Bitter apricot kernel (BAK) is a good source of amygdalin and oil. However, the current production process faces challenges including the inability to simultaneously extract amygdalin and oil, and the risk of toxicity due to amygdalin contamination in the oil. A novel, green and efficient ultrasonication-assisted extraction (UAE) method based on a natural deep eutectic solvent (NADES) composed of choline chloride-lactic acid (CC-LA) was established for simultaneous extraction of amygdalin and oil from BAK for the first time. Under optimizing conditions of a liquid-to-solid ratio of 18 mL/g, ultrasonic power of 420 W, temperature of 42 °C, and extraction time of 32 min, the highest yields of amygdalin and oil were achieved at 10.22 ± 0.04 % and 45.64 ± 0.24 %, respectively. The yield of amygdalin in UAE based on CC-LA was twice and nineteen times that of methanol (90 %, v/v) and water, respectively. Meanwhile, the yield of oil obtained by this new method was comparable to that by organic solvent extraction methods. The fatty acid composition of oil obtained by this method was consistent with that by conventional extraction methods. Scanning electron microscopy (SEM) analysis indicated that CC-LA and UAE had a synergistic effect on the destruction of BAK’s microstructure, which facilitated the recovery of amygdalin and oil. Molecular dynamics simulation results showed that amygdalin in CC-LA reached a higher solvent-accessible surface area (SASA) and formed more hydrogen bonds with longer lifetimes and greater stability. Furthermore, this new method demonstrated potential for industrial scalability.

The potential extractability, effectiveness, and stability of choline chloride/lactic acid (ChCl/HL) eutectics for 18 natural polyhydroxy compounds was investigated. Ultrasound-assisted ChCl/HL extraction exhibited exceptional extraction capabilities for polydatin (26.34 ± 0.44 mg/g) and black plum peel pigment (8.14 ± 0.16 mg/g), enhancements of 68.85 % and 40.34 %, respectively, over corresponding non-ultrasound systems. Certain comparative studies have reported yield 60.86–283.41 % and 71.58–317.04 % increases in glycoside and pigment analog extraction, respectively, demonstrating the broad applicability of ChCl/HL coupled with the ultrasound system. Further study revealed that the recovered pigments in the ChCl/HL system were 1.24– to 4.47–fold more than those in traditional solvents after 25 days of storage at 25 °C. An analysis of structure-function relationship revealed that the ability of ChCl/HL to extract and stabilize compounds depends on its strong hydrogen-bonding ability and the hydrogen bonding network among the H 2 O, extract, DES, and matrix.

Acute liver failure (ALF) is a serious clinical disorder with high fatality rates. Mahuang decoction (MHD), a well-known traditional Chinese medicine, has multiple pharmacological effects, such as anti-inflammation, anti-allergy, anti-asthma, and anti-hyperglycemia. In this study, we investigated the protective effect of MHD against ALF. In the lipopolysaccharide and D-galactosamine (LPS/D-GalN)-induced ALF mouse model, the elevated activities of the serum alanine and aspartate transaminases as well as the liver pathological damage were markedly alleviated by MHD. Subsequently, a metabolomics study based on the ultrahigh performance liquid chromatograph coupled with Q Exactive Orbitrap mass spectrometry was carried to clarify the therapeutic mechanisms of MHD against ALF. A total of 36 metabolites contributing to LPS/D-GalN-induced ALF were identified in the serum samples, among which the abnormalities of 27 metabolites were ameliorated by MHD. The analysis of metabolic pathways revealed that the therapeutic effects of MHD are likely due to the modulation of the metabolic disorders of tricarboxylic acid (TCA) cycle, retinol metabolism, tryptophan metabolism, arginine and proline metabolism, nicotinate and nicotinamide metabolism, phenylalanine metabolism, phenylalanine, tyrosine and tryptophan synthesis, as well as cysteine and methionine metabolism. This study demonstrated for the first time that MHD exerted an obvious protective effect against ALF mainly through the regulation of TCA cycle and amino acid metabolism, highlighting the importance of metabolomics to investigate the drug-targeted metabolic pathways.

The activation of hepatic stellate cells (HSCs) has been considered one of the major events in hepatic fibrosis. Amygdalin has been used to treat cancers and alleviate pain; however, its role and mechanism in HSC activation and hepatic fibrosis remain unclear. In the present study, transforming growth factor-beta 1 (TGF-β1) stimulated the activation of HSCs, as indicated by significantly increased alpha-smooth muscle actin (α-SMA), desmin, collagen I, and tissue inhibitor of metalloproteinase-1 (TIMP-1) protein levels. Amygdalin treatment dramatically suppressed TGF-β1-induced HSC proliferation and activation. Moreover, amygdalin treatment also reduced the TGF-β1-induced secretion of cytokines, including tumor necrosis factor-alpha (TNF-α), interleukin 6 (IL-6), platelet-derived growth factor (PDGF), and chemokine (C-C motif) ligand 2 (CCL2), as well as the phosphorylation of Smad2, Smad3, and p65. In the CCl 4 -stimulated liver fibrosis rat model, amygdalin treatment improved liver fibrosis and liver damage by reducing focal necrosis, collagen fiber accumulation, and the protein levels of α-SMA, desmin, collagen I, and TIMP-1 in hepatic tissue samples and reducing serum alanine transaminase (ALT) and aspartate transaminase (AST) levels. In conclusion, we demonstrated the suppressive effects of amygdalin in TGF-β1-induced HSC activation through modulating proliferation, fibrogenesis, and inflammation signaling in vitro and the antifibrotic effects of amygdalin in CCl 4 -stimulated hepatic fibrosis in rats in vivo .

In this work, relationships between solvent strength of organic phase ( ψ ) for two biphasic solvent systems in high speed countercurrent chromatography, hexane-ethyl-acetate-methanol-water (HEMWat) and ethyl acetate- n -butanol-water (EBuWat), and partition coefficient ( K ) were investigated using four retention models, including Jandera's model (ABM), Neue-Kuss model (NK), linear-solvent-strength model (LSS) and quadratic-solvent-strength model (QSS). Experimental results showed that ABM model had the best fitting results for HEMWat system while NK model and QSS model had good fitting results in EBuWat system. Thus, a mathematical relationship between partition coefficient ( K ) and solvent strength of organic phase ( ψ ) could be obtained by measurement of partition coefficients of the target compounds with three different volume ratios of organic phase. At the same time, a functional map was proposed to construct to get a maneuverable region so that an optimal two-phase solvent system for separation of a target compound could be selected easily, which saved a lot of manpower for high speed countercurrent chromatographic separation. The application of this new method was declared by successful separation of two components, apigenin-6-C- β -D-xylopyranosyl-8-C- α -L-arabinopyranoside and vicenin-3, from dried leaves of Dendrobium officinale Kimura et Migo using high speed countercurrent chromatography.
