Antibacterial and hemostatic bilayered electrospun nanofibrous wound dressings based on quaternized silicone and quaternized chitosan for wound healing

Zhongfei Gao,Changming Su,Chen Wang,Yulong Zhang,Chunhua Wang,Huanhuan Yan,Guige Hou

The design of novel wound dressings with favorable mechanical properties, hemostasis, inherent antibacterial function, and biocompatibility to facilitate skin wound healing and inhibit scar formation has practical significance in clinical applications. Herein, a bilayered nanofibrous membrane (PQCQS) was fabricated by electrospinning poly(ε-caprolactone) (PCL)/quaternized silicone (PQS, outer layer) and polyvinyl alcohol/collagen/quaternized chitosan (PCQC, inner layer) as wound dressings for damaged skin. The PQS nanofiber was utilized as the outer layer of the wound dressing because it combined the good mechanical property of PCL and antibacterial   performance of quaternized silicone to suppress scar formation and provide a stable microenvironment for the wound healing process. The inner membrane (PCQC) exhibited hydrophilicity, biocompatibility, and hemostasis, thus facilitating wound healing. The PQCQS membranes exhibited excellent hydrophilicity, outstanding thermal stability, comparable mechanical properties with human skin, efficient hemostatic performance, and cytocompatibility. Moreover, the PQCQS membranes demonstrated a good balance between antibacterial activity and cell proliferation. Moreover, the PQCQS membranes significantly expedited the wound healing process and inhibited scar hyperplasia better than commercial ointment (MSSG and KELO-COTE) in a rabbit ear full-thickness wounds defect model. That is, the bilayered nanofibrous membranes, with excellent antibacterial activity, hydrophilicity, hemostatic performance, scar inhibition, and wound healing properties, are appropriate candidates for wound dressings.

Control of Surface Wrinkles on Shape Memory PLA/PPDO Micro-nanofibers and Their Applications in Drug Release and Anti-scarring

Wang Lu,Ma Jingyi,Guo Tao,Zhang Fenghua,Dong Aimeng,Zhang Shiqi,Liu Yanju,Yuan Huiping,Leng Jinsong

Micro- and nano-fibers of shape memory polymers (SMP) offer multiple advantages like high specific surface area, porosity, and intelligence, and are suitable for biomedical applications. In this study, biodegradable poly (p-dioxanone) (PPDO) materials were incorporated to improve the brittleness of shape memory polylactic acid (PLA), and plasticizers were used to reduce the transition temperature of SMP composites such that their transitions could be induced close to body temperature. Furthermore, an electrostatic spinning technology was applied to prepare SMP fibers with wrinkled structures and regulate their microstructures and morphologies such that the intelligent transition of wrinkled and smooth morphologies can be achieved on the fiber surface. The application of this controllable-morphology fiber membrane in intelligent controlled drug release and scar inhibition after Ahmed Glaucoma Valve (AGV) implantation was also studied. The drug release from the stretched and deformed drug-loaded fiber membranes was faster than those from membranes with the original shape. This membrane with micro- and nano-fibers had good anti-scarring effects that improved after drug loading. The achievement of intelligent controlled drug release and the evident anti-scarring effects of the membrane broaden the application of SMP fibers in the biomedical field. Graphical Abstract

In vivo study of silk fibroin/gelatin electrospun nanofiber dressing loaded with astragaloside IV on the effect of promoting wound healing and relieving scar

Danping Zhang,Longjian Li,Yinghui Shan,Jie Xiong,Zhongjie Hu,Yan Zhang,Jianqing Gao

This study was to investigate the therapeutic effects of silk fibroin/gelatin (SF/GT) electrospun nanofiber dressing loaded with astragaloside IV (AS) on acute trauma. SF/GT nanofiber dressing was prepared by electrospinning nanotechnology and loaded with AS. Four groups of rats inflicted with full-thickness wounds on the back were treated with saline, blank nanofiber dressing (blank control), AS solution and AS-loaded SF/GT nanofiber dressing, respectively. The results showed that AS-loaded SF/GT nanofiber dressing group had a significantly higher wound healing closure rate compared with control group on the 3rd, 6th, and 9th day after treatment. The VEGF content and the number of macrophages in the wound skin area of AS-loaded SF/GT nanofiber dressing group increased significantly compared to the blank control group 7 days after treatment (36 versus 4; 72 versus 38, p < 0.01, respectively). The expression level of Alpha smooth muscle actin (α-SMA) was inhibited by AS-loaded SF/GT nanofiber dressing treatment compared to the blank control, and the anti-scar effect was further demonstrated by scanning electron microscope and histological analyses. The conclusion of this study was that AS-loaded SF/GT nanofiber dressing with both healing accelerating ability and anti-scaring effect was proved to be a promising therapeutic for acute trauma treatment.

Silk fibroin/gelatin electrospun nanofibrous dressing functionalized with astragaloside IV induces healing and anti-scar effects on burn wound

Ying-Hui Shan,Li-Hua Peng,Xin Liu,Xi Chen,Jie Xiong,Jian-Qing Gao

Functional wound dressing has provided new challenges for researchers who focus on burn to improve skin graft quality, reduce scarring, and develop a pluristratified dermal or epidermal construct of a burn wound. This study aimed to investigate the effect of a silk fibroin/gelatin (SF/GT) electrospun nanofibrous dressing loaded with astragaloside IV (AS) on deep partial-thickness burn wound. AS-loaded SF/GT-blended nanofibrous dressing was prepared by electrospinning nanotechnology. The optimal ratio (25:75) of silk fibroin to gelatin was further optimized by evaluating ATR-FTIR characteristics, mechanical properties, porosity, swelling rate, degradation, and release profile of the AS-loaded SF/GT nanofibrous dressing. In contrast to the blank control, the AS-loaded SF/GT nanofibrous dressing promoted cell adhesion and proliferation with good biocompatibility in vitro ( p   <   0.01). This dressing also accelerated wound healing and inhibited scar formation in vivo by stimulating wound closure ( p   <   0.05), increasing angiogenesis, regulating newly formed types of collagen, and improving collagen organization. These results showed that SF/GT nanofibrous dressing is a promising topical drug delivery system. Furthermore, AS-functionalized SF/GT nanofibrous dressing is an excellent topical therapeutic that could be applied to promote healing and elicit anti-scar effects on partial-thickness burn wound.
