Extraction of chlorogenic acid from Lonicera japonica Thunb. using deep eutectic solvents: Process optimization and mechanism exploration by the combination of experiment and molecular simulation

Zhenyan Fan,Jingyu Li,Junyu Zhou,Yueyuan Chen,Zhifang Cui

The green extraction of chlorogenic acid (CGA) from Lonicera japonica Thunb. using deep eutectic solvents (DESs) was systematically investigated in the study. Quantum chemical calculations (QC) were adopted to evaluate the σ -profiles of CGA and DESs. Subsequent extraction experiments revealed that amongst the 15 DESs studied, those showing better complementarity in polar regions of σ -profile with that of CGA exhibited higher CGA yield, accompanied by lower infinite dilution activity coefficient ( γ ∞ ) of CGA in these solvents. Betaine/glycerol (BET/GLY) demonstrated superior extraction performance and was selected for process optimization via Box-Behnken Design. The established model, validated by ANOVA analysis, indicated that the optimized conditions resulted in a CGA yield 1.9 times higher than that with ethanol extraction. To elucidate the mechanistic basis for performance differences among betaine-based DESs, molecular simulations were conducted, revealing that higher CGA yields correlated with increased interaction energies and greater numbers of hydrogen bonds within the system. Notably, among the six active hydrogen atoms in CGA, three participated in intramolecular H-bonding, while the phenolic hydroxyl group of CGA primarily interacted with the carboxyl group of BET, serving as the dominant contributor during extraction. Furthermore, the highest CGA yield from BET/GLY was likely due to synergistic cooperation between BET, GLY, and H 2 O to interact with CGA. These findings highlight the importance of identifying key interaction sites in both target molecules and extractants to rationally design tailored solvents for enhanced extraction performance. This work provides a strategy integrating computational insights and experimental optimization to advance sustainable extraction of bioproducts.

DNA Methylation Influences Chlorogenic Acid Biosynthesis in Lonicera japonica by Mediating LjbZIP8 to Regulate Phenylalanine Ammonia-Lyase 2 Expression

Zha Liangping,Liu Shuang,Liu Juan,Jiang Chao,Yu Shulin,Yuan Yuan,Yang Jian,Wang Yaolong,Huang Luqi

The content of active compounds differ in buds and flowers of Lonicera japonica (FLJ) and L. japonica var. chinensis (rFLJ). Chlorogenic acid (CGAs) were major active compounds of L. japonica and regarded as measurements for quality evaluation. However, little is known concerning the formation of active compounds at the molecular level. We quantified the major CGAs in FLJ and rFLJ, and found the concentrations of CGAs were higher in the buds of rFLJ than those of FLJ. Further analysis of CpG methylation of CGAs biosynthesis genes showed differences between FLJ and rFLJ in the 5′-UTR of phenylalanine ammonia-lyase 2 (PAL2). We identified 11 LjbZIP proteins and 24 rLjbZIP proteins with conserved basic leucine zipper domains, subcellular localization, and electrophoretic mobility shift assay showed that the transcription factor LjbZIP8 is a nuclear-localized protein that specifically binds to the G-box element of the LjPAL2 5′-UTR. Additionally, a transactivation assay and LjbZIP8 overexpression in transgenic tobacco indicated that LjbZIP8 could function as a repressor of transcription. Finally, treatment with 5-azacytidine decreased the transcription level of LjPAL2 and CGAs content in FLJ leaves. These results raise the possibility that DNA methylation might influence the recruitment of LjbZIP8, regulating PAL2 expression level and CGAs content in L. japonica.

Ionic liquid-based enzyme-assisted extraction of chlorogenic acid from Flos Lonicera Japonicae

Sun Yang,Ding Song,Huang He,Hu Yi

Background In recent years, ionic liquids and enzymes have been widely used in the separation and extraction processes of natural products. Chlorogenic acid (CGA) has important biological and pharmacological activities. It is significant to develop a green and efficient method to extract GCA from Flos Lonicera japonica (FLJ) by integrating the advantages of the ionic liquids and enzymes. Results The optimal type of enzyme and ionic liquid was screened. Pectinase in [C6mim] Br aqueous phase was demonstrated to be an ideal combination. The parameters including extraction time, extraction temperature, pH, enzyme amount, and IL concentration were optimized systematically. Scanning electronic microscopy of FLJ samples demonstrated that pectinase and ionic liquid disposal both obviously facilitated the extraction process by destroying the structure of cell wall. Circular dichroism spectroscopy showed that ionic liquid enhanced the activity of the pectinase by altering its secondary structure. Conclusions Compared with previous reported methods, ionic liquid-based enzyme-assisted extraction of GCA from FLJ was proved to be efficient and practical, offering a higher yield in a shorter time. A novel process was proposed for the extraction of active component from natural resources.

Ultrasound-assisted extraction of polyphenols from Lonicera japonica leaves and their α-glucosidase inhibition

Gang Li,Chunming Bao,Hao Zhang,Lu Bai

Introduction: The search for natural bioactive compounds that act as α-glucosidase inhibitors is a central focus in diabetes treatment research. Methods: This study utilized ultrasonic-assisted extraction to optimize the extraction of polyphenols from honeysuckle leaves through response surface methodology (RSM). Enzyme inhibition and fluorescence quenching experiments were conducted to examine the inhibitory activity and binding behavior of the extracted polyphenols. Results: The results indicated that the optimal conditions for polyphenol extraction were as follows: ethanol concentration, 64%, liquid–solid ratio, 45 mL/g, ultrasonic power, 700 W, ultrasonic time, 6 min. Under these conditions, the yield of polyphenols from honeysuckle leaves was 9.16 ± 0.19%, which closely aligns with the predicted value of 9.18%. The total phenolic content of the polyphenol extracts was 20.6 ± 0.67%, with chlorogenic acid and luteoloside contents measuring 5.65 ± 0.40% and 2.51 ± 0.14%, respectively. The inhibitory effect of polyphenol extracts (IC50, 0.14 ± 0.01 mg/mL) on α-glucosidase was better than that of chlorogenic acid (IC50, 0.55 ± 0.02 mg/mL). Fluorescence quenching experiments indicated that the polyphenol extracts interact with α-glucosidase, resulting in alterations to the microenvironment of amino acid residues.Discussion: This interaction can reduce the binding affinity between the substrate and α-glucosidase, thereby contributing to the objective of lowering postprandial hyperglycemia. Our research results can provide reference for the development and utilization of honeysuckle leaves.

Evaluation of Blue Honeysuckle Berries (Lonicera caerulea L.) Dried at Different Temperatures: Basic Quality, Sensory Attributes, Bioactive Compounds, and In Vitro Antioxidant Activity

Min Yu,Beibei Wang,Zhiqiang Huang,Jinjiao Lv,Yunfei Teng,Tianbo Li,Yu Zhang,Kun Dong,Dong Qin,Junwei Huo,Chenqiao Zhu

This study aims to comprehensively investigate the effects of hot-air dehydration on the quality of blue honeysuckle berries (Lonicera caeruleaL.). The results demonstrated that drying with hot air at 40–65 °C for 7–72 h resulted in blue honeysuckle berries with a moisture content of 0.21–1.10 g H2O/g dry weight. Generally, low to medium temperatures (40–55 °C) showed a better effect on the quality than high temperatures (60–65 °C). Specifically, drying at 40 °C exclusively resulted in better retention of cuticular wax, the best sensory appearance, and the highest total phenolic content. Drying at 45 °C and 50 °C resulted in the highest antioxidant capacity and the optimal sensory flavor. Drying at 55 °C led to the highest soluble solid/acid ratio, ascorbic acid concentration, total flavonoid, and total anthocyanin. The work introduces an innovative raw berry product and provides a comprehensive practical and theoretical framework for convective dehydration of blue honeysuckle berries.

Lonicerin alleviates ovalbumin-induced asthma of mice via inhibiting EZH2/NF-κB signaling pathway

Rui DAI,Yun XIANG,Rui FANG,Hai-Han ZHENG,Qing-Song ZHAO,Yan WANG

Asthma is the most common chronic disease in the respiratory system of children caused by abnormal immunity that responses to common antigens. Lonicerin exerts anti-inflammatory activity in other inflammatory models through targeting enhancer of zeste homolog 2 (EZH2) that is related to asthma. We sought to explore the role and mechanism of lonicerin in regulating allergic airway inflammation. Mice were intraperitoneally injected 10 μg ovalbumin (OVA) on postnatal day 5 (P5) and P10, and then inhaled 3% aerosolized OVA for 10 min every day on P18–20, to establish asthmatic mice model. Lonicerin (10 or 30 mg/kg) was given to mice by intragastric administration on P16–P20. Notably, the administration of lonicerin amended infiltration of inflammatory cells and mucus hypersecretion. OVA-specific IgE level, inflammatory cell count and inflammatory cytokines in asthmatic mice were reduced after lonicerin treatment. Moreover, it suppressed the activity of EZH2 and activation of nuclear factor-kappa B (NF-ĸB) as evidenced by decreasing tri-methylation of histone H3 at lysine 27 and reducing nuclear translocation of NF-κB p65. In a word, Lonicerin may attenuate asthma by inhibiting EZH2/NF-κB signaling pathway.

Rong Zhenji,Yu Chunping,Wang Xin,Wang Ruyue,Gao Yang,Zhang Hailong

Blue honeyberry is a well-known nutritious fruit. Meanwhile, it is also an Oroqen herb in China. However, the chemical constituent of the leaves of the plant is little known, despite having better anti-inflammatory activity. In this study, we investigated the chemical constituents of blue honeyberry leaves and screened their anti-inflammatory effects in RAW264.7 cells. Thirty-nine compounds including a new fernane-type triterpenoid were isolated and identified using various chromatographic approaches and spectroscopic techniques. The new compound and some other components showed a significant anti-inflammatory effect by inhibiting nitric oxide production in RAW264.7 cells, in which compounds 6 and 38 exhibited the inhibition of 40.6 ± 4.8% and 46.0 ± 1.4% at the concentration of 100 μM, respectively, without inducing any cytotoxicity.

Luteolin in Lonicera japonica inhibits the proliferation of white spot syndrome virus in the crayfish Procambarus clarkii

Hai-Feng Jiang,Cheng Chen,Xin-Yuan Jiang,Jing-Lei Shen,Fei Ling,Peng-Fei Li,Gao-Xue Wang

White spot disease caused by the white spot syndrome virus (WSSV) is a highly lethal infectious disease of crustacean aquaculture, with no effective means of control. Active compounds from herbal medicines are promising candidates for the development of antiviral agents for WSSV. We evaluated the efficacy of 12 herbal medicines in inhibiting WSSV proliferation in a WSSV infection model ( Procambarus clarkii ), Lonicera japonica extract had the strongest inhibitory effect on WSSV replication. Luteolin (LUT), the major bioactive compound of L. japonica extract, was further investigated and revealed to have a strong inhibitory effect on WSSV (96.04%, 50 mg/kg). LUT treatment decreased the WSSV load and the expression of important viral genes (immediate-early gene ie1 , DNA polymerase DNApol and envelope protein Vp28 ) in a dose-dependent manner and increased the survival of WSSV-infected crayfish by 60.00% (50 mg/kg). Co-incubation and time-of-addition assays revealed that LUT did not affect the infectivity of viral particles or viral absorption and entry. Expression of immune-related genes such as barrier-to-autointegration ( BAF ), heat shock protein 70 ( Hsp70 ) and signal transducer and activator of transcription ( STAT ), which may be beneficial for viral replication, were significantly decreased ( P  < 0.01) in LUT-treated crayfish. LUT also modified the expression of antioxidants ( SOD , CAT and GST ), pro-inflammatory factor ( COX-1 and COX-2 ), and apoptosis-related ( BAX and BI-1 ) genes ( P  < 0.01) to enhance antioxidant defenses, mitigate inflammation and induce apoptosis, respectively. Overall, LUT could be used as a prophylactic or therapeutic agent against WSSV infection in crustacean aquaculture.

Rapid screening of neuraminidase inhibitors using an at-line nanofractionation platform involving parallel oseltamivir-sensitive/resistant neuraminidase bioassays

Zhixu Chen,Jincai Wang,Jiaming Yuan,Zhen Wang,Zhengchao Tu,Jacques Crommen,Wenhui Luo,Jialiang Guo,Tingting Zhang,Zhengjin Jiang

In this study, an advanced at-line nanofractionation based screening platform was developed to screen potential neuraminidase inhibitors (NAIs) from Lonicera japonica Thunb by involving two parallel bioassays, for determining both oseltamivir-sensitive neuraminidase (NAS) and oseltamivir-resistant neuraminidase (NAR) inhibitory activities. 20 potential NAIs with both NAS and NAR inhibitory effects were screened from Lonicera japonica Thunb and identified by mass spectrometer, including 11 phenolic acids, 8 flavonoids and one iridoid glycoside. The proposed at-line nanofractionation based screening platform for NAIs was also used to rapidly screen nine batches of water extracts of Lonicera japonica Thunb or its similar species. Clear differences in the number and content of active components were easily observed, demonstrating that the proposed method possesses great potential for the quality control of herb medicines.

Separation and enrichment of sibiskoside from Sibiraea angustat with magnetic surface dummy template molecularly imprinted polymers

Zilong Chen,Hongdong Liu,Jie Chen,Weifeng Zhu,Yong Liu,Jianxin Min,Xiaotian Chen,Bin Li,Xin Yang

In this work, a novel strategy was developed for separation and enrichment of sibiskoside by dummy molecular imprinting technology and magnetic separation technology. The structural analogue geniposide was selected as the dummy template, using 4-vinylpyridine as the functional monomer, ethylene glycol dimethacrylate as the cross-linking agent, and acetonitrile as the porogen. The molecularly imprinted layer was formed on the surface of the magnetic carrier to prepare dummy template molecularly imprinted polymers (DMIPs) with a core–shell structure. The DMIPs were characterized by scanning electron microscope (SEM), Fourier transform infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA) and Vibration sample magnetometer (VSM). The results of adsorption kinetics experiments and isothermal adsorption experiments showed that DMIPs can reach adsorption equilibrium in a short period of time and the maximum adsorption capacity can reach 14.67 mg/g. The imprinting factor was 2.08. Compared with the andrographolide, polydatin, arbutin, caffeic acid, neohesperidin dihydrochalcone and quercetin, DMIPs have good adsorption capacity for the sibiskoside. And the reusability was better. After the adsorption of DMIPs, the purity of sibiskoside in the crude extracts from Sibiraea angustata increased to 78%. It provided a basis for the further development and utilization of Sibiraea angustata as well as the separation and enrichment of monoterpenes.

Antidepressant effects of 70% ethanolic extract of Lonicerae japonicae flos and it contained chlorogenic acid via upregulation of BDNF-TrkB pathway in the hippocampus of mice

Lingyu Qin,Junming Wang,Xiaohui Wu,Lingling Song,Yueyue Zhang,Mingzhu Gong,Yanmei Wang,Bingyin Li

ABSTRACT Lonicera japonica flos (LJF) is a common clinical herb with outstanding medicinal and nutritional value. This study aimed to evaluate the antidepressant effects of LJF’s active extract and compound chlorogenic acid (CGA) around BDNF-TrkB pathway. The results showed that LJF extracts and CGA had significant antidepressant effects, and the antidepressant effects of different extracts of LJF was highly positively correlated with the content of CGA (FST, r = 0.998; TST, r = 0.934). Moreover, LJF-70% ethanolic extract and CGA improved CUMS induced depressive behavior, upregulated protein expression levels of BDNF and p -TrkB in the hippocampus, restored the damage of hippocampal neurons, and protect liver from damage. In summary, this study demonstrated for the first time that LJF-70% ethanolic extract was the active extract of LJF antidepressant and CGA was its active antidepressant compound, and the antidepressant mechanism mainly involved the upregulation of BDNF-TrkB signaling pathway in the hippocampus of mice.
