidetes / Firmicutes and reconstruct gut microbiome balance, benefiting to the reduction of colitis induced by irradiation. This work thus presents a digestible nanofiber hydrogel-based radiosensitizer with in vivo micro-CT imaging, intestine-specific decomposition/drug release profiles, and great efficacy in chemo-radiotherapy of cancer.

HA/MgO nanocrystal-based hybrid hydrogel with high mechanical strength and osteoinductive potential for bone reconstruction in diabetic rats

Rui Chen,Hang-Bo Chen,Peng-Peng Xue,Wai-Geng Yang,Lan-Zi Luo,Meng-Qi Tong,Bin Zhong,He-Lin Xu,Ying-Zheng Zhao,Jian-Dong Yuan

Bone repair and regeneration processes are markedly impaired in diabetes mellitus (DM). Intervening approaches similar to those developed for normal healing conditions have been adopted to combat DM-associated bone regeneration. However, limited outcomes were achieved for these approaches. Hence, together with osteoconductive hydroxyapatite (HA) nanocrystals, osteoinductive magnesium oxide (MgO) nanocrystals were uniformly mounted into the network matrix of an organic hydrogel composed of cysteine-modified γ-polyglutamic acid (PGA-Cys) to construct a hybrid and rough hydrogel scaffold. It was hypothesized that the HA/MgO nanocrystal hybrid hydrogel (HA/MgO-H) scaffold can significantly promote bone repair in DM rats via the controlled release of Mg2+. The HA/MgO-H scaffold exhibited a sponge-like morphology with porous 3D networks inside it and displayed higher mechanical strength than a PGA-Cys scaffold. Meanwhile, the HA/MgO-H scaffold gradually formed a tough hydrogel with G′ of more than 1000 Pa after hydration, and its high hydration swelling ratio was still retained. Moreover, after the chemical degradation of the dispersed MgO nanocrystals, slow release of Mg2+ from the hydrogel matrix was achieved for up to 8 weeks because of the chelation between Mg2+ and the carboxyl groups of PGA-Cys. In vitro cell studies showed that the HA/MgO-H scaffold could not only effectively promote the migration and proliferation of BMSCs but could also induce osteogenic differentiation. Moreover, in the 8th week after implanting the HA/MgO-H scaffold into femur bone defect zones of DM rats, more effective bone repair was presented by micro-CT imaging. The bone mineral density (397.22 ± 16.36 mg cm−3), trabecular thickness (0.48 ± 0.07 mm), and bone tissue volume/total tissue volume (79.37 ± 7.96%) in the HA/MgO-H group were significantly higher than those in the other groups. Moreover, higher expression of COL-I and OCN after treatment with HA/MgO-H was also displayed. The bone repair mechanism of the HA/MgO-H scaffold was highly associated with reduced infiltration of pro-inflammatory macrophages (CD80+) and higher angiogenesis (CD31+). Collectively, the HA/MgO-H scaffold without the usage of bioactive factors may be a promising biomaterial to accelerate bone defect healing under diabetes mellitus.
