Flat Silk Cocoon-Based Dressing: Daylight-Driven Rechargeable Antibacterial Membranes Accelerate Infected Wound Healing

Shixiong Yi,Ying Zhou,Jiamei Zhang,Min Wang,Shaohui Zheng,Xiao Yang,Lian Duan,Rui L. Reis,Fangyin Dai,Subhas C. Kundu,Bo Xiao

One of the leading causes of death globally, especially in underdeveloped countries, is bacterial infection. Recently, the prevalence of infections from antibiotic-resistant bacteria has been increasing, which makes the need for innovative antibacterial wound dressings urgent. It is reported that g-C 3 N 4 -based flat silk cocoons (FSCs) with rechargeable antibacterial activity can efficiently generate reactive oxygen species (ROS) under daylight irradiation. The photoactive FSCs store the ROS and then release them in the dark. The engineered FSCs exhibit integrated properties of good biocompatibility, strong mechanical characteristics, robust photoactivity with photostorability, and excellent bactericidal efficiency (99.9% contact killing). In a rat model of infected wounds, the photoactive FSCs induce faster healing and reduce bacterial infections. The successful application of these FSC materials as wound dressings may provide a versatile platform for exploring the use of green photoactive antibacterial materials for accelerated wound healing and prevention of infections.

MXene-based polysaccharide aerogel with multifunctional enduring antimicrobial effects for infected wound healing

Hui Li,Yuanli Yang,Min Mu,Chenqian Feng,Di Chuan,Yangmei Ren,Xiaoxiao Wang,Rangrang Fan,Jiazhen Yan,Gang Guo

Wound infection is a predominant etiological factor contributing to delayed wound healing in open wounds. Hence, it holds paramount clinical significance to devise wound dressings endowed with superior antibacterial properties . In this study, a Schiff base-crosslinked aerogel comprising sodium alginate oxide (OSA), carboxymethyl chitosan (CMCS), and Nb 2 C@Ag/PDA (NAP) was developed. The resultant OSA/CMCS-Nb 2 C@Ag/PDA (OC/NAP) composite aerogel exhibited commendable attributes including exceptional swelling characteristics, porosity, biocompatibility, and sustained antimicrobial efficacy. In vitro antimicrobial assays unequivocally demonstrated that the OC/NAP composite aerogel maintained nearly 100 % inhibition of Staphylococcus aureus and Escherichia coli under an 808 nm laser even after 25 h. Crucially, the outcomes of in vivo infected wound healing experiments demonstrated that the wound healing rate of the OC/NAP composite aerogel group reached approximately 100 % within a span of 14 days, which was significantly greater than that of the blank control group. In vitro and in vivo hemostatic experiments also revealed that the composite aerogel had excellent hemostatic properties. The results of this study demonstrate the remarkable potential of OC/NAP aerogel as a multifunctional clinical wound dressing, especially for infected wounds.

S-Nitrosylated CuS Hybrid Hydrogel Patches with Robust Antibacterial and Repair-Promoting Activity for Infected Wound Healing

Pengpeng Jia,Yu Zou,Jiang Jiang

Development of wound dressing with robust antibacterial and repair-promoting activity has always been an urgent biomedical task during the past years. The therapeutic effect of current hydrogel dressings containing single bioactive agent like nanoparticle or gas is still unsatisfactory for the treatment of infected wound. Herein, a CuS/NO co-loaded hydrogel (CuS/NO Gel) is proposed, which is constructed by sequential polymerization, reduction, and S-nitrosylation of CuS hybrid hydrogel with disulfide bonds. These CuS/NO Gel patches show good mechanical stability, high photothermal activity and excellent biocompatibility. When being applied to treat infected wound, CuS/NO Gel can not only eliminate infection effectively by the synergistic effect of mild photothermal heating and boosted NO release in infection phase, but also promote vascularization and collagen deposition due to the synchronous supply of Cu ion nutrients and low concentration NO signaling molecules in wound repair phase. Compared to hydrogel dressings with individual active agent (CuS Gel or NO Gel), CuS/NO Gel exhibits better antibacterial and repair-promoting activity both in vitro and in vivo. Therefore, this CuS/NO Gel holds great promise in the future clinical treatment against infected wound.

Hemoadhican Fiber Composite with Carbon Dots for Treating Severe Hemorrhage and Infected Wounds

Rui Fang,Ning Yu,Fa Wang,Xi Xu,Jianfa Zhang

Uncontrolled bleeding and infection following trauma continue to pose significant clinical challenges. This study employs hemoadhican (HD) polysaccharide, known for its superior hemostatic properties, as the foundational material to synthesize antibacterial carbon dots (H-CDs) through a hydrothermal method at various temperatures. The H-CDs exhibiting optimal antimicrobial properties were identified via in vitro antimicrobial characterization. The selected H-CDs possess nanoscale dimensions and a positive surface charge. They contain aldehyde groups and generate reactive oxygen species, which effectively eliminate bacteria. Subsequently, H-CDs were integrated into HD fibers (CDs-HD fibers) using a wet-spinning technique. The water vapor transmission rate, blood contact angle, and in vitro antimicrobial efficacy were evaluated. In a rat model of severe femoral artery hemorrhage and a noncompressible hepatic hemorrhage model, CDs-HD fibers demonstrated superior hemostatic performance compared to the commercially available QuikClot Combat Gauze. Furthermore, in a rat model of mixed bacterial wound infection, CDs-HD fibers significantly enhanced epithelial tissue remodeling and collagen deposition. In vivo studies confirmed the excellent biocompatibility of CDs-HD fibers. These findings suggest that CDs-HD fibers hold promise as a potential dressing for managing severe bleeding and preventing wound infections.

Essential Oil Nanoemulsion Hydrogel with Anti-Biofilm Activity for the Treatment of Infected Wounds

Kun Cai,Yang Liu,Yan Yue,Yuancheng Liu,Fengbiao Guo

The formation of a bacterial biofilm on an infected wound can impede drug penetration and greatly thwart the healing process. Thus, it is essential to develop a wound dressing that can inhibit the growth of and remove biofilms, facilitating the healing of infected wounds. In this study, optimized eucalyptus essential oil nanoemulsions (EEO NEs) were prepared from eucalyptus essential oil, Tween 80, anhydrous ethanol, and water. Afterward, they were combined with a hydrogel matrix physically cross-linked with Carbomer 940 (CBM) and carboxymethyl chitosan (CMC) to prepare eucalyptus essential oil nanoemulsion hydrogels (CBM/CMC/EEO NE). The physical-chemical properties, in vitro bacterial inhibition, and biocompatibility of EEO NE and CBM/CMC/EEO NE were extensively investigated and the infected wound models were proposed to validate the in vivo therapeutic efficacy of CBM/CMC/EEO NE. The results showed that the average particle size of EEO NE was 15.34 ± 3.77 nm with PDI ˂ 0.2, the minimum inhibitory concentration (MIC) of EEO NE was 15 mg/mL, and the minimum bactericidal concentration (MBC) againstS. aureuswas 25 mg/mL. The inhibition and clearance of EEO NE againstS. aureusbiofilm at 2×MIC concentrations were 77.530 ± 7.292% and 60.700 ± 3.341%, respectively, demonstrating high anti-biofilm activity in vitro. CBM/CMC/EEO NE exhibited good rheology, water retention, porosity, water vapor permeability, and biocompatibility, meeting the requirements for trauma dressings. In vivo experiments revealed that CBM/CMC/EEO NE effectively promoted wound healing, reduced the bacterial load of wounds, and accelerated the recovery of epidermal and dermal tissue cells. Moreover, CBM/CMC/EEO NE significantly down-regulated the expression of two inflammatory factors, IL-6 and TNF-α, and up-regulated three growth-promoting factors, TGF-β1, VEGF, and EGF. Thus, the CBM/CMC/EEO NE hydrogel effectively treated wounds infected withS. aureus, enhancing the healing process. It is expected to be a new clinical alternative for healing infected wounds in the future. Keywords:eucalyptus essential oil;nanoemulsion;hydrogel;antibacterial;anti-biofilm;infected wounds

Tannic acid-Fe3+ dual catalysis induced rapid polymerization of injectable poly(lysine) hydrogel for infected wound healing

Wenxia Guo,Yang Li,Chenhui Zhu,Zhiguang Duan,Rongzhan Fu,Daidi Fan

Infected wounds are difficult to heal because they are vulnerable to bacterial attacks, inflammatory responses, and oxidative stress . To promote the healing of infected wounds, we developed an injectable dual-network hydrogel TFAEP (TA-Fe, APS, EPL-GMA, PVA) based on ε-poly- l -lysine- graft -glycidyl methacrylate (EPL-GMA), polyvinyl alcohol (PVA), and tannic acid-iron (TA-Fe). TA-Fe formed a stable redox pair, which acted as a dual-autocatalytic system to activate ammonium persulfate , generate free radicals, and subsequently induce EPL-GMA polymerization. Then PVA formed hydrogen bonds with TA molecules. Here, TA-Fe not only simulated peroxidase to convert H 2 O 2 into hydroxyl radicals ( OH), but also exhibited good near-infrared photothermal conversion efficiency, which all endowed the hydrogel with excellent antibacterial ability. In addition, the hydrogel could remove excessive reactive oxygen species and reactive nitrogen species, alleviating oxidative stress and reducing inflammation response due to the presence of TA molecules. Moreover, the hydrogel showed good injectability and tissue adhesion, ensuring the close adhesion of the hydrogel to the wound and achieving the maximum function. In vivo experiments demonstrated that the hydrogel promoted infected wound healing by accelerating epidermal regeneration, promoting angiogenesis and collagen deposition, and facilitating the expression of anti-inflammatory factors.

Bioactive Metal Ion-Coordinated Dynamic Hydrogel with Antibacterial, Immunomodulatory, and Angiogenic Activities for Infected Wound Repair

Xinrui Li,Xu Chen,Lian Guan,Wenbo He,Weiling Yin,Dongcheng Ye,Jia Gao,Miao Wang,Guoqing Pan

The repair of infected wounds is a complex physiopathologic process. Current studies on infected wound treatment have predominantly focused on infection treatment, while the factors related to delayed healing caused by vascular damage and immune imbalance are commonly overlooked. In this study, an extracellular matrix (ECM)-like dynamic and multifunctional hyaluronic acid (HA) hydrogel with antimicrobial, immunomodulatory, and angiogenic capabilities was designed as wound dressing for the treatment of infected skin wounds. The dynamic network in the hydrogel dressing was based on reversible metal–ligand coordination formed between sulfhydryl groups and bioactive metal ions. In our design, antibacterial silver and immunomodulatory zinc ions were employed to coordinate with sulfhydrylated HA and a vasculogenic peptide. In addition to the desired bioactivities for infected wounds, the hydrogel could also exhibit self-healing and injectable abilities. Animal experiments with infected skin wound models indicated that the hydrogel dressings enabled minimally invasive injection and seamless skin wound covering and then facilitated wound healing by efficient bacterial killing, continuous inflammation inhibition, and improved blood vessel formation. In conclusion, the metal ion-coordinated hydrogels with wound-infection-desired bioactivities and ECM-like dynamic structures represent a class of tissue bionic wound dressings for the treatment of infected and chronic inflammation wounds.

A facilely prepared dual-crosslinking adhesive with enhanced adhesive strength for hemostasis and infected wound healing

Ziyuan Guo,Zongliang Wang,Peibiao Zhang

Although biological adhesives have shown advantages in replacing traditional wound suturing techniques, there are still limitations in wound closure, hemostasis, and healing, including insufficient tissue adhesion, potential biological toxicity, and complex preparation processes. In this study, a facile route for preparing injectable dual-crosslinking multifunctional hydrogel adhesive (Gel/EN/FBTA) was developed. The Gel/EN/FBTA adhesive is a dynamic cross-linked network composed of tannic acid (TA), 3-formylphenylboronic acid (3-FPBA) and gelatin, which can provide a large number of bonding sites and strengthen the adhesive cohesion through energy dissipation. The amidation reaction inside gelatin can form stable rigid crosslinks and maintain the structure of the adhesive stably. The balance between adhesion and cohesion can be regulated by adjusting the chemical composition and crosslinking density of the dual-crosslinking network. Under this equilibrium condition, the adhesion strength of Gel/EN/FBTA2 hydrogel is 3 times that of commercial fibrin glue, which shows good hemostatic effects in rat liver injury, rat tail injury, and rabbit liver cross incision models. Furthermore, Gel/EN/FBTA2 hydrogel adhesive can effectively treat wound infection, reduce inflammation level, promote re-epithelialization, accelerate collagen deposition, and achieve the healing of infectious full-thickness wounds. This dual-network design paradigm provides a versatile strategy for developing next-generation bioadhesives with tailored mechanical and bioactive properties, demonstrating significant potential for non-compressible hemorrhage and infected wound management.

A liposomal hydrogel with enzyme triggered release for infected wound

Dandan Li,Xueqin An,Yao Mu

A novel liposomal hydrogel with enzyme triggered release of drug as drug carrier was presented for usage of infected wound. The liposomal hydrogel was prepared by an acombined method of thin-film evaporation and supercritical carbon dioxide technique (TE-scCO 2 ) with freeze-drying. In the liposomal hydrogel, curcumin as a model drug was embedded in the bilayer of the liposomes, and the liposomes were encapsulated in the gelatin-chitosan hydrogel. Phospholipase A 2 (PLA 2 ) riched in wound exudate was served tactfully as a trigger to hydrolyze lecithin in liposome and destroy the liposomes to release drug. Hydrolysis of phospholipids in liposomes was monitored by fluorescence assay of the released curcumin, and relationship between the hydrolysis of lecithin and the release of drug was discussed. The microstructure changes of the liposome after treated by PLA 2 were probed by using florescence probeembedded in the liposomes for the first time. Besides, relationship between the microstructure of liposome and hydrolysis of liposome was revealed. This liposomal hydrogel can not only inhibit the degradation of drug in conveying process and enhance the efficiency of drug release to the infected skin, but also the release of drugs can be on demand by PLA 2 biological activity changed with wound exudate in infected tissue.

Enhanced heterogeneous interface to construct intelligent conductive hydrogel gas sensor for individualized treatment of infected wounds

Linguo Wan,Nanjian Xu,Xiaodong Wu,Mujie Liu,Yong Liu,Jinglong Zhao,Ting Zhang,Jingwei Zhao,Yu Zhou,Qingqing Xie,Yiwei Hu,Xiaoqing Jiang,Chen Tang,Yuping Quan,Shareen Shafique,Ye Tian,Xin Zhang,Yuejun Zhang,Kun Zhou,Jiangbei Cao,Jiawen Jian,Yuheng Wang

In this study, we developed an enhanced heterogeneous interface intelligent conductive hydrogel NH 3 sensor for individualized treatment of infected wounds . The sensor achieved monitoring, self-diagnosis, and adaptive gear adjustment functions. The PPY@PDA/PANI(3/6) sensor had a minimum NH 3 detection concentration of 50 ppb and a response value of 2.94 %. It also had a theoretical detection limit of 49 ppt for infected wound gas. The sensor exhibited a fast response time of 23.2 s and a recovery time of 42.9 s. Tobramycin (TOB) was encapsulated in a self-healing QCS/OD hydrogel formed by quaternized chitosan (QCS) and oxidized dextran (OD), followed by the addition of polydopamine-coated polypyrrole nanowires (PPY@PDA) and polyaniline (PANI) to prepare electrically conductive drug-loaded PPY@PDA/PANI hydrogels. The drug-loaded PPY@PDA/PANI hydrogel was combined with a PANI/PVDF membrane to form an enhanced heterogeneous interfacial PPY@PDA/PANI/PVDF-based sensor, which could adaptively learn the individual wound ammonia response and adjust the speed of drug release from the PPY@PDA/PANI hydrogel with electrical stimulation. Drug release and animal studies demonstrated the efficacy of the PPY@PDA/PANI hydrogel in inhibiting infection and accelerating wound healing . In conclusion, the gas-sensitive conductive hydrogel sensing system is expected to enable intelligent drug delivery and provide personalized treatment for complex wound management.

Sprayable and self-healing chitosan-based hydrogels for promoting healing of infected wound via anti-bacteria, anti-inflammation and angiogenesis

Yujiao Tan,Changliang Xu,Ying Liu,Yun Bai,Xiaoyun Li,Xiaoying Wang

Treatment of infected wound by simultaneously eliminating bacteria and inducing angiogenesis to promote wound tissue regeneration remains a clinical challenge. Dynamic and reversable hydrogels can adapt to irregular wound beds, which have raised great attention as wound dressings. Herein, a sprayable chitosan-based hydrogel (HPC/CCS/ODex-IGF1) was developed using hydroxypropyl chitosan (HPC), caffeic acid functionalized chitosan (CCS), oxidized dextran (ODex) to crosslink through the dynamic imine bond, which was pH-responsive to the acidic microenvironment and could controllably release insulin growth factor-1 (IGF1). The HPC/CCS/ODex-IGF1 hydrogels not only showed self-healing, self-adaptable and sprayable properties, but also exhibited excellent antibacterial ability, antioxidant property, low-cytotoxicity and angiogenetic activity. In vivo experiments demonstrated that hydrogels promoted tissue regeneration and healing of bacteria-infected wound with a rate of approximately 98.4 % on day 11 by eliminating bacteria, reducing inflammatory and facilitating angiogenesis, demonstrating its great potential for wound dressing.

Curcumin-derived water-soluble carbon dots without detectable resistance: Dual potentials for antimicrobial activity and infected wound healing

Jinyan Du,Shiji Liu,Jinrui Hou,Xinyue Wu,Haojie Si,Xingya Guo,Shujuan Zhuo

Due to the abuse of traditional antibiotics, the increasing prevalence of antibiotic resistance poses a significant challenge to the finding of novel antimicrobial materials. Curcumin, a natural polyphenolic compound, exhibits broad-spectrum antibacterial, anti-inflammatory, and antioxidant properties. However, its therapeutic utility is limited by its bioavailability. To improve the biological activity of curcumin, the highly water-soluble carbon dots ( E -CDs) were synthesized by a one-step hydrothermal method using curcumin and ethylenediaminetetraacetic acid disodium salt (Na 2 EDTA) as precursors. It was shown that the surfaces of E -CDs still remain the structural and functional groups of their precursors. The as-obtained E -CDs exhibited a higher antimicrobial efficacy against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ) with the minimum inhibitory concentration (MIC) of only 40 and 150 ppm, respectively. Strikingly, the E -CDs did not induce significant bacterial resistance while exhibiting excellent biocompatibility. In a mouse model of bacterial-induced wound infection, the wound closure had approached 60 % within just four days, highlighting their significant potential for accelerating wound healing. Our experimental results convincingly demonstrated that the E -CDs are a promising candidate for clinical applications in treating infected wounds and combating drug-resistant bacterial infections.
