The objective of this study was to investigate the anti-inflammatory effect of chlorogenic acid methyl ester (CME) and the molecular mechanism involved, through using non-infectious inflammation and infectious inflammation animal models as well as lipopolysaccharide (LPS)-stimulated mouse macrophage RAW264.7 cell models. Our results demonstrated that CME markedly inhibited ear swelling, paw swelling and granuloma swelling, and decreased intraperitoneal capillary permeability in non-infectious inflammation. Moreover, CME significantly alleviated the pathological damage of the lung tissue, reduced the levels of PGE2 and IL-1β in the serum and the protein expression levels of related-inflammatory factors in the lung tissue of LPS-induced mice with acute lung injury (ALI). In addition, CME affected the RAW264.7 cell cycle and inhibited the protein expressions of COX-2 and NLRP3 and prevented the phosphorylation of NF-κB p65 in RAW264.7 cells treated with LPS. These observations not only validated the anti-inflammatory effects of CME, but also revealed the underlying molecular basis, which involves the down-regulation of the expression of inflammatory factors and blockade of the COX-2/NLRP3/NF-κB signaling pathway.

MRSA -induced arthritis is a prevalent and highly debilitating orthopedic condition. The inflammatory response induced by bacterial infection hinders tissue repair and exacerbates bone loss. Traditional antibiotic therapies are limited by low bioavailability, substantial side effects, and narrow efficacy, rendering them inadequate for comprehensive treatment of arthritis. Nitric oxide (NO) has demonstrated considerable potential in overcoming bacterial resistance, modulating immune responses, and facilitating tissue repair. Therefore, a stage-specific NO release strategy, tailored to the distinct phases of bacterial arthritis, is essential for effective treatment. In this study, mesoporous polydopamine nanoparticles were utilized as NO donors (mPDA/NONOate) and encapsulated within a supramolecular hydrogel formed via the host-guest interaction between α-cyclodextrin (α-CD) and Pluronic F127. The injectable nature of the resulting NO/PDA-Gel hydrogel ensured uniform distribution within irregular bone joint infection sites, minimizing NO donor loss and enhancing local bioavailability. Notably, upon near-infrared (NIR) irradiation, the hydrogel induces a rapid increase in local temperature, facilitating rapid NO release. At the same time, the synergistic photothermal effect effectively kills bacteria and rapidly controls the infection. Without light irradiation, NO is sustainably and stably released from the NO/PDA-Gel, modulating the bone immune microenvironment, alleviating inflammation, promoting chondrocyte proliferation and differentiation, and accelerating bone tissue repair, thus significantly shortening the healing time of MRSA -induced arthritis. In conclusion, the injectable self-assembled NO/PDA-Gel offers a precise, stage-matched therapeutic approach for MRSA -induced arthritis and holds promise for the treatment of deep-seated infections caused by other multidrug-resistant pathogens.

The emergence of SARS-CoV-2-Spike mutants not only enhances viral infectivity but also lead to treatment failure. Gaining a comprehensive understanding of the molecular binding mode between the mutant SARS-CoV-2-Spike and human ACE2 receptor is crucial for therapeutic development against this virus. Building upon our previous predictions and verifications regarding heightened viral infectivity of six potential SARS-CoV-2-Spike mutants, this study aims to further investigate the potential disruption of the interaction between these mutants and ACE2 by quercetin, a Chinese herbal compound. Molecular docking and dynamics simulations results reveal that the binding sites of quercetin particularly enriched around a specific “cavity” at the interface of Spike/ACE2 complex, indicating a favorable region for quercetin to interfere with Spike/ACE2 interaction. Virus infection assay confirms that quercetin not only attenuates wild-type virus infectivity but also suppresses the infectivity of all six tested SARS-CoV-2-Spike mutants. Therefore, quercetin represents a promising therapeutic candidate against both wild-type and potential future variants of SARS-CoV-2 exhibiting high viral infectivity.

This study was conducted to design and evaluate a wound dressing based on a polycaprolactone (PCL) nanofiber coated with gamma oryzanol (GO) and chitosan (CS) in mice model. All the dressings were prepared by electrospinning method, and their morphology and physical properties were investigated. The mice were divided into five groups and treated with I) PCL-sole (PCL), III) PCL-mupirocin (PCL-M), IV) PCL-GO, IV) PCL-CS, and V) PCL-CS-GO. Wound area, total bacterial count, histopathological parameters, and expressions of IL-1β, TNF-α, IL-10, MMP-9, EGF, and VEGF were assessed. The fibers were randomly distributed in PCL group, but loading CS and GO increased the complexity and placing on the dressings. PCLs loaded with GO and CS showed lower viscosity, surface tension, and fiber diameter and higher conductivity and water contact angle compared to unloaded PCLs ( P < 0.05). The treatment with PCLs loaded with mupirocin, CS, and GO significantly reduced wound area and total bacterial count ( P < 0.05). Loading PCLs with mupirocin, CS, and GO decreased the expressions of IL-1β, TNF-α, MMP-9, but increased the expressions of IL-10 and VEGF compared to the unloaded PCL group ( P < 0.05). The most optimal responses to wound healing and physical parameters belonged to the PCL-CS-GO group.

Infected bone defects represent one of the most prevalent clinical conditions, affecting millions of patients annually. The local infection and necrosis associated with these defects exacerbate the injury, prolong healing times, and result in significant localized pain, presenting a substantial challenge for clinical repair. In this study, we developed a biomimetic mineralized and antibacterial imCOL1MA hydrogel by employing methacrylated COL1, composite native bone inorganic salts (CNBIS), and Magainin II-PLGA microspheres (mMicrospheres), which was further loaded with bone marrow stem cells (BMSCs) to form osteogenic engineered bone for infected bone defects repair. Briefly, we first optimized the concentration of COL1MA for BMSCs survival, then adjusted proportion of CNBIS to create an appropriate osteoinductive microenvironment, and encapsulated Magainin II in poly (lactic-co-glycolic acid) (PLGA) microsphere for long-term antimicrobial function. Consequently, the promising mineralized and antibacterial imCOL1MA was prepared using 10% COL1MA, 2% CNBIS, and 1% mMicrospheres. The imCOL1MA scaffold served as significant antimicrobial efficacy, excellent biodegradability, good biocompatibility, and osteoinductive microenvironment. As a result, the engineered bone could achieve rapid (only 4 weeks) vascularized and neuralized bone regeneration in a rabbit model of infected bone defects.

Currently, emerging dynamic therapy has gradually become a frequently used strategy for treating infectious bone defects via a rise in reactive oxygen species (ROS) levels, which can bring about oxidative harm to bacteria. However, ROS can be generated only under conditions of exogenous energy, limited by energy penetration or dependence on the existence of internal O2/H2O2. Thus, we designed Na2S2O8-decorated polyetheretherketone implants activated by Fe2+ for infected bone defects. In vitro experiments show that they generate sulfate radical (·SO4–) and hydroxyl radical (·OH) without O2/H2O2 existence, effectively killing bacteria. Additionally, the released Fe2+ enters bacteria and triggers ferroptosis-like death via lipid peroxidation. In vivo experiments show implants achieve an ideal effect of bone integration through a high-efficiency bactericidal effect and enhanced osteogenic activity. As envisioned, our proposed strategy offers a promising approach to halt refractory infection of bone tissue by autonomously catalyzing ROS storms and ferroptosis-like death, facilitating bone-defect recovery.

Maxillofacial trauma is an infection and oxidative stress-induced damage to maxillofacial area related cells. Increasing evidences have suggested that therapeutic strategies with controllable anti-infective and anti-inflammatory capabilities hold great potentials in the clinical treatment of maxillofacial trauma. Herein, we reported an infectious microenvironment responsive bimetallic sulfide (FeCuS x , FCS) combining with near infrared (NIR) irradiation for maxillofacial trauma therapy. In cellular levels, FCS exhibited the most efficient antioxidant and anti-inflammatory, cell migration, and angiogenesis behaviors through down-regulating the inflammatory factor (iNOS) expression level, up-regulating anti-inflammatory factors (Arg-1 and TGF-β) expression level, inducing macrophages M2 polarization (CD86 down-regulation and CD206 up-regulation), and promoting tissue repair factors (CD31) expression level. Besides, it presented the excellent antibacterial ability of Escherichia coli (99.74%) and MRSA (99.78%) for FCS + NIR. Significantly, it also confirmed that FCS + NIR presented the excellent functions of inhibiting wound infections, decreasing wound inflammation, as well as promoting trauma tissue repairing. Altogether, it had developed the strategy of FCS + NIR for the synergistic enhancement of infectious maxillofacial trauma healing, could also serve as a promising therapeutic strategy for the treatment of infectious diseases. Infectious bone defects represent a substantial challenge in clinical practice, necessitating the deployment of advanced therapeutic strategies. This study presents a treatment modality that merges a mild photothermal therapy hydrogel with a pulsed drug delivery mechanism. The system is predicated on a hydrogel matrix that is thermally responsive, characteristic of bone defect sites, facilitating controlled and site-specific drug release. The cornerstone of this system is the incorporation of mild photothermal nanoparticles, which are activated within the temperature range of 40–43 °C, thereby enhancing the precision and efficacy of drug delivery. Our findings demonstrate that the photothermal response significantly augments the localized delivery of therapeutic agents, mitigating systemic side effects and bolstering efficacy at the defect site. The synchronized pulsed release, cooperated with mild photothermal therapy, effectively addresses infection control, and promotes bone regeneration. This approach signifies a considerable advancement in the management of infectious bone defects, offering an effective and patient-centric alternative to traditional methods. Our research endeavors to extend its applicability to a wider spectrum of tissue regeneration scenarios, underscoring its transformative potential in the realm of regenerative medicine.

Quick and effective sterilization of drug-resistant bacteria inevitably became an ever-growing global challenge. In this study, a multifunctional composite (PDA/Cu-CS) hydrogel mainly composed of polydopamine (PDA) and copper-doped calcium silicate ceramic (Cu-CS) was prepared. It was confirmed that PDA/copper (PDA/Cu) complexing in the composite hydrogel played a key role in enhancing the photothermal performance and antibacterial activity. Through a unique “hot ions effect”, created by the heating of Cu ions through the photothermal effect of the composite hydrogel, the hydrogel showed high-efficiency, quick, and long-term inhibition of methicillin-resistant Staphylococcus aureus and Escherichia coli. In addition, the hydrogel possessed remarkable bioactivity to stimulate angiogenesis. The in vivo results confirmed that the “hot ions effect” of the composite hydrogel removed existing infection in the wound area efficiently and significantly promoted angiogenesis and collagen deposition during infectious skin wound healing. Our results suggested that the design of multifunctional hydrogels with “hot ions effect” may be an effective therapeutic approach for the treatment of infectious wounds.

Biomaterial-associated infections can occur any time after surgical implantation of biomaterial implants and limit their success rates. On-demand, antimicrobial release coatings have been designed, but in vivo release triggers uniquely relating with infection do not exist, while inadvertent leakage of antimicrobials can cause exhaustion of a coating prior to need. Here, we attach magnetic-nanoparticles to a biomaterial surface, that can be pulled-off in a magnetic field through an adhering, infectious biofilm . Magnetic-nanoparticles remained stably attached to a surface upon exposure to PBS for at least 50 days, did not promote bacterial adhesion or negatively affect interaction with adhering tissue cells. Nanoparticles could be magnetically pulled-off from a surface through an adhering biofilm, creating artificial water channels in the biofilm. At a magnetic-nanoparticle coating concentration of 0.64 mg cm −2 , these by-pass channels increased the penetrability of Staphylococcus aureus and Pseudomonas aeruginosa biofilms towards different antibiotics, yielding 10-fold more antibiotic killing of biofilm inhabitants than in absence of artificial channels. This innovative use of magnetic-nanoparticles for the eradication of biomaterial-associated infections requires no precise targeting of magnetic-nanoparticles and allows more effective use of existing antibiotics by breaking the penetration barrier of an infectious biofilm adhering to a biomaterial implant surface on-demand.

The clinical treatment of infectious bone defects is difficult and time-consuming due to the coexistence of infection and bone defects, and the simultaneous control of infection and repair of bone defects is considered a promising therapy. In this study, a dual-drug delivery scaffold system was fabricated by the combination of a three-dimensional (3D) printed scaffold with hydrogel for infected bone defects repair. The 3D printed polycaprolactone scaffold was incorporated with biodegradable mesoporous silica nanoparticles containing the small molecular drug fingolimod (FTY720) to provide structural support and promote angiogenesis and osteogenesis. The vancomycin (Van)-loaded hydrogel was prepared from aldehyde hyaluronic acid (AHA) and carboxymethyl chitosan (NOCC) by the Schiff base reaction, which can fill the pores of the 3D-printed scaffold to produce a bifunctional composite scaffold. The in vitro results demonstrated that the composite scaffold had Van concentration-dependent antimicrobial properties. Furthermore, the FTY720-loaded composite scaffold demonstrated excellent biocompatibility, vascularization, and osteogenic ability in vitro. In the rat femoral defect model with bacterial infection, the dual-drug composite scaffold showed a better outcome in both infection control and bone regeneration compared to other groups. Therefore, the prepared bifunctional composite scaffold has potential application in the treatment of infected bone defects.

An ideal and highly desired wound dressing for the repair of infectious wounds should provide a moist environment, close adhesion to the wound site, and self-healing and antibacterial properties. Thus, in the present study, we manufactured multifunctional, mussel-inspired hydrogels CACS-Fe 3+ -CYCS and CACS-Fe 3+ using catechol-grafted-chitosan (CACS) and cysteine-modified-chitosan (CYCS) as raw materials and ferric chloride as a cross-linking agent. The CACS-Fe 3+ -CYCS hydrogel exhibited excellent adhesion because of metal coordination between the ferric ion (Fe 3+ ) and catechol groups and a strong covalent bond between the catechol groups and the amino and sulfhydryl groups. Its self-healing quality was attributed to the dynamic and reversible properties of metal coordination. The loading of the total flavones of Resina Draconis (RDF) provided the hydrogel with good antibacterial properties. The antibacterial rates of the CACS-Fe 3+ -CYCS and RDF-CACS-Fe 3+ -CYCS hydrogels for Escherichia coli and Staphylococcus aureus were 90.62% and 96.45% and 99.98% and 100.00%, respectively. The RDF-CACS-Fe 3+ -CYCS hydrogel at pH 5.0, 6.8, and 8.0 in phosphate-buffered saline exhibited slow, sustained cumulative release rates of 53.94%, 71.97%, and 85.13% at 60 h, respectively. Complete wound healing was observed by day 12 in the CACS-Fe 3+ -CYCS hydrogel group (healing ratio = 96.59%). In the RDF-CACS-Fe 3+ -CYCS hydrogel group, complete healing was achieved by day 9 (healing rate = 94.46%; highest value at all treatment times). Thus, both hydrogels can be used as an ideal dressing for treating infectious wounds. The combination of traditional Chinese medicine and novel wound dressings can work synergistically, providing a novel strategy for designing biomimetic hydrogels for potential applications in wound healing.
