Negative pressure wound therapy (NPWT) offers significant advantages in terms of rate and time for healing through generating sub-vacuum to draw out inflammatory exudate and promote wound closure. However, continuous drainage probably leads to healing delay due to the lack of information about the real status of the wound bed and the potential risk of infection. To address this concern, printed Negative Pressure Smart Patch (NPSP) is reported by integrating smart real-time sensing acidity (infection) and glucose, and anti-infection into NPWT systems. In addition, NPSP delivers vancomycin through chitosan porous microspheres under negative pressure to modulate wound healing. Compared with NPWT, NPSP projects a promising approach to removing bacteria, reducing local inflammation, and accelerating healing in a short period of time.

The rapid detection of Pseudomonas aeruginosa ( P. aeruginosa ) is of great significance for the diagnosis of medical infection. In view of the above, a novel aptasensor based on fluorescence resonance energy transfer (FRET) was developed. It contained aptamer-coupled upconversion nanoparticles (UCNPs-apt) as a donor (excitation 980 nm) and molybdenum disulfide (MoS 2 ) nanosheets as an acceptor. The upconversion fluorescence aptamer system was investigated to obtain the optimal parameters of MoS 2 concentration, the incubation time of UCNPs-apt/MoS 2 and P. aeruginosa , and pH. Based on the optimal parameters, a linear calibration equation (emission 654 nm) with a wide detection range 8.7 × 10 ~ 8.7 × 10 7 cfu/mL, a high coefficient of determination R 2 0.9941, and a low limit of determination (LOD) 15.5 cfu/mL were established. The method was validated with P. aeruginosa infected foci of mouse wound. The advantage of this aptasensor is that analysis results can be obtained within 1.5 h, which was much faster than that of the standard method (18–24 h). Furthermore, combined with a portable instrument, it can be used as a point-of-care testing for the early detection of P. aeruginosa infection, which is useful for selecting the correct antibiotics to achieve good therapeutic effects. Additionally, it also has a broad application prospect in food and environmental areas. Graphical abstract

Fluorescent nanosensors based on ligand-engineered gold nanoclusters (Au NCs), guiding antibiotic therapy, offer a promising approach for rapid bacterial detection. Here, we developed dual-ligand-modified Au NCs with 6-azido-2-thiothymidine (ATT) and L-arginine (Arg), in which Arg modification enhanced the photoluminescence (PL) intensity compared to ATT-Au NCs. This enables a linear response (R2 > 0.97) to bacterial concentrations (105–109 CFU mL−1) within 30 minutes. Targeted metabolomics revealed that bacterial Arg deiminase metabolized Arg to citrulline, triggering PL quenching. Furthermore, it demonstrates the quantitative relationship between bacterial metabolic status and alterations in PL signals. The signal changes were visible under UV light and can also be quantified using a smartphone app (Light Meter). The detection limit reached 105 CFU mL−1. In vivo experiments confirmed that the probe can monitor wounds infected with Staphylococcus aureus (S. aureus), with minimal interference from biological matrices. This work highlights a portable, cost-effective strategy for point-of-care bacterial detection, addressing critical needs in clinical diagnostics.

The development of nanotheranostic agents is attracting attention as an innovative strategy to synergistically detect bacterial infection and eliminate bacteria. A novel MOF (ZIF-67)-armored zinc peroxide core–shell structure with a loaded organic NIR probe (ONP) is reported herein. These nanomaterials serve as “reactive oxygen species factories” that eradicate drug resistant bacteria with image guided and in situ activated photodynamic therapy (PDT). The up-regulated levels of ONOO − at infected sites can be detected by the ONP. ONP could react with ONOO − generating MB as an imaging agent and as a photosensitizer. The ZnO 2 @ZIF-67(ZZ) nanogenerator simultaneously generates O 2 and H 2 O 2 in an infected, weakened acidic microenvironment. The PDT can be applied on-demand to eliminate bacteria depending on illumination of the probe. The in situ activated PDT strategy can effectively exterminate drug-resistant bacteria, using small amounts of nanomaterials, with no obvious adverse effects in vitro and in vivo. The [email protected] 2 @ZIF-67( [email protected] ) nanoagent simultaneously eliminates drug-resistant bacteria, reduces inflammation, and accelerates wound healing. This novel nanotheranostic agent can act as an effective antibacterial platform by integrating infection detection and bacterial eradication synergistically, and bring new light for the increasingly severe challenge of drug-resistant bacteria in clinic.
