Dual-functional antimicrobial coating based on the combination of zwitterionic and quaternary ammonium cation from rosin acid

Chaoqi Chen,Zhaoshuang Li,Xiangzhou Li,Chuntao Kuang,Xiubo Liu,Zhanqian Song,He Liu,Yu Shan

Treatment of implant-associated infection is becoming more challenging, especially when bacterial biofilms form on the surface of implants. To address the challenge of biofilm infection, developing dual-functional antimicrobial coatings (antimicrobial and antifouling) to combat bacterial biofilm infection are superior to those based on a single modality due to avoiding the adverse effects arising from the latter. In this work, we reported an effective dual-functional coating based on the use of synthetic terpolymers, which combined dopamine, zwitterionic 2-methacryloyloxyethyl phosphorylcholine (MPC) and maleopimaric acid quaternary ammonium cation (MPA-N + ). The modified surfaces were characterized by scanning electron microscopy-energy dispersive spectroscopy (SEM-EDS), X-ray spectroscopy (XPS) and water contact angle measurements . The bactericidal efficacies of dual-functional coating were demonstrated by 1.00, 1.09 and 0.94 log reduction of Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli , Pseudomonas aeruginosa , respectively, and effectively inhibited pathogenic biofilm formation. Moreover, the dual-function coating reduced protein and platelet absorption, while showing minimal cytotoxicity to the mammalian cells. Notably, an in vivo implantation model showed that the dual-functional coating eradicated the pathogenic biofilm from the implants, preventing host tissue damage and inflammation. Taken together, this dual-functional coating displayed great therapeutic potential in treating the formidable clinic problems caused by pathogenic biofilm and the accompanying inflammation.

Oxygen-Immunomodulated Nanocatalysts Enhance Anti-Infection by Trained Immunity

Zesong Ruan,Tingwang Shi,Zhao Guo,Yu Zhu,Wenbo Wang,Yihong Ma,Cheng Ding,Yunlong Zhang,Xiaojun Wang,Yunfeng Chen,Han Lin,Min Ge

Biofilm infection microenvironment (BIM) in orthopedic implant-associated infections forms a robust barrier that resists antimicrobial agents and evades the host's immune system, yet there are currently limited effective targeted therapies against the characteristics of the BIM. Here, an emerging nanocatalytic immunotherapy strategy is proposed for treatment based on microenvironmental oxygen regulation. It is reported that the light-activated oxygen immune regulators (LAOIR) can effectively target and disrupt biofilms in the initial phase of phototherapy by generating singlet oxygen (¹O₂). The LAOIR-induced hypoxic microenvironment prolongs neutrophil lifespan and mitigates the immune tolerance induced by lipoteichoic acid (LTA) caused by killed bacteria. As the biofilm disintegrates and oxygen recovers, the neutrophil “immune switch” is triggered, and neutrophils are exhibited enhanced bactericidal activity during immune training with LTA exposure, such as NETosis and phagocytosis. Meanwhile, LAOIR therapy mediates neutrophils to express increased levels of proinflammatory cytokines IL-6, IL-1β, and the chemokine receptors CCR2 and CXCR2, promoting immune recruitment and resulting in a more robust therapeutic effect compared to vancomycin. In vitro and in vivo experiments have demonstrated that LAOIR outperform clinical antibiotics in the treatment of bacterial infections, providing direct evidence for the utilization of oxygen-modulated immune response strategies in the treatment of orthopedic implants.

Transdermal Delivery of Photosensitizer-Catalase Conjugate by Fluorinated Polyethylenimine for Enhanced Topical Photodynamic Therapy of Bacterial Infections

Hairong Wang,Yi Xie,Yanling Chen,He Zhao,Xinjing Lv,Zimu Zhang,Gen Li,Jian Pan,Jian Wang,Zhuang Liu

Pathogenic bacteria induce subcutaneous infections pose serious threats to global public health. Recently, photodynamic therapy (PDT) has been proposed as a non-invasive approach for anti-microbial treatment without the risk to induce drug resistance. However, due to the hypoxic environment of most anaerobiont-infected sites, the therapeutic efficacy of oxygen consuming PDT has been limited. Herein, a transdermal delivery system is reported to allow effective delivery of photosensitizers into infected skin for PDT treatment of skin infections by bacteria. Considering the overproduction of hydrogen peroxide (H 2 O 2 ) in the abscess area, catalase (CAT), an enzyme that triggers H 2 O 2 decomposition to generate O 2 , is conjugated with chlorine e6 (Ce6) to form a photosensitizer conjugate (Ce6-CAT) as an enhanced PDT agent against Staphylococcus Aureus . After screening a series of fluorinated low molecular weight polyethylenimine (F-PEI) with different fluorination degrees, the optimized F-PEI formulation is identified with the best transdermal delivery ability system. Upon mixing, the formed Ce6-CAT@F-PEI nanocomplex shows effective transdermal penetration after being applied to the skin surface. With light exposure of the infected skin, highly effective in vivo anti-bacterial PDT therapeutic effect with Ce6-CAT@F-PEI is observed. This work proposes a transdermal PDT therapeutic nanomedicine particularly promising for the anti-bacterial treatment of skin infections.

Photothermal activated Ni@ZIF-C nanocatalyst boosts triple-mode therapy of skin scaffold against bacterial and biofilm infections

Wenjing Yang,Xujie Zhang,Chenhang Ding,Wurikaixi Aiyiti,Cijun Shuai

Bacterial infections especially bacterial biofilm infections have always been a major obstacle to the reconstruction of deep skin defects. Herein, a photothermal activated multifunctional nanocatalyst (Ni@ZIF-C) is synthesized by doping Ni into zeolitic imidazolate framework-67 (ZIF-67) and carbonizing, following by introducing into polycaprolactone (PCL) to prepare porous scaffolds by selective laser sintering. Especially, the Ni@ZIF-C nanocatalyst endows scaffold favorable chemo/photodynamic/photothermal triple-mode antibacterial activity under near-infrared light irradiation. On the one hand, the highly conductive Ni@ZIF-C improves the separation efficiency of photogenerated electron-hole pairs to trigger reactive oxygen species (ROS) generation, achieving photodynamic sterilization. On the other hand, the superior photothermal conversion efficiency of Ni@ZIF-C in scaffold not only directly destruct bacterial biofilms by hyperthermia but also enhance its own peroxidase-like activity to induce more ROS production, and the latter is bactericidal by chemical mode. Importantly, the destruction of the biofilms enables the pre-generated ROS to kill deep bacteria, achieving a triple-mode combined or even synergistic antibacterial efficacy. Results demonstrated that the PCL/Ni@ZIF-C scaffold with triple bactericidal modalities presents stronger bactericidal ability than PCL/ZIF-67 and PCL scaffolds. Encouragingly, it remarkably destroyed S. aureus and E. coli biofilms and inhibited biofilm formation, effectively triggered massive ROS generation and ultimately eradicated the bacteria deep within the biofilms. Taken together, the scaffold exhibited tremendous potential in utilizing photothermal activated multimodal combined antibiofilm and antibacterial therapy.

A bacterially engineered macrophage sponge as a neutralization decoy to treat bacterial infection

Cheng Gao,Cheryl H.T. Kwong,Mian Tang,Jinwei Liu,Hiotong Kam,Shengke Li,Simon M.Y. Lee,Chunhai Fan,Hua-Zhong Yu,Ruibing Wang

Antibiotic-resistant strains have emerged, and endotoxins vary greatly in their structural motifs across bacterial species, limiting the clearance efficiency of structure-specific anti-endotoxin antibodies. This study constructs intracellularly gelated macrophages (GMs) with an intact membrane structure to neutralize bacteria and endotoxins via receptor-ligand interaction and share the bacterial burden to ameliorate in vivo infection without considering their molecular structure. Furthermore, GMs derived from bacterially infected macrophages (here referred to as bacterially engineered macrophages) are found to have upregulated membrane pathogen-related receptors for strengthened bacterium and endotoxin recognition and increased the membrane zeta potential to reduce the electrostatic repulsion force for facile GM-bacterium surface interaction. Bacterially engineered GMs achieve efficient management of sepsis and wound infection and targeted alleviation of bacterial pneumonia. The top-down fabrication of GMs provides a universal neutralization strategy against bacterial infection, and bacterially engineered host immune cells, upon intracellular gelation, demonstrate a natural cell engineering venue to obtain comprehensive neutralization decoys.

Baicalin Downregulates RLRs Signaling Pathway to Control Influenza A Virus Infection and Improve the Prognosis

Peng Pang,Ke Zheng,Sizhi Wu,Huachong Xu,Li Deng,Yucong Shi,Xiaoyin Chen

The objective of this study is to investigate the effects of baicalin on controlling the pulmonary infection and improving the prognosis in influenza A virus (IAV) infection. PCR and western blot were used to measure the changes of some key factors in RLRs signaling pathway. MSD electrochemiluminescence was used to measure the expression of pulmonary inflammatory cytokines including IFN-γ, TNF-α, IL-1β, IL-2, IL-4, IL-5, IL-6, IL-10, IL-12p70, and KC/GRO. Flow cytometry was used to detect the proportion of Th1, Th2, Th17, and Treg. The results showed that IAV infection led to low body weight and high viral load and high expression of RIG-I, IRF3, IRF7, and NF-κB mRNA, as well as RIG-I and NF-κB p65 protein. However, baicalin reduced the rate of body weight loss, inhibited virus replication, and downregulated the key factors of the RLRs signaling pathway. Besides, baicalin reduced the high expression inflammatory cytokines in lung and decreased the ratios of Th1/Th2 and Th17/Treg to arouse a brief but not overviolent inflammatory response. Therefore, baicalin activated a balanced host inflammatory response to limit immunopathologic injury, which was helpful to the improvement of clinical and survival outcomes.

Novel functionalized Ti6Al4V scaffold for preventing infection and promoting rapid osseointegration

Xianxing Xu,Hailun Xu,Qihao Chai,Ziyang Li,Zhentao Man,Wei Li

Total hip arthroplasty (THA) is the most common and radical treatment for end-stage hip disease. Prosthesis loosening and periprosthetic infection are the most serious complications after THA, which may lead to prosthetic dysfunction. The early prevention of infection and the promotion of fast osseointegration between the prosthesis and host bone are essential to the success of THA. However, Ti6Al4V does not have biological function, such as promoting osseointegration and antibacterial properties, which limits its widespread use. Therefore, it is necessary to design a functional hip prosthesis to simultaneously reduce the incidence of loosening and periprosthetic infection. In this study, we developed a novel functionalized coating via micro-arc oxidation modification combined with vancomycin to treat a prosthetic surface for the early postoperative period to resist bacterial infection and promote osseointegration. The functionalized coating simultaneously prevents periprosthetic infections and promotes osseointegration at the prosthesis–host bone interface. We successfully constructed a prosthetic coating with the dual function of promoting osseointegration and preventing infection, which provides a practical and effective method for the treatment of periprosthetic infection and prosthetic loosening after THA.

A self-assembling peptide inhibits the growth and function of fungi via wrapping strategy

Gao-Feng Qi,Xin Cui,Xue-Feng Gong,Xu Cui,Huan-Ge Xu,Qi-Lin Liang,Kuo Zhang,Xiao-Ling Sha,Litao Li,Gui-Yuan Wang,Hong-Wen Liang,Lei Wang

Fungal infections contribute substantially to human morbidity and mortality. Particular concern is the high rate of mortality associated with invasive fungal infections, which often exceeds 50.0% despite the availability of several antifungal drugs. Herein, we show a self-assembling antifungal peptide (AFP), which is able to bind to chitin on the fungal cell wall and in situ form AFP nanofibers, wrapping fungi. As a result, AFP limits the proliferation of fungi, slows the morphological transformation of biphasic fungi, inhibits the adhesion of fungi to host cells and the formation of biofilms. Compared to the broadspectrum antifungal fluconazole, AFP achieved the comparable inhibitory effect (MIC50 = 3.5 µM) on fungal proliferation. In addition, AFP significantly inhibits the formation of fungal biofilms with the inhibition rate of 69.6% at 1 µM, better than fluconazole (17.2% at 1 µM). In skin infection model of mice, it was demonstrated that AFP showed significantly superior efficacy than fluconazole. In systemic candidiasis mice model, AFP showed similar efficacy with first-line antifungal amphotericin B (AmpB) and anidulafungin (AFG). This study provides a promising wrapping strategy for the anti-fungal infection.

Dual inactivation mechanism of photocatalysis and metal ions induces broad-spectrum antibacterial polyethylene fabric to resist bio-infection

Jinqi Wang,Qianqin Zhu,Xiaolong Su,Jing Zhao,Wenjuan Wang,Tianqi Jiang,Senlong Yu,Gongxun Zhai,Hengxue Xiang,Meifang Zhu

Hospital-acquired infections (HAIs) remain a persistent challenge in healthcare settings, driving the urgent need for advanced protective textiles that offer both barrier and antimicrobial capabilities. Polyethylene (PE) fiber-based protective suits present a promising solution to mitigate HAIs, owing to their lightweight nature, high tensile strength, and chemical resistance—properties essential for extended wear in clinical environments. However, the intrinsic lack of antimicrobial activity in PE fibers restricts their efficacy in preventing cross-infections. In this work, we report the development of a scalable, environmentally sustainable PE@ZIF-8 antibacterial nonwoven fabric, fabricated via a basic chemical immersion process. This method involves -COOH grafting through a diazonium reaction, enabling the uniform growth of ZIF-8 nanocrystals on the PE fibers. The resulting fabric demonstrates a broad-spectrum antibacterial efficacy of 99.9 % against E. coli and B. subtilis , attributed to a dual mechanism involving photocatalysis and Zn²⁺ release. Furthermore, the PE@ZIF-8 fabric retains favorable hydrophobicity, permeation resistance, and mechanical properties, all of which meet practical performance standards for protective textiles. This work provides a significant advancement in infection control, addressing both antimicrobial protection and the practical wearability of PE-based protective gear, offering a viable solution for infection-prone medical environments.

Polysaccharide-Based Coating with Excellent Antibiofilm and Repeatable Antifouling-Bactericidal Properties for Treating Infected Hernia

Qifen Li,Leixiang Wang,Lu Yu,Chengbo Li,Xianrui Xie,Huanhuan Yan,Wenjuan Zhou,Chunhua Wang,Zhonghao Liu,Guige Hou,Yu-Qing Zhao

In recent years, the utilization of medical devices has gradually increased and implantation procedures have become common treatments. However, patients are susceptible to the risk of implant infections. This study utilized chemical grafting to immobilize polyethylenimine (QPEI) and hyaluronic acid (HA) on the surface of the mesh to improve biocompatibility while being able to achieve antifouling antimicrobial effects. From the in vitro testing, PP–PDA-Q-HA exhibited a high antibacterial ratio of 93% against S. aureus, 93% against E. coli, and 85% against C. albicans. In addition, after five rounds of antimicrobial testing, the coating continued to exhibit excellent antimicrobial properties; PP–PDA-Q-HA also inhibits the formation of bacterial biofilms. In addition, PP–PDA-Q-HA has good hemocompatibility and cytocompatibility. In vivo studies in animal implantation infection models also demonstrated the excellent antimicrobial properties of PP–PDA-Q-HA. Our study provides a promising strategy for the development of antimicrobial surface medical materials with excellent biocompatibility.

Natural polyphenolic antibacterial bio-adhesives for infected wound healing

Hengjie Zhang,Yuqi Feng,Tianyou Wang,Jianhua Zhang,Yuxian Song,Jing Zhang,Yiwen Li,Dingzi Zhou,Zhipeng Gu

Bio-adhesives used clinically, commonly have the ability to fill surgical voids and support wound healing, but which are devoid of antibacterial activity, and thus, could not meet the particular needs of the infected wound site. Herein, a series of natural polyphenolic antibacterial bio-adhesives were prepared via simple mixing and heating of polyphenols and acid anhydrides without any solvent or catalyst. Upon the acid anhydride ring opening and acylation reactions, various natural polyphenolic bio-adhesives could adhere to various substrates (i.e., tissue, wood, glass, rubber, paper, plastic, and metal) based on multi-interactions. Moreover, these bio-adhesives showed excellent antibacterial and anti-infection activity, rapid hemostatic performance and appropriate biodegradability, which could be widely used in promoting bacterial infection wound healing and hot burn infection wound repair. This work could provide a new strategy for strong adhesives using naturally occurring molecules, and provide a method for the preparation of novel multifunctional wound dressings for infected wound healing.

Platensimycin-Encapsulated Liposomes or Micelles as Biosafe Nanoantibiotics Exhibited Strong Antibacterial Activities against Methicillin-Resistant Staphylococcus aureus Infection in Mice

Zhe Wang,Xingyun Liu,Ying Peng,Meng Su,Saibin Zhu,Jian Pan,Ben Shen,Yanwen Duan,Yong Huang

Platensimycin (PTM) is a promising natural product drug lead against Gram-positive bacteria, including methicillin-resistant Staphylococcus aureus (MRSA), while the clinical development was hampered by problems related to its poor solubility and pharmacokinetic properties. In this study, we used liposomes and micelles as carriers of PTM to prepare PTM nanoformulations for the treatment of MRSA infection in mice. PTM-loaded nanoparticles could effectively reduce residual bacteria in the MRSA-infected macrophage cell model, comparing to free PTM. More importantly, in vivo studies showed that encapsulation of PTM by liposomes or micelles effectively improved the pharmacokinetic properties of PTM in Sprague–Dawley rats and the survival rate of MRSA-infected C57BL/6J mice. Our study has thus suggested that the clinically used nanocarriers, such as liposome and micelle, might also be useful to improve the efficacy of other natural product drug leads to accelerate their in vivo evaluation and preclinical development.
