Safety assessment of copper- and zinc-based metal-organic frameworks (MOFs) for embryonic development and adult fish of Oryzias melastigma

Mohammad Sadiq Nikzad,Jiangbing Qiu,Guixiang Wang,Xue Wang,Natnael Sisay Demo,Aifeng Li

The increasing prevalence of harmful algal blooms (HABs), specifically those resulted by toxigenic Alexandrium spp., notably threats human health and aquatic ecosystem. Metal organic frameworks (MOFs), particularly copper- and zinc-based MOFs have emerged as promising materials to control HABs. However, their application safety needs to be assessed because of poor knowledge of their ecotoxicity to marine organisms. In this study, the toxicity of Cu-MOF and Zn-MOF was evaluated to Oryzias melastigma at embryonic and adult stages. The Cu-MOF exhibited a concentration-dependent mortality and hatching rate at concentrations ≥ 2.0 mg L −1 . The Zn-MOF exhibited similar effects when the exposure doses ≥ 80 mg L −1 . Additionally, adult fish after exposure to Cu-MOF showed significant mortality rate at higher concentrations (≥ 4.0 mg L −1 ), beside elevated oxidative stress markers. In contrast, Zn-MOF did not show mortality and oxidative stress although bioaccumulation of Zn 2 + occurred in tissues. Based on the safety assessment of MOFs to marine medaka, the dosages for application of Cu-MOF (≤ 1.0 mg L −1 ) and Zn-MOF (≤ 20 mg L −1 ) are safe for controlling A. minutum -forming HABs. This study hints that the application of MOFs in mitigation of HABs should be carefully evaluated to balance their benefits in contrast to ecological risks.

Toxic effects and comparison of common amino antioxidants (AAOs) in the environment on zebrafish: A comprehensive analysis based on cells, embryos, and adult fish

Chenyang Rao,Fuhao Chu,Fang Fang,Dongfang Xiang,Bo Xian,Xiaying Liu,Shaopan Bao,Tao Fang

The ubiquity of amino antioxidants (AAOs) in the environment has attracted increasing attention, given their potential toxicity. This investigation represents a pioneering effort, systematically scrutinizing the toxicological effects of four distinct AAOs across the developmental spectrum of zebrafish, encompassing embryonic, larvae, and adult stages. The results indicate that four types of AAO exhibit varying degrees of cell proliferation toxicity. Although environmentally relevant concentrations of AAOs exhibit a comparatively circumscribed impact on zebrafish embryo development, heightened concentrations (300 μg/L) of N -(1,3-dimethylbutyl)- N′ -phenyl- p -phenylenediamine (6PPD) and N -isopropyl- N′ -phenyl- p -phenylenediamine (IPPD) distinctly evoke developmental toxicity. Behavioral analysis results indicate that at concentrations of 20 and 300 μg/L, the majority of AAOs significantly reduced the swimming speed and activity of larvae. Moreover, each AAO triggers the generation of reactive oxygen species (ROS) in larvae, instigating diverse levels of oxidative stress. The study delineates parallel toxicological patterns in zebrafish exposed to 300 μg/L of 6PPD and IPPD, thereby establishing a comparable toxicity profile. The comprehensive toxicity effects among the four AAOs is as follows: IPPD >6PPD >  N -Phenyl-1-naphthylamine (PANA) > diphenylamine (DPA). These findings not only enrich our comprehension of the potential hazards associated with AAOs but also provide data support for structure-based toxicity prediction models.

Screening on the differentially expressed miRNAs in zebrafish (Danio rerio) exposed to trace β-diketone antibiotics and their related functions

Jieyi Li,Jinfeng Liu,Yuhuan Zhang,Xuedong Wang,Weijun Li,Hongqin Zhang,Huili Wang

The toxicity of β-diketone antibiotics (DKAs) to larval and adult zebrafish ( Danio rerio ) was investigated by miRNA sequencing and bioinformatics analyses. In control and DKA-exposed groups, 215 differentially expressed miRNAs were screened, and 4076 differential target genes were predicted. Among 51 co-differentially expressed genes, 45 were annotated in KOG functional classification, and 34 in KEGG pathway analysis. The homology analysis of 20 miRNAs with human hsa-miRNAs demonstrated 17 high homologous sequences. The expression levels of 12 miRNAs by qRT-PCR were consistent with those by sRNA-seq. A regulatory network for 4 positive miRNA genes (dre-miR-10, ⿿96, ⿿92 and ⿿184) was plotted, and the high-degree of connectivity between miRNA-gene pairs suggests that these miRNAs play critical roles during zebrafish development. The consistent expression of dre-miR-184 and dre-miR-96 was proved in 120-hpf zebrafish brain, gill, otoliths and lateral line neuromast by qRT-PCR, miRNA-seq, W-ISH and ISH. DKA-exposure led to vacuolation of interstitial cells, reduced number of neurons, glial cell proliferation and formation of glial scar, and the obvious abnormality of cell structure might result from abnormal expression of differentially expressed miRNAs. In general, chronic DKA-exposure resulted in comprehensively toxic effects on larval and adult zebrafish tissues, especially for nervous system.
