Fabrication of a bionic asymmetric wettable Cu-doped chitosan-laponite-PCL wound dressing with rapid healing and antibacterial effect

Zini Liu,Xueqi Chen,Chen Li

Asymmetrical dressings, which are composed of a compact top layer and a porous bottom layer, are commonly used to mimic the characteristics and structure of the epidermis and dermis layers, and overcome the flaws of traditional dressings such as wound dryness and bacterial penetration. Herein, a bio-inspired double-layer asymmetric wettable wound dressing was prepared by low-temperature 3D printing coupled with electrospinning technology. The hydrophobic top layer of poly(caprolactone)(PCL) film produced by electrospinning was used to simulate the compact and air-permeable epidermis. The hydrophilic bottom layer of the dressing, a scaffold composed of chitosan and copper ions doped Laponite (Cu@CS-Lap) was used to kill bacteria and speed up wound healing. Additionally, the composite dressings also showed excellent cytocompatibility and antibacterial properties in vitro experiments. The migratory area of Cu-doped group human umbilical vein endothelial cells increased by about 48.19% compared to the control group, as revealed by the results of the cell scratch experiment. Furthermore, in vivo experiments in rats showed that wound closure at the 0.5Cu@CS5-PCL dressing reached 98.24% after 12 days, indicating the enormous potential of asymmetric double dressings in boosting wound healing.

Rapid bacteria capturing and killing by AgNPs/N-CD@ZnO hybrids strengthened photo-responsive xerogel for rapid healing of bacteria-infected wounds

Bo Huang,Xiangmei Liu,Zhaoyang Li,Yufeng Zheng,Kelvin Wai Kwok Yeung,Zhenduo Cui,Yanqin Liang,Shengli Zhu,Shuilin Wu

Rapid bacteria killing and tissue repair are critical for the fast healing of bacteria-infected wounds, especially under poor sanitation without antibiotics. Herein, we prepared a novel xerogel constructed by thioether as a main chain and embedded by Ag nanoparticles (NPs) and ZnO NPs decorated by N-doped carbon dots (N-CD@ZnO). This hybrid xerogel not only has good mechanical properties without swelling but also has the ability to capture bacteria rapidly by the disulfide group through electrostatic interaction. Under 808 nm near-infrared (NIR) light irradiation for 15 min, this hybrid xerogel killed 99.9% Escherichia   coli   and 99.85% Staphylococcus aureus by the synergistic action of released Ag + and reactive oxygen species (ROS) produced by N-CD@ZnO using up-conversion technology (N-CD reduces the biotoxicity of ZnO and changes its photocatalytic response region from the 368 nm ultraviolet (UV) region to the 808 nm NIR region). In addition, the biotoxicity of Ag + is limited by the Ag-S covalent bond, which ensures good biocompatibility for the xerogel. Finally, in wound repair experiments in vivo , this xerogel is able to completely repair the wound within 10 days. This work will open up a new avenue for xerogel antibacterial materials.

A molybdenum sulfide based nitric oxide controlled release oral gel for rapid healing of oral mucosal ulcers

Haoxin Cheng,Guangqi Tian,Hongmei Liu,Danmeng Bai,Yue Zhang,Qingqing Wang,Mengzhen Zhao,Shuangyuan Cao,Dan Deng,Xiaolei Wang

Oral mucosal ulcer is the most prevalent oral mucosal lesion, affecting over 25 % of general population. The current treatment regimens lack efficacy in addressing challenges such as wound bleeding, bacterial infection and inflammation on a continuous basis. Hence, a multi-functional oral gel (termed MPCST) with a long-acting duration is designed. It is based on a tannic acid-thioctic acid (TATA) supramolecular hydrogel which absorbs tissue exudate while exhibiting robust tissue adhesion properties. To form MPCST, TATA is loaded with MPCS, which are composed of polydopamine (PDA)-coated molybdenum disulfide (MoS 2 ) nanoflakes (MoS 2 @PDA) with high photothermal conversion efficiency, nitric oxide (NO) precursor nitroprusside (SNP) and cerium oxide (CeO 2 ) with high reactive oxygen species (ROS) scavenging rate. Upon exposure to 808 nm near-infrared (NIR) irradiation, MPCS rapidly heats up and releases NO to promote angiogenesis, while exhibiting strong ROS scavenging, antibacterial (including oral common Streptococcus mutans ), and anti-inflammatory properties. Animal experiments show that the MPCST oral gel, composed of MPCS and TATA hydrogel, exhibits superior therapeutic efficacy compared to the commonly used dexamethasone patch.

Antimicrobial peptide-modified AIE visual composite wound dressing for promoting rapid healing of infected wounds

Yi Chen,Hongjin Qian,Dandan Peng,Yan Jiang,Qiaolin Liu,Yan Tan,Longbao Feng,Biao Cheng,Guilan Li

Wound infection is a major problem faced during wound healing. Therefore, it is necessary to develop wound dressings with excellent antimicrobial properties. Here, a smart response system of PVA-TPE/HA-AMP/SF/ALG wound dressing was prepared by a combination of chemical cross-linking and freeze-drying methods. We grafted AMP onto HA to endow the wound dressing with bacterial resistance and slow release of AMP. At the same time, the system detects bacterial activity in real time for precise antimicrobial activity (through the use of PVA-TPE) and modulates inflammation to reduce bacterial infection (through the use of AMP). In addition, the PVA-TPE/HA-AMP/SF/ALG wound dressing has a good three-dimensional mesh structure, which promotes cell proliferation, enhances collagen deposition and angiogenesis, and thus effectively promotes rapid healing of infected wounds. Moreover, it can induce the expression of inflammatory factors such as VEGF, TNF-α, IFN-γ, IL-4 and TGF-β1 in infected wounds through the Wnt/CAMK/p-PKC signaling pathway, inhibit inflammatory responses, promote wound healing and reduce scar formation. Therefore, the PVA-TPE/HA-AMP/SF/ALG wound dressing smart response system shows great promise in infected wound healing.
