The black soldier fly is a valuable resource insect capable of transforming organic waste while producing antimicrobial peptides (AMPs). The inhibition zone assay (IZA) is a method used to demonstrate the antimicrobial activity of AMPs. This study aimed to examine the experimental principles and establish a standardized IZA method. Results indicated that the AMPs extract consisted of proteins ranging in molecular weights from 0 to 40 kDa. The AMPs diffused radially on an agar plate through an Oxford cup. The diffusion radius was influenced by the concentration and volume of the AMPs but ultimately determined by the mass of the AMPs. The swabbing method was found to be effective for inoculating bacteria on the agar plate. Among the conditions tested, the plate nutrient concentration was the most sensitive factor for the IZA, followed by bacterial concentration and incubation time. Optimal conditions for the IZA included a nutrient plate of 0.5× TSA, a bacterial concentration of 106CFU/mL, and an incubation time of 12 h, with oxytetracycline (OTC) at 0.01 mg/mL serving as the positive control. The antimicrobial-specific activity of AMPs could be standardized by the ratio of inhibition zone diameters between AMPs and OTC. These findings contribute to the standardization of the IZA method for profiling the antimicrobial activity of AMPs.

Polyethyleneimine (PEI) was immobilized on SBA-15 for obtaining polyethyleneimine-modified silica [email protected] ( [email protected] , [email protected] , and [email protected] ). With the coordination from PEI, zinc ions were impregnated on the [email protected] , and then, Zn 2+ [email protected] were calcined at 550 ℃ to obtain the nano zinc oxide loaded on the mesoporous silica, ZnO-SBA-15s (ZnO-SBA-1.0, ZnO-SBA-1.5, and ZnO-SBA-2.0). The anti-bacterial activity of ZnO-loaded SBA-15 was; thereafter, assessed by the zone of inhibition and enzyme labeling methods. The results confirmed that the coordination ability of the imino groups in PEI helped adsorb more zinc ions during the process of impregnation so that more nano-ZnO could be supported on the surface of SBA-15. The modification did not change the mesostructure of the two-dimensional hexagonal phase and orderliness of SBA-15. At the mass ratio of PEI:SBA-15 = 1.0:1, 1.5:1, and 2.0:1, the concentration of zinc element (wt.%) was measured by inductively coupled plasma optical emission spectrometry as 2.5, 2.56, and 3.5% for ZnO-SBA-1.0, ZnO-SBA-1.5, and ZnO-SBA-2.0, respectively. The size of inhibition of SBA-15, ZnO-SBA-0, ZnO-SBA-1.0, ZnO-SBA-1.5, and ZnO-SBA-2.0 were 0, 1.14 ± 0.01, 2.56 ± 0.01, 2.58 ± 0.01, and 2.69 ± 0.02 cm, respectively. ZnO-SBA-15s suppressed the growth of Escherichia coli compared to blank SBA-15 in a ZnO dose-dependent manner.

This work describes the fabrication and characterization of antibacterial poly(vinylidene fluoride) (PVDF) hollow fibre membranes via phase inversion using dry-jet wet-spinning technique. Ag-loaded NaY zeolites were particularly synthesized and then doped with PVDF spinning solution to produce a novel antibacterial hollow fibre with enhanced antibacterial efficiency. The morphologies, crystal structures, and Ag + loading of synthesized Ag-loaded NaY zeolites were analyzed by field emission scanning electron microscope (FE-SEM), X-ray diffraction (XRD) and inductively coupled plasma-atomic emission spectrometry (ICP-AES), respectively. The antibacterial efficiencies of the prepared hollow fibre membranes against Escherichia coli ( E. coli ) were evaluated by halo zone test, E. coli suspension filtration test and adhesion test. The surface morphologies , pure water flux, hydrophilicity, tensile strength , through-pore size, resistance to BSA of antibacterial PVDF hollow fibre membrane were also investigated. All results demonstrated that PVDF hollow fibre fabricated by doping Ag-loaded zeolites showed excellent antibacterial property, high permeability and mechanical strength , indicating the promising application as the antibacterial ultra-filtration membranes.
