Bone defect repair is a hot topic in tissue engineering, and the development of economical and effective bone scaffold materials to achieve the goal of repairing bone defects is an important task for researchers. In this experiment, fresh bovine cancellous bone was used as a xenogeneic
bone material. After two high-temperature calcinations, β-tricalcium phosphate (β-TCP) material was obtained. An in-situ co-precipitation method was employed to prepare hydroxyapatite+graphene oxide (HA+GO) composite material. Then, the β-TCP material
was immersed in the HA+GO composite material solution to prepare β-TCP+HA+GO composite material. The surface modification of β-TCP material with HA and GO improved the elastic modulus of the raw material. In vitro cell showed that the composite material was non-toxic,
biocompatible, and promoted the proliferation of MC3T3-E1 cells on the β-TCP+HA+GO material. Finally, it was found that β-TCP+HA+GO composite material effectively stimulated the actin and mineralization behavior of MC3T3-E1 cells, and promoted the expression of bone-related
factors such as Alkaline phosphatase (ALP), Collagen I (COLI), Osteocalcin (OCN), Bone Morphogenetic Protein-2 (BMP-2), and Bone Morphogenetic Protein-7 (BMP-7) in MC3T3-E1 cells In conclusion, β-TCP+HA+GO composite material can be applied in the biomedical field as a bone substitute
or repair material.

The Kirschner wire (K-wire) is widely used in orthopedic external fixation due to its versatility and clinical effectiveness. However, a significant challenge associated with its use is the potential for bacterial migration, subsequent infection, and dislodgement as the wire penetrates the skin and bone. This study introduces a novel bioactive material, selenium/calcium silicate (Se/β-CS), achieved by integrating selenium—an essential trace element in the human body—into bioceramic calcium silicate. This integration was accomplished using a combined chemical co-deposition method and redox reaction. Furthermore, a uniform and controllable Se/β-CS coating was applied to the K-wire's surface using the Langmuir-Blodgett technique. This coating gradually releases active components—Si, Ca, and Se—that effectively eliminate bacterial infections and promote osteointegration. The findings of this study offer promising opportunities for the use of robust and multifunctional coating materials on implantable devices, particularly within the fields of orthopedics, transplantation, and surgery.

Calcium sulfate (CaS) is a commonly used synthetic bone graft material, but it lacks antimicrobial properties. Magnesium oxide (MgO) has been reported to possess antimicrobial and osteogenic effects in vitro . The present study aimed to develop and evaluate MgO-CaS composites for their antimicrobial and biological properties. MgO nanoparticles were mixed with α-calcium sulfate hemihydrate at 0–25% (w/w). The resultant composites were characterized for setting time, compressive strength, degradation, pH change, and reactive oxygen species (ROS) generation. In vitro inhibition of three pathogens ( E. coli , S. aureus , C. alb .), disruption of bacterial biofilm formation, biocompatibility, alkaline phosphatase (ALP) expression, and angiogenic potential were studied. In vivo antibacterial ( S. aureus ) effects were also evaluated in a rat muscle model. The addition of MgO shortened the setting time and reduced compressive strength. In vitro , the composites exhibited a stable pH plateau and nearly linear degradation profiles; they inhibited bacterial biofilm formation by 18.1–62.0% and reduced the masses of pre-formed biofilms by 38.9–86.5%; they also reduced the formation of bacterial colonies by 87.5–99.0% and fungal colonies by 81.6–97.9%. In vitro biocompatibility decreased with increasing MgO content, but composites with 0–7.5% MgO were non-cytotoxic. ALP levels and angiogenic potential peaked at 2.5% and 7.5% MgO, respectively. In vivo , the composites significantly reduced the retrieved bacterial counts and white blood cell infiltration compared with CaS. These suggest that CaS-MgO composites are effective antimicrobial bone grafting materials with reasonable biocompatibility and potential to reduce infection rates.

Hydrogel-based materials exhibit great potential in tissue engineering. However, their mechanical weakness limits applications in hard tissue regeneration, especially under load-bearing conditions. Although various strengthening strategies have been applied, the achieved mechanical response of hydrogels still lags behind the mechanics of natural bone. In this study, we present a novel mineralization approach to fabricate mechanically robust and highly osteogenic mineralized hydrogels. Cross-linking between deprotonated chains of poly(acrylic acid) (PAA) and divalent cations has led to formation of hydrogels with a compressive strength and elastic modulus of 0.3±0.1 kPa and 1.3±0.2 kPa, respectively. Subsequent in situ formation of nano-calcium hydroxide crystals remarkably increased the compressive strength and modulus to 7.9±0.6 MPa and 339.3±31.4 MPa, respectively, surpassing those of trabecular bone. Moreover, the mineralized hydrogels demonstrated remarkable osteogenic potential in vivo, exhibiting immunoregulatory activity, promoting early angiogenesis, and accelerating fracture healing at weeks 4 and 8. The mechanism of osteogenesis was further revealed by transcriptome sequencing, indicating that the mineralized hydrogels regulated the translation of extracellular matrix and biomineralization. Overall, our study presents a pioneering and cost-effective method for fabricating materials with exceptional mechanical strength and strong osteogenic properties, offering a promising avenue for load-bearing bone repair applications of hydrogel-based materials.

Biomimetic hydrogels with satisfactory mechanical properties and osteogenic/angiogenic abilities have attracted extensive attention in recent years. Injectable hydrogels with the advantages of minimally invasive implantation and adaptability to irregular defects exhibit superior application potential in the field of bone regeneration. However, poor mechanical properties and limited self-healing ability hinder their potential as supporting materials. Furthermore, the absence of osteogenic and angiogenic properties significantly affect their application in bone regeneration. For injectable hydrogels, simulating natural bone tissue ECM presents an opportunity for developing innovative materials for bone repair. In this work, a strategy to prepare biomimetic nanofiber-reinforced dually cross-linked (DC) injectable hydrogels with self-healing, osteogenic and angiogenic properties was proposed. The PLGA/MDex/CaP@PBLG nanofiber hydrogels were crosslinked through two reversible dynamic interactions including Schiff base reaction and physical chelation, thereby demonstrating good self-healing property. Meanwhile, the incorporation of CaP@PBLG mineralized nanofibers and alendronate sodium (BP) grafted on PLGA could endow the hydrogels with favorable osteogenic function. Moreover, the introduction of antioxidant ferulic acid (FA) could significantly stimulate angiogenic activity of the hydrogels. The successful regeneration of cranial defects in rats demonstrated a potential application of PLGA/MDex/CaP@PBLG nanofiber hydrogels in the field of bone regeneration.

Piezoelectricity is reported to be able to promote bone scaffolds with excellent osteogenic performance. Herein, barium titanate/β-tricalcium phosphate (BTO/β-TCP) piezoelectric composite scaffolds were 3D printed, and their osteogenic performances were investigated in detail. The fabrication of BTO/β-TCP piezoelectric composite scaffolds employed cutting-edge DLP 3D printing technology. The scaffolds, featuring a triply periodic minimal surface (TPMS) design with a porosity of 60%, offered a unique structural framework. A comprehensive assessment of the composition, piezoelectric properties, and mechanical characteristics of the BTO/β-TCP scaffolds was conducted. Notably, an increase in the BTO volume fraction from 50 to 80 vol % within the scaffolds led to a reduction in compressive strength, decreasing from 2.47 to 1.74 MPa. However, this variation was accompanied by a substantial enhancement in the piezoelectric constant d33, soaring from 1.4 pC/N to 21.6 pC/N. Utilizing mouse osteoblasts (MC3T3-E1) in a live/dead cell staining assay, under the influence of external ultrasound, demonstrated the commendable biocompatibility of these piezoelectric composite ceramic bone scaffolds. Furthermore, thorough analyses of alkaline phosphatase (ALP) activity and polymerase chain reaction (PCR) findings provided compelling evidence of the scaffolds’ superior osteogenic properties, underpinning their effectiveness at the cellular protein and gene levels. In conclusion, this study offers a groundbreaking strategy for the employment of BTO/β-TCP piezoelectric composite scaffolds in bone implant applications, harnessing their unique blend of biocompatibility, piezoelectricity, and osteogenic potential.

Zirconia ceramics have become increasingly used as dental implant material which is mainly due to their “tooth-like” color, superior biocompatibility, and excellent mechanical properties . However, the bio-inertness of the zirconia surface limits its use for a wider range of clinical applications. To enhance the surface bioactivity, a combination of magnetron sputtering , and anodization has in the present study been used to deposit a homogeneous nano-porous coating (ZrNP) on the zirconia surface (forming ZrO 2 /ZrNP with a ZrNP pore diameter of 20–30 nm, and a ZrNP layer thickness of ∼2 μm). EDS and XRD analysis showed that the distribution of the surface chemical components, as well as the crystalline structure of the zirconia (ZrO 2 ), remained unchanged. On the other hand, the hydrophilicity of the ZrO 2 /ZrNP surface became significantly improved (as compared with a control group). In addition, the bond strength of the coating was decreased as a result of the anodization treatment. Moreover, both the Zr and ZrNP nanolayers exhibited improved proliferation and osteogenic properties in the in vitro cell experiment. Also, the ZrNP nanolayer was found to promote the biocompatibility and osteogenic ability to the largest extent. The ZrNP nanolayer did also provide numerous adhesion sites for MC3T3-E1 whereby the cell morphology could be enhanced. Additionally, the nano porous ZrNP layer improved the new bone formation around the zirconia implant and did robustly enhance the push out bond strength, thereby indicating the best osteointegration ability. In conclusion, a nano porous zirconium layer has been found to be very promising for the improvement of the bioactivity of the zirconia implant material.

Biphasic calcium phosphate (BCP) ceramics are valued for their osteoconductive properties but have limited osteogenic and angiogenic activities, which restricts their clinical utility in bone defect repair. Silicon doping has emerged as an effective strategy to enhance these biological functions of BCP. However, the biological impact of BCP is influenced by the level of silicon doping, necessitating determination of the optimal concentration to maximize efficacy in bone repair. This study investigated the effects of silicon doping on both the physicochemical and biological properties of BCP, with a specific focus on osteogenic and angiogenic potentials. Results indicated that silicon doping exceeding 4 mol.% led to the formation of α-TCP, accelerating BCP degradation, enhancing silicon ion release, and promoting mineralization product formation. Simultaneously, silicon doping increased the porosity of BCP scaffolds, which typically reduces their compressive strength. Nevertheless, scaffolds doped with ≤4 mol.% silicon maintained compressive strengths exceeding 2 MPa. In vitro biological experiments indicated that higher levels of silicon doping (≥6 mol.%) partially inhibited the successful differentiation of stem cells and the vascularization of endothelial cells. Optimal conditions for promoting osteogenic differentiation and angiogenesis were identified between 2 and 4 mol.% silicon doping, with an optimal level of approximately 4 mol.%. Subsequent in vivo experiments confirmed that BCP scaffolds doped with 4 mol.% silicon effectively promoted vascularization and new bone formation, highlighting their potential for clinical bone defect repair.

Titanium and its alloys have been widely used as orthopedic and dental implants. However, during practical use, wear debris and surface corrosion will cause the overproduction of reactive oxygen species leading to an inflammatory reaction, which has detrimental effects on osteogenesis around the titanium implants. To solve this problem, in this work, cerium modified titanium surface was constructed by plasma immersion ion implantation (PIII) technology. The mechanical property, corrosion resistance, antioxidant capability, and osteogenic properties of modified titanium were systematically studied. The results indicate that surface hardness and corrosion resistance of modified titanium are markedly improved and the reactive oxygen species level is reduced. Moreover, cell spreading, proliferation, alkaline phosphatase, and collagen secretion of rBMSCs on modified titanium surfaces are improved.

In order to improve early osseointegration and long-term survival rate of implants, a multifunctional titanium surface that promotes osteogenesis and antibacterial properties is expected. Incorporation of bioactive trace elements such as magnesium ions was proved a promising method to improve osseointegration of titanium. Phytic acid has strong chelating ability with multivalent cations, which has been used in surface modification. Moreover, phytic acid was proved antibacterial potential. Herein, to improve the osteogenic and antibacterial properties, a phytic acid-magnesium (PA-Mg) layer was introduced on titanium using phytic acid as a cross-linker molecule. No obvious changes of the surface characterization were observed by scanning electron microscopy and atomic force microscopy. X-ray photoelectron spectroscopy confirmed that the PA-Mg layer covalently bond to the Ti surface, and the thickness of the PA-Mg layer was about 150 nm. Besides, improved hydrophilic and more protein adsorption were observed on Ti-PA-Mg. Notably, a relatively controlled magnesium release was also observed on Ti-PA-Mg. Human bone mesenchymal stem cells showed better adhesion, proliferation, and osteogenic differentiation on Ti-PA-Mg samples, indicating improved biocompatibility and osteoinductivity. Moreover, Ti-PA-Mg had better antibacterial properties against porphyromonas gingivalis than Ti. Overall, the PA-Mg layer on Ti surface improved the osteogenic and antibacterial properties, which may have promise for use in dental implantation.

Biomaterial-associated infection (BAI) is a kind of serious post-operative complication in orthopaedic surgery. Antibiotic-loaded bone cement shines a light on BAI prevention for convenient manipulation and complex filling. To this aim, we designed an antibacterial bone cement based on Nano-hydroxyapatite/Polyurethane (PUHA) loading with antibiotic Enoxacin (EN). The distinct shear-thinning behavior of the prepolymers was observed, indicating a good injectability. The PUHA bone cement possessed a suitable curing speed, and the addition of EN might slightly expedite the curing process and enhance the mechanical properties. The EN release profile indicated that the EN-loaded bone cement could reach the minimum inhibitory concentration in 2 h, and sustainedly released EN for almost 8 days, exhibiting an antibacterial delivery potential. Antibacterial test further confirmed the antibacterial ability of EN-loaded bone cement is in a dose-dependent manner. However, the osteogenic performance of drug-loaded bone cement with high dosage is not as good as antibacterial activity. When the EN concentration of antibacterial cement was lower than 32 μg⋅mL −1 , the proliferation and osteogenic differentiation of rat mesenchymal stem cells could be significantly promoted. Overall, this study verified the potential of the EN-loaded PUHA bone cement in anti-infection and osteogenesis for bone repairing.

In the present study, it is found that the prepared alumina ceramics has better mineralization ability in a newly revised simulated body fluid. With the extension of mineralization time, the amount of hydroxyapatite (HA) crystals deposited on the surface of alumina ceramics also increased gradually. The results of cell biological experiments of alumina ceramics with hydroxyapatite surface layer demonstrate that the mineralized materials have better biological activity and osteogenesis properties in vitro. In the meanwhile, the ALP activity and expression of osteogenesis-related genes (OPN, ALP, Col-I, and OCN) of mouse bone marrow stromal stem cells on the samples were significantly promoted by increasing the formation of HA on the surface of alumina ceramics. Our research concluded that alumina ceramics with HA phase on surface had great potential to be developed as a sort of bioactive material in the bone repair field.
