Materials made from bile acids can benefit from the excellent biocompatibility and rigid skeleton of these natural compounds. To address concerns of the biocompatibility of shape-memory polymers (SMPs) in biomedical applications, a series of polyurethanes (PUs) is developed using bile acid as the hard segment and oligo(ethylene glycol) as the soft segment through a one-pot polymerization. These polymers can be easily processed into skin-like fibers, tubes, strips, etc., with tunable mechanical properties, e.g., elongation at break (483%) and high tensile strength (40.9 MPa). Their glass transition temperatures spread across a wide range, enabling them to exhibit dual, triple, and quadruple shape memory effects (SMEs). Due to intermolecular interactions, these PUs display a maximum self-healing efficiency of 96% in tensile strength for 1.5 h. In vitro cytotoxicity tests, hemolysis evaluations and healing efficiency in vivo all indicate their good biocompatibility and biosafety. These bio-derived PUs offer an option of shape memory polymers easy to make, with good tissue compatibility for applications as surgical sutures, artificial muscles, and soft materials.

Surgical threads are of great importance to prevent wound infection and accelerate tissue healing in surgical treatment. Cellulose nanofibrils (CNF) and chitosan (CS) are attracting increasing attention to be employed as biomedicine materials due to their nontoxicity, cytocompatibility, and biodegradability. However, a robust and absorbable cellulose-based surgical thread has not been explored. Therefore, in this work, a bioinspired CNF/CS composite thread containing 5% cationic polyacrylamide (CPAM) by the mass of CS was prepared, and the obtained CNF/CS-5C thread exhibited excellent mechanical properties and low swelling ratio in water due to the high cross-link degree. Especially, the tensile strength (1877 ± 107 MPa) of this thread was much higher than that of most reported CNF-based threads. Meanwhile, compared with commercial silk and Vicryl surgical threads, the CNF/CS-5C thread exhibited better in vitro cytocompatibility toward endothelial and fibroblast cells and lower inflammatory response in vivo to subcutaneous tissues of rats. In addition, the obtained thread could be regarded as a promising absorbable suture, which exhibited excellent wound healing performances in vivo. Therefore, the prepared absorbable thread will open a new window to prepare novel and advanced cellulose-based threads for medical applications.

The surgical suture has long been used to reconnect the injured tissues to restore their structure and function. However, its utility remains challenging in many areas, such as surgical site infections and minimally invasive surgeries. Herein, we report a novel surgical suture that possesses both antibacterial activity and shape memory effect to address these issues. In detail, natural antibacterial berberine was incorporated directly into the spinning solution of shape memory polyurethane with a near body transition temperature, and then berberine-containing polyurethane (BP) fibers were prepared by a facile one-step wet-spinning method for surgical suture. The prepared BP fibers were micro-sized and characterized by their transition temperature, morphology, water contact angles, mechanical properties, in vitro shape memory effect, drug release, and antibacterial activity. The results showed that with the increasing amount of the incorporated berberine, the transition temperatures of the fibers were not significantly affected, remains at near body temperature, while the contact angles of the fibers were significantly decreased and the mechanical properties of the fibers were significantly weakened. The optimized fiber was selected to evaluate the cytotoxicity and in vivo biocompatibility before in vivo shape memory effect and wound healing capacity in a mouse skin suture–wound model was tested. Besides the shape memory effect, it was demonstrated that the fiber is capable of antibacterial activity and anti-inflammatory effect, and promoting wound healing. The mechanism of the antibacterial activity and anti-inflammatory effect of the fiber was discussed. Overall, it is expected that by the berberine added to the fiber for surgical suture, it will be more popular and extend the utility of the sutures in a wide range of clinical applications.

Development of biocompatible and bioactive drug-loaded sutures is considered as an effective but challenging strategy for the wound healing process by delivering biological drugs (e.g., antibiotics) or growth factors (e.g., bFGF) at the surgical wound sites. Conventional offline suture strategies often lead to fast and uncontrollable release of drugs at wound sites, rendering wound healing to become a longer and painful process for patients. Herein, we propose an online suture strategy to fabricate electrospun polycaprolactone (PCL) fibrous yarns, incorporated with both collagen (COL) and bFGF, to produce bFGF-COL@PCL sutures. Upon demonstrating the well-oriented and aligned fibrous microstructure, high mechanical properties, and controlled release of bFGF from bFGF-COL@PCL sutures in vitro, we then applied bFGF-COL@PCL sutures to an incision wound healing mouse model in vivo. Further in vivo study showed that as compared to the commercialized Vicryl suture, bFGF-COL@PCL sutures significantly promoted the wound healing at different stages by accelerating granulation tissue formation, collagen deposition, and re-epithelialization. The enhanced wound healing efficiency of bFGF-COL@PCL sutures is likely attributed to two synergistic factors: (i) the well-oriented nanofibrous structure reduces tissue drag to minimize their trauma and (ii) the presence of both collagen and bFGF enhances the basement membrane (BM) reconstruction, cell proliferation, and angiogenesis. This work demonstrates an effective suture strategy and system for surgical suture applications.

Natural polymer-based sutures have attractive cytocompatibility and degradability in surgical operations. Herein, anionic cellulose nanofibrils (ACNF) and cationic guar gum (CGG) are employed to produce nontoxic CGG/ACNF composite filament with a unique core–shell structure via interfacial polyelectrolyte complexation (IPC) spinning. The comprehensive characterization and application performance of the resultant CGG/ACNF filament as a surgical suture are thoroughly investigated in comparison with silk and PGLA (90% glycolide and 10% l -lactide) sutures in vitro and in vivo, respectively. Results show that the CGG/ACNF filament with the typical core–shell structure and nervation pattern surface exhibits a high orientation index (0.74) and good mechanical properties. The tensile strength and knotting strength of CGG/ACNF suture prepared by twisting CGG/ACNF filaments increase by 69.5%, and CGG/ACNF suture has a similar friction coefficient to silk and PGLA sutures. Moreover, CGG/ACNF suture with antibiosis and cytocompatibility exhibits better growth promotion of cells than silk suture, similar to PGLA suture in vitro. In addition, the stitching experiment of mice with the CGG/ACNF suture further confirms better healing properties and less inflammation in vivo than silk and PGLA sutures do. Hence, the CGG/ACNF suture with a simple preparation method and excellent application properties is promising in surgical operations.

Aliphatic polycarbonates gels have received unprecedented attention due to their environmentally friendly and sustainable properties. However, traditional aliphatic polycarbonates gels lacking functionalization properties are not able to meet the requirements of future innovations. Based on this situation, in this study, novel functionalized aliphatic polycarbonate gels PEG 2000 - b -P(TMC-co-MCC) n are developed. The formation of hydrogen bonds network endows the gel with good comprehensive performances. Accordingly, the gel possesses a tensile strength and stretchability up to 2.62 MPa and 397.7%, presents a shape recovery rate of 97.7% in 9 s at 30 °C and also can repair crack at room temperature for 4.5 h without any external stimulation. Furthermore, these performances can be regulated by the ratio of the flexible segment, and the specific relationships between them are also presented. Notably, the cross-linking mechanism of the gels PEG 2000 - b -P(TMC-co-MCC) n is also proposed. Significantly, the transition states, energy barriers and reaction enthalpies of each step in the formation of oligomers by ring-opening polymerization of small molecules are calculated by density functional theory to deepen the understanding of the reaction mechanism. This aliphatic polycarbonate gel with excellent shape memory and self-healing properties provides prospects for developing the biomaterials such as surgical sutures and stents, as well as flexible electronics.
