The emergence and prevalence of multidrug-resistant (MDR) bacteria have posed a serious threat to public health. Of particular concern are methicillin-resistantStaphylococcus aureus(MRSA) andblaNDM,mcr-1andtet(X)-positive Gram-negative pathogens. The fact that few new antibiotics have been approved in recent years exacerbates this global crisis, thus, new alternatives are urgently needed. Antimicrobial peptides (AMPs) originated from host defense peptides with a wide range of sources and multiple functions, are less prone to achieve resistance. All these characteristics laid the foundation for AMPs to become potential antibiotic candidates. In this study, we revealed that peptide WW307 displayed potent antibacterial and bactericidal activity against MDR bacteria, including MRSA and Gram-negative bacteria carryingblaNDM-5,mcr-1ortet(X4). In addition, WW307 exhibited great biofilm inhibition and eradication activity. Safety and stability experiments showed that WW307 had a strong resistance against various physiological conditions and displayed relatively low toxicity. Mechanistic experiments showed that WW307 resulted in membrane damage by selectively targeting bacterial membrane-specific components, including lipopolysaccharide (LPS), phosphatidylglycerol (PG), and cardiolipin (CL). Moreover, WW307 dissipated membrane potential and triggered the production of reactive oxygen species (ROS). Collectively, these results demonstrated that WW307 represents a promising candidate for combating MDR pathogens. Keywords:antimicrobial peptide;antibiotic resistance;multidrug-resistant bacteria;membrane damage

The colonization of Enterococcus in the female vagina leads to neonatal and pediatric enterococcal septicemia. Linezolid (LZD) is a kind of mainstream drug for treating multidrug-resistant Gram-positive infections. OptrA is the main LZD-resistance gene at Enterococci in human isolates. It is essential to explore the prevalence of optrA-carrying Enterococcus in vaginal secretions of late pregnant women and the drug resistance of optrA. From May to June 2023, this study recruited 340 volunteers in late pregnancy (35–40 weeks of pregnancy) to provide non-repetitive vaginal discharge samples. Luria-Bertani broth and florfenicol (10 µg/mL) were used to enrich bacteria. Enterococci was identified through time-of-flight mass spectrometry. Additionally, antimicrobial susceptibility, polymerase chain reaction, and next-generation sequencing assays were applied to this study. Fifty-four optrA-carrying Enterococcus strains were obtained, the proportion of the whole vagina of late pregnant women was 15.88% (54 out of 340), and Enterococcus faecalis account the highest proportion. All optrA-carrying Enterococcus were resistant to at least three drugs. Tetracycline, chloramphenicol, erythromycin, and LZD have higher bacterial resistance rates. Genetic environment analysis revealed that IS1216E, fexA, and erm(A) may synergistically exert multidrug resistance with optrA. It is necessary to strengthen the surveillance of optrA-carrying Enterococcus in pregnant women. This study provides scientific support for controlling hospital infections and managing antibiotic-resistant bacteria, and provides a scientific basis for rational clinical medication.

Disinfectants are widely used in food production and environmental sanitation to prevent illness, but bacteria resistance to these disinfectants and co-resistance to antibiotics pose a threat to public health. This study investigated the impact of commonly used disinfectants on the resistance of Salmonella Typhimurium (ST) to disinfectants and antibiotics, and explored the metabolic mechanisms underlying the resistance changes. The results showed that subinhibitory concentrations of disinfectants had a minor impact on the resistance of ST to four disinfectants. However, chlorine-containing disinfectants stress enhanced bacteria resistance to ampicillin, while quaternary ammonium compounds stress increased resistance to tetracycline and gentamicin. Untargeted metabolomics analysis revealed significant changes in glutathione metabolism and lysine and valine degradation pathways after disinfectant exposure. Specifically, ST activated lysine decarboxylation, leading to a significant decrease in lysine levels after benzalkonium chloride exposure, while valine and leucine degradation pathways were activated by sodium hypochlorite stress. The addition of downregulated L-lysine and L-valine increased the sensitivity of ST to antibiotics, providing further evidence for the findings of metabolomics. This study provides guidance for the proper use of disinfectants in food processing and establishes a strategy based on metabolomics to control antibiotic-resistant bacteria.

Cu 3 P nanoparticles (NPs) were proposed as an efficient antibacterial agent for impeding bacterial resistance. Both sensitive and resistant strains of E. coli and S. aureus (3 × 10 6 CFU mL −1 ), and the microbial populations from a fishery water were completely inactivated by Cu 3 P NPs (≤ 1.5 µg mL −1 ) within 30 min with no regrowth. The antibacterial mechanism of Cu 3 P NPs relied on their own multiple enzyme-like activities, to elucidate, the action of ROS produced by their oxidase- and peroxidase- like activities was facilitated by their inherent activities for glutathione depletion and the lipid peroxidation. Comparing with that traditional antibiotics induced bacterial resistance (6 out of 7) within 3 passages, Cu 3 P NPs kept their initially inactivated efficiency, demonstrating that they can delay the onset of bacterial resistance. Furthermore, the Cu 3 P NPs shown no obviously toxic effect on aquatic organisms and a negligible side-effect toward mammalian tissues. This strategy is insightful for struggle with resistant bacteria.

The existence of antibiotic resistant bacteria (ARB) and antibiotic resistance genes (ARGs) has been a global public environment and health issue. Due to the different cell structures, gram-positive/negative ARB exhibit various inactivation mechanisms in water disinfection. In this study, a gram-negative ARB Escherichia coli DH5α ( E. coli DH5α) was used as a horizontal gene transfer (HGT) donor, while a gram-positive ARB Bacillus as a recipient. To develop an efficient and engineering applicable method in water disinfection, ARB and ARGs removal efficiency of Fe(VI) coupled peroxydisulfate (PDS) or peroxymonosulfate (PMS) was compared, wherein hydroxylamine (HA) was added as a reducing agent. The results indicated that Fe(VI)/PMS/HA showed higher disinfection efficiency than Fe(VI)/PDS/HA. When the concentration of each Fe(VI), PMS, HA was 0.48 mM, 5.15 log E. coli DH5α and 3.57 log Bacillus lost cultivability, while the proportion of recovered cells was 0.0017 % and 0.0566 %, respectively, and HGT was blocked. Intracellular tetA was reduced by 2.49 log. Fe(IV) and/or Fe(V) were proved to be the decisive reactive species. Due to the superiority of low cost as well as high efficiency and practicality, Fe(VI)/PMS/HA has significant application potential in ARB, ARGs removal and HGT inhibition, offering a new insight for wastewater treatment.

Introduction To effectively minimise the emergence and dissemination of antibiotic resistant bacteria, a holistic One Health approach is called for. The Sino-Swedish Integrated Multisectoral Partnership for Antibiotic Resistance Containment is a cross-sectoral and integrated project on antibiotic resistance, conducted in Shandong Province in China. This paper outlines the overall study protocol for the project. To our knowledge, this is the first research programme aiming to take a true holistic approach across multiple sectors simultaneously in China, and the first to incorporate both antibiotic use and infection prevention and control in addition to antibiotic resistance patterns. The project aims to address gaps in current knowledge and seeks to improve the situation through a system-wide intervention. By using a One Health approach we can address important research questions that individual discipline investigations are unable to. The results obtained should thus more closely reflect the world in which human health, animal health and the environment are inextricably and intimately interlinked. Methods and analysis Both quantitative and qualitative studies are included for households from 12 villages, their surrounding environment and a tertiary care hospital in a nearby town. The studies include analyses of antibiotic consumption for humans and pigs; qualitative and quantitative data on perceptions, knowledge and attitudes; faecal carriage of extended spectrum β-lactamase and carbapenemase-producing Enterobacteriaceae from pigs and humans, and occurrence in household drinking water, surface water, waste water and clinical bacterial isolates from the hospital. Carriage of methicillin-resistant Staphylococcus aureus in humans, household pigs and clinical bacterial isolates is also investigated. Furthermore, potential inter-relationships between these sources are analysed. A multifaceted One Health intervention is designed and implemented in 6 of the 12 villages. Repeated and continuous data collections take place over 2 years, where the repeated data collection is performed after 1 year of intervention. Comparisons are made between intervention and control villages, before and after the intervention. Ethics Ethics approval was obtained from the first Affiliated Hospital, College of Medicine, Zhejiang University, China, reference number 2015#185 and 2015#283.

The human stress hormones catecholamines play a critical role in communication between human microbiota and their hosts and influence the outcomes of bacterial infections. However, it is unclear howM. tuberculosissenses and responds to certain types of human stress hormones. In this study, we screened several human catecholamine stress hormones (epinephrine, norepinephrine, and dopamine) for their effects onMycobacteriumgrowth. Our results showed that epinephrine significantly stimulated the growth ofM. tuberculosisin the serum-based medium as well as macrophages. In silico analysis and molecular docking suggested that the extra-cytoplasmic domain of the MprB might be the putative adrenergic sensor. Furthermore, we showed that epinephrine significantly enhancesM. tuberculosisbiofilm formation, which has distinct texture composition, antibiotic resistance, and stress tolerance. Together, our data revealed the effect and mechanism of epinephrine on the growth and biofilm formation ofM. tuberculosis, which contributes to the understanding of the environmental perception and antibiotic resistance ofM. tuberculosisand provides important clues for the understanding of bacterial pathogenesis and the development of novel antibacterial therapeutics.Keywords:M. tuberculosis;M. smegmatis;epinephrine;mprB;biofilm;antibiotic resistance

Escherichia colicontamination in poultry is a significant concern due to its potential to cause foodborne illness. The presence of extraintestinal pathogenicE. coli(ExPEC) strains in chicken carcasses can lead to severe human infections. This study investigates the prevalence, virulence, and antibiotic resistance ofE. coliisolates from chicken carcasses processed in both wet market and industrial environments, with a focus on the detection capabilities of MALDI-TOF MS. A total of 119E. coliisolates were obtained. Only a small proportion (5/119) carried enteropathogenic virulence genes. In contrast, 71.42% (85/119) of the isolates harbored multiple extraintestinal virulence genes. Among these, iucCandsitA, which are associated with systemic infections, were present in 68.24% (58/85) and 43.53% (37/85) of the isolates, respectively. Furthermore, 47.06% (56/119) of the isolates carrying at least two extraintestinal virulence genes were classified as ExPEC. Additionally, 94.6% (54/56) of ExPEC isolates were multidrug resistant (MDR), showing resistance to over three antibiotic classes, raising concerns about the spread of antibiotic resistance. MALDI-TOF MS profiling revealed significant heterogeneity among the ExPEC isolates, with no distinct clustering patterns based on processing environment or sampling site. These findings underscore the public health risks posed by ExPEC in poultry and emphasize the need for improved surveillance, stringent hygiene practices, and responsible antibiotic use in poultry production.

With the increase of antibiotic resistance currently available at an alarming rate, new improved films of Chitosan (CS) have been achieved with different levels of Tigecycline (TIGE) using traditional casting technology. The structural, optical, and morphological characterizations of the prepared (CS/TIGE) composites were investigated using XRD, FT-IR, UV–visible, and FE-SEM. Also, the antibacterial characterization was investigated. XRD pattern shows a semi-crystalline peak at 2θ = 21° begins to appear at Cs/TIGE2 concentration and its intensity increase with increasing TIGE concentration which indicate the interaction between TIGE and Cs. FT-IR reveals conservation of the distinctive bands with change in both intensity and position that attributed to increase the ratios of TIGE in Chitosan. UV–vis spectra confirmed the interaction of the CS and the drug TIGE. FE-SEM photos confirm the increasing of TIGE particles on the surface of CS/TIGE films by increasing the ratios of TIGE. The antimicrobial effect of Chitosan and five different ratios of Chitosan with TIGE have been identified against four different bacterial species, two gram-negative ( Escherichia coli , Bacillus subtilis) and two gram-positive bacteria ( Staphylococcus aureus , Pseudomonas aeruginosa ). The obtained results indicated that the composite Cs/TIGE can be used in many antimicrobial applications because of its greatly antimicrobial activity .

Introduction: Salmonella Typhimurium is the leading cause of foodborne illnesses in China, resulting in major epidemics and economic losses in recent years. Uridine diphosphate–glucose pyrophosphorylase galU plays an important role in thebiosynthesis of the bacterial envelope. Herein, we evaluated the role of galU in S. Typhimurium infection in chicken. Methods: A galU gene mutant was successfully constructed by red homologous recombination technology, and biological characteristics were studied. Results: The galU mutant strain had a rough phenotype;was defective in biofilm formation, autoagglutination, and motility; exhibited greater sensitivity to most antibiotics, serum, and egg albumen; and had lowercapacity for adhesion to chicken embryo fibroblasts cell line (DF-1). The galU mutant showed dramatically attenuated pathogenicity in chicken embryos (100,000-fold), BALB/c mice (420-fold), and chicks (100-fold).Discussion: The results imply that galU is an important virulence factor in the pathogenicity of S. Typhimurium, and it may serve a target for the development of veterinary drugs, providing a theoretical basis for the prevention and control of S. Typhimurium.

In aquatic environments, antibiotics degrade into byproducts, potentially enhancing bacterial resistance. However, the specific mechanisms by which these byproducts induce bacterial resistance remain elusive. This study conducted experimental evolution experiments to explore how E. coli adapts to tetracycline (TC) and its primary degradation products—anhydrotetracycline (ATC), epitetracycline (ETC), and 4-epianhydrotetracycline (EATC)—through evolution experiments. Prolonged exposure to TC and its byproducts significantly increased frequency of resistant mutants in E. coli ATCC25922, with a maximum 10 6 -fold increase. Resistant mutants exhibited markedly elevated minimum inhibitory concentrations (MICs) for TC, ampicillin (AMP), and ciprofloxacin (CIP), indicating multidrug resistance. Transcriptomic analysis showed that the antibiotic resistance phenotype could be related to enhanced target protection, metabolic adaptations, and reduced membrane permeability. The induction pathways between TC and its byproducts were distinct. Specifically, TC 20d (where TC 20d represents the mutants collected after 20 days of continuous exposure to TC) was associated with more alterations in ribosome-associated genes, which was correlated with an enhanced defensive response as shown by the data. Moreover, variations in energy metabolism gene expression suggest a robust metabolic defense in ATC 20d and ETC 20d . When TC and its byproducts—ATC, ETC, and EATC—act together, they induce antibiotic resistant mutants at rates of 29.8 %, 18.9 %, 18.3 %, and 31.9 %, respectively. This study provides a descriptive overview of the possible adaptive mechanisms and pathways that may be involved in antibiotic resistance due to environmental exposure.

The emergence of antibiotic-resistant bacteria necessitates the development of novel, sustainable, and biocompatible antibacterial agents. This study addresses cytotoxicity and environmental concerns associated with traditional silver nanoparticles (AgNPs) by exploring lignin, a readily available and renewable biopolymer, as a platform for AgNPs. We present a novel one-pot synthesis method for lignin-based AgNPs (AgNPs@AL) nanocomposites, achieving rapid synthesis within 5 min. This method utilizes various organic solvents, demonstrating remarkable adaptability to a wide range of lignin-dissolving systems. Characterization reveals uniform AgNP size distribution and morphology influenced by the chosen solvent. This adaptability suggests the potential for incorporating lignin-loaded antibacterial drugs alongside AgNPs, enabling combined therapy in a single nanocomposite. Antibacterial assays demonstrate exceptional efficacy against both Gram-negative and Gram-positive bacteria, with gamma-valerolactone (GVL)-assisted synthesized AgNPs exhibiting the most potent effect. Mechanistic studies suggest a combination of factors contributes to the antibacterial activity, including direct membrane damage caused by AgNPs and sustained silver ion release, ultimately leading to bacterial cell death. This work presents a straightforward, adaptable, and rapid approach for synthesizing biocompatible AgNPs@AL nanocomposites with outstanding antibacterial activity. These findings offer a promising and sustainable alternative to traditional antibiotics, contributing to the fight against antibiotic resistance while minimizing environmental impact.
