Inorganic nanomaterials that mimic enzymes are fascinating as they potentially have improved properties relative to native enzymes, such as greater resistance to extremes of pH and temperature and lower sensitivity to proteases. Although many artificial enzymes have been investigated, searching for highly-efficient and stable catalysts is still of great interest. In this paper, we first demonstrated that bovine serum albumin (BSA)-stabilized MnO2 nanoparticles (NPs) exhibited highly peroxidase-, oxidase-, and catalase-like activities. The activities of the BSA-MnO2 NPs were evaluated using the typical horseradish peroxidase (HRP) substrates o-phenylenediamine (OPD) and 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of either hydrogen peroxide or dissolved oxygen. These small-sized BSA-MnO2 NPs with good dispersion, solubility and biocompatibility exhibited typical Michaelis–Menten kinetics and high affinity for H2O2, OPD and TMB, indicating that BSA-MnO2 NPs can be used as satisfactory enzyme mimics. Based on these findings, BSA-MnO2 NPs were used as colorimetric immunoassay tags for the detection of goat anti-human IgG in place of HRP. The colorimetric immunoassay using BSA-MnO2 NPs has the advantages of being fast, robust, inexpensive, easily prepared and with no HRP and H2O2 being needed. These water-soluble BSA-MnO2 NPs may have promising potential applications in biotechnology, bioassays, and biomedicine.

Integrating surface plasmon resonance (SPR) structures on fiber end facets enables the sensing system to be miniature, label-free, real-time, and remote detection. Yet, the wave vector matching condition for SPR excitation is hard to be satisfied by regulating the wave vector of light emitted from fiber ends. Here, we demonstrate a novel SPR microsensor realized by integrating a 3D micro waveguide onto the fiber tip. The mechanism of SPR excitation is illustrated by wave vector matching between the in-plane component of waveguide mode and surface plasmon polaritons . Using two-photon polymerization (2PP) and metallization process, the designed 3D waveguide probe with a length of 10 µm is fabricated on the fiber tip. Then, the microsensor was applied for the immune-sensing of anti-human IgG and human IgG interactions, and a LOD of 1.11 nM was achieved. The ultracompact structure endows with the capability for high-performance detection in the micrometre scale through a convenient “dip-and-read” manner. As a proof-of-concept for this “dip-and-read” small volume sensing, immunoassay detection in a small sample volume of 5 μL was also successfully demonstrated. The obtained results indicated that our 3D fiber-probe SPR microsensor may provide a powerful tool for small-volume biosensing applications.

Lab-on-tip technology opens a new door for optical fiber surface plasmon resonance (SPR) sensors. In this paper, we first demonstrate an ultra-compact reflective SPR microtip probe on the flat fiber end in experiment, which has a length of tens of microns and a diameter of several microns. Simulations and experiments show that this new fiber SPR microtip probe coated with a nano-goldfilm has excellent refractive index (RI) sensing characteristic and great potential for biochemical detection at single cell level. Based on the high RI sensitivity, a bio-probe assembled with Chitosan/Polysodium Styrene Sulfonate (Cs/PSS) nano-polyelectrolyte film and the sheep anti-human IgG-HRP is prepared to recognize human IgG specifically. Due to the high surface sensitivity, the SPR microtip probe exhibits an ultra-low detection limit for human IgG. Due to the high bulk sensitivity and high thermo-optical coefficient (TOC) of polydimethylsiloxane (PDMS), a reference temperature probe based on SPR microtip is proposed, which is used to compensate for the temperature cross-sensitivity of the bio-probe. This new SPR microtip probe can be used not only in optical fiber sensor, but also in Tip-Enhanced Near-Field Raman Microscopy and Scanning Near-Field Optical Microscope.

In response to the urgent requirement for high-throughput multiplexed detection, a novel dual-encoded suspension array (SA) based on thickness and digital fluorescent peak is proposed, along with the corresponding optical system. The SAs are prepared by thickness encoded micro-glass chips (MGCs) with electrostatically adsorbed quantum dots (QDs). Using the thickness characteristics of microcarrier itself and digital fluorescence peak (DFP) as the encoded signals ensure the high encoding stability with simple fabrication process at a low cost. A spectral-domain phase and fluorescence spectroscopy microscopy (SDPFSM) system is designed for the first time to decode the thickness information and DFP with ultrahigh accuracy. The high selectivity and sensitivity of the dual-encoded SAs are verified by the multiplexed immunoassays . The quantitative detection of anti-human IgG, anti-mouse IgG, and anti-rabbit IgG was realized with the limits of detection (LOD) of 0.19, 0.35 and 0.31 pM/mL and the sensitivity of 713, 470 and 456 a.u./(pM/mL), respectively. The coding number increases by a factor of 458 compared with the traditional fluorescence coding method due to the additional use of the thickness information of the MGCs itself as a coding channel. The aforementioned properties demonstrate that the dual-encoded SA and the corresponding decoding platform are promising and robust in multiplexed detection.

A reliable, rapid and ultrasensitive immunoassay is described for determination of immunoglobulin G (IgG). It is making use of biofunctional magnetite (Fe 3 O 4 ) superparticles coated with SiO 2 and serving as an infrared (IR) probe. The unique IR fingerprint signals originating from the transverse and longitudinal phonon modes, respectively, of the asymmetric stretching of the Si–O–Si bridges display a satisfactory resistance to optical interference from the environment. The adoption of Fe 3 O 4 superparticles instead of Fe 3 O 4 nanoparticles as the magnetic core warrants a controllable structure and a strong magnetic response. This facilitates the efficient purification of the probes and the alleviation of the interfacial resistance between the liquid-solid interfaces by using a magnet. The gold-coated substrate was used to immobilize goat-anti-human IgG. The analyte (human IgG) was incubated with the IR probes, and then captured by the substrate immobilized antibody with the assistance of an external magnetic field. The integral area of the IR absorption band between 1250 cm −1 – 900 cm −1 was chosen for quantitative assay. The limit of detection is 95 fM, which is two orders of magnitude better than that without the magnetic field. The assay time was shortened from 2 h to 1 min. High selectivity, specificity, and long-term stability of the immunoassay were achieved. The performance of the assay when analyzing blood samples confirmed the practicability of the method.
Schematic presentation of the infrared (IR) immunoassay based on Fe 3 O 4 superparticle@SiO 2 nanocomposites. The assistance of an external magnetic field reduces the incubation time and improves the detection sensitivity.

We developed a three-dimensional (3D) polymer-brush substrate for protein and peptide microarray fabrication, and this substrate was facilely prepared by copolymerization of glycidyl methacrylate (GMA) and 2-hydroxyethyl methacrylate (HEMA) monomers via surface-initiated atom transfer radical polymerization (SI-ATRP) on a glass slide. The performance of obtained poly(glycidyl methacrylate-co-2-hydroxyethyl methacrylate) (P(GMA-HEMA)) brush substrate was assessed by binding of human IgG with rabbit antihuman IgG antibodies on a protein microarray and by the determination of matrix metalloproteinase (MMP) activities on a peptide microarray. The P(GMA-HEMA) brush substrate exhibited higher immobilization capacities for proteins and peptides than those of a two-dimensional (2D) planar epoxy slide. Furthermore, the sensitivity of the P(GMA-HEMA) brush-based microarray on rabbit antihuman IgG antibody detection was much higher than that of its 2D counterpart. The enzyme activities of MMPs were determined specifically with a low detection limit of 6.0 pg mL–1 for MMP-2 and 5.7 pg mL–1 for MMP-9. By taking advantage of the biocompatibility of PHEMA, the P(GMA-HEMA) brush-based peptide microarray was also employed to evaluate the secretion of MMP-2 and MMP-9 by cells cultured off the chip or directly on the chip, and satisfactory results were obtained.
