Hydrogel-assisted paper-based lateral flow sensor for the detection of trypsin in human serum

Jiantao Ping,Wenli Wu,Lubin Qi,Jie Liu,Jinpeng Liu,Binglu Zhao,Quanbo Wang,Li Yu,Jin-Ming Lin,Qiongzheng Hu

The detection of trypsin and its inhibitor is significantly important for both clinical diagnosis and disease treatment. Herein, we demonstrate a hydrogel-assisted paper-based lateral flow sensor for the detection of trypsin and its inhibitor for the first time. The gelatin hydrogel is hydrolyzed based on the gel-to-sol transition in the presence of trypsin, which results in the release of the trapped water molecules in the gelatin hydrogel. By placing one end of a pH indicator strip onto the hydrolyzed gelatin hydrogel, water is flowing along the pH indicator strip. However, in the absence of trypsin, water cannot flow along the pH indicator strip as the water molecules are trapped in the gelatin hydrogel. The detection limit of the system reaches as low as 1.0 × 10 −6  mg/mL, and it is also applied to the quantitative detection of trypsin in human serum. In addition, the detection of a clinical drug aprotinin that is an inhibitor of trypsin is also successfully achieved. Noteworthy, only the gelatin hydrogel, pH indicator strip, and PS substrate are needed to fulfill the detection of trypsin without the need of other chemicals or reagents. Overall, we develop a particularly simple, elegant, robust, competitive, high-throughput, and low-cost approach for the rapid and label-free detection of trypsin and its inhibitor, which is very promising in the development of commercial products for sensing, diagnostic, and pharmaceutical applications . Besides, the hydrogel-assisted paper-based lateral flow sensor can also be employed to detect other analytes of interest by use of different stimuli-responsive hydrogel systems.

Expression of ectopic trypsin in atherosclerotic plaques and the effects of aprotinin on plaque stability

Jia-Yu Shi,Hai-Yan Pan,Kun Liu,Min Pan,Guo-Jun Si

Background Our previous research revealed that trypsin is abundantly expressed in atherosclerotic plaques and its distribution overlaps with that of matrix metalloproteinase-9 (MMP-9). This study was performed to explore the possible roles of trypsin in vulnerable atherosclerotic plaque formation. Methods and results Twenty-four rabbits were randomly assigned to a normal (control) group, an atherosclerosis (experimental) group and a trypsin inhibitor (aprotinin) group. In the 13th feeding week, the aprotinin group was treated with 5 mg/kg/day aprotinin via ear vein for 4 weeks. At the end of the 16th week, coronary arterial and aortic expression of trypsin, proteinase-activated receptor-2 (PAR-2), activated MMP-9, and pro-inflammatory cytokines were significantly greater in the experimental group than in the control group. Aprotinin decreased trypsin expression and activation in plaques, blocked PAR-2 and MMP-9 activation, and decreased cytokine expression; it also increased fibrous cap thickness, decreased the intima-media thickness and intimal/medial ratio, thus significantly ameliorating plaque vulnerability. Upregulated trypsin, MMP-9 and PAR-2 were also found in coronary intimal atherosclerotic plaques of patients undergoing coronary artery bypass grafting. Conclusions Ectopic trypsin was significantly upregulated in atherosclerotic plaques, which increased pro-inflammatory cytokine levels by activating PAR-2 and promoted plaque instability by activating proMMP-9, thereby promoting atherosclerosis and plaque vulnerability. In addition, the high trypsin expression in human coronary intimal atherosclerotic plaques suggests that targeting trypsin may be a new strategy for acute coronary syndrome prevention.

Equipment-free determination of trypsin and its inhibitor based on the distance-readout via viscosity change on paper substrates

Xiaoxia Liu,Fanghua Xia,Guojuan Zhang,Wenmei Gao,Xiaodi Guo,Jinzhong Zhao,Wuyong Zhao,Miaomiao Tian

Herein, a simple, convenient, and cost-effective distance-readout paper-based sensor is developed for the detection of trypsin. Trypsin hydrolyzes the gelatin hydrogel via the gel-to-sol transition, resulting in a change of viscosity and leading to a change in diffusion behavior. The concentration of trypsin is thus quantitatively determined by measuring the difference in diffusion diameter of the spot on the paper. The linear range for trypsin detection is from 0.01 to 10.0 mg/mL with a limit of detection of 0.0019 mg/mL. It is further applied to the quantitative detection of trypsin in human serum and the standard inhibitor aprotinin. In addition, the inhibitors screening in 10 traditional Chinese medicines has been successfully achieved. It only requires the use of the gelatin hydrogel, MCE filter paper, and a Vernier caliper. Therefore, this convenient method is very promising in point-of-care testing (POCT) applications and the development of commercial products.

Cryo-induced clarification of Termitomyces albuminosus hydrolysates: An emerging clean-label strategy

Minbo Li,Xianbing Xu,Min Fan,Junjie Yi,Ming Du

Clean-label clarification of polysaccharide-rich mushroom hydrolysates is critical for producing high-quality natural seasonings. In this study, we developed a green polysaccharide-targeted clarification strategy termed cryo-induced molecular flocculation (CIMF), which offers a remarkable advantage of being additive-free and highly efficient compared with conventional clarification methods such as natural sedimentation, ethanol-assisted flocculation, and chitosan-assisted flocculation for clarifying Termitomyces albuminosus hydrolysates (TAH). CIMF achieved a high clarification efficiency (60.41%) without the use of external flocculants by selectively aggregating and removing insoluble polysaccharides (41.25%) while efficiently retaining peptides (96.15%). Structural analyses and molecular dynamics simulations further revealed that low temperature facilitated monosaccharide aggregation by reducing the solvent-accessible surface area and enhancing sugar–sugar interactions leading to compact cluster formation. In summary, CIMF represents a clean-label and polysaccharide-oriented clarification strategy that provides a sustainable and green approach for the manufacture of high-quality functional food ingredients.

Nanoparticle-mediated targeting of PGC-1α reveals critical metabolic pathways in bladder cancer metastasis

Liu Zhenghua,Xin Peng,Wu Weiwei,Jin Mingyue,Du Yang,Jiang Yuanjun,Liu Tao,Zhang Hao

Metastatic bladder cancer is characterized by its aggressive behavior and complex molecular mechanisms that remain largely undefined. This study explores the therapeutic potential of targeting peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α) using liposomal nanoparticles to deliver PGC-1α siRNA to bladder cancer cells. We employed comprehensive transcriptomic, proteomic, and metabolomic analyses to investigate the impact of PGC-1α silencing. Our in vitro results demonstrate that targeting PGC-1α significantly impairs mitochondrial function and disrupts energy metabolism, affecting critical pathways such as glycolysis and the citric acid cycle, as well as altering mRNA nuclear export. In vivo experiments in animal models show that nanoparticles loaded with si-PGC-1α effectively reduce lung metastasis, indicating a significant impact on metastatic progression. These findings highlight PGC-1α as a key regulator of metabolic reprogramming in metastatic bladder cancer and suggest that its inhibition could serve as a promising therapeutic strategy. By elucidating the role of PGC-1α in cancer metabolism, this study provides new insights into the molecular underpinnings of bladder cancer metastasis and offers potential avenues for developing targeted therapies aimed at the metabolic vulnerabilities of this malignancy.

A peptidomics-based investigation of the synergistic effects of neutral and Alcalase proteases on release of antioxidant peptides from soybean protein isolates

Shulong Cai,Liqi Fan,Dan Liu,Changxing Wei,Na Yang,Xichao Fu,Rong Tang,Jucai Xu,Sheng Yin,Xin Chen

Alcalase and neutral protease are widely employed for producing soybean antioxidant peptides, but their synergy mechanisms remain unclear. In this work, soybean protein isolate (SPI) was hydrolyzed synergistically by the two proteases. The products by Alcalase exhibited a larger proportion of low molecular-weight (MW, <1 kDa) peptides compared to those from neutral protease (87.74 % vs 76.08 %). After synergistic hydrolysis, an “averaging” effect on free amino acid composition and MW distribution was observed, but the peptide length distribution shifted evidently towards dipeptides (57.07 % with simultaneous enzyme use, compared to 20.43 % and 33.70 % for individual enzymes). Similarly, the N/C-terminal residue distribution also displayed an ‘averaging’ trend. Furthermore, synergistic processing increased the proportion (approximately 7 %) of potential antioxidant peptides with predicted probability >0.7, aligning with observed non-additive antioxidant activities and suggesting a synergistic mechanism beyond simple additivity. We hope the work could provide guidance and references for the target peptide release from SPI.

Synthesis of Formononetin Derivatives and Their Protective Effects Against Oxidative Stress

Luo Zeping,Pan Liwei

Objective: With the molecular formula C 16 H 12 O 4 , formononetin (FMN) is an appealing synthon exhibiting a wide range of biological activities, including antioxidant, anti-infective, and anti-apoptotic effects. Due to increasing interest in the mechanisms of action of antioxidants, our group aimed to identify the cellular target sites of these compounds. Methods: Cell Counting Kit-8 (CCK-8), enzyme-linked immunosorbent assay (ELISA), transmission electron microscopy (TEM), TdT-mediated dUTP nick end labeling (TUNEL) staining, real-time quantitative polymerase chain reaction (RT-qPCR), and Western blot were used to analyze the effects of oxidative stress on PC12 cell survival, antioxidant enzyme activity, mitochondrial morphology, apoptosis, key intracellular proteins, and signaling pathways. The potential roles of candidate targets were further investigated using proteomics, molecular docking, kinetic simulation, and cellular thermal shift assay (CETSA). Results and Discussion: One of the compounds, ( VI ) (5-(8-(((1-(benzyloxy)-3-hydroxy-1-oxopropan-2-yl)amino)methyl)-7-hydroxy-4-oxo-4 H -chromen-3-yl)-2-methoxybenzenesulfonic acid), was highly active. It enhanced PC12 cell proliferation, reduced lactate dehydrogenase (LDH) and malondialdehyde (MDA) levels, and increased superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) activities, effectively inhibiting apoptosis and ameliorating mitochondrial damage. Meanwhile, ( VI ) upregulated the expression of PI3K/Akt and Nrf2/HO-1 proteins. The study also demonstrated that ( VI ) binds significantly to the oxidative stress-related protein HO-1. Conclusions: Compound ( VI ) effectively protects PC12 cells against H 2 O 2 -induced oxidative damage. Its protective mechanism may involve activation of the PI3K/Akt and Nrf2/HO-1 signaling pathways, with HO-1 serving as a potential key target of its action.

Effects of high-pressure and CaCl2 pretreatments on the salt taste-enhancing activity of hydrolysate derived from spent hen meat

Ruixia Chen,Xiao-Chen Liu,Xianqi Yao,Wei Wang,Junyi Xiang,Igor Tomasevic,Weizheng Sun

BACKGROUND High-sodium intake has been proven to bring serious risks to public health. A potential sodium substitute of salt taste-enhancing hydrolysate (STEH) of protein has been focused on recently. The salt taste-enhancing activity (STEA) of STEH still needs to be improved. High-pressure and calcium chloride (CaCl 2 ) pretreatments were reported to affect proteolysis and promote the release of bioactive peptides. Hence, we investigated effects of high-pressure and CaCl 2 pretreatments on hydrolysis and STEA of STEH derived from spent hen. RESULTS The pretreatments significantly influenced STEA of spent hen meat hydrolysate (SHH), especially 200 MPa pressure and 80 mmol L −1 CaCl 2 pretreatments increased 27.1% salt taste intensity of SHH compared to that of blank (without pretreatments) according to sensory evaluation, the SHH umami also increased after pretreatments. In SHH, the proportion of peptides < 1000 Da increased up to 79.37% after the pretreatments compared to 73.68% of the blank. The degree of hydrolysis (DH) increased to 19.45% for moderate high-pressure (200 MPa) from 18.02% for blank, and the DH decreased after higher high-pressure and CaCl 2 pretreatments, especially for CaCl 2 in 80 mmol L −1 . The change in particle size distribution of SHH has similar trends to DH. CONCLUSION High-pressure and CaCl 2 pretreatments increased STEA of SHH by affecting hydrolysis process. The STEA increase may be related to increased small-peptide proportion in SHH. Meanwhile, moderate high-pressure may promote protein unfolding and further increase DH according to particle size distribution of SHH. The combination of proteolysis and pretreatments of high-pressure and CaCl 2 is a promising method to produce STEH. © 2024 Society of Chemical Industry.

Construction of Integral Decellularized Cartilage Using a Novel Hydrostatic Pressure Bioreactor

Xiaoxiao Li,Weikang Zhao,Dandan Zhou,Pei Li,Chen Zhao,Qiang Zhou,Yiyang Wang

The decellularized extracellular matrix (ECM) of cartilage is a widely used natural bioscaffold for constructing tissue-engineered cartilage due to its good biocompatibility and regeneration properties. However, current decellularization methods for accessing decellularized cartilaginous tissues require multiple steps and a relatively long duration to produce decellularized cartilage. In addition, most decellularization strategies lead to damage of the microstructure and loss of functional components of the cartilaginous matrix. In this study, a novel decellularization strategy based on a hydrostatic pressure (HP) bioreactor was introduced, which aimed to improve the efficiency of producing integral decellularized cartilage pieces by combining physical and chemical decellularization methods in a perfusing manner. Two types of cartilaginous tissues, auricular cartilage (AC) and nucleus pulposus (NP) fibrocartilage, were selected for comparison of the effects of ordinary, positive, and negative HP-based decellularization according to the cell clearance ratio, microstructural changes, ECM components, and mechanical properties. The results indicated that applying positive HP improved the efficiency of producing decellularized AC, but no significant differences in decellularization efficiency were found between the ordinary and negative HP-treated groups. However, compared with the ordinary HP treatment, the application of the positive or negative HP did not affect the efficiency of decellularized NP productions. Moreover, neither positive nor negative HP influenced the preservation of the microstructure and components of the AC matrix. However, applying negative HP disarranged the fibril distribution of the NP matrix and reduced glycosaminoglycans and collagen type II contents, two essential ECM components. In addition, the positive HP was beneficial for maintaining the mechanical properties of decellularized cartilage. The recellularization experiments also verified the good biocompatibility of the decellularized cartilage produced by the present bioreactor-based decellularization method under positive HP. Overall, applying positive HP-based decellularization resulted in a superior effect on the production of close-to-natural scaffolds for cartilage tissue engineering.Impact statementIn this study, we successfully constructed a novel hydrostatic pressure (HP) bioreactor and used this equipment to produce decellularized cartilage by combining physical and chemical decellularization methods in a perfusing manner. We found that positive HP-based decellularization could improve the production efficiency of integral decellularized cartilage pieces and promote the maintenance of matrix components and mechanical properties. This new decellularization strategy exhibited a superior effect in the production of close-to-natural scaffolds and positively impacts cartilage tissue engineering.

Real-time global coagulation assay via ordered porous layer interferometry using silica colloidal crystal film

Feng Wu,Yili Xu,Chenyang Ji,Xue Li,Jue Shen,Yejie Fei,Guanghan Yuan,Hui Qian

Background The coagulation process is inherently complex, and conventional diagnostic techniques are typically only capable of detecting single coagulation factors, lacking capacity to assess the overall coagulation function of the patient. Therefore, it is thus imperative to develop a novel, global coagulation assay for the diagnosis of hemorrhagic and thrombotic disorders and to evaluate the therapeutic efficacy of anticoagulant drugs. The ordered porous layer interferometry technology, developed using silica colloidal crystal (SCC) film as sensing substrate, is characterized by low cost, real-time, and convenient operation. Furthermore, entire biological events process can be detected by migration of the SCC film interference spectra. Results In this study, we developed a global coagulation assay with ordered porous layer interferometry by SCC film, a model of coagulation was created using fibrinogen and thrombin. The results demonstrated that this method could analyze the effect of different thrombin activities and different fibrinogen concentrations on the coagulation process. The performance of the coagulation assay was further validated using real blood samples and the method was found to have excellent detection capacity for different concentrations of CaCl 2 , which could trigger coagulation process. Hypercoagulable and hypocoagulable samples were tested by setting different biochemical conditions, and it could distinguish blood samples from different states significantly. Clinical samples were collected and the approach was compared with standard coagulation assays with satisfactory clinical correlation. Significance This global coagulation assay is simple, rapid and real-time, and does not require various biochemical reagents. The whole process of coagulation reaction can be recorded, and multiple coagulation indicators can be obtained in one test. It is anticipated to provide a novel low-cost and powerful method for clinical coagulation testing.

A novel approach for engineering DHCM/GelMA microgels: application in hepatocellular carcinoma cell encapsulation and chemoresistance research

Dandan Zhou,Xiaoxiao Li,Wencun Liu,Mingjun Zhang,Ying Cheng,Zhousong Xu,Jian Gao,Yiyang Wang

Liver cancer, a highly aggressive malignancy, continues to present significant challenges in therapeutic management due to its pronounced chemoresistance. This resistance, which undermines the efficacy of conventional chemotherapy and targeted therapies, is driven by multifaceted mechanisms, with increasing emphasis placed on the protective role of the tumor microenvironment (TME). The hepatocellular carcinoma extracellular matrix (ECM), a primary non-cellular component of the TME, has emerged as a critical regulator in cancer progression and drug resistance, particularly in hepatocellular carcinoma cell (HCC). In this study, a hybrid biomimetic hydrogel was engineered by integrating decellularized hepatocellular carcinoma matrix (DHCM) with gelatin methacrylate (GelMA) precursors. This composite DHCM/GelMA hydrogel was designed to replicate the physicochemical and functional properties of the hepatocellular carcinoma ECM, thereby offering a biomimetic platform to explore the interactions between HCCs and their microenvironment. Leveraging a custom-designed microfluidic 3D printing platform, we achieved high-throughput fabrication of HCC-encapsulated DHCM/GelMA microgels, characterized by enhanced uniformity, biocompatibility, and scalability. These microgels facilitated the construction of hepatocellular carcinoma microtissues, which were subsequently employed for chemoresistance studies. Our findings revealed that DHCM/GelMA microgels closely mimic the hepatocellular carcinoma tumor microenvironment, effectively recapitulating key features of ECM-mediated drug resistance. Mechanistic studies further demonstrated that DHCM significantly upregulates the expression of Aquaporin 3 (AQP3) in the encapsulated HCCs. This upregulation potentially activates mTOR signaling-associated autophagy pathways, thereby enhancing chemoresistance in HCCs. These biomimetic models provide a robust and versatile platform for studying the underlying mechanisms of drug resistance and evaluating therapeutic interventions. This innovative approach highlights the potential of DHCM/GelMA microgels as a transformative tool in cancer-associated tissue engineering and anticancer drug screening. By enabling detailed investigations into the role of ECM in chemoresistance, this study contributes to advancing therapeutic research and offers promising strategies to overcome drug resistance, ultimately improving clinical outcomes in liver cancer treatment.

Tanshinone IIA attenuates estradiol‑induced polycystic ovarian syndrome in mice by ameliorating FSHR expression in the ovary

Jing Jin,Qiao‑Yun Hu,Wen‑Wen Xu,Wen‑Jia Zhu,Bei Liu,Jing Liu,Wei Wang,Hui‑Fang Zhou

Tanshinone IIA (TSIIA) is a major component of Salvia miltiorrhiza, a Chinese herb that exhibits a therapeutic effect on polycystic ovary syndrome (PCOS). The present study replicated PCOS via the neonatal treatment of estradiol in mice. Estrous cycles, body and ovarian weight, serum levels of testosterone and estradiol were determined. Histological examination of ovaries was performed. The mRNA and protein levels of aromatase luteinizing hormone receptor and follicle‑stimulating hormone (FSHR) in ovaries and granule cells were assayed by reverse transcription‑quantitative polymerase chain reaction and western blotting, respectively. TSIIA was revealed to reverse all disorders induced by estradiol treatment, including prolonged estrous cycles, increased body and ovarian weight, increased atretic cyst‑like follicles and decreased corpus luteum, large antral follicles and preovulatory follicles. These improvements in PCOS as a result of TSIIA treatment are likely due to the revised testosterone/estradiol balance, as TSIIA reversed the decrease in aromatase mRNA, the enzyme that converts androgen to estrogen. As the expression of aromatase is regulated by the FSH pathway, TSIIA‑mediated elevation in FSHR expression may lead to the upregulation of aromatase. Therefore, TSIIA revises the balance of androgen and estrogen by rescuing the reduced expression of FSHR and aromatase, thus attenuating murine PCOS. The current study aimed to further the application of natural drugs in the treatment of PCOS to confront the side effects of hormone drugs and expand the use of TSIIA.
