Insights into the metabolite profiles of Rubus chingii Hu at different developmental stages of fruit

Yu-Jiao Hua,Fen Xie,Kun-jun Mao,Yi-Yuan Luo,Yong-Juan Ding

The fruits of Rubus chingii Hu have high medicinal and nutritional values. However, the metabolite profiles of R. chingii , especially the alterations during different development stages of fruit, have not been comprehensively analyzed, hindering the effective utilization of the unique species. In this study, we comprehensively analyzed the metabolites of R. chingii fruit at four developmental stages using systematic untargeted and targeted liquid chromatography-mass spectrometry metabolomics analysis and identified 682 metabolites. Significant changes were observed in metabolite accumulation and composition in fruits during the different developmental stages. The contents of the index components, kaempferol-3-O-rutinoside and ellagic acid, were the highest in immature fruit. The analysis identified 64 differentially expressed flavonoids and 39 differentially expressed phenolic acids; the accumulation of most of these differentially expressed metabolites decreased with the developmental stages of fruit from immaturity to maturity. These results confirmed that the developmental stages of fruit are a critical factor in determining its secondary metabolite compositions. This study elucidated the metabolic profile of R. chingii fruit at different stages of development to understand the dynamic changes in metabolites.

Identification of polyphenolic composition in the fruits of Rubus chingii Hu and its antioxidant and antiproliferative activity on human bladder cancer T24 cells

Li Kuangyu,Zeng Maolin,Li Qiaoling,Zhou Benhong

Rubus chingii Hu, namely “Fu-pen-zi” in Chinese, is commonly used as a functional food in China. A rapid and sensitive High performance liquid chromatography (HPLC) coupled with linear ion trap-OrbiTrap hybrid mass spectrometer (LTQ OrbiTrap MS) was employed to identify the polyphenolic compounds in R. chingii fruits. A total of 18 compounds were identified in R. chingii polyphenols (RCPs), including five phenolic acids, four ellagitannins, and nine flavonoids. Additionally, the total phenolics, total flavonoids and total tannins content as well as antioxidant and antiproliferative activity were evaluated for the first time. The contents of total phenolics, total flavonoids, and total tannins were 356.42 mg GAE g −1 of extract, 201.94 mg RUE g −1 of extract and 155.12 mg GAE g −1 of extract, respectively. Antioxidant assays showed RCPs had favorable DPPH and OH − radical scavenging capacity and strong ferric ion reducing power. Moreover, the results also indicated the RCPs could inhibit cell proliferation and induce the cell apoptosis in T24 bladder cancer cells. Altogether, the results suggest R. chingii possesses potent antioxidant properties and could inhibit the growth of T24 cells by inducing the cell apoptosis in a dose-dependent manner.

Exploring the potential mechanism of Rubus corchorifolius L. fruit polyphenol-rich extract in mitigating non-alcoholic fatty liver disease by integration of metabolomics and transcriptomics profiling

Yin Qin,Ruyan Fan,Yingxin Liu,Shuyi Qiu,Lu Wang

Nonalcoholic fatty liver disease (NAFLD), as the commonest chronic liver disease, is accompanied by liver oxidative stress and inflammatory responses. Herein, the extract obtained from Rubus corchorifolius fruits was purified and characterized for its polyphenol composition. The liver protective effect of the purified R. corchorifolius fruit extract (RCE) on mice with high-fat-diet (HFD)-induced NAFLD were investigated, and the potential mechanisms were explored through the integration of transcriptomics and metabolomics. Results showed that the polyphenolic compounds in RCE mainly included (−)-epigallocatechin, procyanidin B2, keracyanin, vanillin, dihydromyricetin, and ellagic acid. In addition, RCE intervention ameliorated liver and mitochondrial damage, which was evidenced by decreased indices of oxidative stress, liver function markers, and lipid profile levels. The liver metabonomics research revealed that RCE intervention affected the metabolic pathways of metabolites, including linoleic acid metabolism, galactose metabolism, alanine, aspartate and glutamate metabolism, retinol metabolism, glycine, serine and threonine metabolism, tryptophan metabolism, aminoacyl-tRNA biosynthesis, riboflavin metabolism, starch and sucrose metabolism, and arachidonic acid metabolism. Additionally, liver transcriptomics research indicated that pathways like fatty acid degradation, circadian rhythm, valine, leucine and isoleucine degradation, primary bile acid biosynthesis, cytokine–cytokine receptor interaction, adipocytokine signaling pathway, glutathione metabolism, lipid and atherosclerosis were significantly enriched. The transcriptomics and metabolomics analysis demonstrated that RCE intervention had significant modulatory effects on the metabolic pathways associated with glycolipid metabolism. Moreover, RT-PCR results verified that RCE intervention regulated liver mRNA levels associated with the inflammatory response. Therefore, our findings suggest that the intake of RCE might be an effective strategy to alleviate liver damage.

The Evaluation of the Phytochemical Profiles and Antioxidant and α-Glucosidase Inhibitory Activities of Four Herbal Teas Originating from China: A Comparative Analysis of Aqueous and Ethanol Infusions

Jin Zhang,Jinling Lv,Guodong Zhuang,Junjia Zhang,Feng Hu,Yongsheng Chen

Recent research has demonstrated the positive impact of herbal tea consumption on postprandial blood glucose regulation. This study conducts a comparative analysis of aqueous and ethanol extractions on four herbal teas (Mallotus, Cyclocarya, Rubus, and Vine) to assess their phytochemical profiles and functional attributes. Phytochemical contents, antioxidant activities, α-glucosidase inhibitory activities, and chemical compositions are investigated via colorimetric analyses and UPLC-Q-Orbitrap HRMS/MS, respectively. Results indicate that Vine, among the teas studied, exhibits the most pronounced glucose-regulating effects under both extraction methods. While ethanol extractions yield higher phytochemical content overall, the compositions vary. Conversely, aqueous extracts demonstrate unexpectedly potent antioxidant activities and comparable α-glucosidase inhibitory activities to ethanol extracts. Phytochemical contents correlate positively with antioxidant activities and α-glucosidase inhibitory activities. However, antioxidant activities exhibit a weak positive correlation with α-glucosidase inhibitory activities. These findings provide evidence that aqueous extracts from herbal teas contain valuable phytochemical compositions beneficial for antioxidants and individuals with hyperglycemia, suggesting their potential as functional ingredients to enhance the nutritional value of herbal food products.

Screening of Monoamine Oxidase B Inhibitors from Fragaria nubicola by Ligand Fishing and Their Neuroprotective Effects

Yi-Kao Hu,Yi-Ming Liu,Xiao-Lin Bai,Chao Ma,Xun Liao

Fragaria nubicola, known as Tibetan strawberry, is an edible plant possessing various health-promoting effects. However, its functional compositions were rarely studied. In this work, monoamine oxidase B (MAO-B) inhibitors in this plant were rapidly screened using the enzyme-functionalized magnetic nanoparticles coupled with UPLC-QTOF-MS. Two inhibitors, quercetin-3-O-β-d-glucuronide-6″-methyl ester (1) and kaempferol-3-O-β-d-glucuronide-6″-methyl ester (2), were identified from this plant with the IC50 values of 19.44 ± 1.17 and 22.63 ± 1.78 μM, respectively. Enzyme kinetic analysis and molecular docking were carried out to investigate the mechanism of inhibition. Contents of both compounds as well as those of total phenolics and flavonoids were quantified to be 24.76 ± 1.26, 35.59 ± 1.17, 837.67 ± 10.62, and 593.46 ± 10.37 μg/g, respectively. In addition, both compounds exhibited significant neuroprotective effects on 6-hydroxydopamine-induced PC12 cells. This is the first report on the neuroprotective components of F. nubicola, suggesting its potential for developing neuroprotective functional food.
