Catheter-associated urinary tract infections (CAUTIs) are one of the most common hospital-acquired infections. In this study, a new Fe 3+ -carboxymethylcellulose (CMC)-polyacrylamide (PAAm) - Ag nanoparticles (AgNPs) interpenetrating hydrogel coating with antibacterial properties on the surfaces of latex urinary catheters was prepared with a simple dipping method. The morphology, chemical composition , surface roughness , and contact angles of the coated urinary catheters were characterized by using SEM, EDS, ATR-FTIR, XPS , AFM , and contact angle instruments, respectively. The antibacterial performance of the coated urinary catheters was characterized by the inhibition zone tests. Furthermore, the toxicity profile of the coated urinary catheters was also evaluated in vitro via MTT assay and hemolysis test. The experimental results showed that the Fe 3+ -CMC-PAAm-AgNPs hydrogel-coated urinary catheters had strong antibacterial effects . The concentration of AgNPs after the release of 30 days was still higher than the effective antibacterial concentration of the total Ag (0.1 ppb) and was lower than the toxic concentration of human cells (10 ppm). The inhibition zone tests of the coated urinary catheter against E. coli (ATCC25922) showed that both the CMC-PAAm-AgNPs hydrogel coatings and the Fe 3+ -CMC-PAAm-AgNPs hydrogel coatings had clear inhibition zones in the culture medium. The addition of Fe 3+ exhibited synergistic antibacterial effects with AgNPs. Moreover, the coatings also showed admirable biocompatibility and hemocompatibility. The synergistic antibacterial mechanism was also proposed. The novel Fe 3+ -CMC-PAAm-AgNPs hydrogel coatings for urinary catheters would have great potential in reducing CAUTIs.

Benzalkonium chloride (BC) is a quaternary ammonium antimicrobial agent used in a variety of applications. In this work, BC was prepared into deep eutectic solvent (DES) with acrylic acid (AA) or methacrylic acid (MA). Within the newly prepared DES, BC is responsible for antimicrobial properties, while AA and MA are responsible for polymerization. Three types of microorganisms, E. coli (gram-negative bacilli), S. aureus (gram-positive cocci) and C. albicans (fungi), were assessed for antimicrobial properties through agar diffusion test. DES viscosity measurements and polymerizations were also conducted to assist the antimicrobial performance analysis. From this study, stronger antimicrobial effectiveness of BC-AA DES towards S. aureus and C. albicans was observed, while smaller inhibition zone widths were obtained for BC-AA DES polymer compared to BC-AA DES monomer which may due to the limited active component transportation after polymerization. When changing AA to MA, increased structural complexity and decreased linearity may limit the molecule movement thus reduce the inhibition zone width, which could be proved by the calculated activation energy results. Accurately determined eutectic ratio of DES is recommended to get optimized drug release control. This work offers a new sight for preparation of antimicrobial materials with stronger effectiveness and limited release.

Diabetic wound healing remains a major challenge due to its vulnerability to bacterial infection, as well as the less vascularization and prolonged inflammatory phase. In this study, we developed a hydrogel system for the treatment of chronic infected wounds, which can regulate inflammatory (through the use of antimicrobial peptides) and enhance collagen deposition and angiogenesis (through the addition of platelet-rich plasma (PRP)). Based on the formation of Schiff base linkage, the ODEX/HA-AMP/PRP hydrogel was prepared by mixing oxidized dextran (ODEX), antimicrobial peptide-modified hyaluronic acid (HA-AMP) and PRP under physiological conditions, which exhibited obvious inhibition zones against three pathogenic bacterial strains ( E. coli, S. aureus and P. aeruginosa ) and slow release ability of antimicrobials and growth factors. Moreover, CCK-8, live/dead fluorescent staining and scratch test confirmed that ODEX/HA-AMP/PRP hydrogel could facilitate the proliferation and migration of L929 fibroblast cells. More importantly, in vivo experiments further demonstrated that the prepared hydrogels could significantly improve wound healing in a diabetic mouse infection by regulating inflammation, accelerating collagen deposition and angiogenesis. In addition, prepared hydrogel showed a significant antibacterial activity against S. aureus and P. aeruginosa, inhibited pro-inflammatory factors (TNF-α, IL-1β and IL-6), enhanced anti-inflammatory factors (TGF-β1) and vascular endothelial growth factor (VEGF) production. The findings of this study suggested that the composite hydrogel with AMP and PRP controlled release ability could be used as a promising candidate for chronic wound healing and infection-related wound healing.
