While polyethersulfone (PES) represents outstanding oxidative, thermal and hydrolytic stability as well as good mechanical and film-forming properties, the hemocompatibility of PES membranes must be dramatically enhanced to reduce injections of anticoagulants during hemodialysis. In this study, a series of biological macromolecules with heparin-like chains were synthesized via reversible addition fragmentation chain transfer (RAFT) polymerization to design anticoagulant membrane surfaces. When the synthesized copolymers were used as additives to modify the PES membrane using the phase separation method, the functional groups of the copolymers migrated and formed a negatively charged coating on the membrane surface. The modified PES membrane blended with a heparin-like block copolymer showed prolonged blood coagulation time and thereby good hemocompatibility. In addition, the clotting time of the modified membrane was enhanced with increasing amounts of the heparin-like amphiphilic tri-block copolymer. Furthermore, the results of the cell morphology and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay suggest that the cytocompatibility increases due to the addition of heparin-like additives. Thus, the heparin-like surface modification method seemed to be a promising approach for application in the biomedical field.

Chronic kidney disease caused by functional impairment of glomerular endothelial cells (GECs) is one of the etiologies of hemodialysis treatment. As a critical component of the hemodialysis process, hemodialysis membranes having been suffering from intricated modification methods. In order to solve a series of problems caused by blood incompatibility, usually triggered by rough surfaces, an anticoagulant coating with a similar physical structure and physiological properties to GECs was prepared on the polyethersulfone (PES) membrane. The smooth glomerular endothelium-like coating was obtained by a hydrated layer formed by tannic acid (TA) and α-lipoic acid (α-LA). We explored two possible reaction mechanisms of TA and α-LA, which were characterized and discussed by ATR-FTIR and XPS. Besides, the hydrophilic and negatively charged coating has filtration and anticoagulant properties similar to those of GECs. The results show that the modified membranes have appreciable hydrophilicity (the water contact angle is 29.7°) as well as high urea removal rate (96.5%) and BSA protein retention (99.7%). In addition, the modified membranes show favorable hemocompatibility and free radical scavenging ability. The reduction of platelet adhesion number is reduced by 98.7% and PRT is prolonged to 297 s. DPPH and ABTS radical scavenging capacity reach 1.18 and 0.89 μmol/cm 2 , respectively. In this work, a surface coating mimicking GECs was fabricated for the potential candidates of new hemodialysis membranes, which will significantly enhance the dialysis efficiency and alleviate the coagulation.

Blood contact medical devices require a lubricant and anticoagulant surface to reduce tissue trauma, thrombosis, and other complications. Hydrogel coating is a promising strategy but usually shows poor adhesion and weak mechanical properties, leading to the failure of the coating by delamination or fracture during clinical applications. Herein, we report a tough and slippery hydrogel coating through the procedures of photoinitiator entrapment, pregel solution dip-coating, and UV-curing. The surface-embedded initiator and in situ polymerization attribute to the robust interfacial bonding, and double-network (DN) hydrogel formulation improves the toughness of the hydrogel coating itself. We characterize the morphology, composition, thickness, lubrication, and anticoagulant capabilities of the coating. Furthermore, we confirm the toughness of the coating by sonicating, soaking, repeated friction, tension, and the bending test. Results show that the DN hydrogel coating has a low coefficient of friction (∼0.03) after 3000 cycles of friction with a 20 N load (∼33 kPa). This coating method may bring clinical benefits in the use of vascular catheters as well as other interventional devices.

Anticoagulant surface modification of blood-contacting materials has been shown to be effective in preventing thrombosis and reducing the dose of anticoagulant drugs that patients take. However, commercially available anticoagulant coatings, that is, both bioinert and bioactive coatings, are typically based on a single anticoagulation strategy. This puts the anticoagulation function of the coating at risk of failure during long-term use. Considering the several pathways of the human coagulation system, the synergy of multiple anticoagulation theories may provide separate, targeted effects at different stages of thrombosis. Based on this presumption, in this work, negatively charged poly(sodium p-styrenesulfonate-co-oligo(ethylene glycol) methyl ether methacrylate) and positively charged poly(lysine-co-1-adamantan-1-ylmethyl methacrylate) were synthesized to construct matrix layers on the substrate by electrostatic layer-by-layer self-assembly (LBL). Amino-functionalized β-cyclodextrin (β-CD-PEI) was subsequently immobilized on the surface by host–guest interactions, and heparin was grafted. By adjusting the content of poly(oligo(ethylene glycol) methyl ether methacrylate) (POEGMA), the interactions between modified surfaces and plasma proteins/cells were regulated. This multistage anticoagulant surface exhibits inertness at the initial stage of implantation, resisting nonspecific protein adsorption (POEGMA). When coagulation reactions occur, heparin exerts its active anticoagulant function in a timely manner, blocking the pathway of thrombosis. If thrombus formation is inevitable, lysine can play a fibrinolytic role in dissolving fibrin clots. Finally, during implantation, endothelial cells continue to adhere and proliferate on the surface, forming an endothelial layer, which meets the blood compatibility requirements. This method provides a new approach to construct a multistage anticoagulant surface for blood-contacting materials.
