Pilot-scale production of selenium-enriched Nostoc sphaeroides colonies and polysaccharides using two-phase cultivation strategy

Jingjing Hu,Peihua Liu,Qiangqiang Wang,Xuyuan Nie,Jianhua Tan,Junchao Shu,Junjian Mai,Yi Cao,Yongdong Zou,Zebo Huang

Nostoc sphaeroides , originally discovered in selenium-rich fields, is a photosynthetic, nitrogen-fixing cyanobacterium that forms edible spherical macrocolonies. However, prolonged selenium supplementation negatively impacts colony quality in culture. To address this, a two-phase cultivation strategy was developed to spatiotemporally separate biomass accumulation from selenium enrichment, resulting in high-quality selenium-enriched N. sphaeroides colonies. The first phase focused on colony growth in nitrogen-free BG-11 0 medium, while the second phase emphasized selenium enrichment in selenium-supplemented, phosphorus-reduced, and nitrogen-containing BG-11 medium (“selenium enrichment medium”). Scale-up to 300 L confirmed the robustness of this process, achieving desired colony hardness of ∼3 newtons, selenium content of ∼350 μg/g, and yield of ∼0.5 g/L (dry weight). Additionally, polysaccharides containing ∼110 μg/g selenium were isolated from the colonies, exhibiting biochemical properties similar to selenium-free polysaccharides from control colonies. These results provide valuable insight into the potential of N. sphaeroides as a cell factory chassis for producing selenium bioproducts.

Effects of Spontaneous Fluorescent Polystyrene on Nostocaceae and Daphnia Magna

Shaolin Qiu,Wenjiao Yuan,Yujie Qin,Xin Feng,Meitong Li,Yuhong Xie

Microplastic pollution is widespread around the world and inevitably comes into contact with organisms. With the accumulation of microplastics in the environment, the negative impact of microplastics on organisms has become the main focus in the field of microplastics. In this study, the different particle and concentration effects of fluorescent polystyrene microplastics (PS-MPs) onNostocaceaeandDaphnia Magnawere researched. The results indicate that PS-MPs adhered toNostocaceaethrough static electricity, which hindered the absorption of photons and CO2byNostocaceae, resulting in a decrease in chlorophyll, a low growth rate and high mortality forNostocaceae. PS-MPs with very small particles may be integrated into the blood ofDaphnia Magna, leading to an increasing trend of mortality and a decrease in spawning rate. The research provides basic data and a reference for the effect of PS-MPs on freshwater organisms and has implications for the further study of microplastics. Keywords:microplastics;fluorescent polystyrene;Nostocaceae;Daphnia Magna;photosynthesis;reproductive effect

Ultrasonic-assisted three-phase partitioning with deep eutectic solvents for extraction of Nostoc commune Vauch polysaccharides and their anti-ulcerative colitis activity

Shaojie Zhang,Zhongnan Wu,Qian Zhang,Han Zhao,Junhao Li

This study explores the extraction of polysaccharides from Nostoc commune Vauch. using ultrasonic-assisted three-phase partitioning with deep eutectic solvents (UA-TPP-DES). Response surface methodology was used to determine the optimized UA-TPP-DES conditions as follows: a 1: 2 M ratio of lauric acid to terpineol, 30 min of ultrasonication at 60 °C with 100 W power, 20 % moisture content, 20 % w/w (NH 4 ) 2 SO 4 concentration, and a 2: 1 top-to-bottom phase volume ratio. The UA-TPP-DES method significantly improved the polysaccharide extraction efficiency of N. commune (yielding NCP-DES-3, 7.31 %) compared to conventional water extraction (3.35 %). The structural features of NCP-DES-3 were characterized through various physical and chemical methods, including molecular weight determination, monosaccharide composition analysis, Fourier Transform Infrared Spectroscopy (FT–IR), Gas Chromatography-Mass Spectrometry (GC–MS), and Nuclear Magnetic Resonance (NMR) spectroscopy. Additionally, the dextran sulfate sodium (DSS)-induced ulcerative colitis model in Drosophila was used to study the anti-ulcerative colitis (UC) activity of NCP-DES-3. In vivo experiments showed that NCP-DES-3 improved the intestinal barrier by modulating intestinal epithelial cell (IEC) and intestinal stem cell (ISC) growth and differentiation. Our results demonstrated that the UA-TPP-DES approach is an eco-friendly and efficient strategy for polysaccharide extraction, while the bioactive polysaccharide NCP-DES-3 shows promising potential as a natural therapeutic agent for UC.

Ultrasonic-assisted extraction of a low molecular weight polysaccharide from Nostoc commune Vaucher and its structural characterization and immunomodulatory activity

Ying Pan,Chunjuan Liu,Shuo Jiang,Lili Guan,Xinyao Liu,Liankui Wen

In the current study, a novel crude polysaccharide (cNCEP) was extracted from N. commune Vaucher utilizing ultrasonic-assisted extraction (UAE) with 60 % ethanol, employing response surface methodology. The optimal yield of cNCEP was determined to be 8.07 ± 0.08 mg/g, achieved through ultrasonic-assisted extraction under the conditions of a material-to-liquid ratio of 1:22, temperature of 56 °C, power of 570 W, and duration of 147 min. Subsequent purification of NCEP via Sephadex G75 resulted in a novel polysaccharide with a molecular weight of 20.466 kDa. NCEP exhibited significant scavenging activites against DPPH and hydroxyl radicals, as well as notable in vitro immunomodulatory properties. Furthermore, the mechanisms underlying the immunomodulatory effects of NCEP, involving enhancement of immunity, were investigated, revealing potential regulation of MAPK and TLR4-IRF7-NF-κB signaling pathways through RNA-Seq and Western blot analyses. These findings highlight the promising potential of NCEP as an organic immunomodulatory agent and functional food ingredient.

Polysaccharides isolated from Nostoc commune Vaucher inhibit colitis-associated colon tumorigenesis in mice and modulate gut microbiota

Min Guo,Zhuoyu Li

Accumulating evidences have reported that the gut microbiota plays an important role in the occurrence and progression of colorectal cancer. Commonly known as a kind of constituent macromolecules, non-digestible polysaccharides have always been fermented by the intestinal microbiota. Nostoc commune Vaucher (N. commune) has always been appreciated as a healthy food and supplementation worldwide as it is rich in nutrients. Particularly, polysaccharides account for 60% of the dry weight of N. commune. In this study, we examined the protective effects of the polysaccharides isolated from N. commune (NVPS) against colitis-associated colon tumorigenesis in mice treated with azoxymethane (AOM) and dextran sulfate sodium salt (DSS) and the impact of these polysaccharides on the intestinal microbiota. NVPS were administered to mice through an intragastric gavage for 14 weeks. Our results demonstrated that the treatment with NVPS significantly decreased the number and sizes of tumors and reduced the incidence of intestinal tumors. Using 16S rRNA gene sequencing and qPCR, we analyzed the bacterial composition of the fecal samples obtained from mice. The results demonstrated that the alterations in the abundance of Firmicutes and Bacteroidetes caused by the AOM/DSS treatment were significantly reversed in response to the NVPS treatment. Moreover, the short-chain fatty acid (SCFA)-producing genera, including butyric acid-producing genera (Butyricicoccus, Butyrivibrio and Butyricimonas) and acetic acid-producing genera (Lachnospiraceae UCG 001, Lachnospiraceae UCG 006, and Blautia), were drastically enriched following the NVPS intervention. These compositional alterations induced by the NVPS were associated with the suppressed colonic inflammation and carcinogenesis. In conclusion, our results demonstrated an appreciable capability of NVPS to restore the gut microbiota profile altered by AOM/DSS, indicating the potential of NVPS as a promising prebiotic candidate for the prevention and treatment of colorectal cancer.

Selective production of singlet oxygen for harmful cyanobacteria inactivation and cyanotoxins degradation: Efficiency and mechanisms

Bingzhi Yu,Xizi Li,Mengfan He,Yan Li,Jiafeng Ding,Yuchi Zhong,Hangjun Zhang

Knowledge about the impact of singlet oxygen ( 1 O 2 ) on the characteristics and inactivation of harmful cyanobacterial organic matter is limited. In this study, the feasibility of using an improved single-iron doped graphite-like phase carbon nitride catalyst (FeCN) to activate peroxymonosulfate (PMS) catalytic production of 1 O 2 to inactivate four harmful cyanobacteria was investigated. The inactivation efficiencies at 30 min were 92.77%, 66.84%, 91.06%, and 93.45% for Microcystis aeruginosa ( M. aeruginosa ), Nodularia harveyana , Oscillatoria sp., and Nostoc sp . , respectively. This was associated with adjusting experimental parameters, such as the FeCN and PMS doses and initial pH, to obtain the maximum 1 O 2 yield. The quenching experiment results and electron paramagnetic resonance spectra showed that 1 O 2 generated via the non-radical pathway might play a dominant role in inactivating harmful cyanobacteria and degrading harmful algal toxins (Microcystin-LR and Nodularin). In addition, the FeCN-PMS system not only effectively destroyed the integrity of harmful cyanobacterial cells but also effectively degraded cyanobacterial toxins , thereby preventing severe secondary contamination by cell rupture. A possible removal mechanism was proposed. This reveals the potential of 1 O 2 to simultaneously inactivate harmful cyanobacteria and degrade harmful cyanobacterial toxins.

Carbon nanomaterial-treated cell cultures of Nostoc flagelliforme produce exopolysaccharides with ameliorative physio-chemical properties

Xiaolong Yuan,Xiang Gao,Tao Zheng,Jing Wang,Yibei Dong,Huidan Xue

The feasibility and efficiency of carbon nanomaterials (CNMs) in algal biotechnology are less known. In this study, the influences of four CNMs, graphene (G), graphene oxide (GO), multiwalled carbon nanotube (MWCNT), and aminated multiwalled carbon nanotube (MWCNT-NH 2 ), on cell growth and exopolysaccharide (EPS) production, as well as the physiochemical properties of EPS, were investigated in cell culture of Nostoc flagelliforme . A proper concentration (15 mg L −1 ) of four CNMs was chosen for use after a preliminary test. Upon GO treatment, the biomass was improved by 11.1 % and the EPS production was increased by 36.1 % on day 16 compared to the nontreated control. Four CNM treatments significantly improved cellular O 2 · − and H 2 O 2 levels as well as superoxide dismutase and catalase activities. The monosaccharide compositions and functional groups of the EPSs were obviously altered by the CNM treatments. Particularly, the GO treatment-resulting EPS showed obviously improved flocculating ability, water absorption ability, and reactive oxygen species scavenging ability. In general, four CNMs exerted distinct influences on the production and physio-chemical property alteration of the EPS in N. flagelliforme culture. This work expands our understanding of the application of CNMs in the induced production and functional modification of polysaccharides during algal cultivation.

Structure elucidation and molecular mechanism of an immunomodulatory polysaccharide from Nostoc commune

Yiting Yang,Ping Ren,Ying Sun,Junyi Li,Xinjun Zhou,Haipeng Zhang,Chengguang He,Huining Dai,Lili Guan

Nostoc commune Vaucher, a terrestrial and benthic blue-green alga, widely used in food and medicine worldwide. N. commune Polysaccharides (NCVP) have excellent biological activities, especially immunomodulatory, hypoglycemic and anti-tumor activities. However, the mechanism and structure-activity relationship of NCVP has been less studied. In this study, based on methylation and NMR results, a novel polysaccharide NCVP2 with 135 kDa, containing→4)-α-D-Gal p -(1→, → 4)-β-D-Glc p -(1→, and →4)-α-D-Xyl p -(1→ residues as the backbon, was sequentially purified from N.commune by DEAE-52 and Sephadex G-100 column. NCVP2 (50 μg/mL) exhibited the strong in vitro immunomodulatory activity by promoting the generation of nitric oxide ( NO ) and reactive oxygen species (ROS). A total of 2048 differentially expressed genes (DEGs) were identified by RNA-seq, including 1019 down-regulated genes and 1065 up-regulated genes. These DEGs were mainly enriched in the immune-related biological processes, involving in Mitogen-activated protein kinase (MAPK) and Toll-like receptor (TLR) signaling pathways by GO and KEGG enrichment analysis. Furthermore, Western blot results proved NCVP2 could recognize TLR2 and TLR4/MD2, and regulate TLR7/IRF7, MAPK and PI3K/AKT signaling pathways. In summary, a novel polysaccharide NCVP2 from N.commun e was proposed to exhibit significant immunomodulatory effects with multiple-paths and targets, and has great potential in the development of healthy foods such as immunomodulators.
