Characterization of CYP3A genes and the impact of letrozole on gonadal development in Apostichopus japonicus

Kong Lingnan,Abulizi Abudula,Cong Jingjing,Meng Weihan,Chang Yaqing,Sun Zhihui

The cytochrome P4503A ( CYP3A ) subfamily has been well characterized in vertebrates, where it plays a pivotal role in xenobiotic detoxification and the regulation of steroid hormone levels. However, their molecular features and functions in invertebrates remain poorly understood. The sea cucumber ( Apostichopus japonicus ) is not only a commercially significant mariculture species but also an excellent model for studying organ development and evolutionary biology. In this study, we identified seven CYP3A genes in A. japonicus using gonadal transcriptomic data. These genes exhibited ubiquitous expression across adult tissues, with significantly higher levels in ovarian tissues compared to testes tissues. Protein sequence analysis revealed that the catalytic residues Ala 306 -Thr 310 -Cys 437 , which are critical for aromatase activity, are highly conserved. This suggests that CYP3A proteins may mediate aromatization-like reactions similar to CYP19A in vertebrates. Additionally, dietary administration of the aromatase inhibitor letrozole led to histological changes in the testes and a reduction in the diameter of large oocytes in the ovaries. These findings provide valuable insights into the molecular mechanisms underlying gonadal development in sea cucumbers and lay the groundwork for future research on the artificial induction of sex reversal in echinoderms.

17α-Methyltestosterone Affected Growth, Gonadal Development, and Intestinal Microbial Analysis in the Giant Freshwater Prawn (Macrobrachium rosenbergii)

Bai Liufu,Qiyao Su,Kunhao Hong,Jie Wei,Yakun Wang,Zhiqiang Han,Lingyun Yu

Simple Summary17α-methyltestosterone (MT) is known to suppress ovaries and induce spermatogenesis; yet, its effects in crustaceans remain underexplored. This study investigates the impact of dietary MT on gonad development and intestinal microbiota in juvenileMacrobrachium rosenbergiithrough sex ratio determination, histological observation, the expression of reproductive development-related genes, and an analysis of the intestinal microbiota structure. The results showed that an. optimal MT concentration and treatment duration promoted gonadal differentiation and growth. However, excessive MT concentrations inhibited male gonad development and slowed growth rates. MT supplementation significantly increased microbial abundance and altered the intestinal microbial community structure. These findings clarify the effects of adding MT to feed on the reproductive development and intestinal microbiota of juvenileM. rosenbergii, providing a reference for MT application in crustaceans.17α-methyltestosterone (MT) is known for its ability to suppress ovaries and induce spermatogenesis; yet, its effects in crustaceans are underexplored. This study investigates the impact of varying MT concentrations on the gonadal development and intestinal microbiota of juvenileMacrobrachium rosenbergii. Feeds containing different MT doses were provided, and the sex ratios, histological observations, reproductive gene expression, and intestinal microbial composition were analyzed. The results revealed short-term feeding (60 days) of 1000 mg/kg MT resulted in the highest male ratio, while long-term feeding (150 days) of 500 mg/kg MT achieved the same outcome. Conversely, long-term feeding of 1500 mg/kg MT led to the lowest male ratio and retarded male germ cell development. An intestinal microbiota analysis showed that MT supplementation significantly increased microbial abundance and altered the intestinal microbial community structure. Additionally, MT suppressed the expression of female reproductive-related genes. This study provides insights into the effects of MT on reproductive development and gut microbiota in juvenile prawns, offering a valuable reference for the application of MT in crustacean aquaculture.

Effects of copper oxide nanoparticles on reproductive system of zebrafish

Guizhu Wu,Lu Gao,Shaoming Zhang,Daolin Du,Yonglai Xue

Copper oxide nanoparticles (CuO NPs) were regarded as the versatile materials in daily life and the in-depth evaluation of their biological effects is of great concern. Herein the female and male zebrafishes were chosen as the model animals to analyze the reproductive toxicity caused by CuO NPs at low concentration (10, 50 and 100 μg/L) After 20-days exposure, the structure of zebrafish ovary and testis were impaired. Moreover, the contents of 17β-estradiol (E2) in both females and males were increased, while the contents of testosterone (T) were decreased, indicating the imbalanced sex hormones caused by CuO NPs. The expression of genes along the hypothalamic pituitary-gonad (HPG) axis, were examined with quantitative real-time PCR to further evaluate the toxic mechanisms. Meanwhile, the levels of erα/er2β and cyp19a in female zebrafishes and erα/er2β , lhr , hmgra/hmgrb , 3βhsd and 17βhsd in male zebrafishes were obviously up-regulated. While, the level of αr was obviously down-regulated in female and male zebrafishes. Thus, the obtained data uncovered that long-term exposure of CuO NPs with low dose could trigger the endocrine disorder, resulting in the disturbance of E2 and T level, inhibition of gonad development, and alteration of HPG axis genes. In brief, this study enriched the toxicological data of NPs on aquatic vertebrates and provided the theoretical support for assessing the environmental safety of NPs.

Comprehensive transcriptome analysis of hypothalamus reveals genes associated with disorders of sex development in pigs

Shuwen Tan,Yi Zhou,Haiquan Zhao,Jinhua Wu,Hui Yu,Yin Yang,Yalan Yang,Huabin Zhao,Hua Li

XX sex reversal, also called XX disorders of sex development (XX-DSD), is a condition affecting the development of the gonads or genitalia, and is relatively common in pigs. However, its genetic etiology and transcriptional regulation mechanism in the hypothalamic-pituitary-gonadal axis (HPGA) remain mostly unknown. XX-DSD ( SRY -negative) pigs and normal sows were selected by external genitalia observation. The hypothalamus, which is the integrated center of the HPGA was sampled for whole-transcriptome RNA-seq. The role of DEmiRNA was validated by its overexpression and knockdown in vitro. A total of 1,258 lncRNAs, 1,086 mRNAs, and 61 microRNAs differentially expressed in XX-DSD pigs compared with normal female pigs. Genes in the hormone biosynthesis and secretion pathway significantly up-regulated, and the up-regulation of GNRH1 , KISS1 and AVP may associate with the abnormal secretion of GnRH. We also predicted the lncRNA-miRNA-mRNA co-expression triplets and constructed three competing endogenous RNA (ceRNA) potentially associated with XX-DSD. Functional enrichment studies suggested that TCONS_00340886 , TCONS_00000204 and miR-181a related to GnRH secretion. Further, miR-181a inhibitor up-regulated GNRH1 , PAK6 , and CAMK4 in the GT1-7 cells. Conversely, transfection of miR-181a mimics obtained the opposite trends. The expression levels of FSHR , LHR , ESR1 and ESR2 were significantly higher in XX-DSD gondas than those in normal sows. Taken together, we proposed that the balance of endocrine had broken in XX-DSD pigs. The current study is the first to examine the transcriptomic profile in the hypothalamus of XX-DSD pigs. It provides new insight into coding and non-coding RNAs that may be associated with DSD in pigs.
