Functional polymers bear specified chemical groups and have specified physical, chemical, biological, pharmacological, or other uses. To adjust the properties while keeping material usage low, a method for direct synthesis of functional polymers is indispensable. Here we show that various functional polymers can be synthesized by in situ cross-linked polymerization/copolymerization. We demonstrate that the polymers synthesized by the facile method using different functional monomers own outstanding pH-sensitivity and pH-reversibility, antifouling property, antibacterial and anticoagulant property. Our study opens a route for the functionalization of commodity polymers, which lead to important advances in polymeric materials applications.

The development of microorganisms and formation of thrombus on a biomaterial surface can seriously lead to device failure and threaten human health. Nonetheless, a surface that has both antibacterial and anticoagulant properties has scarcely been developed. Herein, a novel dual-action membrane composed of polyethersulfone (PES) bulk material and a hydrophilic anionic poly-2-acrylamido-2-methylpropanesulfonic acid (PAMPS) polymer has been prepared via the cationic antibacterial agent poly(hexamethylene biguanide) (PHMB)-induced phase separation technique. Interestingly, the resultant membrane can offer tunable antibacterial and anticoagulant properties, while maintaining satisfactory permeability and greatly increasing selectivity. The membrane also shows excellent hydrophilicity, a well-defined porous surface, and cross section with a sponge gradient structure. Furthermore, the PHMB–PAMPS complex formed on the membrane surface displays outstanding long-term stability, which is crucial for further practical applications. More importantly, the hollow fiber membrane fabricated by the cationic polyelectrolyte-induced phase separation technique confirms its capability to control the membrane permeability (257.4 L·m–2·h–1·bar–1) and selectivity (95.9%) without destroying the membrane structure. The present work opens a straightforward and efficient avenue for the rational design of a functional surface to fight biomedical material-associated infections.

This study aimed to improve the efficiency of decellularization and enhance the functional properties of vascular grafts to optimize their application in vascular repair. Rabbit abdominal aortas were used as the decellularization target, and ultrasound-assisted decellularization was performed using intermittent ultrasound at 100 W power, 20 kHz frequency, and 4 °C. Rabbit abdominal aortas were subjected to three different decellularization techniques. Based on comparative evaluation, ultrasound-assisted decellularization was implemented to enhance cell removal efficiency. In addition, dual-factor surface modification was performed using sodium heparin (HEP) and vascular endothelial growth factor 165 (VEGF165) to investigate anticoagulant and endothelialization potential. Ultrasound optimization enhanced decellularization efficiency by 1.5 times, increased matrix integrity to 85%, and decreased chemical residues by 30%. Dual-factor functionalization with HEP and VEGF165 improved anticoagulant properties by 40%, prolonged thrombus formation time by 45%, and enhanced endothelialization by 68%. In vivo animal studies demonstrated a 93% blood flow patency rate post-implantation, with superior tissue repair compared to the control group. This study presents an innovative approach that integrates ultrasound optimization and functional modification, addressing the limitations of traditional decellularization methods. It offers a high-performance, low-toxicity strategy for developing vascular grafts with significant clinical potential, particularly for small-diameter vascular applications.

Although dressing blood-contacting devices with robust and synergistic antibacterial and antithrombus properties has been explored for several decades, it still remains a great challenge. In order to endow materials with remarkable antibacterial and antithrombus abilities, a stable and antifouling hydrogel coating was developed via surface-initiated polymerization of sulfobetaine methacrylate and acrylic acid on a polymeric substrate followed by embedding of antimicrobial peptides (AMPs), including WR (sequence: WRWRWR-NH2) or Bac2A (sequence: RLARIVVIRVAR-NH2) AMPs. The chemical composition of the AMP-embedded hydrogel coating was determined through XPS, zeta potential, and SEM–EDS measurements. The AMP-embedded antifouling hydrogel coating showed not only good hemocompatibility but also excellent bactericidal and antiadhesion properties against Gram-positive and Gram-negative bacteria. Moreover, the hydrogel coating could protect the AMPs with long-term bioactivity and cover the positive charge of the dotted distributed AMPs, which in turn well retained the hemocompatibility and antifouling capacity of the bulk hydrogels. Furthermore, the microbiological results of animal experiments also verified the anti-infection performance in vivo. Histological and immunological data further indicated that the hydrogel coating had an excellent anti-inflammatory function. Therefore, the present study might provide a promising approach to prevent bacterial infections and thrombosis in clinical applications of blood-contacting devices and related implants.

The catheters coating can be effective in reducing bloodstream infection and thrombosis, which are the major complications in blood contact catheters. However, the surface functional coating is difficult to be implemented due to the high surface stretching force from the minor-caliber. In this work, we propose a covalent bonding coating of polydopamine/titanium dioxide quantum dots (PDA/TiO 2 QDs) on polyurethane (PU) catheters, which can fulfill a dual-function of antibacterial and antithrombosis. The PDA/TiO 2 QDs layer was prepared by dip-coating, where the intermediate transition layer of PDA was reacted with the internal hydroxyls of PU surface by pre-oxidation and bonds with the external TiO 2 QDs coating. The surface microstructures are analyzed by SEM, TEM and XPS methods, and the antimicrobial and anticoagulant performances are investigated by bacterial plate count and platelet adhesion tests. The oxidizing and hydrophilic effect of the top layer of TiO 2 QDs were enhanced by the QD-sized particles. The antibacterial activities of the PDA/TiO 2 QDs coating on PU catheters against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ), especially to S. aureus , are evidenced by bacterial plate count test, reaching good bactericidal rates of 49.9 % against E. coli and 83.7 % against S. aureus , respectively. Platelet adhesion test and whole blood dynamic circulation modeling demonstrate that the PDA/TiO 2 QDs coating effectively inhibits platelet adhesion due to an excellent hydrophilicity of TiO 2 QDs surface, and thereafter reduce thrombus formation.
