Bile acids are endogenous amphiphilic steroids from the metabolites of cholesterol. Studies showed that they might contribute to the pathogenesis of cardiopathy in cholestatic liver diseases. Chenodeoxycholic acid (CDCA) and deoxycholic acid (DCA) is associated with colon cancer, gallstones, and gastrointestinal disorders. However, little information is available regarding their cardiac effects. Here, we reported that CDCA (100 μM) and DCA (100 μM) significantly increased the left ventricular developed pressure of the isolated rat hearts to 122.3 ± 5.6% and 145.1 ± 13.7%, and the maximal rate of the pressure development rising and descending (± dP/dt max ) to 103.4 ± 17.6% and 124.4 ± 37.7% of the basal levels, respectively. They decreased the heart rate and prolonged the RR, QRS, and QT intervals of Langendorff-perfused hearts in a concentration-dependent manner. Moreover, CDCA and DCA increased the developed tension of left ventricular muscle and the cytosolic Ca 2+ concentrations in left ventricular myocytes; these functions positively coordinated with their inotropic effects on hearts. Additionally, CDCA (150 μM) and DCA (100 μM) decreased the sinoatrial node beating rate to 80.6 ± 3.0% and 79.7 ± 0.9% of the basal rate (334.2 ± 10.7 bpm), respectively. These results were consistent with their chronotropic effects. In conclusion, CDCA and DCA induced positive inotropic effects by elevating the Ca 2+ in left ventricular myocytes. They exerted negative chronotropic effects by lowering the pace of the sinoatrial node in rat heart. These results indicated that the potential role of bile acids in cardiopathy related to cholestasis.

Hollow microspheres as the filler material of syntactic foams have been adopted in extensive practical applications, where the physical parameters and their homogeneity have been proven to be critical factors during the design process, especially for high-specification scenarios. Based on double-emulsion droplet templates, hollow microspheres derived from microfluidics-enabled soft manufacturing have been validated to possess well-controlled morphology and composition with a much narrower size distribution and fewer defects compared to traditional production methods. However, for more stringent requirements, the innate density difference between the core–shell solution of the double-emulsion droplet template shall result in the wall thickness heterogeneity of the hollow microsphere, which will lead to unfavorable mechanical performance deviations. To clarify the specific mechanical response of microfluidics-derived hollow silica microspheres with varying eccentricities, a hybrid method combining experimental nanoindentation and a finite element method (FEM) simulation was proposed. The difference in eccentricity can determine the specific mechanical response of hollow microspheres during nanoindentation, including crack initiation and the evolution process, detailed fracture modes, load-bearing capacity, and energy dissipation capability, which should shed light on the necessity of optimizing the concentricity of double-emulsion droplets to improve the wall thickness homogeneity of hollow microspheres for better mechanical performance.

Continuous real-time human motion and heart-rate monitoring systems can provide vital clinical information for disease diagnosis, preventive healthcare and rehabilitation care. However, it is still a challenge to design wearable pressure/strain sensors with great flexibility and sustainable power for motion detection and physiological signal monitoring. Here, a self-powered laminated fabric sensor based on Schottky contact was successfully fabricated. This laminated fabric sensor was constructed by sandwiching the polypyrrole (PPy)-coated fabric with the Ni-coated fabric and Al plastic film. The electrical outputs under strain were attributed to the single Schottky contact between PPy and Al. This fabric sensor exhibited excellent stability under continuous quick cycling operation and at high relative humidity, which could be adapted to different human body parts for continuous motion detection and heart-rate monitoring. This work is of great significance to demonstrate a promising strategy toward self-powered wearable pressure sensors used for human motion detecting and hear-rate monitoring.

Flexible near-infrared (NIR) organic photodetectors (OPDs) are promising for health monitoring and wearable electronic applications, but suffer from poor sensitivity related to the low work function anode. In this study, we developed flexible NIR OPDs based on a high work function PEDOT:PSS anode. The optimized PEDOT:PSS anode (SFD-PH1000) exhibits a high conductivity of 4400 S cm−1 on the PEN substrate and a high work function of 5.07 eV, which effectively blocks reverse electron injection in OPDs. As a result, the flexible NIR OPD based on SFD-PH1000 anode exhibits one order of magnitude reduction in dark current density and a three-time increase in specific detectivity compared with that based on the traditional ITO anode. The SFD-PH1000-based flexible OPD also shows markedly superior mechanical flexibility, and can monitor heart rate and blood oxygenation in photoplethysmography test with an error less than 3%.

The early monitoring of cardiovascular biomarkers is essential for the prevention and management of some cardiovascular diseases. Here, we present a novel, compact, and highly integrated skin electrode as a mechanical–electrochemical dual-model E-skin, designed for the real-time monitoring of heart rate and sweat ion concentration, two critical parameters for assessing cardiovascular health. As a pressure sensor, this E-skin is suitable for accurate heart rate monitoring, as it exhibits high sensitivity (25.2 pF·kPa−1), a low detection limit of 6 Pa, and a rapid response time of ~20 ms, which is attributed to the iontronic sensing interface between the skin and the electrode. Additionally, the electrode functions as a potassium ion-selective electrode based on chemical doping, achieving an enhanced response of 11 mV·mM−1. A test based on the real-time monitoring of a subject riding an indoor bike demonstrated the device’s capability to monitor heart rate and sweat potassium ion levels reliably and accurately. This advancement in wearable technology offers significant potential for enhancing patient care based on the early detection and proactive management of cardiovascular conditions.

Flexible pressure sensors have gained attention for their comfort, portability, and potential in long-term pulse monitoring and early cardiovascular disease diagnosis. However, stretching stress during daily activities affects sensor accuracy, causing motion artifacts (MAs) that hinder precise pulse signal detection. To address this challenge, the anti-motion artifact iontronic pressure sensor (S-smooth sensor), featuring a soft-hard stretchable interface with energy dissipation properties is developed. By regulating the local modulus of the encapsulation layer, this structure dissipates stretching stress, achieving an MAs suppression rate of up to 90%, significantly improving pulse signal accuracy and reliability. Additionally, the sensor incorporates a dielectric layer and double electrode layer (EDL) sensing interface, with a low-friction design that ensures high sensitivity (92.76 kPa−¹) and stability, maintaining performance over millions of cycles. The sensor accurately captures heart rate (HR) and pulse peak time differences (Δt) under various finger-bending conditions. When integrated into a portable wireless pulse monitoring system, it shows a heart rate loss rate of only 2.9% during intense physical activity. This approach avoids complex chemical processes and material restrictions, offering a novel solution for motion artifact suppression in sensors, with significant potential for real-time health monitoring and assisted diagnosis.

Mental stress, a human's common emotion that is difficult to recognize and describe, can give rise to serious psychological disorders. Skin and sweat are easily accessible sources of biomarkers and bio-signals that contain information about mental stress. It is challenging for current wearable devices to monitor psychological stress in real-time with a non-invasive manner. Therefore, we have developed a smartwatch integrated with a sweat cortisol sensor and a heart rate variation (HRV) sensor. This smartwatch can simultaneously record the cortisol levels in sweat and HRV index in real time over a long period. The cortisol sensors based on organic electrochemical transistor (OECT) are fabricated by utilizing the Prussian-blue (PB) doped molecular imprinting polymer (MIP) modified gate electrode. The sensor signal current will decrease following the combination of sweat cortisol, due to the blocking of the PBMIP conductive path, demonstrating good sensitivity, selectivity, and stability. The HRV sensor is manufactured by a photoplethysmography method. We have integrated the two sensors into a wearable smartwatch that can match well with the mobile phone APP and the upper computer software. Through the use of this smartwatch, we have observed a negative correlation between cortisol levels in sweat and the HRV index in short-term stressful environments. Our research presents a great progress in real-time and non-invasive monitoring human's stress levels, which promotes not only the stress management, but also better psychological research.

We have found that niclosamide induced relaxation of constricted artery. However, niclosamide is insoluble, the low bioavailability and the resultant low plasma concentration limit its potential exertion in vivo . The aim of the present study is to synthesize a soluble poly (methacrylic acid-niclosamide) polymer (PMAN) and study the effects of PMAN on arterial function in vitro and the blood pressure and heart rate of rats in vivo . We synthesized the poly (methacrylic acid-niclosamide) polymer (PMAN), the chemical structure of which was identified by FTIR and 1 H NMR spectra. The average molecular weight and polydispersity index of PMAN were 5138 and 1.193 respectively. Compared with niclosamide, the water solubility of niclosamide in PMAN was significantly increased. PMAN showed dose-dependent vasorelaxation effect on rat mesenteric arteries with intact or denuded endothelium in phenylephrine (PE) and high K + (KPSS)-induced vasoconstriction models in vitro . The efficacy of vasorelaxant effect and the cytotoxic effect of PMAN on vascular smooth muscle cells (A10) were lower than that of niclosamide. The LD 50 of PMAN in mice (iv) was 80 mg/kg. Venous injection of PMAN (equivalent 5 mg niclosamide per kg) showed acute reduction of the rat blood pressure and heart rate in vivo . In conclusion, the solubility of niclosamide was increased in the way of poly (methacrylic acid-niclosamide) polymer, which relaxes the constricted arteries in vitro and reduces the rat blood pressure and heart rate in vivo , indicating that modifying niclosamide solubility through polymerization is a feasible approach to improve its pharmacokinetic profiles for potential clinic application.

Simultaneous monitoring of the body’s biochemical and biophysical signals via wearable devices can provide a comprehensive assessment of an individual’s health state. However, current multifunctional sensors for synchronous biochemical and biophysical sensing rely on discrete sensing units, posing a limitation in increased complexity in device assembly, signal processing, and system integration. In this study, we report a dual-mode and self-powered wearable sensor with ion and pressure-sensing capabilities by interfacing a hydrogel film with a solid ion-selective electrode. The hydrogel film can not only collect natural sweat from the skin but also offer a piezoionic response to pressure. We show that wrist pulse-induced pressure response can be incorporated into the noise of the response to sweat sodium ions, allowing for the simultaneous measurement of heart rate and sweat electrolytes. This work provides an example of simplifying the development of wearable multimode sensors through the rational design of classic electrochemical sensors.

Amitriptyline (AMI), one of the widely used tricyclic antidepressants (TCAs), has become a pharmaceutical contaminant frequently detected in aquatic ecosystems. However, the impacts of AMI exposure and underlying mechanisms on fish are still limited. In this study, adult zebrafish (F0) were exposed to AMI at 0 (control), 0.8, and 8 μg/L for 14 days. Subsequently, the exposed zebrafish were paired for spawning, and their offspring (F1) were reared in an AMI-free medium until 5 days post-fertilization (dpf). This study aimed to assess variations in behaviors and neurotransmitter levels in both the F0 (at the end of the 14-day exposure) and F1 generations (at 5 dpf). As a result, waterborne AMI exposure significantly reduced the locomotor activity, frequency of body contact, and duration of chase in F0 zebrafish, and resulted in notable changes in monoamine neurotransmitter levels in their brains. Parental exposure to AMI significantly elevated the heart rate and eye movement but reduced the locomotor activity in the F1 zebrafish, also along with significant changes in monoamine neurotransmitters and acetylcholine. Furthermore, significant correlations between the changes in behavioral traits and neurotransmitter levels were identified in both F0 and F1 generations. Our findings confirm the critical role of monoamine modulation in the neurobehavioral toxicity of AMI on zebrafish and their offspring, and emphasize the importance of paying attention to its multigenerational effects on fish.

Deltamethrin is a classical pyrethroid insecticide that is frequently detected in aquatic environments and organisms. Furthermore, deltamethrin has been detected in samples related to human health and is a potential risk to public health. This study aimed to investigate the mechanism of cardiotoxicity induced by deltamethrin. Zebrafish were exposed to 0.005, 0.05, or 0.5 μg/L deltamethrin for 28 days. The results showed a significant reduction in male reproduction compared to female reproduction. Additionally, the heart rate decreased by 15.75 % in F1 after parental exposure to 0.5 μg/L deltamethrin. To evaluate cardiotoxicity, deltamethrin was administered to the zebrafish embryos. By using miRNA-Seq and bioinformatics analysis, it was discovered that miR-29b functions as a toxic regulator by targeting dnmts . The overexpression of miR-29b and inhibition of dnmts resulted in cardiac abnormalities, such as pericardial edema, bradycardia, and abnormal expression of genes related to the heart. Similar changes in the levels of miR-29b and dnmts were also detected in the gonads of F0 males and F1 embryos, confirming their effects. Overall, the results suggest that deltamethrin may have adverse effects on heart development in early-stage zebrafish and on reproduction in adult zebrafish. Furthermore, epigenetic modifications may threaten the cardiac function of offspring.

Bitter taste receptors (Tas2rs), the members of the G-protein-coupled receptors, mediate the bitter taste and express in extra-oral tissues. Previous studies have shown that Tas2r mRNAs are expressed in the whole heart and cultured cardiomyocytes of neonatal rats. This study aimed to determine the expression of Tas2rs and their function in the adult rat hearts by using RT-qPCR techniques, Langendorff-perfused isolated hearts, and isolated sinoatrial (SA) nodes. The data presented here revealed the mRNA expression of Tas2rs and their coupled G-protein subunits in the SA node and left ventricle of adult rat hearts. Tas2r agonists, quinine and chloroquine , decreased the heart rate and increased the RR interval and QRS duration in Langendorff-perfused isolated rat hearts; they reduced the spontaneous beating rate of isolated SA nodes with pEC 50 values of 4.907 ± 0.045 and 4.968 ± 0.030, respectively. The blockade of Tas2r108 with abscisic acid , the inhibition of phosphodiesterases (PDEs) with 3-isobutyl-1-methylxanthine (IBMX), or the selective inhibition of PDE3 and PDE4 with a cocktail of cilostamide and rolipram , attenuated the negative chronotropic effects of quinine and chloroquine on the SA node. Furthermore, quinine and chloroquine suppressed the tachycardia effect of isoprenaline on the SA node and shifted the concentration-response curve of isoprenaline rightward. In summary, we provided a few lines of evidence that Tas2r agonists, quinine and chloroquine, decreased the heart rate by prolonging ventricular depolarization, and by attenuating the SA node pace in a PDE-dependent manner; they can counteract with β -adrenergic receptor activation and eliminate isoprenaline-induced tachycardia.
