Mussel-inspired hydrogel with injectable self-healing and antibacterial properties promotes wound healing in burn wound infection

Sun Ao,Hu Danrong,He Xinye,Ji Xiao,Li Tao,Wei Xiawei,Qian Zhiyong

Burn wound infections cause serious problems for public health. More than 180,000 patients die from burns every year worldwide. In addition, the difficulty of healing wounds and wound infections caused by burns affects the mental health of patients. Therefore, it is very important to develop a wound dressing that can promote wound repair and exhibits good antibacterial effects. Here, we used oxidized konjac glucomannan (OKGM), γ-poly(glutamic acid) modified with dopamine and L -cysteine (γ-PGA-DA-Cys) and ε-polylysine (ε-PL) to produce an OKGM/γ-PGA-DA-Cys/ε-PL (OKPP) hydrogel. This hydrogel was produced by thiol-aldehyde addition and Schiff-base reactions and has the ability to be injected and self-heal. The results showed that the hydrogel exhibits good antibacterial effects on Pseudomonas aeruginosa and Staphylococcus aureus and has antioxidant effects in vitro. Moreover, the hydrogel also exhibits good adhesion. In a burn wound infection model, the hydrogel promoted wound healing and reduced the production of inflammation. These results proved that the hydrogel has clinical potential as a wound dressing for burn wound infection.

Silkworm cocoon bionic design in wound dressings: A novel hydrogel with self-healing and antimicrobial properties

Dong Zhang,Lei Zhao,Xiaohu Cui,Xinpeng Li,Zhisong Qian,Xueyan Zhou,Zhongren Ma,Mohd Sobri Takriff,Zhiqiang Li,Ying Niu,Guilan Ma,Gongtao Ding,Zifan Wang

Hydrogels with rapid wound-healing capabilities and antimicrobial effects are gaining significant interest in related fields. Nonetheless, developing a multifunctional hydrogel wound dressing with injectable self-assembling, self-healing, antimicrobial properties, and efficient skin wound-healing capabilities remained a formidable challenge. In this experiment, we drew inspiration from silkworm cocoons' natural formation and protective mechanisms, employing a novel physical cross-linking method to create an injectable and self-healing quaternary hydrogel successfully. The hydrogel is based on a matrix of silk fibroin/silk sericin (SF/SS), with 1,2-dimyristoyl-sn-glycero-3-phosphate sodium salt (DMPG) serving as a physical cross-linking agent to form the hydrogel network structure, and the incorporation of silver nanoparticles (AgNPs) further enhances its antimicrobial capabilities. Our biomimetic hydrogel, which replicated the chemical properties of silkworm cocoons, demonstrated excellent hydrophilicity with a water contact angle that ranged from 37 to 52°. Its tensile and compressive resistance was approximately four times greater than that of a pure SF hydrogel, and its swelling performance was about three times higher than that of a pure SF hydrogel. Furthermore, the hydrogel exhibited an impressive bacterial inhibition rate of over 98 % in bacterial growth and inhibition experiments, which provided a solid foundation for accelerating wound healing. Likewise, experiments with mice and histological analyses revealed that on day 7, the expression of TNF- α and IL-1 β in the wound tissues treated with the SF/SS/AgNPs hydrogel was significantly reduced by >25 % compared to the blank control group. This reduction indicates that the hydrogel could decrease the production of inflammatory cytokines, potentially aiding in the acceleration of wound healing and mitigation of inflammation-related adverse reactions. By day 14, the wounds were healed mainly, with the wound area reduced by 17 % compared to that of the blank group. This demonstrates the significant potential of this cocoon-mimetic hydrogel in accelerating wound healing and providing wound protection.

Injectable and self-healing dual-network chitosan/hyaluronic acid/polypeptide (CHP) antibacterial hydrogels for wound healing

Peng He,Guanghui Gu,Youyin Xu,Gang Wei,Mingjin Xu

Skin wound healing faces challenges of bacterial infection and environmental stimulation, and hence the design of biocompatible wound dressings with antibacterial and self-healing abilities is promising. Herein, we report the design and synthesis of bioactive composite hydrogels (CHP) with chitosan (CS), hyaluronic acid (HA), and polypeptide (PP). Ascribing to dynamic molecular cross-linking, the fabricated CHP hydrogels exhibit good stability, excellent biocompatibility, injectability, self-healing ability, and antimicrobial activity. Microscopic analysis indicates that the CHP hydrogels maintain a 3D porous dual-network structure, and the porosity of hydrogels is related to the content of PP as the reinforcement. Antimicrobial evaluation of the CHP hydrogels demonstrates significant efficacy against E. coli and S. aureus , with improved performance at higher PP concentration. Furthermore, the hydrogels show enhanced cytocompatibility, minimal hemolysis, and improved in vitro cell proliferation. The tests with wound mouse models indicate that the CHP hydrogels can accelerate wound healing effectively, enhance angiogenesis, and promote tissue regeneration. Histological evaluation confirms complete epidermal regeneration, improved collagen deposition, and increased protein expression. The designed CHP hydrogels present potential clinical applications as a wound dressing material to meet the needs of rapid and effective wound healing.

An injectable, self-healing composite hydrogel with enhanced near-infrared photo-antibacterial therapeutic effects for accelerated wound healing

Jiameng Wang,Hao Cheng,Weiyi Chen,Peide Han,Xiaohong Yao,Bin Tang,Wangping Duan,Pengcui Li,Xiaochun Wei,Paul K. Chu,Xiangyu Zhang

Antibacterial hydrogels with injectable and self-healing properties have attracted much attention in the field of wound dressings because they not only can prevent bacterial infections, but also meet the basic needs for wound healing. However, how to achieve high antibacterial efficacy and low drug resistance for hydrogels is still a challenge. Herein, an injectable, self-healing, near-infrared (NIR) photosensitive antibacterial hydrogel composed of CuS-grafted-curcumin ( [email protected] ) and carboxymethyl cellulose modified with aldehyde groups and hydroxypropyl trimethyl ammonium chloride chitosan is prepared for wound dressings. The p-n junction formed between curcumin and CuS promotes separation of electron-hole pairs, enhances the mobility of photogenerated charges, and destroys the conjugated structure of curcumin simultaneously consequently increasing the photocatalytic activity. [email protected] is immobilized and distributed uniformly in the hydrogel via the π-π conjugation ring. The hybrid hydrogel exhibits excellent antibacterial activity after irradiation with 808 NIR light for 10 min due to the enhanced photodynamic and photothermal antibacterial effects. In addition, the hybrid structure improves the biocompatibility of CuS and expedites infected-wound healing at a mild temperature of 45 ℃. This organic/inorganic hybrid is shown to be an excellent wound dressing for the treatment of bacterial-infected wounds.

Injectable chitosan hydrogels tailored with antibacterial and antioxidant dual functions for regenerative wound healing

Yajie Zhong,Farzad Seidi,Yilin Wang,Ling Zheng,Yongcan Jin,Huining Xiao

Herein, an injectable self-healing hydrogel with inherent antibacterial activity was fabricated based on the dynamic covalent bond formation between boronic acid and catechol groups in quaternized chitosan as building blocks in conjunct with the in-situ encapsulation of epigallocatechin-3-gallate (EGCG, a green tea derivative). Benefiting from the catechol groups and therapeutic effect of EGCG, the resulting natural-based injectable hydrogels showed excellent antibacterial and antioxidant dual functions in preventing bacterial infection and scavenging radicals. The hydrogels loaded with EGCG also exhibited good biocompatibility along with contact-active antibacterial activity via a “capture and kill” strategy. The dynamic covalent bonding enables shear-thinning delivery of the gels via syringe, followed by rapid self-healing. Additionally, the in vivo wound healing evaluation in a full-thickness skin defect model revealed the appreciable and regenerative wound healing performance of the hydrogels, demonstrating their great potential as novel wound dressings for wound management.

Self-Healing Nanozyme Hydrogel with Nitric Oxide Production and Photothermal Effect to Promote Wound Healing

Huiyuan Bai,Zihan Ding,Maorong Jiang,Dengbing Yao

Bacterial-infected skin wounds are a severe global healthcare problem. Bacterial invasion and an immoderate inflammatory response are major obstacles in the wound healing process. To mitigate these issues, a multifunctional nanozyme hydrogel dressing (FeCu/S/OxC/G) was prepared by loading sodium nitroprusside and iron–copper nanoparticles into a hydrogel network that was made of sodium periodate oxidized chondroitin sulfate, gelatin, and borax. Owing to different types of dynamic bonds, including Schiff base bonds and hydrogen bonds, the hydrogel showed self-healing ability, good injectability, and excellent adhesive performance. In vitro results demonstrated that the FeCu/S/OxC/G + H2O2 + NIR group exhibited a higher inactivation rate of Staphylococcus aureus and fewer biofilms than other groups, indicating the combined antibacterial effects of POD-like activity, nitric oxide, and photothermal therapy. In vivo results verified that FeCu/S/OxC/G hydrogel together with NIR laser irradiation could maximally increase the wound healing rate of S. aureus-infected mice via promoting epidermal formation, accelerating collagen deposition, and reducing the expression of inflammatory factors. This study provided a promising therapeutic strategy with combined antibacterial and anti-inflammatory effects for wound healing.

Glucose-responsive, self-healing, wet adhesive and multi-biofunctional hydrogels for diabetic wound healing

Zhuo Huang,Min Wang,Langjie Chai,Hang Chen,Danyang Chen,Yulin Li,Hongtao Liu,You Wu,Xuxia Yang,Lu He,Longjian Xue,Yifeng Lei,Liang Guo

Diabetic wounds are serious clinical complications which manifest wet condition due to the mass exudate, along with disturbed regulation of inflammation, severe oxidative stress and repetitive bacterial infection. Existing treatments for diabetic wounds remain unsatisfactory due to the lack of ideal dressings that encompass mechanical performance, adherence to moist tissue surfaces, quick repair, and diverse therapeutic benefits. Herein, we fabricated a wet adhesive, self-healing, glucose-responsive drug releasing hydrogel with efficient antimicrobial and pro-healing properties for diabetic wound treatment. PAE hydrogel was constructed with poly(acrylic acid- co -acrylamide) (AA-Am) integrated with a dynamic E-F crosslinker, which consisted of epigallocatechin gallate (EGCG) and 4-(2-acrylamidoethylcarbamoyl)-3-fluorophenylboronic acid (AFPBA). Due to the dynamic crosslinking nature of boronate esters, abundant catechol groups and hydrogen bonding, PAE hydrogel demonstrated excellent mechanical properties with about 1000 % elongation, robust adhesion to moist tissues, fast self-healing, and absorption of biofluids of 10 times of its own weight. Importantly, PAE hydrogel exhibited sustained and glucose-responsive release of EGCG. Together, the bioactive PAE hydrogel had effective antibacterial, antioxidative, and anti-inflammatory properties in vitro , and accelerated diabetic wound healing in rats via reducing tissue-inflammatory response, enhancing angiogenesis, and reprogramming of macrophages. Overall, this versatile hydrogel provides a straightforward solution for the treatment of diabetic wound, and shows potential for other wound-related application scenarios.

A biomimetic nanofiber composite hydrogel with tissue adhesion, self-healing and antibacterial ability for infected wound healing

Yinghao Tian,Xiaogang Bao,Shunmin Wang,Chen Tang,Nianqi Wu,Guifei Li,Kaixuan Ren,Jingbo Yin,Shifeng Yan,Guohua Xu

Skin injuries represent a significant clinical challenge, as conventional dressings frequently induce secondary trauma and microbial infiltration due to suboptimal barrier properties, ultimately delaying tissue repair. Ideal wound dressings should not only replicate the structure of native skin tissue but also create an environment conducive to cell viability. In this study, an injectable nanofiber composite self-healing hydrogel was developed for treating infected wounds. The antimicrobial properties of the hydrogel were achieved through the adsorption of branched polyethyleneimine (PEI) on gelatin fibers, while its self-healing capabilities were enhanced via Schiff base reactions and its tissue adhesion was strengthened by the incorporation of dopamine. Results demonstrated that the hydrogel exhibited strong biocompatibility and antimicrobial activity, promoted macrophage polarization towards the M2 phenotype, effectively suppressed inflammation, and facilitated wound healing in an infected wound model. Statement of Significance Wound infections pose a significant clinical challenge, often impeding healing and, in severe cases, leading to ulceration or life-threatening complications. In this study, a gelatin nanofiber composite hydrogel (PGF@ALG/PLGA hydrogel) functionalized with branched polyethyleneimine (PEI) was developed to address infected wounds through a biomimetic structure and enhanced pro-healing properties. The gelatin nanofibers within the hydrogel matrix facilitated electrostatic immobilization of PEI, effectively mitigating its inherent cytotoxicity by restricting free cationic charge exposure while ensuring localized surface enrichment. The resulting hydrogel exhibited robust tissue adhesion and autonomous self-healing capability. In infected wound models, the PEI-modified nanofibers within PGF@ALG/PLGA hydrogels demonstrated obvious antibacterial efficacy and promoted macrophage polarization to the M2 phenotype, synergistically accelerating the transition from the inflammatory phase to tissue regeneration. These findings underscore the therapeutic potential of PGF@ALG/PLGA hydrogel as a multifunctional platform for managing chronic infected wounds.

Injectable self-healing hydrogels loaded with Crinis Carbonisatus nanoparticles for rapid hemostasis and wound healing

Rui Tian,Fazhen Luo,Yilin Yu,Jinxia Mi,Xiaoya Gao,Zhengtao Wang,Yan Xie

Uncontrolled hemorrhage and infection are leading causes of post-traumatic deaths in both clinical and battlefield settings. Although several wound dressings have been developed, rapid and effective hemostasis and wound healing promotion in irregular and non-compressible wounds remain serious challenges. In this study, a multifunctional hydrogel (HPCS-C/ODEX/CC hydrogel) was developed using two natural polysaccharide derivatives, catechol-modified hydroxypropyl chitosan (HPCS-C) and oxidized dextran (ODEX), as well as Crinis Carbonisatus (CC) nanoparticles for rapid hemostasis and wound healing. The hydrogels exhibited favorable injectability and self-healing properties due to dynamic Schiff bonds between HPCS-C and ODEX. The introduction of catechol molecules in HPCS-C enhanced tissue adhesion, while the incorporation of CC nanoparticles also promoted the aggregation of blood cells to form a thrombus, enabling the hydrogel to rapidly establish a robust hemostatic barrier in various complex wounds. As a result, the HPCS-C/ODEX/CC hydrogel reduced bleeding in liver and tail hemorrhage models by 84.11-94.34% and 70.12-92.37%, respectively. The hydrogels also exhibited excellent biocompatibility, antibacterial, antioxidant, and wound healing promoting properties. Overall, HPCS-C/ODEX/CC hydrogels represent a promising dressing for rapidly and effectively stopping bleeding and promoting wound healing, offering a viable strategy for the treatment of uncontrollable bleeding and improving the survival of trauma patients.

Bio-inspired Lotus-fiber and Mussel-based Multifunctional Hydrogels for Wound Healing: Super-stretchability, Self-healing, Adhesion and Antibacterial Properties

Yang Xiaoling,Li Chenchen,Li Bo,Zhang Yuanyuan,Li Jinping,Liu Na,Nie Xin,Zhang Dawei,Zhou Ming,Liao Xiaoling

Hydrogel-based wound dressings, which facilitate rapid wound closure and healing, are essential for effective wound management. However, the development of an ideal hydrogel that possesses excellent mechanical properties, effective self-healing capabilities, tissue adherence and antimicrobial characteristics for wound dressing presents a significant challenge in clinical settings. Inspired by lotus-fiber and mussel, we synthesized a novel multifunctional hydrogel composed of bacterial cellulose-reinforced dopamine-grafted oxidized hyaluronic acid/polyacrylamide (OHA-DA/PAM/BC). This was achieved through a one-pot reaction that employed free radical polymerization of acrylamide, dynamic Schiff bonding and intermolecular hydrogen bonding. Compared with the pure PAM hydrogels, which exhibited an elongation at break of 4022% and a maximum tensile strength of 26.42 kPa, the OHA-DA/PAM hydrogel demonstrated significantly enhanced stretchability at 9949% and an increased tensile strength of 34.73 kPa when 0.3% OHA-DA was incorporated during hydrogel formulation. Notably, the addition of 0.8% BC significantly enhanced the tensile strength to 57.04 kPa and super-stretchability to 10679%. The OHA-DA/PAM/BC hydrogel also exhibited remarkable self-healing capabilities, achieving a mechanical recovery of 84.74% within 12 hours. Additionally, its adhesive and injectable properties are advantageous for dynamic wound repair. Furthermore, the OHA-DA/PAM/BC hydrogel exhibited minimal hemolytic activity and potent intrinsic antibacterial properties against both E. coli and S. aureus. In a mouse model of wound healing, this hydrogel reduced the healing duration to 14 days while enhancing the regeneration of both skin structure and function. Histological analyses further revealed that the hydrogel significantly promoted the development of well-organized granulation tissue, angiogenic tissue and collagen accumulation in the wound region. This study successfully developed an OHA-DA/PAM/BC multifunctional hydrogel characterized by exceptional stretchability, self-healing, adhesiveness, injectability and antibacterial activity, demonstrating a significant impact on wound healing in vivo. These findings indicated that the OHA-DA/PAM/BC hydrogel holds substantial potential as wound dressings for future clinical applications.

Dynamic covalent constructed self-healing hydrogel for sequential delivery of antibacterial agent and growth factor in wound healing

Mingmao Chen,Jia Tian,Yan Liu,Huan Cao,Ruyue Li,Jianhua Wang,Jiulin Wu,Qiqing Zhang

The conventional injectable hydrogels as wound dressing materials frequently encounter bacterial infection and lack self-healing ability. Besides, the hydrogels injected into the irregular wound sites are easily damaged by external forces, which further hinder wound healing. To make up for these deficiencies, we developed a novel hydrogel with the unique properties including dynamic covalent based self-healing, antibacterial and sequential drug release. This injectable and self-healing hydrogel was constructed via the dynamic reaction of aminated gelatin (NGel), adipic acid dihydrazide (ADH) and oxidized dextran (ODex). After injection or damage, the broken hydrogel pieces performed excellent self-healing capability to generate an integral hydrogel owing to dynamic imine and acylhydrazone bonds. Meanwhile, bFGF@PLGA microspheres and chlorhexidine acetate (CHA) were embedded into the hydrogels. This bFGF@PLGA/CHA/hydrogel presented a good biodegradability, and it could retain the morphological stability after swelling. Moreover, bFGF@PLGA/CHA/hydrogels achieved the sequential release of CHA and bFGF during the drug release assay. When topically applied onto the wound skin of rats, the hydrogels can effectively prevent infection at the early stages due to the rapid release of CHA, then further accelerate cell proliferation and wound healing through the sustained release of bFGF. These results indicate that the proposed self-healing hydrogel with sequential release properties is a highly promising dressing material for wound healing.
