Antidepressant and immunosuppressive activities of two polysaccharides from Poria cocos (Schw.) Wolf

Wuxia Zhang,Lu Chen,Peng Li,Jinzhong Zhao,Jinyou Duan

Pursuit of novel effective antidepressants is an urgent task. Poria cocos ( Schw .) Wolf (PCW) is a traditional herb with antidepressant effects . Polysaccharides designated PCWPW and PCWPS were purified from PCW by DEAE-52 cellulose chromatography. Their structures were characterized using physicochemical and spectral methods. Chemical analysis indicated that PCWPW (37,154 Da) and PCWPS (186,209 Da) were mainly composed of mannose , glucose, galactose and fucose . Their antidepressant activities were evaluated by tail suspension test (TST), forced swimming test (FST) with chronic unpredictable mild stress (CUMS) model mice. To investigate the antidepressant mechanism, the neuroprotective and immunomodulation effects were assessed by MTT method. Results showed that PCWPW and PCWPS possess obvious antidepressant effects and can suppress ConA-stimulated T cell proliferation in a dose-dependent manner. In addition, PCWPS could protect the PC12 cells from H 2 O 2 -induced damage and suppress LPS-induced B cell proliferation . These findings implied that PCWPW and PCWPS might be a candidate for developing antidepressant or immunosuppressive agents in food and pharmaceutical industries.

Antidepressant potential of Uncaria rhynchophylla and its active flavanol, catechin, targeting melatonin receptors

Chang-An Geng,Tong-Hua Yang,Xiao-Yan Huang,Yun-Bao Ma,Xue-Mei Zhang,Ji-Jun Chen

Ethnopharmacological relevance Traditional Chinese medicines (TCMs) are fascinating sources for natural drug candidates. Uncaria rhynchophylla (Gouteng) is a famous TCM used for alleviating central nervous system (CNS) disorders, while its antidepressant constituents are still disputed. Aim of the study The present study was designed to assess the antidepressant property of U. rhynchophylla and characterize the active constituents targeting melatonin receptors which are closely related to CNS diseases. Materials and methods The total extract and each fraction of U. rhynchophylla were extensively assessed for their agonistic activity on melatonin receptors in vitro . The following bioassay-guided fractionation yielded the active constituents, whose activity was confirmed by dose-dependent bioassay and antagonistic experiment on HEK293 cells. Their antidepressant effects were evaluated on forced swimming test (FST), tail suspension test (TST) and open-field test (OFT) mice models in vivo . Their metabolic profiles in mice plasma were analyzed by LCMS-IT-TOF. Results The stems and hooks of U. rhynchophylla were revealed with agonistic activity on melatonin receptors (MT 1 and MT 2 ). Under the guidance of bioassay, two flavanols , catechin and epicatechin were obtained and showed obviously activity agitating MT 1 (EC 50 = 25.8 and 156.1 μM) and MT 2 (EC 50 = 47.3 and 208.8 μM) receptors. The agonistic activity of catechin on melatonin receptors can be antagonized by luzindole at the concentrations of 1.57–100 μM. Catechin could significantly reduce the immobility time in both FST and TST mice models at doses of 80 and 40 mg/kg, without obvious effect on locomotor activity in OFT mice model. Five phase II (M1-M5) and one phase I (M6) metabolites of catechin were detected in mice plasma after intragastric ( i.g .) administration. Conclusion Catechin is a potent antidepressant candidate from U. rhynchophylla by targeting melatonin receptors. The main metabolic pathways of catechin in mice plasma are glucuronidation (M3) and methylated glucuronidation (M4 and M5). This study provides valuable information for understanding the antidepressant potency of Gouteng and its active constituents.

Pipette-tip solid-phase extraction coupled with matrix-assisted laser desorption/ionization mass spectrometry enables rapid and high-throughput analysis of antidepressants in rat serum

Sun Zhi,Wang Fangfang,Li Wenxuan,Ren Ruobing,Zhou Peipei,Jia Qingquan,Zhao Lingguo,Chen Di,Zuo Lihua

Therapeutic drug monitoring is essential for ensuring the efficacy and safety of medications. This study introduces a streamlined approach that combines pipette-tip solid-phase extraction (PT-SPE) with matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS), facilitating rapid and high-throughput monitoring of drug concentrations. As a demonstration, this method was applied to the extraction and quantification of antidepressants in serum. Utilizing Zip-Tip C18, the method enabled the extraction of antidepressants from complex biological matrices in less than 2 min, with the subsequent MALDI-MS analysis yielding results in just 1 min. Optimal extraction recoveries were achieved using a sampling solution at pH 9.0 and a 10 μL ethanol desorption solution containing 0.1% phosphoric acid. For MALDI analysis, 2,5-dihydroxybenzoic acid was identified as the most effective matrix for producing the highest signal intensity. The quantification strategy exhibited robust linearities ( R 2  ≥ 0.997) and satisfactory limits of quantification, ranging from 0.05 to 0.5 μg/mL for a suite of antidepressants. The application for monitoring dynamic concentration changes of antidepressants in rat serum emphasized the method’s efficacy. This strategy offers the advantages of high throughput, minimal sample usage, environmental sustainability, and simplicity, providing ideas and a reference basis for the subsequent development of methods for therapeutic drug monitoring. Rapid, green and semi-automated PT-μSPE for antidepressants analysis in urine and plasma using melt-blown polypropylene from disposable face masks as sorbent

Qianwei Li,Bowen Deng,Ru Feng,Di Chen,Linlin Hua

Extracting antidepressants from biological samples is challenging due to their low concentrations and complex matrices. Pipette-tip solid-phase microextraction (PT-μSPE) simplifies the process, reducing solvent use and enabling automation, but challenges like custom adsorbents and toxic solvents limit its broader application. Melt-blown polypropylene nonwoven, sourced from the middle layer of 3-ply disposable non-woven face masks, was employed as the sorbent material. The extraction tip was easily assembled by combining a 200 μL pipette tip, the melt-blown polypropylene nonwoven, and a 1 mL pipette tip. This method provides a cost-effective and sustainable alternative to conventional adsorbents. A 90% ethanol solution was selected as the desorption solvent to comply with green chemistry principles, with the volume minimized to 100 μL to reduce waste. The pH adjustment was achieved using low-toxicity sodium carbonate (Na₂CO₃). The developed PT-μSPE method proved excellent linearity, sensitivity, accuracy, precision, and robustness, making it suitable for routine antidepressant monitoring in plasma and urine. It offers a practical, scalable, and environmentally friendly solution for therapeutic drug monitoring.

Antidepressant-like action of agmatine in the acute and sub-acute mouse models of depression: a receptor mechanism study

Chen Zhao-Di,Chen Wen-Qiang,Wang Zhi-Yuan,Cao Dan-Ni,Wu Ning,Li Jin

Previous studies have shown that agmatine, a potential neuromodulator or co-transmitter, exhibited antidepressant-like action in animal models, yet its mechanism, especially the receptor mechanism, remains unclear. In the present study, using efaroxan, a preferential antagonist of I 1 imidazoline receptor (I 1 R) and yohimbine, an antagonist of α 2 adrenergic receptor (α 2 AR), we investigated the roles of I 1 R and α 2 AR in agmatine’s antidepressant-like effect in acute and sub-acute depression models in mice. We found that in the tail-suspension test (TST) and the forced swimming test (FST), acute administration of agmatine (20 and 40 mg/kg, p.o.) significantly shortened the immobility time. Concurrent administration of efaroxan (1 mg/kg, i.p.) completely abolished the antidepressant-like effects of agmatine (40 mg/kg, p.o.) whereas yohimbine (5 mg/kg, i.p.) failed to exert similar effects, suggesting that the acute antidepressant-like effects of agmatine was mainly mediated by I 1 R but not α 2 AR. Additionally, in the learned helplessness (LH) test, repeated administration of agmatine (20 mg/kg, p.o., q.d.) for 5 days significantly decreased the escape latency and the number of escape failure, and these effects were respectively abolished by concurrent administration of efaroxan (0.5 mg/kg,i.p., q.d.) and yohimbine (3 mg/kg, i.p., q.d.) for 5 days, suggesting that the antidepressant-like action of agmatine in the LH test was achieved via the activation of both I 1 R and α 2 AR. In summary, we found that the antidepressant-like effects of agmatine in the TST and the FST were mediated by activating I 1 R and in the sub-acute LH test were mediated by activating both I 1 R and α 2 AR.

Antidepressant Properties of a Four-compound Cocktail Identified from Si-Ni-San by HIF-1 Pathway Modulation

Na An,Dongxing Zhang,Jile Xin,Xinyi Zhang,Zhijuan Zhang,Ligang Ma,Le Zhao,Huimin Wu,Weisheng Feng,Xiaoke Zheng

Background: Si-Ni-San (SNS) is the formula prescription of Traditional Chinese Medicine (TCM) with anti-depression properties, but its underlying mechanisms remain unclear. Objective: This study provides novel approaches for the study of Traditional Chinese Medicine (TCM) and offers new opportunities for exploring the pharmacological properties of SNS. Methods: The ingredients in SNS implicated in the treatment of depression were identified and studied using network pharmacology. SwissTargetPrediction and molecular docking were used to study the interaction of SNS ingredients and their targets. The protective effect of these ingredients and their cocktail in rat pheochromocytoma cells (PC12) exposed to corticosterone (Cor) were evaluated using the CCK-8 assay, Hoechst 33342 staining, 2',7'-dichlorodihydro fluorescein diacetate (H2DCFDA) staining, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay, and in-cell Western analysis. Results: The network pharmacology study showed that the HIF-1 signaling pathway was the most crucial pathway implicated in the anti-depressive property of SNS. MAPK1 (ERK2), MAPK3 (ERK1), AKT1, VEGFA, STAT3, and EGF were identified as hub target proteins in the HIF-1 signaling pathway. Quercetin, naringenin, licochalcone A, and kaempferol from SNS, which targeted the six proteins mentioned above, were used to create a cocktail. This cocktail exerted protective properties, decreased the oxidative stress in PC12 exposed to Cor, and successfully regulated the expressions of AKT1, p-AKT1, ERK1, ERK2, p-ERK1/2, STAT3, p- STAT3, and VEGFA induced by Cor exposure. The SwissTargetPrediction and molecular docking study showed that the cocktail may regulate the HIF-1 signaling pathway by directly binding with AKT1 and MAPK1. Conclusion: The cocktail from SNS comprised of quercetin, naringenin, licochalcone A, and kaempferol exerts anti-depression potentiality by modulating the HIF-1 signaling pathway via direct interactions with AKT1 and MAPK1.

Lingjiao Wang,Mengqiang Zhao,Jiao Li,Hongtao Xu,Yang Song,Yuanyuan Zhao,Jing Yu,Chunhua Zhou

Background Therapeutic drug monitoring for antidepressants (ADs) is vital due to the potentially serious consequences and disputes related to medical events. Therefore, we created a quick and convenient analysis way for separation and quantification of ADs. Methods To ensure quantitative stability, we divided the 16 ADs or their metabolites into 4 pools (AD1-AD4), considering the hospital frequency that the clinician prescribed, the physicochemical properties of medicines, and the calibration range of selected ADs. After precipitation with methanol, the analytes were eluted for at least 3.5 min on a BEH C 18 analytical column by different gradient elution methods. Results: The LLOQ and LOD were 1.25–10 ng/mL and 0.42–5 ng/mL, respectively. High precision (<12 %) and accuracy (87.07–111.47 %) were demonstrated by quality control samples both within and between days. All the compounds were stable at room temperature and within −80 °C. Conclusion The method is of wide clinical and laboratory interest due to simpler sample cleanup, shorter chromatographic run times, and wider calibration range compared to other methods.

Yumei Gan,Linlin Hua,Yuanyuan Zheng,Bin Wang,Di Chen

Effective antidepressant monitoring is crucial for safe treatment, with liquid chromatography being the primary detection method. Due to the complexity of biofluids, meticulous sample preparation is vital to remove interferences and concentrate analytes. In this study, a novel solid-phase extraction method utilizing unprocessed kapok fiber (KF) as an eco-friendly and efficient sorbent , ingeniously incorporated within an in-syringe device for the extraction of antidepressants from biofluids was introduced. The extraction process displayed several advantages, including simplicity and rapidity (less than 3 min with minimal manual intervention), as well as the potential for automation, which represents significant advancements in sample preparation techniques. The method demonstrated excellent analytical performance when coupled with high-performance liquid chromatography-ultraviolet detection, achieving good linearity (R 2  ≥ 0.991), excellent relative recovery (92.6–107.5 %), high precision in intraday and interday analyses (RSDs within 0.8–6.0 % and 1.2–7.9 %, respectively), and good sensitivity with limits of quantification between 5–10 ng/mL. Notably, given the use of a renewable and biodegradable material with KF, and a low-toxicity desorption solvent with ethanol, the developed method stands out for its environmental sustainability, aligning with green analytical chemistry principles. Its application to real biological samples validates its efficacy as a rapid and reliable tool for clinical and forensic monitoring of antidepressants. Moreover, the low cost of this method facilitated by the economical nature of KF and the reusability of syringes further enhances its practicability. This study presents a unique approach by employing natural and renewable KF as adsorbents, thereby eliminating the need for complex material synthesis and making a pioneering contribution to the field of analytical chemistry for drug analysis in complex matrices.

Influence of antidepressants on stream microbiota: Consequences for leaf itter breakdown and nutrient cycling

Xilin Wang,Tianying Tao,Keying Hu,Yangyang Lv,Qian Zhang,Luyao Yu,Baodan Jin,Yuanqian Xu,Xia Cao,Jingjing Du

As an emerging pollutant, antidepressants in wastewater have received extensive attention due to their metabolic stability and antimicrobial activity in aquatic systems. However, the scarcity of experimental data limits the validation of their potential impacts on ecosystem functionality. This study examined the effects of fluoxetine and amitriptyline (0–100 ng L −1 ) on microbial decomposers in stream ecosystems. These two antidepressants exhibited different effects on the process of leaf litter decomposition. Fluoxetine at 1 ng L −1 inhibited the dominance of functional bacteria ( Caulobacter and Flavobacterium ) and cellobiohydrolase activity, significantly reducing the leaf decomposition rate by 11.5%. Notably, amitriptyline at 10 ng L −1 promoting this ecological process by enhancing fungal biomass and most enzyme activities, and increasing the abundance of functional fungi ( Anguillospora and Setophaeosphaeria ). Nevertheless, when amitriptyline concentrations exceeded 10 ng L −1 , nitrogen-limitation was observed in microbial decomposers. These findings illustrate the complexity of the aquatic microbial community in responding to external factors, underscoring the importance of further research into the effects of antidepressants on the nutrient cycling and organic matter dynamics of ecosystems.

Zhanwu Li,Bowen Deng,Jiajia Chen,Ru Feng,Shuling Wang,Shu Li,Di Chen,Linlin Hua

Therapeutic drug monitoring is essential for ensuring the safe and effective administration of medications, for example antidepressants, which require precise dosing to avoid adverse effects. Developing efficient and reliable methods for extracting and determining antidepressants in biofluids is crucial for advancing personalized medicine. This study presents the development of a one-pot synthesis of magnetic adsorbent with integrated pH regulation for the efficient extraction and determination of antidepressants in biofluids. Magnetic particles (Fe 3 O 4 ), multi-walled carbon nanotubes functionalized with hydroxyl groups (MWCNTs-OH), and calcium oxide (CaO) as a pH regulator were combined in a single-step grinding process to synthesize the magnetic adsorbent. The resulting adsorbent not only exhibit a high surface area and strong magnetic properties, enabling the efficient and selective adsorption of target compounds, but they also directly adjust the solution’s pH to alkaline conditions. This built-in pH regulation enhances the extraction efficiency of antidepressants without requiring external pH adjustment, significantly simplifying the sample preparation process. Coupled with high-performance liquid chromatography-ultraviolet detection, the synthesized magnetic adsorbent were successfully applied to the extraction and quantification of antidepressants in human plasma and urine samples. The validated method demonstrated good linearity, with a coefficient of determination (R 2 ) greater than 0.99, along with high accuracy and precision. Analysis of real samples further confirmed the method’s reliability and robustness. This approach offers a rapid, convenient, and cost-effective solution for therapeutic drug monitoring, demonstrating significant potential for widespread clinical and bioanalytical applications.

Junhui He,Dongbo Han,Chunlian Jia,Jiaxiu Xie,Fucui Zhu,Jie Wei,Dongmei Li,Dongmei Wei,Yi Li,Li Tang,Guining Wei,Jing Yan,Yuanming Tong,Lifang Yang,Xuecai Tan

Purpose To investigate the mechanisms of antidepressant action of active fraction of Polyrhachis vicina Rogers (AFPR) through network pharmacology, molecular docking and experimental validation. Methods GC-MS was used to predict chemical compounds, corresponding databases were used to predict chemical compound targets and depression targets, Cytoscape software was used to construct and analyze the protein interaction network map, DAVID database was used to analyze gene ontology (GO) and KEGG signaling pathway, and AGFR software was used to perform molecular docking. Subsequently, the underlying action mechanisms of AFPR on depression predicted by network pharmacology analyses were experimentally validated in a CORT-induced depression model in vitro and in vivo. Results A total of 52 potential targets of AFPR on antidepressant were obtained. GO is mainly related to chemical synaptic transmission, signal transduction and others. KEGG signaling pathways are mainly related to cAMP signaling pathway and C-type lectin receptor signaling pathway. The experiment results showed that AFPR significantly increased the expression of PRKACA, CREB and BDNF in mouse brain tissue and PC12 cells. Furthermore, after interfered of cAMP in PC12 cells, the decreased expression of PRKACA, CREB and BDNF was reversed by AFPR.Conclusion AFPR may exert antidepressant effects through multiple components, targets and pathways. Furthermore, it could improve neuroplasticity via the cAMP signaling pathway to improve depression-like symptoms.

Magnetic phytic acid-modified kapok fiber biochar as a novel sorbent for magnetic solid-phase extraction of antidepressants in biofluids

Bin Wang,Yongyue Chen,Wenxuan Li,Yuwei Liu,Xudong Xia,Xia Xu,Yongli Yang,Di Chen

Background Therapeutic drug monitoring (TDM) of antidepressants is essential for monitoring patient medication to avoid drug toxicity, complications, or nonadherence. Chromatographic techniques with high sensitivity and reproducibility are the main detection method for antidepressants. Effective pretreatment of biological sample processes is necessary prior to instrumental analysis. Magnetic solid-phase extraction (MSPE) has received much attention for its advantages of simple operation, rapidity, cost-effectiveness and low organic solvent consumption. Therefore, the development of a suitable and green magnetic sorbent for the detection of antidepressants in plasma and urine is apparently necessary. (88) Results A magnetic phytic acid-modified kapok fiber biochar sorbent (Fe 3 O 4 /PAKFBC) was successfully synthesized by pyrolytic impregnation and physical milling methods. Fe 3 O 4 /PAKFBC exhibited a large specific surface area (214 m 2  g −1 ) and a rich pore structure (5–10 nm). The extraction equilibrium, using 10 mg Fe 3 O 4 /PAKFBC, can be completed in about 1 min. The density functional theory (DFT) results showed that the adsorption mechanism of Fe 3 O 4 /PAKFBC on the six antidepressants mainly included electrostatic interactions, van der Waals interactions, π-π interactions and weak hydrogen bonding. Examination using the greenness assessment tools showed that the developed method exhibited excellent greenness. By combining with liquid chromatography-ultraviolet (LC-UV), a quantitative method with good linearity (R 2  > 0.993) and relative recoveries (92.4–107.7%) and negligible matrix effect (−11.5–6.0%) was developed. The Fe 3 O 4 /PAKFBC successfully detected six antidepressants in plasma and urine samples, requiring no pH adjustment with buffer salts. (142) Significance The environmental sustainability of the proposed methods was affirmed by six greenness evaluation tools, all indicating exceptional eco-friendliness. The Fe 3 O 4 /PAKFBC demonstrated outstanding greenness in both its creation and analytical application, proving highly effective in real sample applications and showcasing potential for broader use. This study contributes to a deeper and broader understanding of the microscopic adsorption mechanism, which can help in the optimization and development of more green sorbents. (69).
