Full-length normalization subtractive hybridization analysis provides new insights into sexual precocity and ovarian development of red swamp crayfish Procambarus clarkii

Hong-Yue Tan,Guang-Ming Shao,Peng-Fei Kang,Yu-Feng Wang

The red swamp crayfish Procambarus clarkii is an important freshwater economic crayfish in Chinese inland due to its high market value and consumer demand. The sexual precocity in this species has received considerable attention in recent years because more and more individuals matured at small sizes, which devalues the commercial production. In this study, by the full-length normalization subtractive hybridization (FNSH) method we identified 16 differently expressed sequences between precocious and normal grown P . clarkii ovaries. Of the down-regulated sequences in precocious ovaries, one encodes a peptide which shared the highest identity to C1q-like family and contained a typical C1q domain at the C-terminal, thus it was named PcC1q-like . QRT-PCR analysis showed that PcC1q-like was transcribed specifically in ovary, with higher expression level in vitellogenic stages. In situ hybridization exhibited specific expression of PcC1q-like in follicular cells around the vitellogenic oocytes . Knockdown of PcC1q-like by injection of double strand RNA (dsRNA) resulted in disruption of follicular cells, decreased transcripts of vitellogenin ( Vg ) and vitellogenin receptor ( VgR ), and reduced progesterone content. These results suggest that PcC1q-like may promote ovarian development by stimulating the synthesis and transportation of vitellogenin into oocytes. These data provide insights into the molecular regulatory mechanisms of sexual precocity and ovarian development in crustaceans. Statement of relevance The red swamp crayfish Procambarus clarkii is one of the most important freshwater aquaculture species in Chinese inland. We screened 16 sequences that are potentially involved in sexual precocity of P. clarkii . One candidate was demonstrated to be a novel ovary-specific C1q-like gene of P. clarkii , thus named PcC1q-like . Knockdown of PcC1q-like resulted in disruption of follicular cells, decreased transcripts of Vg and VgR , and reduced progesterone, suggesting its essential role in vitellogenic ovarian development of P. clarkii .

The Regulatory Effect of Insulin-Like Growth Factor-2 on Hypothalamic Gonadotropin-Releasing Hormone Neurons during the Pubertal Period

Xiaoli Dai,Yuanyuan He,Xinghui Han,Wen Sun,Jian Yu,Yating Lin,Yonghong Wang

Background: The insulin-like growth factor (IGF) system plays a vital role in regulating gonadotropin-releasing hormone (GnRH), whether the IGF2 can act on the GnRH neurons during the pubertal period is unclear. Methods: Central precocious puberty (CPP) rats were induced by danazol, and when the rats met the first diestrus, they were euthanized and tissues were collected. GT1-7 cells were cultured and treated with 0, 1, 10 ng/mL IGF2 for 4 hours and the changes in GnRH were measured. Mice were injected intracerebroventricularly with IGF2 (15 ng/g, 5 μL) or with the same dose of phosphate buffered saline (PBS), after eight hours, they were euthanized and tissues collected. Results: CPP rats had increased expression of IGF2 and GnRH mRNA and their respective proteins in the preoptic area (POA) of the hypothalamus. Treatment of GT1-7 cells with 10 ng/mL of IGF2 increased GnRH mRNA and protein expression, and GnRH concentration in the culture medium. Injection of IGF2 protein into the lateral ventricle of mice increased the expression of GnRH mRNA and protein in the POA. Conclusions: IGF2 may upregulate the synthesis of GnRH during the pubertal period, and may also take part in the pathology of CPP.

Maternal oral exposure to low-dose BPA accelerates the onset of puberty by promoting prepubertal Kiss1 expression in the AVPV nucleus of female offspring

Chenyan Jiang,Wenke Dong,Guanglin Gao,Wen Sun,Yonghong Wang,Bowen Zhan,Yanyan Sun,Jian Yu

As the incidence of precocious puberty has risen in recent years and the age at puberty onset is younger, children may be at increased risk for health consequences associated with the early onset of puberty. Bisphenol A (BPA) is recognized as an endocrine disruptor chemical that is reported to induce precocious puberty. The effect of BPA exposure modes, times, and doses (especially low dose) were controversial. In the present study, we evaluated the potential effects of maternal exposure to low-dose BPA on the hypothalamus, particularly on the arcuate (ARC) nucleus and anteroventral periventricular (AVPV) nucleus during peri-puberty in offspring of BPA-treated rats. Pregnant rats were exposed to corn oil vehicle, 0.05 mg·kg −1 ·day −1 BPA, or 5 mg·kg −1 ·day −1 from gestation day 1 (GD1) to postnatal day 21 (PND21) by daily gavage. Body weight (BW), vaginal opening (VO), ovarian follicular luteinization, and relevant hormone concentrations were measured; hypothalamic Kiss1 and GnRH1 levels by western immunoblot analysis were also assessed as indices of puberty onset. During or after exposure, low-dose BPA restricted BW after birth (at PND1 and PND5), and subsequently accelerated puberty onset by promoting the expression of prepubertal Kiss1 and GnRH1 in the AVPV nucleus on PND30, leading to advanced VO, an elevation in LH and FSH concentrations (on PND30). We also noted increased BW on PND30 and PND35. Maternal oral exposure to low-dose BPA altered the BW curve during the neonatal and peripubertal periods, and subsequently accelerated puberty onset by promoting prepubertal Kiss1 expression in the AVPV nucleus.

Multi-omic analysis of precocious puberty girls: pathway changes and metabolite validation

Zhou Fang,Mao Jianhong

Objective: Precocious puberty (PP) is a prevalent endocrine disorder affecting the physical and mental wellbeing of children. Identifying the triggering factors of PP has become a central issue. This study seeks to investigate the metabolomic and transcriptomic alterations in PP.Material and methods: First, 37 school-aged girls diagnosed with PP and 25 age-matched prepubertal control girls were recruited, and the fecal samples were collected for non-targeted metabolomic analysis to screen for differentially expressed metabolites (DEMs). Subsequently, an animal model of PP was constructed by danazol administration to neonatal female rats, and both fecal non-targeted metabolomics and serum next-generation transcriptomic sequencing were performed to screen DEMs and differentially expressed genes (DEGs) in PP. Moreover, the DEM co-existing in clinical and animal models was administrated to PP rats to explore the role of the target metabolite in PP.Results: A total of 24 DEMs in PP clinical samples and 180 DEMs and 425 DEGs in PP animal samples were identified. The Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis showed that these DEMs and DEGs were enriched in disease-associated pathways, including fatty acid synthesis, glycerolipid metabolism, pyrimidine metabolism, steroid hormone biosynthesis, progesterone-mediated oocyte maturation, and gonadotropin-releasing hormone (GnRH) signaling pathway, forming a tight DEM–DEG pathway regulatory network. Further DEM validation demonstrated that thymine supplementation delayed the opening of the vagina and development of PP in model rats. Conclusion: This study reveals that the metabolomic and transcriptomic changes, along with enriched pathways, are implicated in PP based on clinical and animal analyses. The findings may provide new strategies and research avenues for PP treatment.
