Analysis of antioxidants in Chrysanthemum indici flos by online gradient extraction and HPLC-FRAP

Weifeng Fan,Jianqiao Zhou,Zi Wu,Guoying Tan,Haoxiang Li,Quanxi Mei,Zhengming Qian

Chrysanthemum indici flos (CIF), a traditional functional beverage, has been used in China for thousands of years. However, research on the antioxidant properties and compounds in CIF is insufficient. In the current study, an online gradient extraction (OGE) coupled with a high performance liquid chromatography-ferric reducing antioxidant power (HPLC-FRAP) system for rapid identification of antioxidants in CIF was proposed. The CIF sample (0.3 mg) was extracted online by a gradient mobile phase (acetonitrile and 0.1% formic acid), separated on a Poroshell 120 SB-Aq column, and then detected for the anti-oxidative activity with the online FRAP assay. 36 peaks were found and 28 compounds were identified. Among them, 18 peaks showed antioxidant activity, six of which were detected in CIF for the first time. The total analytical time was less than 45 min, including sample extraction, separation, and antioxidant identification. The results indicate that the OGE-HPLC-FRAP system is a simple, efficient and rapid tool for screening antioxidants in CIF, which provides the scientific data of CIF's antioxidant research and is helpful to improve the quality evaluation of CIF.

Rapid identification of pathogenic bacteria from clinical positive blood cultures via virus-like magnetic bead enrichment and MALDI-TOF MS profiling

Zhirou Zhang,Enyun Xing,Wenzhuo Zhao,Minghui Song,Cuiping Zhang,Hong Liu,Xiaomin Li,Hongxiu Yu

Reducing the time required for the detection of bacteria in blood samples is a critical area of investigation in the field of clinical diagnosis. Positive blood culture samples often require a plate culture stage due to the interference of blood cells and proteins, which can result in significant delays before the isolation of single colonies suitable for matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis. In this study, we developed a non-specific enrichment strategy based on SiO2-encapsulated Fe3O4 nanoparticles combined with MALDI-TOF MS for direct identification of bacteria from aqueous environments or positive blood culture samples. Three distinct types of Fe3O4@SiO2 magnetic nanoparticles (MNPs) with unique surface morphologies were developed: spherical MNPs with smooth surfaces (Fe3O4@SN), mesoporous silica coated MNPs (Fe3O4@MSN), and MNPs exhibiting a viral spiked structure (Fe3O4@VSN). These MNPs exhibited excellent binding affinity towards both Staphylococcus aureus and Klebsiella pneumoniae in PBS and artificial saliva solutions. Furthermore, the strategy of using Fe3O4@VSN, which involves non-specific interactions between bacterial cells and the virus-like surface, resulted in a dramatic reduction in the minimum detectable concentrations of target pathogens by up to 1000-fold compared to conventional methods. Our results demonstrate that the use of Fe3O4@VSN has the potential to significantly reduce the processing time required after blood culture and may be useful for enrichment and identification of microorganisms in complex clinical samples.

Rapid identification of electricigens via silver-plated photonic crystal filters

Wang Delong,Zhao Xiangwei,Liu Xing,Mu Zhongde,Gu Zhongze

Short acquisition time and small volumes of incubated bacterial cultures are ideal for the routine and rapid identification or screening of electricigens in research and applications of microbial fuel cells. In this study, a functional substrate based on colloidal photonic crystals (PCs) was developed both for the filtration and identification of electricigens by surface-enhanced Raman scattering (SERS). The fabrication of the substrate was simplified by electroless plating of silver on filtration-based self-assembled PCs on a filter membrane. The silver-plated ordered PC structure provided a 10 7 -fold enhancement of Raman intensity compared to that obtained with a bare PC substrate. The substrate allowed for a “drop and measure” method of bacterial identification within 5 min with a 5 μL sample volume only. The results showed that not only the electricigens Geobacter sp . and Shewanella sp . could be discriminated with species and strain specificity, but also Geobacter sp . and pilus-mutated Geobacter sp . strains. The developed silver-plated PC filter offers tremendous opportunities in energetic, environmental, and clinical applications.

Rapid in vitro identification of the pathogenic bacteria in actual urine samples by proton transfer reaction mass spectrometry

Qi Zhang,Xue Zou,Wei Xu,Yueting Ding,Jie Jin,Yulan Song,Ziyi Wang,Huihui Wang,Zhou Liu,Qiangling Zhang,Xun Bao,Qu Liang,Chaoqun Huang,Chengyin Shen,Yannan Chu

The speed of bacterial identification is crucial for the treatment efficiency of patients with infections. However, the clinical method typically requires more than 24 h to obtain identification results. Recent studies have revealed that bacteria can emit or absorb characteristic volatile organic compounds (VOCs) in the early stage of cultivation. Based on this discovery, our study tried to develop a rapid identification method using these bacteria-derived VOCs. The studied subjects are five common urinary tract infection (UTI) bacteria: Klebsiella pneumoniae ( Kpn ), Escherichia coli ( Eco ), Pseudomonas aeruginosa ( Pae ), Enterococcus faecalis ( Efs ) and Enterococcus faecium ( Efm ). Within six hours of cultivation, characteristic VOCs specific to these bacteria emerged and were detected by a proton transfer reaction mass spectrometry (PTR-MS). Using a hierarchical classification tree method, we constructed several discriminant models based on different combinations of these VOCs, all achieving 100 % accuracy. To validate the clinical applicability of this method, we collected and analyzed 119 actual urine samples. A classification model utilizing VOCs with m/z values of 63, 65, and 71 achieved 95.0 % accuracy. These three VOCs were identified as dimethyl sulfide, ethanol, and 3-methyl-1-butanol, respectively, using gas chromatography-proton transfer reaction mass spectrometry (GC-PTR-MS). Our results demonstrate that this novel method can rapidly and accurately identify UTI bacteria, requiring only one-fourth of the time compared to clinical method. This method has the potential to help promptly formulate antibiotic treatment plans, which could shorten the treatment process and enhance therapeutic effects.
