Amomum tsaoko is widely distributed in south-west China as a spice. In present study, the antibacterial activity of Amomum tsaoko essential oil (AEO) against foodborne pathogens was evaluated. The antibacterial activity was determined by measuring the zones of inhibition (ZOI), minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and the time-kill assay. Results showed the susceptibility of foodborne bacteria Escherichia coli ( E. coli ) was excellent with the lowest MIC and MBC values of 3.13 and 6.25 mg/mL, respectively. The probable antibacterial mechanism of AEO was the change in permeability and integrity of the cell membranes leading to leakage of nucleic acids and proteins. Detection of the kinetics of E. coli deactivation in situ showed that AEO has good preservative activities in foods. It is necessary to consider that AEO will become a promising antibacterial additive for food preservative.

BACKGROUND In order to prolong the storage and inhibit microorganisms of pakchoi, the antibacterial activity and mechanism of ultrasound combined with sodium hypochlorite (NaClO-US), the efficiency of NaClO-US in reducing Escherichia coli , Staphylococcus aureus and Pseudomonas aeruginosa as well as preserving quality of pakchoi were investigated. RESULTS Ultrasound treatment could significantly reduce the usage of NaClO solution from 800 ppm to 500 ppm. NaClO-US decreased the counts of E. coli , S. aureus and P. aeruginosa , which disrupted the bacterial cell membrane with cytoplasmic leakage. In addition, NaClO-US significantly increased cell membrane permeability, while cell membrane integrity decreased, the secondary structure of bacterial proteins showed several obvious changes, such as the increase of random coil content, as well as the decrease of α -helix content. The bacterial counts, E. coli , S. aureus and P. aeruginosa population in pakchoi treated with NaClO-US reduced by 1.89, 1.40, 1.60, 1.72 log CFU g −1 , respectively compared to control sample after storage for 15 days. NaClO-US resulted in minimum chlorophyll depletion, flavor and sensory deterioration. CONCLUSION NaClO-US solution treatment inhibited microorganisms and prolonged storage of pakchoi. © 2022 Society of Chemical Industry.

The previous study indicated that cuminaldehyde (CUM) could be used as an antibacterial agent in sauced beef to reduce the propagation of Staphylococcus aureus ( S. aureus ). This research took sauced beef treated with 0.4 μL/mL CUM as the research object. Transcriptomic and proteomic methods were used to comprehensively analyze the changes in genes and proteins of S. aureus under CUM stress. A total of 258 differentially expressed genes (DEGs, 178 up-regulated and 80 down-regulated) and 384 differentially expressed proteins (DEPs, 61 up-regulated and 323 down-regulated) were found. It was observed that CUM destroyed the cell wall and cell membrane by inhibiting the synthesis of peptidoglycan and fatty acid. Low energy consumption strategies were formed by reducing glycolysis and ribosome de novo synthesis. The levels of genes and proteins associated with the glycine, serine , threonine , methionine , cysteine, and branched-chain amino acids were dramatically changed, which impaired protein synthesis and reduced bacterial viability . In addition, the up-regulated DEGs and DEFs involved in DNA replication , recombination and single-stranded DNA-binding contributed to DNA repair . Moreover, ATP-binding cassettes (ABC) transporters were also perturbed, such as the uptake of betaine and iron were inhibited. Thus, this study revealed the response mechanism of S. aureus under the stress of CUM, and provided a theoretical basis for the application of CUM in meat products.

Botryosphaeria dothidea, a notorious plant pathogen, is responsible for causing chestnut rot during postharvest storage. This research aimed to assess the antifungal properties of perilla essential oil (PEO) and perillaldehyde (PAE) againstB. dothidea. PEO’s and PAE’s inhibitory effects onB. dothideawere investigated using an agar dilution method, a fumigation method, and an in vivo assay in chestnuts and shell buckets. Based on the results of gas chromatography-mass spectrometry, it was confirmed that the main component of PEO was elemicin. The antifungal mechanism of PEO and PAE againstB. dothideawas investigated by conducting staining experiments of the fungal cell wall and cell membrane. PEO and PAE strongly inhibit the mycelial growth ofB. dothideain a dose-dependent manner. The inhibitory mechanism is mainly related to the destruction of the integrity of the fungal cell wall and plasma membrane. Notably, PEO retains its antifungal efficacy againstB. dothideain chestnuts, effectively prolonging their storage life. These findings indicate that PEO and PAE are nontoxic, eco-friendly botanical fungicides, holding promise for controlling postharvest chestnut rot.

This study investigated the effects of shade drying (SD), hot air drying (HD), vacuum drying (VD), and vacuum freeze-drying (VFD) on the drying characteristics, bioactive compound content, and biological activities of pomegranate peel. Additionally, preliminary research was conducted on the antimicrobial mechanisms of pomegranate peel polyphenols. The results demonstrated that VFD achieved the shortest drying time while maintaining superior appearance quality and the highest levels of bioactive compounds: total phenolic content (TPC, 86.78 mg/g), total flavonoid content (TFC, 19.37 mg/g), total tannin content (TTC, 7.6 mg/g), total anthocyanin content (TAC, 0.07 mg/g), and ABTS antioxidant capacity (383.10 mg TE/g). The VFD treatment also showed superior •OH radical scavenging activity (122.50 mg TE/g) and α-ALA inhibition (5.18 mg/mL), as well as maximum inhibition zones against S. aureus (21.33 ± 0.04 mm), B. subtilis (16.23 ± 0.04 mm), and A. tenuissima (83.89%). The HD demonstrated slightly lower but comparable quality parameters to VFD. 22 major phenolic compounds were identified in the VFD. Molecular docking studies revealed four compounds, including punicalagin, punicalin, rutin, and ellagic acid 4-O-xylopyranoside, that effectively bound to fungal cell wall functional proteins. Among these, punicalagin exhibited primary interactions through hydrogen bonding and hydrophobic interactions with the target proteins.

Meat spoilage caused by Salmonella Typhimurium ( S. typhimurium ) contamination has become a major concern worldwide. Herein, the antibacterial mechanisms of cinnamaldehyde (CIN) against S. typhimurium were evaluated, and the alginate dialdehyde (ADA)-gelatin (Gel) crosslinked hydrogels loaded with CIN nanoparticles prepared by zein and carboxymethyl chitosan (ZCC/CINNPs) were fabricated for meat preservation. Results showed the efficient antibacterial effects of CIN via interfering with the S. typhimurium cell membrane, increasing the membrane permeability, disrupting the membrane integrity, thereby leading to the intracellular materials leakage and cell death. ZCC/CINNPs were prepared and characterized, showing potent antibacterial and antioxidant activities, and excellent stabilities. ZCC/CINNPs addition into ADA/Gel hydrogels markedly improved the barrier, antibacterial and antioxidant properties, while decreased the water loss. Furthermore, the release behaviors of CIN from the composite hydrogels in different food systems suggested the suitability for the preservation of various food products. Practical application results showed the composite hydrogels with 25 % ZCC/CINNPs (AGH-ZC 25 %) enhanced the storage stability of chicken and pork meat at 4 °C, which decreased the weight loss rate, maintained the pH, color, and textural stabilities, retarded the microbial growth and lipid oxidation of meat samples. Findings here suggested AGH-ZC 25 % hydrogels had great potential in meat preservation.

Background : Streptococcus suis is a zoonotic pathogen that causes meningitis, septicaemia, endocarditis, arthritis, and pneumonia in human beings. With the increasing prevalence of S. suis infections and a general decline in the effectiveness of antibiotics, the development of novel drugs that have effect on S. suis is extremely urgent. Emodin, a natural anthraquinone derivative of Rheum palmatum L., Reynoutria japonica Houtt., Polygonum multiflorum Thunb. and Cassia obtusifolia L., has been reported to exert anti- S. suis effect; however, the specific mechanism of the anti- S. suis action by targeting β-ketoacyl-acyl carrier protein synthase Ⅱ (FabF) in the fatty acid synthesis pathway remains unexplored. Purpose: We sought to reveal the potential role of emodin to prevent S. suis infection, investigate its mechanism of anti- S. suis action, and provide further evidence of emodin as an alternative to traditional antibiotic agents. Methods : The in vitro anti- S. suis properties of emodin were assessed through minimum inhibitory concentration (MIC) assays, and time-kill assays. Subsequently, the mechanisms underlying emodin’s mode of action at the molecular level by targeting FabF were elucidated using molecular docking, site-directed mutagenesis, bio-layer interferometry assays, and cellular thermal shift assays. Finally, metabolomics, cell membrane phospholipid content assay and biochemical parameters assays were used to detect emodin disrupting cell membrane integrity and function by affecting fatty acid biosynthesis. Results : In this study, we have identified that emodin inhibits S. suis by suppressing free fatty acids (FFAs) synthesis and disrupting phospholipid production by targeting FabF, a key enzyme in the fatty acid biosynthesis pathway. This interference compromises the integrity and functionality of the cell membranes of S. suis . Emodin also triggers the dissipation of the proton motive force, accelerates the tricarboxylic acid cycle, and enhances cellular respiration, ultimately leading to S. suis cell death. Conclusion : This study suggested that emodin inhibits the growth of S. suis via targeting FabF and the inhibition of fatty acid biosynthesis through enzyme-targeted drug design. This represents a novel strategy for developing antimicrobial agents against S. suis and addressing the challenge of antibiotic resistance.

A multitude of serious chronic infections are involved in bacterial biofilms that are difficult to eradicate. Here, a water-soluble galactose-functionalized cationic 4,4-difluoro-4-bora-3a,4a-diaza-s-indacene (BODIPY)-based photodynamic therapy agent was synthesized for selectively eliminating the bacterial biofilm. These conjugates can capture bacteria to form aggregations through electrostatic interaction and then generate a large number of reactive oxygen species (ROS) under visible light irradiation to kill the bacteria without the emergence of bacterial resistance. Simultaneously, this agent could effectively inhibit and eradicate both Gram-positive and Gram-negative bacterial biofilms. The in-depth analysis of the antimicrobial mechanism confirmed that the conjugates can quickly bind on the bacterial surface, irreversibly disrupt the bacterial membrane, and distinctly inhibit intracellular enzyme activity, ultimately leading to the bacterial death. Importantly, these conjugates are highly selective toward bacterial cells over mammalian cells as well as no cytotoxicity to A549 cells and no discernible hemolytic activity. Collectively, this water-soluble galactose-decorated cationic BODIPY-based photodynamic therapy agent design provides promising insights for the development of therapy for antibiotic-resistant bacteria.

The destructive effect and mode of action of lemon verbena essential oil on cells were investigated, taking the isolatedPseudosciaena D4as the research object. The extracellular absorbance of thePseudosciaena D4increased at OD260 and OD280 after being treated with lemon verbena essential oil, which destroyed the integrity ofPseudosciaena D4cells, showing a significant effect on preventing biomembrane formation and destroying the formed biomembrane. With an increased concentration of lemon verbena essential oil, extracellular polysaccharide showed a significant decrease in content and a significant increase in inhibition rate, indicating that the secretion of extracellular polysaccharide byPseudosciaena D4cells could be inhibited by lemon verbena essential oil during the process of biomembrane formation. Cell introcession and shrinkage appeared after the treatment with essential oil, and a transparent cavity was formed by the out-flowed cell content. Lemon verbena essential oil destroyed the cell wall, resulting in an enhanced permeability of the cell membrane and leakage of the contents, thereby causing cell death. Keywords:lemon verbena essential oil;antibacterial activity;antibacterial mechanism;Pseudosciaena crocea

Carbon dots have attracted much attention in the field of antibacterial because of their good biocompatibility, easy preparation, and basically not inducing bacterial resistance. At present, studies have shown that many antibacterial carbon dots have high positive surface potential, but few researchers have been carried out on how to design carbon dots with high positive surface potential by using nontoxic precursors. In this paper, three structurally similar amino acids with different isoelectric points were selected as carbon sources, and three kinds of carbon dots based on amino acid were synthesized with hydrothermal method. The structural characteristics of the three carbon dots were characterized by XRD, FT-IR, XPS, etc. The content of nitrogen-containing groups of the three carbon dots was compared with the Kaiser test. The relationship between the Zeta potential and the antibacterial effect was also investigated. And it shows that the arginine carbon dots, which have the highest positive charge, also have the best antibacterial performance. So, the arginine carbon dots were further doped with nitrogen-containing molecules (diethylenetriamine, DETA) to obtain high surface positive potential carbon dots (named DETA-Arg-CDs, Zeta potential = + 21.7 mV) with the minimum inhibitory concentration (MIC) against E. coli and S. aureus being 8 and 4 ug/mL, respectively. The antibacterial mechanism of high surface positive potential carbon dots was demonstrated by a series of experiments and computational simulations. Cytotoxicity tests showed that the carbon dots were almost nontoxic within the range of antibacterial concentration. Finally, it was applied to the preparation of antibacterial masks. The bacteriostatic zone experiment showed that the mask sprayed with DETA-Arg-CDs had an obvious bacteriostatic zone, while the blank control group had no bacteriostatic zone. In summary, this study synthesized carbon dots with excellent antibacterial properties by using simple and safe amino acids and small nitrogen-containing molecules and explored the antibacterial mechanism of carbon dots with high surface positive charge.

Geotrichum citri-aurantii(G. citri-aurantii) is one of the most important postharvest pathogens leading to a postharvest loss of citrus by causing sour rot. In this study, the antifungal activity oftrans-2-hexenal, a natural component of essential oil, againstG. citri-aurantiiwas evaluated.Trans-2-hexenal treatment inhibited the mycelia growth ofG. citri-aurantiiwith a minimum inhibitory concentration and minimum fungicidal concentration oftrans-2-hexenal at 0.50 and 1.00 μL/mL, respectively. Moreover,trans-2-hexenal efficiently reduced the incidence of sour rot of Satsuma fruit inoculated withG. citri-aurantii. Ultrastructural observations and Fourier transform infrared (FT−IR) results showed thattrans-2-hexenal treatment affected the cell wall and cell membrane instructions ofG. citri-aurantii. The content ofβ-1,3-glucan was significantly decreased aftertrans-2-hexenal treatment, but the cell wall permeability was not changed. The decrease in lipid and ergosterol contents might be responsible for this antifungal activity. Several important genes,FKS1,ERG1,ERG7, andERG11, showed decreasing expression levels aftertrans-2-hexenal treatment. Molecule-docking results also indicated thattrans-2-hexenal could join with the protein of FKS1, ERG1, ERG7, and ERG11 to impact enzyme activities. These results demonstrated thattrans-2-hexenal is a promising fungicide for controlling sour rot of harvested citrus fruit by damaging the membrane integrity ofG. citri-aurantii.Keywords:G. citri-aurantii;trans-2-hexeanl;antifungal mechanism;cell wall;cell membrane

This study aimed to prepare sodium alginate-linalool emulsion (SA-LE) to overcome the low solubility of linalool and explore its inhibitory activity against Shigella sonnei . The results indicated that linalool significantly reduced the interfacial tension between SA and oil phase ( p < 0.05). Droplet sizes of fresh emulsions were uniform with sizes from 2.54 to 2.58 μm. The ζ-potential was between −23.94 and -25.03 mV, and the viscosity distribution was 973.62 to 981.03 mPa·s at pH 5–8 (near neutral pH) without significant difference. In addition, linalool could be effectively released from SA-LE in accordance with the Peppas-Sahlin model, mainly described by Fickian diffusion. In particular, SA-LE can inhibit S. sonnei with a minimum inhibitory concentration of 3 mL/L, which was lower than free linalool. The mechanism can be described as damaging the membrane structure and inhibiting respiratory metabolism accompanied by oxidative stress based on FESEM, SDH activity, ATP and ROS content. These results suggest that SA is an effective encapsulation strategy to enhance the stability of linalool and its inhibitory effect on S. sonnei at near neutral pH. Moreover, the prepared SA-LE has the potential to be developed as a natural antibacterial agent to address the growing food safety challenges.
