Panax ginsengis a valuable traditional Chinese medicine in Northeast China. Ginsenoside, the active component of ginseng, has not been investigated much for its effects on aging and its underlying mechanism(s) of action. Here, we investigated the effects of total ginsenoside (TG), a mixture of the primary active ginsenosides fromPanax ginseng, on the lifespan ofCaenorhabditis elegans(C. elegans). We found that TG extended the lifespan ofC. elegansand reduced lipofuscin accumulation. Moreover, TG increased the survival ofC. elegansin response to heat and oxidative stress via the reduction of ROS. Next, we used RNA-seq to fully define the antiaging mechanism(s) of TG. The KEGG pathway analysis showed that TG can prolong the lifespan and is involved in the longevity regulating pathway. qPCR showed that TG upregulated the expression ofnrh-80,daf-12,daf-16,hsf-1and their downstream genes. TG also reduced the fat accumulation and promoted lipid metabolism. Moreover, TG failed to extend the lifespan ofdaf-16andhsf-1mutants, highlighting their role in the antiaging effects of TG inC. elegans. The four main constitution of TG were then confirmed by HPLC and included ginsenoside Re, Rg1, Rg2and Rd. Of the ginsenosides, only ginsenoside Rd prolonged the lifespan ofC. elegansto levels comparable to TG. These findings provided mechanistic insight into the antiaging effects of ginsenoside inC. elegans. Keywords:Panax ginseng;lifespan;lipid metabolism;stress-resistant;Caenorhabditis elegans

Ganoderma atrum is an edible and medicinal fungi in oriental countries with various bioactivities, such as antioxidant, anti-inflammatory and anti-aging properties. The study was to evaluate the effects of polysaccharide from Ganoderma atrum (PSG) on lifespan, healthspan (locomotion, pumping, lipofuscin, ROS, chemotaxis and intestinal permeability) and defense system (stress resistance, antioxidant enzymes activities), as well as to investigate its anti-aging mechanism in Caenorhabditis elegans . The findings indicated that PSG prolonged the lifespan (optimal concentration at 400 μg/mL, with an increased average lifespan by 23.10 %) and improved senescence-related physiological indicators of worms. Furthermore, the lipid accumulation was reduced, while stress resistance and antioxidant enzymes activities were enhanced after PSG intervention. Meanwhile, studies with mutants demonstrated that DAF-16/FOXO and SKN-1/Nrf-2 were required for lifespan-extension, which might be attributed to the IIS and MAPK pathways. Overall, these demonstrated that PSG was a potential dietary supplement to promote health and ameliorate aging-associated pathologies.

Phytoremediation is an effective solution to treat pollution with antibiotic compounds in aquatic environments; however, the underlying mechanisms for plants to cope with antibiotic pollutants are obscure. Here we used cell suspension culture to investigate the distribution and transformation of ciprofloxacin (CIP) in common reed ( Phragmites australis ) plants, as well as the accompanying phenotypic and metabolic responses of plants. By means of radioactive isotope labelling, we found that in total 68 % of CIP was transformed via intracellular Phase I transformation (reduction and methylation), Phase Ⅱ conjugation (glycosylation), and Phase Ⅲ compartmentalization (cell-bound residue formation mainly in cell walls, 23 %). The reduction and glycosylation products were secreted by the cells. To mitigate stress induced by CIP and its transformation products, the cells activated the defense system by up-regulating both intra- and extra-cellular antioxidant metabolites ( e.g. , catechin, l -cystine, and dehydroascorbic acid), anti-C/N metabolism disorder metabolites ( e.g. , succinic acid), secreting signaling ( e.g. , nicotinic acid), and anti-stress ( e.g. , allantoin) metabolites. Notably, the metabolic reprogramming could be involved in the CIP transformation process ( e.g. , glycosylation). Our findings reveal the strategy of wetland plants to cope with the stress from CIP by transforming the xenobiotic compound and reprogramming metabolism, and provide novel insights into the fate of antibiotics and plant defense mechanisms during phytoremediation.

Microglial activation plays an important role in neurodegenerative diseases associated with oxidative stress. tert-Butyl hydroperoxide (t-BHP), an analog of hydroperoxide, mimics the oxidative damage to microglial cells. It has been reported that ginsenoside Rg1 (G-Rg1), an active ingredient of Panax ginseng, has anti-stress and anti-inflammatory properties. The present study aims to investigate the ability of G-Rg1 to decrease the t-BHP-mediated cell damage of BV2 microglial cells. We performed flow cytometry assays to facilitate the detection of reactive oxygen species as well as Western blotting analyses and immunofluorescence assays using specific antibodies, such as antibodies against phospho-mitogen-activated protein kinases (p-MAPKs), phospho-nuclear factor-κB (p-NF-κB), B-cell lymphoma 2 (Bcl-2), Bcl-2-associated X (Bax), Caspase-3, autophagy marker light chain 3 (LC3), and Becline-1. We found that treatment with 50 μM G-Rg1 protected microglial cells against oxidative damage induced by 10 μM t-BHP.
