Nanomaterial technology has attracted much attention because of its antibacterial and drug delivery properties, among other applications. Metal-organic frameworks (MOFs) have advantages, such as their pore structure, large specific surface area, open metal sites, and chemical stability, over other nanomaterials, enabling better drug encapsulation and adsorption. In two examples, we used the common pathogenic bacteriumStaphylococcus aureusand highly infectious influenza A virus. A novel complex MIL-101(Fe)-T705 was formed by synthesizing MOF material MIL-101(Fe) with the drug favipiravir (T-705), and a hot solvent synthesis method was applied to investigate the in vitro antibacterial and antiviral activities. The results showed that MIL-101(Fe)-T705 combined the advantages of nanomaterials and drugs and could inhibit the growth ofStaphylococcus aureusat a concentration of 0.0032 g/mL. Regarding the inhibition of influenza A virus, MIL-101(Fe)-T705 showed good biosafety at 12, 24, 48, and 72 h in addition to a good antiviral effect at concentrations of 0.1, 0.2, 0.4, 0.8, 1.6, and 3 μg/mL, which were higher than MIL-101(Fe) and T-705.Keywords:MIL-101(Fe);favipiravir;antibacteria;antiviral

Largemouth bass ranavirus (LMBRaV), also known as largemouth bass virus (LMBV), is a highly damaging viral pathogen that causes widespread mortality in cultured largemouth bass and poses a significant threat to the farming industry in China. However, there is a paucity of effective preventive methods for this disease by LMBRaV. Research has established that herbal extracts possess antiviral activity, potentially exerting a controlling effect on viruses in aquaculture. Therefore, the identification of simple and structurally safe herbal extract immunomodulators is critical for largemouth bass health and development. Rhein, an herbal compound, exhibits a broad spectrum of pharmacological effects in herbal medicine. This study aimed to evaluate the properties and effects of rhein against LMBRaV in epithelioma papilloma cyprinid (EPC) and largemouth bass. In vitro CCK-8 assays were performed to determine the highest safe concentrations. Significantly, 50 mg/L rhein effectively inhibited cytopathic effects (CPE) in EPC induced by LMBRaV, resulting in a 97 % reduction in viral replication rates. Furthermore, a six-point dose-response experiment revealed that rhein doses as low as 6.4 mg/L inhibited LMBRaV and suppressed LMBRaV infection with a dose-dependent manner. Subsequent experiments demonstrated that sustained rhein activity effectively alleviated CPE and nuclear damage caused by LMBRaV infection. Additionally, rhein exerted protective effects against declines in LMBRaV-induced mitochondrial membrane potential. In vivo experiments validated that largemouth bass diets supplemented with 0.075 % rhein exerted a protective effect against LMBRaV infection. Moreover, LMBRaV MCP gene expression levels were lower in several organs of fish in the rhein supplementation group. Immunogene analysis determined that rhein upregulated relevant gene expression levels in multiple organs of largemouth bass, with higher activities observed for the Mx, INF-γ, and IRF3 genes. Histopathological results displayed that a supplemental dosage of 0.075 % rhein significantly mitigated LMBRaV infection-induced pathological damage in the liver, spleen, and kidney. Taken together, these in vivo and in vitro results support the effective use of rhein as a promising candidate for the development of antiviral treatment against LMBRaV infection, highlighting its role in aquatic animal husbandry as a wide-spectrum antiviral and immune response booster.

Micropterus salmoidesrhabdovirus (MSRV) is a primary viral pathogen in largemouth bass aquaculture, which leads to tremendous economic losses yearly. Currently, there are no approved drugs for the treatment and control of this virus. Our previous studies screened the herbMagnolia officinalisfrom many traditional Chinese medicines, and we isolated and identified magnolol as its main active compound against multiple rhabdoviruses, including MSRV. On the basis of the structure–activity relationship and pharmacophore model of magnolol, two new magnolol derivatives, namely, hydrogenated magnolol and 2,2′-dimethoxy-magnolol, were designed and synthesized. Their anti-MSRV activities were systematically investigated both in vitro and in vivo. By comparing the half-maximal inhibitory concentration (IC50), it was found that hydrogenated magnolol possessed a higher anti-MSRV activity than magnolol and 2,2′-dimethoxy-magnolol, with an IC50of 13.37 μM. Furthermore, hydrogenated magnolol exhibited a protective effect on the grass carp ovary (GCO) cell line by reducing the cytopathic effect induced by MSRV. Further studies revealed that hydrogenated magnolol did not directly impact virions or interfere with MSRV adsorption. It worked within the 6–8 h of the phase of virus replication. In vivo treatment of MSRV infection with magnolol and hydrogenated magnolol showed that they significantly improved the survival rate by 44.6% and 62.7%, respectively, compared to MSRV-infected groups. The viral load measured by the expression of viral glycoprotein in the organs including the liver, spleen, and kidney also significantly decreased when fish were intraperitoneally injected at a dose of 20 mg/kg. Altogether, the structural optimization of magnolol via hydrogenation of the propylene groups increased its anti-MSRV activity both in vitro and in vivo. These results may provide a valuable reference for anti-MSRV drug discovery and development in aquaculture. Keywords:Micropterus salmoidesrhabdovirus (MSRV);magnolol;largemouth bass;antiviral activity

Transmission through the respiratory tract is one of the most important ways for bacteria and viruses to infect the human body; the use of high-performance antibacterial and antiviral protective equipment is the most effective way to prevent the spread of respiratory diseases. However, at present, most personal protective equipment lacks the ability to kill pathogens. In this paper, a kind of polytetrahydropyrimidine–polyethylene terephthalate functional fiber (PET–PTHP fibers) with highly sustained antibacterial and antiviral properties was prepared. The inactivation rate of the fibers against Staphylococcus aureus and Escherichia coli was as high as 99.99%, and the antibacterial time was more than 72 h. The fibers have an obvious destructive effect on lentiviruses and can reduce the infection rate of lentiviruses in BxPC-3 cells from 25.4 to 9.7%. The cytotoxicity test, cell live/dead staining test, and cell proliferation test all confirmed that PET–PTHP fibers have no obvious cytotoxicity and good cytocompatibility. By applying the functional fibers to the inner layer of the masks, a new type of mask with adsorption, filtration, and killing properties against pathogens was prepared. The filtration efficiency of the new masks was 99.3%, and the pressure drop was 104 Pa. The new masks have excellent air permeability and filtration effect, meet the practical application conditions, and are of grade A; therefore, these masks provide medical protection as well as kill pathogens at the same time, further reducing the risk of human infection.

The unprecedented pandemic has highlighted the need for textiles that rapidly inactivate pathogens to protect public health. However, how to achieve rapid, effective and durable pathogen inactivation with the minimum antimicrobial dose loading has become a major challenge for cotton fabric modification. Here, a surface engineering cotton fabric (Co-CMC@Cu 2+ ) was prepared by grafting carboxymethyl chitosan (CMC) onto cotton fabric and then loading with Cu 2+ via coordination. The Co-CMC@Cu 2+ with minimal copper ion dosage achieved rapid inactivation, excellent comfort, good biosafety, and durability. Specifically, Co-CMC modified with 12.5 mmol/L copper ion solutions achieved impressive bacterial reduction (BR) rates of 94.3 % and 89.3 % against E. coli and S. aureus , respectively, within 10 min of contact. Moreover, the bacteria were completely eradicated after 2 h of continuous contact. Additionally, Co-CMC@Cu 2+ achieved excellent phage reduction (PR) rates of 98.9 % and 100 % against phi6 and phi-x174 bacteriophages, respectively, within 30 min of exposure, highlighting its potential for viral inactivation. Furthermore, Co-CMC@Cu 2+ demonstrated exceptional durability for pathogen inactivation, with BR and PR values above 80 %, even after 150 washing cycles. This novel antimicrobial and antiviral cotton fabric will be a promising candidate textile to protect public health during a pandemic.

The emerging COVID-19 pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has raised a global catastrophe. To date, there is no specific antiviral drug available to combat this virus, except the vaccine. In this study, the main protease (M pro ) required for SARS-CoV-2 viral replication was expressed and purified. Thirty-six compounds were tested as inhibitors of SARS-CoV-2 M pro by fluorescence resonance energy transfer (FRET) technique. The half-maximal inhibitory concentration (IC 50 ) values of Ebselen and Ebsulfur analogs were obtained to be in the range of 0.074–0.91 μM. Notably, the molecules containing furane substituent displayed higher inhibition against M pro , followed by Ebselen 1i (IC 50 = 0.074 μM) and Ebsulfur 2k (IC 50 = 0.11 μM). The action mechanism of 1i and 2k were characterized by enzyme kinetics, pre-incubation and jump dilution assays, as well as fluorescent labeling experiments, which suggested that both compounds covalently and irreversibly bind to M pro , while molecular docking suggested that 2k formed an S S bond with the Cys145 at the enzymatic active site. This study provides two very potent scaffolds Ebsulfur and Ebselen for the development of covalent inhibitors of M pro to combat COVID-19.

Previous researches suggested that polysaccharides from brown algae had anti-virus activity. We hypothesized that nature polysaccharide from marine plants might have the effect on anti-SARS-CoV-2 activity. By high throughput screening to target 3CLpro enzyme using polysaccharides library, we discover a crude polysaccharide 375 from seaweed Ecklonia kurome blocked 3CLpro enzymatic activity and shows good anti-SARS-CoV-2 infection activity in cell. Further, we show that homogeneous polysaccharide 37502 from the 375 may bind to 3CLpro well and disturb spike protein binding to ACE2 receptor. The structure characterization uncovers that 37502 is alginate. These results imply that the bioactivities of 375 on SARS-CoV-2 may target multiple key molecules implicated in the virus infection and replication. The above results suggest that 375 may be a potential drug candidate against SARS-CoV-2.

Largemouth bass ranavirus (LMBRaV) is a highly lethal virus that has devastated the largemouth bass aquaculture sector. Therefore, it is imperative to develop therapeutic strategies for the control of this pathogen. In this work, the antiviral activity of Saikosaponin C was examined both in vitro and in vivo . According to the results of the in vitro antiviral activity assay, 30 mg/L of Saikosaponin C significantly down-regulated the expression of the LMBRaV major coat protein (MCP) gene in epithelioma papilloma cyprinid (EPC) cells, with an inhibitory rate exceeding 84%, and prevented LMBRaV replication in a dose-dependent manner. It also significantly limited its cytopathic effect (CPE), thereby playing a protective role on infected cells. Notably, Saikosaponin C has a certain preventative impact on the virus by influencing the mechanisms by which viral particles connect to cell receptors, which prevents LMBRaV from adsorbing on EPC cells. Simultaneously, Saikosaponin C also directly inactivated LMBRaV. The results of exploring the viral invasion cycle demonstrated that Saikosaponin C primarily exerted its antiviral effect during the middle stage (2–6 h) of viral infection. In addition, Saikosaponin C inhibited LMBRaV-induced apoptosis of EPC cells, maintained physiological cellular morphology and structure, and protected cells from loss of cellular morphology, formation of apoptotic vesicles, and cell fragmentation. Moreover, Saikosaponin C considerably improved the survival of virus-infected largemouth bass, according to the findings of in vivo investigations. In summary, Saikosaponin C demonstrated excellent anti-LMBRaV activity and holds considerable promise for development in the field of aquaculture.

Human enteroviruses cause many diseases; however, there is no specific therapeutic drug. G-quadruplex is an atypical secondary structure formed in the guanine rich region of DNA or RNA, which can exist in the viral genome. The different positions of G-quadruplex play an important role in the regulation of virus replication and infection. Whether G-quadruplexes are present in human enteroviruses is unknown. In current study, we analyzed the potential quadruplex forming sequences of human enteroviruses, especially EV-A71 virus, which causes hand, foot, and mouth disease. The results showed that there were a certain number of potential quadruplex-forming sequences in human enteroviruses. Through a variety of experimental methods, we evaluated the formation potential of EV-A71 encoded G-quadruplex and analyzed the binding ability of G-quadruplex ligands, including BRACO-19, pyridostatin and TMPyP4 to virus encoded G-quadruplexes. G-quadruplex ligands BRACO-19, PDS and TMPyP4 could inhibit the transcription of constructs containing EV-A71 G-quadruplex sequences. Moreover, we found that BRACO-19 was able to inhibit the replication of EV-A71, suggesting that targeting G-quadruplexes in EV-A71 genome by G-quadruplex ligands could be a novel antiviral way against EV-A71. Our finding not only uncovered the G-quadruplexes in human enteroviruses, but also would provide a new strategy for human enteroviruses therapy.IMPORTANCE G-quadruplex is a stable nucleic acid secondary structure formed by the folding of guanine rich nucleic acid. The important regulatory function of G-quadruplex makes it an attractive target of antiviral effect. Human enteroviruses cause a variety of human diseases, including common cold, nervous system diseases, cardiovascular damage, and diabetes. Enterovirus A71 (EV-A71) is one of pathogens causing hand, foot, and mouth disease; however, whether G-quadruplexes are present in the genomes of human enteroviruses is unknown. The function of G-quadruplexes in the EV-A71 genomes is not clear. We predicted and characterized G-quadruplex sequences in EV-A71. G-quadruplex ligands were identified to stabilize EV-A71 G-quadruplexes with high affinities. We also demonstrated G-quadruplex ligand BRACO-19 inhibited EV-A71 replication. Our studies provide a framework for targeting G-quadruplexes in the enteroviruses genome, which will be a new way to develop antiviral agents against human enteroviruses.

The recent outbreak of a coronavirus disease (COVID-19) has posed a great threat to public health and financial system. Most current masks used to prevent the spread of COVID-19 are typically absence of biocidal properties. We designed a novel polymer, polystyrene grafted by 5, 5-dimethylhydantoin and trimethylamine (PSDT), which possesses halamine site and cationic quaternary ammonia salt site. Furthermore, PSDT/PU [email protected] membranes (PSDT/PU NNMs) were obtained by electrospinning technology. Our strategy enables inherent N-halamine and quaternary ammonia salt (QAS) group to be covalently integrated into membranes, realizing the efficient and stable biocidal properties. Meanwhile, the introduction of nanonets endows electrospun membranes with prominent air filtration performance. The resulting membranes exhibit integrated properties of high interception of fine particles (96.7%) and low pressure drop (95.4 Pa). Besides, chlorinated PSDT/PU [email protected] membranes (with active chlorine content of 0.60 wt% and quaternary ammonia salt content of 2.20 wt%) exhibited superior bactericidal (>99.9999%) and virucidal (>99.999%) efficiency in a short time (2 min), which enables chlorinated PSDT/PU NNMs to be served as the filtration material by providing bacterial interception (99.77%) and contact killing against pathogens. The successful synthesis of PSDT/PU NNMs provide innovative insights for exploring filtration materials in a nanonet and biocidal form.

Micropterus salmoides rhabdovirus (MSRV) is a prominent pathogen that mainly endangers largemouth bass fry, which often causes devastating diseases to largemouth bass and leads to huge economic losses in aquaculture. So far, there is no approved effective measure to control MSRV infection. In the study, 24 magnolol derivatives were used to conduct anti-MSRV activity screening by RT-qPCR, and the results indicated that 5,5′-diallyl-3,3′-dinitro-[1,1′-biphenyl]-2,2′-diol (M3) exhibited the best anti-MSRV efficiency with IC 50 of 0.92 μM. Moreover, further study showed that the CPE induced by MSRV was obviously weaken by M3, and the viral titers were also significantly decreased by M3 within 96 h after MSRV infection ( p < 0.05). Next, the anti-MSRV mechanism of M3 was preliminarily investigated, and the findings illustrated that M3 did not directly act on MSRV virion which had no effect on the infectivity of MSRV. Furthermore, the adsorption of MSRV onto GCO cells was not hindered by M3. The results of cell cycle detected by flow cytometry demonstrated that M3 altered the state of cell cycle arrest in S phase induced by MSRV, influencing the environment required for virus replication. In addition, observation of survival rate and determination of viral load were conducted to further verify the antiviral activity of M3 against MSRV. The results illustrated that administration with M3 at the dose of 20 mg/kg via intraperitoneal injection significantly enhanced the survival rate of largemouth bass infected with MSRV by 35.98 % ( p < 0.01). Moreover, the viral loads of liver, spleen and kidney were significantly decreased by M3 on day 3 and day 5 post MSRV infection ( p < 0.05). Finally, the antiviral effects of M3 against aquatic rhabdoviruses spring viremia of carp virus (SVCV) and infectious hematopoietic necrosis virus (IHNV) were explored, and the results demonstrated that M3 evidently lessened the production of CPE induced by SVCV and IHNV, and inhibited the proliferation of SVCV and IHNV in EPC cells in a dose-dependent manner, with IC 50 of 2.92 μM and 2.78 μM, respectively. To sum up, M3 exhibits a high antiviral activity against MSRV, and is expected to be developed into antiviral specific drugs against rhabdoviruses in aquaculture.

This study delves into the antiviral efficacy of Formononetin (FMN) and Mizoribine (MZR) against the Porcine Reproductive and Respiratory Syndrome Virus (PRRSV), a virus with a considerable economic impact and a current void in effective treatments. FMN and MZR were found to inhibit various PRRSV strains in vitro, predominantly in the early stages of viral infection. Noteworthy was the observation of their synergistic effects when combined with Ribavirin. The study underscores the antiviral potential of FMN and MZR, particularly emphasizing their low cytotoxicity at specific concentrations. These results position FMN and MZR as promising antiviral agents against PRRSV, underscoring their low cytotoxicity and efficacy in early-stage viral inhibition. Such findings pave the way for their potential inclusion in future PRRSV management strategies.
